Lipid-Inspired Low Melting Ionic Liquids via Synergistic Cyclopropanation and Branching of Terpenoids.

Bacteria employ cyclopropane motifs as bioisosteres for unsaturations to modulate lipid bilayer fluidity and protect cellular membranes under environmental stress. Drawing inspiration from this biological strategy, we investigated how cyclopropanation impacts the thermophysical properties of lipid-inspired ionic liquids. We synthesized a series of imidazolium-based ionic liquids incorporating cyclopropanated derivatives of three renewable terpenoids: phytol, farnesol, and geraniol. Through an integrated approach combining property-driven design, thermophysical analysis, X-ray crystallography,

Socialane, a Nonaprenyl Terpene Hydrocarbon Surface Lipid from the Collembola Hypogastrura socialis.

Springtails use unique compounds for their outermost epicuticular wax layer, often of terpenoid origin. We report here the structure and synthesis of socialane, the major cuticular constituent of the Collembola Hypogastrura socialis. Socialane is also the first regular nonaprenyl terpene with a cyclic head group. The saturated side chain has seven stereogenic centers, making the determination of the configuration difficult. We describe here the identification of socialane and a synthetic approach using the building blocks farnesol and phytol, enantioselective hydrogenation, and α-alkylation of

Hizikia fusiforme functional oil (HFFO) prevents neuroinflammation and memory deficits evoked by lipopolysaccharide/aluminum trichloride in zebrafish.

BACKGROUND: Oxidative stress, cholinergic deficiency, and neuroinflammation are hallmarks of most neurodegenerative disorders (NDs). Lipids play an important role in brain development and proper functioning. Marine-derived lipids have shown good memory-improving potentials, especially those from fish and microalgae. The cultivated macroalga Hizikia fusiforme is healthy food and shows benefits to memory, but the study is rare on the brain healthy value of its oil. Previously, we had reported that the Hizikia fusiforme functional oil (HFFO) contains arachidonic acid, 11,14,17-eicosatrienoic acid

Isolation and characterization of antiplasmodial and antimicrobial compounds from Tetracera alnifolia using a bioassay-guided approach.

A preliminary biological screening demonstrated that leaf extracts from Tetracera alnifolia exhibit promising activity against Plasmodium falciparum and Candida albicans. To identify the constituents responsible for these effects, a bioassay-guided fractionation strategy was subsequently employed. Fractions were purified using flash chromatography, followed by semi-preparative HPLC-DAD-MS and LC-SPE-NMR. Structural elucidation of the isolated compounds was accomplished through comprehensive 1D and 2D NMR analyses in combination with HR-ESI-MS. The purified metabolites were subsequently evaluat

Heredopathia atactica polyneuritiformis (Refsum's Disease): a second trial of dietary therapy in two patients.

Two sibs with heredopathia atactica polyneuritiformis (Refsum's Disease) each had a relapse after ceasing to follow a special diet. On being placed on a very restricted low phytol, low phytanic acid diet both had marked subjective improvement, and marked objective improvement both in physical findings and in quantitative tests of strength, dexterity, and motor nerve conduction velocities, concomitant with a fall in serum phytanic acid levels. No relapse occurred in 22 months on this diet.

Heredopathia atactica polyneuritiformis (phytanic acid storage disease). A new case with special reference to dietary treatment.

A 33-year-old man with Refsum disease exhibited clinical features of night blindness, dysequilibrium, hearing loss, itchy dry skin, symmetrical polyneuropathy, distal muscle weakness, pes cavus, and hammer toe. His total serum protein was increased, nerve conduction velocities were slow, and serum phytanic acid levels were high. Dietary restriction of phytol resulted in a decrease in serum phytanic acid without any visual and autitory changes; however, coordination, skin lesions, and nerve conduction velocities definitely improved.

A mouse model for alpha-methylacyl-CoA racemase deficiency: adjustment of bile acid synthesis and intolerance to dietary methyl-branched lipids.

alpha-Methylacyl-CoA racemase (Amacr) deficiency in humans leads to sensory motor neuronal and liver abnormalities. The disorder is recessively inherited and caused by mutations in the AMACR gene, which encodes Amacr, an enzyme presumed to be essential for bile acid synthesis and to participate in the degradation of methyl-branched fatty acids. To generate a model to study the pathophysiology in Amacr deficiency we inactivated the mouse Amacr gene. As per human Amacr deficiency, the Amacr(-/-) mice showed accumulation (44-fold) of C27 bile acid precursors and decreased (over 50%) primary (C24)

Disorders related to the metabolism of phytanic acid.

The phytanic acid found in man stems from exogenous sources, mainly as minor parts of fish and animal fats. Free phytol, which is easily converted to phytanic acid in mammals, is present in fats of vegetable origin. Healthy individuals are able to degrade the small amounts of phytanic acid and phytol which are ingested. Accumulation of phytanic acid has been considered diagnostic for Refsum's disease, and a prerequisite for this diagnosis. However, a few patients with proven Refsum's disease have eliminated their phytanic acid stores by dietary means. Two healthy mothers of patients with Refsu

Chemical defenses of Trifolium glanduliferum against redlegged earth mite Halotydeus destructor.

Trifolium glanduliferum Boiss. was resistant to redlegged earth mite, Halotydeus destructor Tucker (Acari: Penthaleidae), feeding compared with Trifolium subterraneum L. Extracts from T. glanduliferum leaves, obtained with dichloromethane and methanol, acted as feeding deterrents to the redlegged earth mite. Bioassay-directed fractionation of the extracts led to the isolation of several metabolites, coumarin, alpha-and beta-ionone, linalool, medicarpin, that were shown to be deterrent in laboratory experiments.
