Phototoxicity of TiO₂ nanoparticles to a freshwater benthic amphipod: are benthic systems at risk?

This study investigated phototoxicity of TiO₂ nanoparticles (nano-TiO₂) to a freshwater benthic amphipod (Hyalella azteca) using 48-h and 96-h bioassays. Thorough monitoring of particle interactions with exposure media (Lake Superior water, LSW) and the surface of organisms was performed using dynamic light scattering, UV/Vis spectroscopy, and Scanning Electron Microscopy. Large agglomeration and sedimentation (>77%) in LSW was observed after 0.5h. A simulated solar radiation (SSR)-favored surface attachment of nanoparticles was observed, indicating enhanced phototoxicity with the increased at

PLDT (planarian light/dark test): an invertebrate assay to quantify defensive responding and study anxiety-like effects.

BACKGROUND: Planarians, like rodents, instinctively spend more time in dark versus light environments when given a choice. This behavioral phenomenon is called negative phototaxis, which may reflect defensive responding related to an anxiety-like phenotype. NEW METHOD: We propose a planarian light/dark test, designated PLDT, to predict anxiogenic- or anxiolytic-like effects. Experimentally, we placed a planarian at the midline of a Petri dish, containing test compound or water, that was split evenly into light and dark compartments and determined time spent in the light over 10min. RESULTS:

Improvement of non-motor and motor behavioral alterations associated with Parkinson-like disease in Drosophila melanogaster: Comparative effects of treatments with hesperidin and L-dopa.

Non-motor alterations such as anxiety and memory deficit may represent early indications of Parkinson's disease (PD), and therapeutic strategies that reduce non-motor alterations are promising alternatives for the treatment. Therefore, the search for natural compounds that act on motor and non-motor complications is highly relevant. In this sense, we demonstrated the role of hesperidin (Hsd) as a citrus flavonoid and its pharmacological properties as an antioxidant and neuroprotective agent. Our objective was to evaluate Hsd in developing motor and non-motor alterations in a Drosophila melanog

The impacts of plastics on aquatic insects.

Environmental contamination by plastics and its negative effect on biodiversity have been well-documented in several types of organisms, especially in marine environments. Therefore, it is necessary to assess the impacts of plastic on other organisms such as aquatic insects, which predominantly inhabit freshwaters. It is widely known that these organisms are sensitive to environmental change, especially by contamination. Therefore, this study aimed at testing the hypothesis that aquatic insects are impacted by plastic contamination. We made a systematic search for international papers related

Effects of TiO2, SiO2, Ag and CdTe/CdS quantum dots nanoparticles on toxicity of cadmium towards Chlamydomonas reinhardtii.

Nanoparticles (NPs) are inevitably released into the aquatic environment for being widely used and may affect the toxicity of other contaminants already present in the environment, such as trace metals. However, the effects of NPs on the ecotoxicity of cadmium (Cd), a common environmental trace metal pollutant, are not well explored. In this study, effects of four widely used NPs TiO2 (n-TiO2), SiO2 (n-SiO2), Ag (n-Ag) and CdTe/CdS core/shell quantum dots (QD) on the toxicity of Cd to the freshwater algae Chlamydomonas reinhardtii were assessed respectively. Cd reduced the algae biomass, impai

Heteromeric p97/p97R155C complexes induce dominant negative changes in wild-type and autophagy 9-deficient Dictyostelium strains.

Heterozygous mutations in the human VCP (p97) gene cause autosomal-dominant IBMPFD (inclusion body myopathy with early onset Paget's disease of bone and frontotemporal dementia), ALS14 (amyotrophic lateral sclerosis with or without frontotemporal dementia) and HSP (hereditary spastic paraplegia). Most prevalent is the R155C point mutation. We studied the function of p97 in the social amoeba Dictyostelium discoideum and have generated strains that ectopically express wild-type (p97) or mutant p97 (p97(R155C)) fused to RFP in AX2 wild-type and autophagy 9 knock-out (ATG9(KO)) cells. The discovery of stromatolites developing at 3570 m above sea level in a high-altitude volcanic lake Socompa, Argentinean Andes.

