Multifunctional PDA-graphene family nanocomposites for antibacterial and anticancer photothermal therapy.

The increasing demand for multifunctional, biocompatible nanomaterials has spurred the exploration of hybrid systems with synergistic antimicrobial, anticancer, antioxidant, and regenerative properties. In this study, polydopamine (PDA)-based nanocomposites incorporating graphene oxide (PDA-GO) and graphene quantum dots (PDA-GQD) were synthesized and systematically characterized for their physicochemical and biological functionalities. The nanocomposites raised temperature > 50 °C within 5 min under 808 nm laser irradiation (1.5 W/cm2, 250 μg/mL). Both composites showed antibacterial activity

Triple-Responsive Cross-Linked Micelles for Synergistic Chemo-Photothermal Cancer Therapy.

We have developed a novel triple-responsive (pH/reduction/near-infrared (NIR) light) cross-linked polymer micelle PEG-P(LL/LL-LA)-PCL-IR820 for enhanced synergistic cancer therapy. This micelle was synthesized using a triblock amphiphilic polymer based on polylysine, functionalized with maleic anhydride (LA) as a cross-linking site and polycaprolactone (PCL) for encapsulating the anticancer drug doxorubicin (DOX). The photosensitizer IR820 was grafted onto PCL to enable photothermal and photodynamic effects under NIR irradiation. The DOX-loaded cross-linked micelles (DCMs) demonstrated excepti

Glucose-functionalized gold nanoparticles for effective photothermal therapy in lung cancer.

Lung cancer remains the leading cause of cancer-related mortality worldwide, largely due to late-stage diagnoses that severely limit therapeutic interventions. In this context, nanoparticle-mediated photothermal therapy (PTT) has emerged as a promising and minimally toxic modality for solid tumors. We synthesized gold nanoparticles (AuNPs) with three distinct morphologies-spheres, rods, and stars-and functionalized them with polyethylene glycol (AuNPs-PEG) or polyethylene glycol conjugated with 2-deoxy-D-glucose (AuNPs-Gluc). In vitro analyses using human (A549, H1299) and murine (LLC) lung ca

Composite Microparticles from Microfluidics for Chemo-/Photothermal Therapy of Hepatocellular Carcinoma.

Hydrogel microcarrier-based drug delivery systems are of great value in the combination therapy of tumors. Current research directions concentrate on the development of more economic, convenient, and effective combined therapeutic platforms. Herein, we developed novel adhesive composite microparticles (MPPMD) with combined chemo- and photothermal therapy ability via microfluidic electrospray technology for local hepatocellular carcinoma treatment. These composite microparticles consisted of doxorubicin (DOX)-loaded and polydopamine-wrapped mesoporous silicon and alginate. Benefiting from such

Polymeric pH-Responsive Metal-Supramolecular Nanoparticles for Synergistic Chemo-Photothermal Therapy.

Stimuli-responsive drug delivery carriers, particularly those exhibiting pH sensitivity, have attracted significant scholarly interest due to their promising potential in anticancer therapeutic applications. This phenomenon can primarily be ascribed to the inherently acidic nature of tumor microenvironments. However, pH-responsive carriers frequently require the incorporation of functional groups or materials sensitive to pH changes. Given the pH-sensitive characteristics of metal coordination with natural small-molecule drugs, organometallic supramolecules present a facile and effective strat

Recent advances in Prussian blue-based photothermal therapy in cancer treatment.

Malignant tumours are a serious threat to human health. Traditional chemotherapy has achieved breakthrough improvements but also has significant detrimental effects, such as the development of drug resistance, immunosuppression, and even systemic toxicity. Photothermal therapy (PTT) is an emerging cancer therapy. Under light irradiation, the phototherapeutic agent converts optical energy into thermal energy and induces the hyperthermic death of target cells. To date, numerous photothermal agents have been developed. Prussian blue (PB) nanoparticles are among the most promising photothermal age

Photothermal nanozyme-ignited Fenton reaction-independent ferroptosis for breast cancer therapy.

Ferroptosis is a type of programmed cell death caused by the iron-dependent lipid hydroperoxide pathway and has attracted significant interest. However, Fenton reaction-dependent ferroptosis has shown unsatisfactory therapeutic effects in tumor therapy, mainly due to inadequate reaction conditions in the tumor microenvironment. Here, we report a new strategy for Fenton-independent pathway by employing photothermal nanozyme to overcome limitations of the low efficiency of Fenton reaction. Specifically, we used iron redox pair (Fe2+/Fe3+)-containing hollow mesoporous Prussian blue (HMPB) nanocub

Cellular nanovesicles with bioorthogonal targeting enhance photodynamic/photothermal therapy in psoriasis.

The interaction between over-proliferating keratinocytes and excessive activated immune cells contributes to the development and progression of psoriasis. Photodynamic therapy (PDT)/photothermal therapy (PTT) is a promising method to treat skin diseases due to their synergistic effect and low side effect. Cell membrane is an ideal cell-mediated drug delivery platform with inherent biocompatibility, but lacks of specific targeting properties to increase drug accumulation in the psoriatic site. Here, we develop a new PDT/PTT strategy to treat psoriasis, based on cell membrane functionalization b

Nanoscale Covalent Organic Framework for Combinatorial Antitumor Photodynamic and Photothermal Therapy.

