Plants encounter diverse pathogens and have evolved a 2-layered innate immune system to detect pathogen molecules and activate defense mechanisms that restrict infection. Most cloned plant Resistance (R) genes encode NLR immune receptors. NLR genes are often found in clusters of paralogs with sequence and copy number variation; whether these NLR clusters evolve in response to single or multiple pathogens has been unclear. We report here the isolation of a Phytophthora capsici resistance gene, Rpc2, along with a novel P. infestans resistance gene, Rpi-amr5, from 2 Solanum americanum accessions.

BACKGROUND: Pepper is a crucial vegetable crop with significant economic value, yet its yield and quality are severely affected by pepper Phytophthora Blight. Our previous research indicated that CaSBP12 negatively regulates pepper resistance to Phytophthora capsici (P. capsici) infection. However, the precise role of CaSBP12 in the defense response against P. capsici remains unclear. Thus, it is essential to investigate the defense mechanisms by which CaSBP12 contributes to pepper resistance against P. capsici infection.
RESULTS: In this study, silencing CaSBP12 significantly increased the e

The elicitor arachidonic acid in combination with jasmonic acid (JA) induced a higher level of defense against the late blight agent in potato (Solanum tuberosum L.) tissues than in combination with salicylic acid (SA). On the contrary, the elicitor chitosan displayed a higher inductive effect in combination with SA as compared with JA. The optimal concentrations of tested compounds were selected for designing the compositions activating wound repair, induction of proteinase inhibitors, and resistance to the biotrophic pathogen Phytophthora infestans (Mont.) de Bary. It was demonstrated that t

Phytophthora plurivora has become a major driver of alder decline across Europe, posing a critical threat to Alnus lusitanica populations in Northern Spain. Effective and sustainable control measures for this oomycete pathogen remain lacking, particularly in riparian ecosystems where chemical options are unsuitable. This study evaluated the biocontrol potential of native endophytic fungi associated with symptomatic A. lusitanica. P. plurivora and 43 endophytic fungi were isolated from declining trees across six river systems. Nine (9) out of the 43 endophytic fungi were promising and used for

Potato late blight is an important disease in potato production, which causes serious damage. Salicylic acid (SA) is a plant hormone involved in the regulation of potato (Solanum tuberosum) resistance to Phytophthora infestans. In this study, it was found that exogenous methyl salicylate (MeSA) treatment could significantly enhance the resistance of potato to P. infestans. RNA-seq results confirmed that SA was important for potato resistance to P. infestans. Salicylic acid binding protein 2 (SABP2) is a member of α/β hydrolase family, which can convert MeSA into SA to regulate the steady state

Phytophthora infestans is a notorious oomycete pathogen that causes potato late blight. It secretes numerous effector proteins to manipulate host immunity. Understanding mechanisms underlying their host cell manipulation is crucial for developing disease resistance strategies. Here, we report that the conserved RXLR effector Pi05910 of P. infestans is a genotype-specific avirulence elicitor on potato variety Longshu 12 and contributes virulence by suppressing and destabilizing host glycolate oxidase StGOX4. By performing co-immunoprecipitation, yeast-two-hybrid assays, luciferase complementati

Phytophthora capsici Leonian causes destructive economical losses in pepper production, and a promising source of natural fungicides- Helianthus tuberosus leaves was reported. The antifungal activities of different extracts and compounds from H. tuberosus leaves against the phytopathogen, P. capsici Leonian, were examined by chemometric analysis, including HPLC-MS/MS and multivariate data analyses. Principal component analysis and orthogonal partial least squares-discriminate analysis were applied to examine the four groups of H. tuberosus leaves samples, including crude extracts obtained by d

The oomycete genus Phytophthora includes numerous pathogens, some causing severe tree diseases. In forestry, their impact is usually assessed on adult trees, while their effects on seed germination and seedling development-crucial phases in forest regeneration-remain poorly investigated. Here, we tested the effects of the two main causal agents of ink disease in sweet chestnut, P. cinnamomi and P. x cambivora, along with three other Phytophthora species commonly found in European sweet chestnut forests (P. plurivora, P. cactorum, P. cryptogea), on seed germination and seedling development of f

Phytophthora species are among the most destructive plant pathogens worldwide, causing severe losses in agricultural, forest, and natural ecosystems. In recent years, the management of Phytophthora diseases has increasingly shifted toward eco-sustainable strategies, with growing interest in plant-derived extracts, particularly essential oils, as low-risk alternatives to synthetic fungicides. In this study, a structured review was combined with new in vitro assays to assess the antifungal activity of essential oils from Thymus vulgaris (TV-EO) and T. serpyllum (TS-EO) against P. cinnamomi, P. d