We describe stromatolites forming at an altitude of 3570 m at the shore of a volcanic lake Socompa, Argentinean Andes. The water at the site of stromatolites formation is alkaline, hypersaline, rich in inorganic nutrients, very rich in arsenic, and warm (20-24°C) due to a hydrothermal input. The stromatolites do not lithify, but form broad, rounded and low-domed bioherms dominated by diatom frustules and aragonite micro-crystals agglutinated by extracellular substances. In comparison to other modern stromatolites, they harbour an atypical microbial community characterized by highly abundant re

The photochemical reaction cycle and photoinduced proton transfer of sensory rhodopsin II (Phoborhodopsin) from Halobacterium salinarum.

Sensory rhodopsin II (HsSRII, also called phoborhodopsin) is a negative phototaxis receptor of Halobacterium salinarum, a bacterium that avoids blue-green light. In this study, we expressed the protein in Escherichia coli cells, and reconstituted the purified protein with phosphatidylcholine. The reconstituted HsSRII was stable. We examined the photocycle by flash-photolysis spectroscopy in the time range of milliseconds to seconds, and measured proton uptake/release using a transparent indium-tin oxide electrode. The pKa of the counterion of the Schiff base, Asp(73), was 3.0. Below pH 3, the

Elucidation of the life cycle of Galactosomum nagasakiense (Heterophyidae), the causative parasite of trematode whirling disease in marine fish, with discovery of congeneric species in the gastropod first intermediate host Cerithium dialeucum.

Trematode whirling disease (TWD), caused by Galactosomum nagasakiense (Heterophyidae), affects a wide range of wild and cultured marine fish, with occasional mass mortalities reported in aquaculture since the 1960s. Fish serve as the second intermediate host, while the black-tailed gull Larus crassirostris has been identified as the definitive host. However, the first intermediate host has remained unidentified for decades. Field surveys were conducted in Tsushima, Nagasaki Prefecture, Japan, an endemic region of TWD, aiming to identify the gastropod first intermediate host. Among 1341 snails

Heterozygous mutation of Drosophila Opa1 causes the development of multiple organ abnormalities in an age-dependent and organ-specific manner.

Optic Atrophy 1 (OPA1) is a ubiquitously expressed dynamin-like GTPase in the inner mitochondrial membrane. It plays important roles in mitochondrial fusion, apoptosis, reactive oxygen species (ROS) and ATP production. Mutations of OPA1 result in autosomal dominant optic atrophy (DOA). The molecular mechanisms by which link OPA1 mutations and DOA are not fully understood. Recently, we created a Drosophila model to study the pathogenesis of optic atrophy. Heterozygous mutation of Drosophila OPA1 (dOpa1) by P-element insertion results in no obvious morphological abnormalities, whereas homozygous

Regulation of cyanobacterial type IV pilus-dependent functions by interaction between a c-di-GMP receptor and two transcription factors.

Cyanobacteria utilize type IV pili for many behavioural responses, such as phototaxis, aggregation, floating, and DNA uptake. Type IV pilus-dependent functions are regulated by the nucleotide second messengers, c-di-GMP and cAMP. In this study, we investigated the role of a recently identified c-di-GMP receptor (CdgR) in cyanobacteria that harbours a ComFB domain. ComFB-domain proteins are widespread in cyanobacteria and are also present in heterotrophic bacteria. We demonstrated that the CdgR homolog from the cyanobacterium Synechocystis sp. PCC 6803, a model organism for studying type IV pil

Exposure to a sublethal concentration of CdO nanoparticles impairs the vision of the fruit fly (Drosophila melanogaster) by disrupting histamine synthesis and recycling mechanisms.

While there is substantial literature on potential risks associated with exposure to emerging nanomaterials, less is known about the potential effects of hazardous metallic nanoparticles on vision, as well as the mechanisms that underpin them. The fruit fly (Drosophila melanogaster) was used as an in vivo model organism to investigate the effects of exposure to a sublethal concentration (0.03 mg CdO NPs/mL, which was 20% of the LC50) on fly vision and compound eye ultrastructure. First, we observed a reduction in phototaxis response in treated flies but no change in locomotor activity. Because

Origin and early evolution of neural circuits for the control of ciliary locomotion.

