Lipid- and Polymer-Based Nanoscale Delivery of Anti-Aging Phytochemicals.

Recently, nano-scale delivery approaches have been a field of focus to improve the limitations regarding current traditional delivery systems of phytochemicals to treat age-related disorders. Nanotechnology can help to overcome both the large and small-scale barriers associated with traditional drug and vitamin delivery techniques, such as poor biodistribution, instability, and intracellular trafficking, through cell-targeting and other methods. The main forms of nano-drug delivery systems can be classified under lipid-based nanocarrier systems, polymer-based nanocarrier systems, and inorganic

Molecular docking analysis of DprE1 from M. tuberculosis with phytochemicals.

Tuberculosis remains a major global health problem due to the emergence of drug-resistant strains of Mycobacterium tuberculosis. Mycobacterium tuberculosis DprE1, a key enzyme involved in cell wall biosynthesis, was targeted to identify potential anti-tuberculosis agents. Hence, molecular docking and molecular dynamics simulations were performed to screen Indian phytochemicals against DprE1. Among the tested compounds, Nimbolide exhibited one of the strongest binding affinities (-10.3 kcal/mol), outperforming standard antibiotics. Simulation results further confirmed the stability of the DprE1

Nanoengineered phytochemicals overcome blood-brain barrier constraints in neurodegenerative disorders.

Neurodegenerative disorders represent a growing global health burden and remain largely incurable, with current therapies providing only symptomatic relief and limited disease modifications. A major obstacle to effective treatment is the inability of many neuroprotective agents to reach the brain at therapeutically relevant concentrations due to poor bioavailability and the restrictive nature of the blood-brain barrier. Plant-derived phytochemicals possess well-documented antioxidant, anti-inflammatory, anti-apoptotic, and neuromodulatory activities; however, their clinical translation has bee

Emerging trends of phytochemicals as ferroptosis modulators in cancer therapy.

Ferroptosis, a novel form of regulated cell death characterized by dependence on iron and lipid peroxidation, has been implicated in a wide range of clinical conditions including neurological diseases, cardiovascular disorders, acute kidney failure, and various types of cancer. Therefore, it is critical to suppress cancer progression and proliferation. Ferroptosis can be triggered in cancer cells and some normal cells by synthetic substances, such as erastin, Ras-selective lethal small molecule-3, or clinical pharmaceuticals. Natural bioactive compounds are traditional drug discovery tools, an

Anti-Eryptotic Activity of Food-Derived Phytochemicals and Natural Compounds.

Human red blood cells (RBCs), senescent or damaged due to particular stress, can be removed by programmed suicidal death, a process called eryptosis. There are various molecular mechanisms underlying eryptosis. The most frequent is the increase in the cytoplasmic concentration of Ca2+ ions, later exposure of erythrocytes to oxidative stress, hyperosmotic shock, ceramide formation, stimulation of caspases, and energy depletion. Phosphatidylserine (PS) exposed by eryptotic RBCs due to interaction with endothelial CXC-Motiv-Chemokin-16/Scavenger-receptor, causes the RBCs to adhere to vascular wal

Promising Perspective to Facioscapulohumeral Muscular Dystrophy Treatment: Nutraceuticals and Phytochemicals.

Facioscapulohumeral Muscular Dystrophy (FSHD) is in the top three list of all dystrophies with an approximate 1:8000 incidence. It is not a life-threatening disease; however, the progression of the disease extends over being wheelchair bound. Despite some drug trials continuing, including DUX4 inhibition, TGF-ß inhibition and resokine which promote healthier muscle, there is not an applicable treatment option for FSHD today. Still, there is a need for new agents to heal, stop or at least slow down muscle wasting. Current FSHD studies involving nutraceuticals as vitamin C, vitamin E, coenzyme Q

Neuroinflammation in pathogenesis of Alzheimer's disease: Phytochemicals as potential therapeutics.

Accumulation of neurotoxic forms of amyloid-β proteins in senile plaques and hyperphosphorylated tau proteins in neurofibrillary tangles is a well-known pathophysiological hallmark of Alzheimer's disease (AD). However, clinical trials with drugs targeting amyloid-β and tau have failed to demonstrate efficacy in treating AD. All currently FDA-approved anti-AD drugs have symptomatic effects only and are not able to cure this disease. This makes necessary to search for alternative therapeutic targets. Accumulating evidence suggests that systemic inflammation and related vascular dysfunction play

The anti-borreliae efficacy of phytochemicals and micronutrients: an update.

Naturally occurring substances have been used for centuries to fight against various pathogens. They serve as a source for new chemical entities or provide options to already existing therapeutics. While there is an increasing interest in studying antimicrobial properties of naturally derived agents, little is known about their effects against Borrelia burgdorferi sensu lato, the causative pathogens of Lyme disease. A better understanding of this aspect could advance knowledge about pathophysiology of these bacteria and help improve the efficacy of current approaches against Lyme disease. Here