Phytophthora blight (PB), caused by Phytophthora cajani, is a key constraint limiting pigeonpea productivity across the tropics and subtropics. Frequent PB outbreaks under erratic rainfall and warm temperatures intensifies the need for a holistic understanding of pathogen variability and stable host resistance. This study evaluated the morpho-cultural, physiological, and molecular diversity of P. cajani and investigated pigeonpea genotypes for resistance against representative isolates. Twenty-two isolates collected from diverse agro-ecological zones of India were purified, and six representat

Nonexpressor of pathogenesis-related 1 (NPR1) is crucial for activating the plant immune system through the signaling molecule salicylic acid (SA), which triggers systemic acquired resistance (SAR) in Arabidopsis. In this study, three putative genes associated with NPR1 from Arabidopsis have been identified in the genome of Theobroma cacao, namely, TcNPR1, TcNPR2, and TcNPR3, suggesting a functional diversification among the three gene entities. Phylogenetic analysis revealed that TcNPR1 and TcNPR2 branched alongside their Arabidopsis orthologs, NPR1 and NPR2, indicating that these genes maint

Biopesticide fungicides are naturally derived compounds that offer protection from plant diseases through various modes of action, including antimicrobial activity and upregulation of defense responses in host plants. These plant protectants provide a sustainable and safe alternative to conventional pesticides in integrated disease management programs and are especially salient in the management of high-risk and economically important diseases such as late blight of tomato and potato, for which host resistance options are limited. In this study, a commercially available biopesticide, EF400 com

BACKGROUND: Oospores are the most persistent propagules of Phytophthora capsici. The viability of oospores is determined by staining methods such as the tetrazolium bromide (MTT) test.
OBJECTIVES: The aim was to assess the MTT test and a plasmolysis method for their utility in determining viability of oospores of P. capsici.
METHODS: For either procedure the reactions of non-treated (viable) and lethally heat-treated (non-viable) oospores were assessed and compared with different matings of isolates.
RESULTS: The plasmolysis method provided lower estimates of oospore viability relative to M

Phytophthora blight caused by Phytophthora capsici severely limits pepper production, and chemical fungicides induce pathogen resistance and reduce fruit quality. Nano‑selenium (nano-Se) shows biocontrol potential but suffers from poor stability. Here, metabolomic analyses were performed to explore the mechanism by which pectin-modified nano-Se (nano-Se@PT) acts in pepper. Results showed that nano-Se@PT enhanced plant resistance by modulating the phenylpropanoid pathway (elevating 6-nitrocoumarin by 30%) and the tricarboxylic acid cycle. Nano-Se@PT recruited beneficial rhizosphere microorganis

Phytophthora species are eukaryotic plant pathogens that cause root rot and dieback diseases in thousands of plant species worldwide. Despite their significant economic and ecological impacts, fundamental molecular tools such as DNA transformation methods are not yet established for many Phytophthora species. In this study, we have established a PEG/calcium chloride (CaCl2)-mediated protoplast transformation method for Phytophthora agathidicida, the causal agent of kauri dieback disease. Adapting a protocol from Phytophthora sojae, we systematically optimized the protoplast digesting enzymes,

Plant growth-promoting rhizobacteria (PGPR) have proven to be a sustainable alternative for managing plant pathogens. Developing inoculants with these PGPRs requires a rigorous selection of highly effective native bacterial isolates. In the present study, 29 bacterial isolates were obtained from the rhizosphere of 'Zutano' avocado (Persea americana Mill.), a rootstock widely used for the commercial cultivar 'Hass', from a commercial orchard on the northern coast of Peru. Two antagonistic isolates showing the highest in vitro inhibition capacity were selected to evaluate their efficacy against

Phytophthora and allied oomycete pathogens pose a perennial challenge to global food security through their devastating impact on crop systems. While metalaxyl has demonstrated remarkable efficacy in controlling Phytophthora diseases since its introduction decades ago, the persistent emergence of metalaxyl-resistant strains has severely compromised its field efficacy. Elucidating the genetic determinants underlying resistance mechanisms is critical to developing surveillance strategies and sustainable countermeasures against evolving oomycete resistance. Through experimental evolution, we gene
