The development of multifunctional deep-red phosphors for optical thermometry and plant lighting still faces considerable challenges. In this work, a series of novel red-emitting phosphors LuMg3SbO7:xMn4+ (0 ≤ x ≤ 0.030) were synthesized by a conventional high-temperature solid-state reaction. XRD and SEM results confirm the high phase purity and uniform element distribution, with negligible effect of Mn4+ doping on the host structure. The phosphors show broad Mn4+ absorption (200-600 nm) suitable for UV LED excitation and yield intense deep-red emission (600-800 nm) under UV irradiation. Conc

ELONGATED HYPOCOTYL 5 (HY5) and PHYTOCHROME INTERACTING FACTOR 4 (PIF4) compete for binding to common promoter elements of growth-related genes. While HY5 suppresses these genes to inhibit elongation, under high ambient temperature, PIF4 promotes their expression. The molecular regulation of this differential binding is critical for adaptive growth. Under elevated temperature (28°C), BBX28 expression and accumulation are enhanced, and bbx28 seedlings exhibit hyposensitivity to temperature-mediated hypocotyl elongation. Our genetic analyses indicate that BBX28 and PIF4 act in the same pathway a

Canopy shade enhances the activity of PHYTOCHROME INTERACTING FACTORs (PIFs) to boost auxin synthesis in the cotyledons. Auxin, together with local PIFs and their positive regulator CONSTITUTIVELY PHOTOMORPHOGENIC 1 (COP1), promotes hypocotyl growth to facilitate access to light. Whether shade alters the cellular redox status thereby affecting growth responses, remains unexplored. Here, we show that, under shade, high auxin levels increased reactive oxygen species and nitric oxide accumulation in the hypocotyl of Arabidopsis. This nitroxidative environment favored the promotion of hypocotyl gr

Iron (Fe) deficiency affects global crop productivity and human health. However, the role of light signaling in plant Fe uptake remains uncharacterized. Here, we find that light-induced Fe uptake in tomato (Solanum lycopersicum L.) is largely dependent on phytochrome B (phyB). Light induces the phyB-dependent accumulation of ELONGATED HYPOCOTYL 5 (HY5) protein both in the leaves and roots. HY5 movement from shoots to roots activates the expression of FER transcription factor, leading to the accumulation of transcripts involved in Fe uptake. Mutation in FER abolishes the light quality-induced c

Understanding processes such as blood-brain barrier (BBB) disruption and tumor progression can greatly benefit from simultaneous molecular, functional, and hemodynamic imaging in deep tissue, yet few existing imaging modalities can provide all three in a single system. Here, we present an integrated imaging platform that combines 3D photoacoustic tomography with ultrasound localization microscopy (3D-PAULM) to enable intrinsically co-registered, multiparametric imaging. 3D-PAULM unifies multispectral photoacoustic molecular imaging, ultrasound B-mode imaging, microbubble-enhanced power Doppler

Sensory photoreceptors abound in nature and enable organisms to adapt behavior, development, and physiology to environmental light. In optogenetics, photoreceptors allow spatiotemporally precise, reversible, and non-invasive control by light of cellular processes. Notwithstanding the development of numerous optogenetic circuits, an unmet demand exists for efficient systems sensitive to red light, given its superior penetration of biological tissue. Bacteriophytochrome photoreceptors sense the ratio of red and far-red light to regulate the activity of enzymatic effector modules. The recombinati

Viscosity is one of the important microenvironmental parameters of abiotic stress. Abnormal changes in viscosity have been shown to be correlated with pathological activities and diseases of plants. However, current probes may not be suitable for sensing in plants due to their hydrophobicity and the phytochromes found in plants. With this research, we successfully constructed a structurally simple, water-soluble near-infrared fluorescent probe HJA-MQ-D, which can simultaneously monitor dynamic viscosity changes in plants and animal systems. The probe exhibits a relatively long emission wavelen

Mitochondrial alternative oxidase (AOX), the unique respiratory terminal oxidase in plants, catalyzes the energy wasteful cyanide (CN)-resistant respiration and plays a role in optimizing photosynthesis. Although it has been demonstrated that leaf AOX is upregulated after illumination, the in vivo mechanism of AOX upregulation by light and its physiological significance are still unknown. In this report, red light and blue light-induced AOX (especially AOX1a) expressions were characterized. Phytochromes, phototropins and cryptochromes, all these photoreceptors mediate the light-response of AOX

Fiber cells are a primary component of vascular tissue in angiosperms, offering mechanical support for most aerial structures, reinforcing vascular transport functions, and enabling vertical growth. Our previous research discovered light as a vital signal in modulating fiber cell-wall thickening and thus length and strength during maturation. Here, we reveal that the red light-sensing Phytochrome B (PhyB)/Phy-Interacting Factor-4 (PIF4) module plays a major photoregulatory role in Arabidopsis stem fiber growth upon activation through its negative influence on the downstream transcription facto

Double perovskite oxides have emerged as pivotal host matrices for Mn4+-activated deep-red phosphors in recent years owing to their exceptional chemical stability and versatile structural tunability. In this study, a series of Mn4+-doped double perovskite La2(Al0.5MgTa0.5)O6 (LAMT) deep-red phosphors were synthesized via the high-temperature solid-state method. Their crystal structure, luminescent properties, and potential applications in plant lighting were systematically investigated. Within the LAMT structure, the [MgO6] and [Al/TaO6] octahedra undergo distortion because of disparities in c