Behaviour evolved before nervous systems. Various single-celled eukaryotes (protists) and the ciliated larvae of sponges devoid of neurons can display sophisticated behaviours, including phototaxis, gravitaxis or chemotaxis. In single-celled eukaryotes, sensory inputs directly influence the motor behaviour of the cell. In swimming sponge larvae, sensory cells influence the activity of cilia on the same cell, thereby steering the multicellular larva. In these organisms, the efficiency of sensory-to-motor transformation (defined as the ratio of sensory cells to total cell number) is low. With th

Separating Chromera velia Zoospores From Culture and Estimating Their Average Motility Speed and Lifespan.

Chromera velia is an apicomplexan alga uniquely positioned as the closest photosynthetic relative to apicomplexan parasites (Sporozoa), which include the human pathogens that cause malaria (Plasmodium) and toxoplasmosis (Toxoplasma). Under favorable conditions, C. velia forms motile zoospores that contribute to dispersal and possibly host interaction. However, zoospores coexist with other developmental stages in culture, making their isolation technically challenging. Previous studies characterized the phototactic behavior of zoospores in several taxa, yet this response has not been used to se

Temperature and light effects on Trichobilharzia szidati cercariae with implications for a risk analysis.

BACKGROUND: Cercarial dermatitis (swimmer's itch) caused by bird schistosome cercariae, released from intermediate host snails, is a common disorder also at higher latitudes. Several cases were observed in the artificial Danish freshwater Ringen Lake frequently used by the public for recreational purposes. The lake may serve as a model system when establishing a risk analysis for this zoonotic disease. In order to explain high risk periods we determined infection levels of intermediate host snails from early spring to late summer (March, June and August) and elucidated the effect of temperatur

Shedding light on amphipod behaviour: Baseline locomotion behaviour for laboratory ecotoxicology experiments.

Behavioural endpoints offer a sensitive, non-invasive alternative to traditional mortality assays. Yet, they remain underutilized in regulatory frameworks due to a lack of standardised methodologies and baseline behavioural data. This study investigates the baseline locomotion of the marine amphipod Marinogammarus marinus to external light triggers to support standardised laboratory behavioural assays. Using an automated observation chamber, here we tracked swimming behaviour in response to light stimuli, quantifying locomotion as distance travelled. Groups of eight amphipods were tested in la

Aged nanoplastics exacerbate CPPD-induced ocular toxicity via the thyroxine pathway in zebrafish.

The coexistence of naturally aged nanoplastics (NPs) and the antioxidant N-cyclohexyl-N'-phenyl-p-phenylenediamine (CPPD) in aquatic environments poses combined risks to ecosystems. This study evaluated the developmental and ocular toxicity of CPPD alone and in co-exposure with two types of aged polystyrene nanoplastics (PS) including ultraviolet-irradiated (PS-UV) and ozonated (PS-O₃) in embryonic zebrafish. Results showed that the presence of aged PS significantly exacerbated CPPD-induced developmental toxicity, manifested as synergistic inhibition of spontaneous movement, touch response, an

Postembryonic development and lifestyle shift in the commensal ribbon worm.

BACKGROUND: Various morphological adaptations are associated with symbiotic relationships between organisms. One such adaptation is seen in the nemertean genus Malacobdella. All species in the genus are commensals of molluscan hosts, attaching to the surface of host mantles with a terminal sucker. Malacobdella possesses several unique characteristics within the order Monostilifera, exhibiting the terminal sucker and the absence of eyes and apical/cerebral organs, which are related to their adaptation to a commensal lifestyle. Nevertheless, the developmental processes that give rise to these mo

Tadpoles rely on mechanosensory stimuli for communication when visual capabilities are poor.

The ways in which animals sense the world changes throughout development. For example, young of many species have limited visual capabilities, but still make social decisions, likely based on information gathered through other sensory modalities. Poison frog tadpoles display complex social behaviors that have been suggested to rely on vision despite a century of research indicating tadpoles have poorly-developed visual systems relative to adults. Alternatively, other sensory modalities, such as the lateral line system, are functional at hatching in frogs and may guide social decisions while ot