Despite the excellent photodynamic and photothermal properties of organic molecular photosensitizers (PSs) and photothermal agents (PTAs), such as porphyrin and naphthalocyanine, their poor water solubility severely impedes their biological applications. Covalent organic frameworks (COFs), as an emerging class of organic crystalline porous materials, possess free active end groups (bonding defects) and large inner pores, which make them an ideal type of nanocarriers for loading hydrophobic organic molecular PSs and PTAs by both bonding defect functionalization (BDF) and guest encapsulation app

Laparoscopic fluorescence image-guided photothermal therapy enhances cancer diagnosis and treatment.

Endoscopy is the gold standard investigation in the diagnosis of gastrointestinal cancers and the management of early and pre-malignant lesions either by resection or ablation. Recently gold nanoparticles have shown promise in cancer diagnosis and therapeutics (theranostics). The combination of multifunctional gold nanoparticles with near infrared fluorescence endoscopy for accurate mapping of early or pre-malignant lesions can potentially enhance diagnostic efficiency while precisely directing endoscopic near infrared photothermal therapy for established cancers. The integration of endoscopy

Polyphosphoester-Based Nanocarrier for Combined Radio-Photothermal Therapy of Breast Cancer.

Recently, clinical research on tumor therapy has gradually shifted from traditional monotherapy toward combination therapy as tumors are complex, diverse, and heterogeneous. Combination therapy may be essential for achieving the optimized treatment efficacy of tumors through distinct tumor-inhibiting mechanisms. At the same time, nanocarriers are emerging as an excellent strategy for delivering both drugs simultaneously. This work presents utilization of a polyphosphoester-based nanocarrier (NPIR/Cur) to achieve the codelivery of hydrophobic photothermal agent IR-780 and radiosensitizer curcum

Photothermal therapy synergizes with bacterial azurin to suppress CT26 tumor growth.

Photothermal therapy (PTT) has emerged as a promising antitumor modality; however, the inflammatory response triggered by PTT promotes the infiltration and activation of immune cells within the tumor microenvironment, leading to elevated IFN-γ secretion. This inflammatory cytokine subsequently upregulates PD-L1 on tumor cells, facilitating tumor immune evasion and limiting the long-term efficacy of PTT. To address these challenges, we engineered a living platform (PMEA) by coating Escherichia coli Nissle 1917(ECN)with a polydopamine-metformin(MET) layer and incorporating a heat-inducible azuri

Photothermal-gas combination therapy promotes checkpoint blockade immunotherapy in colon cancer.

Checkpoint blockade immunotherapy emerges as a potential cure of cancer, but the monotherapy suffers from a low response rate in clinic. Photothermal therapy (PTT) that harvests light energy to ablate tumor is reported to activate tumor-specific immune response, meanwhile nitric oxide (NO) is considered to involve in immune regulation. Herein, we designed a multifunctional nanoplatform that enables photothermal-gas combination therapy by conjugating indocyanine green-thiol (ICG-SH) and s-nitrosoglutathione (GSNO) onto polyvinyl pyrrolidone (PVP)-coated gold nanoparticles (AIG). Urease-Powered Black TiO2 Micromotors for Photothermal Therapy of Bladder Cancer.

Urease-powered nano/micromotors can move at physiological urea concentrations, making them useful for biomedical applications, such as treating bladder cancer. However, their movement in biological environments is still challenging. Herein, Janus micromotors based on black TiO2 with urease asymmetric catalytic coating were designed to take benefit of the optical properties of black TiO2 under near-infrared light and the movement capability in simulated bladder environments (urea). The black TiO2 microspheres were half-coated with a thin layer of Au, and l-Cysteine was utilized to attach the ur

Research progress of organic photothermal agents delivery and synergistic therapy systems.

Cancer is currently one of the leading causes of mortality worldwide. Due to the inevitable shortcomings of conventional treatments, photothermal therapy (PTT) has attracted great attention as an emerging and non-invasive cancer treatment method. Photothermal agents (PTAs) is a necessary component of PTT to play its role. It accumulates at the tumor site through appropriate methods and converts the absorbed light energy into heat energy effectively under near-infrared light irradiation, thus increasing the temperature of the tumor area and facilitating ablation of the tumor cells. Compared to

Nanomaterials-based advanced systems for photothermal / photodynamic therapy of oral cancer.

The traditional clinical approaches for oral cancer consist of surgery, chemotherapy, radiotherapy, immunotherapy, and so on. However, these treatments often induce side effects and exhibit limited efficacy. Photothermal therapy (PTT) emerges as a promising adjuvant treatment, utilizing photothermal agents (PTAs) to convert light energy into heat for tumor ablation. Another innovative approach, photodynamic therapy (PDT), leverages photosensitizers (PSs) and specific wavelength laser irradiation to generate reactive oxygen species (ROS), offering an effective and non-toxic alternative. Targeted Delivery of Quinoxaline-Based Semiconducting Polymers for Tumor Photothermal Therapy.

Photothermal therapy (PTT) holds great potential in the field of cancer treatment due to its high specificity and low invasiveness. However, the low conversion efficiency, inadequate tumor accumulation, and limited cellular uptake continue to impede PTT effectiveness in treating tumors. The present study focuses on the utilization of quinoxaline and its nanoparticles to develop an organic semiconducting photothermal agent (PAQI-BDTT) for tumor photothermal therapy. To achieve this, PAQI-BDTT was encapsulated within liposomes modified with cyclic Arg-Gly-Asp (cRGD) peptide targeting tumors (nam