Nuclear bodies are discrete suborganelle structures that perform specialized functions in eukaryotic cells. In plant cells, light can induce de novo formation of nuclear bodies called photobodies (PBs) composed of the photosensory pigments, phytochrome (PHY) or cryptochrome (CRY). The mechanisms of formation, the exact compositions, and the functions of plant PBs are not known. Here, we have expressed Arabidopsis CRY2 (AtCRY2) in mammalian cells and analyzed its fate after blue light exposure to understand the requirements for PB formation, the functions of PBs, and their potential use in cell

The influence of temperature on plants is essential. However, our knowledge on the intricate regulation process underlying heat stress (HS) response in plants is limited. Recently, information about thermal sensors in vivo has begun to emerge. In this study, another primary environmental stimulus, light, was verified once again to work with temperature synergistically on plants, through the modulation of numerous biological processes. With the application of transcriptomic analysis, a substantial number of heat-responsive genes were detected involved in both light- and phytohormone-mediated pa

Methylotrophic yeasts such as Pichia pastoris are widely used for heterologous protein production because they contain strong and tightly regulated promoters. However, the use of methanol as an inducer presents several practical challenges, including toxicity, flammability, high oxygen demand during fermentation, and increased production costs. To overcome these limitations, researchers have been working on redesigning the AOX1 regulatory system and developing alternative induction strategies that do not rely on methanol. One promising approach is optogenetics, which uses light to control gene

Phyllanthus emblica is valued for its nutritional and medicinal properties, yet the genomic divergence between localized and widespread cultivars remains poorly understood. We investigated the genomes of two individuals from the endemic cultivar 'Hongguang' (HG), propagated via regional grafting, and the commercially widespread 'Dongkeng' (DK), known for its superior protein content. Using whole-genome sequencing, we reconstructed phylogenies from two nuclear markers, profiled genome-wide variations, assembled chloroplast genomes, and verified relative plastid copy numbers via real-time quanti

As a master bZIP transcription factor, ELONGATED HYPOCOTYL 5 (HY5) coordinates plant growth with environmental acclimation across developmental stages. Beyond its role in photomorphogenesis, HY5 integrates diverse environmental and endogenous signals, linking environmental perception with developmental and metabolic programs to ensure coordinated physiological adaptation. This review synthesizes current understanding of the multilayered regulatory networks controlling HY5, ranging from constitutive photomorphogenic 1/suppressor of phytochrome A (COP1/SPA)-mediated ubiquitin-dependent degradati

Light is a vital environmental signal that regulates the expression of plastid genes. Plastids are crucial organelles that respond to light, but the effects of light on plastid RNA processing following transcription remain unclear. In this study, we systematically examined the influence of light exposure on plastid RNA processing, focusing on RNA splicing and RNA editing. We demonstrated that light promotes the splicing of transcripts from the plastid genes rps12, ndhA, atpF, petB, and rpl2. Additionally, light increased the editing rate of the accD transcript at nucleotide 794 (accD-794) and

Light quality affects mutualisms between plant roots and arbuscular mycorrhizal fungi (AMFs), which modify nutrient acquisition in plants. However, the mechanisms by which light systemically modulates root colonization by AMFs and phosphate uptake in roots remain unclear. We used a range of approaches, including grafting techniques, protein immunoblot analysis, electrophoretic mobility shift assay, chromatin immunoprecipitation, and dual-luciferase assays, to unveil the molecular basis of light signal transmission from shoot to root that mediates arbuscule development and phosphate uptake in t

The switch from dark- to light-mediated development is critical for the survival and growth of seedlings, but the underlying regulatory mechanisms are incomplete. Here, we show that the steroids phytohormone brassinosteroids play crucial roles during this developmental transition by regulating chlorophyll biosynthesis to promote greening of etiolated seedlings upon light exposure. Etiolated seedlings of the brassinosteroids-deficient det2-1 (de-etiolated2) mutant accumulated excess protochlorophyllide, resulting in photo-oxidative damage upon exposure to light. Conversely, the gain-of-function

Many fungal and oomycete pathogens differentiate a feeding structure named the haustorium to extract nutrition from the plant epidermal cell. The atypical resistance (R) protein RPW8.2 activates salicylic acid (SA)-dependent, haustorium-targeted defenses against Golovinomyces spp., the causal agents of powdery mildew diseases on multiple plant species. How RPW8.2 activates defense remains uncharacterized. Here, we report that RPW8.2 interacts with the phytochrome-associated protein phosphatase type 2C (PAPP2C) in yeast and in planta as evidenced by co-immunoprecipitation and bimolecular fluore

Plant cells characterized by apical growth, for example, root hairs and apical cells of moss protonema, are a convenient model to address the problem of gravity response mechanisms including initiation of cell polarity. The fluorescent calcium probe, chlorotetracycline, allowed us to display the calcium distribution gradient in these cells. Irradiation by red light led to a sharp decrease in the Ca2+ ion activity in cells. During clinostatting in darkness the pattern of calcium influx and distribution changes inconsiderably as compared with control; in root hairs calcium is detected mainly in
