Effects of propranolol, pindolol and atenolol after multiple administration in rats on the duration of hexobarbital sleeping time and on the activity of some drug-metabolizing enzyme systems.

Three beta-adrenergic blocking agents propranolol (50 mg/kg), pindolol (5mg/kg) and atenolol (150mg/kg) were administered orally in rats for 21 days. The duration of hexobarbital (HB) sleeping time and the activities of some liver monooxygenases and synthetases were studied. The HB sleeping time was not changed. Propranolol and pindolol decreased the ethylmorfin-N-demethylase (EMND) and benzphetamine-N-demethylase (BND) activities. The three beta-adrenergic blocking agents did not change the activities of anilinehydroxylase (AH), ethoxycoumarine-O-demethylase (ECOD), and glutathione-S-transfer

Both acute and subchronic treatments with pindolol, a 5-HT(1A) and β₁ and β₂ adrenoceptor antagonist, elevate regional serotonin synthesis in the rat brain: an autoradiographic study.

Antidepressant treatments, including those that increase serotonin (5-HT) neurotransmission, require several weeks or months until the onset of the therapeutic effect in depressed patients. The negative feedback on 5-HT transmission exhibited by the 5-HT(1A) and 5-HT(1B) autoreceptors has been postulated as a possible delaying factor. The aim of the present study was to assess the effect of the acute and subchronic treatment with pindolol, a 5-HT(1A/1B,) β₁ and β₂ adrenoceptor antagonist, on 5-HT synthesis, one of the key parameters of 5-HT neurotransmission. Male Sprague-Dawley (SPD) rats (18

Treatment of hypertension with beta-blockers with and without intrinsic sympathomimetic activity.

In a randomized double-blind trial 36 patients with essential hypertension were treated with either metoprolol or pindolol for 6 months following a 6-week placebo period. At the end of the placebo period and after 6 weeks and 6 months of active therapy peripheral hemodynamics at rest and during maximal vasodilatation were studied. Exercise heart rate was reduced to the same extent with both metoprolol and pindolol, indicating that the doses used (metoprolol average 179 mg/day; pindolol average 12 mg/day) were equipotent as regards beta-adrenoceptor blocking effect. The antihypertensive effect

Chronotropic cardiac effects of falipamil in conscious dogs: interactions with the autonomic nervous system and various ionic conductances.

The chronotropic cardiac effects of falipamil were studied in conscious dogs with chronic atrioventricular (AV) block. Falipamil (0.5-2 mg/kg) initially increased atrial rate dose dependently. After atropine and atropine-pindolol, falipamil (2 mg/kg) decreased atrial rate, but after pindolol, it did not modify atrial rate. After atropine-pindolol-phenoxybenzamine, atropine-pindolol-yohimbine, atropine-pindolol-verapamil, and atropine-pindolol-quinidine pretreatment, falipamil produced atrial bradycardia. Falipamil dose-relatedly decreased ventricular rate. Falipamil (2 mg/kg) decreased ventric

Modification of the discriminative stimulus effects of 8-OH-DPAT, buspirone and the beta-adrenoreceptor antagonist pindolol after chronic administration of the 5-HT(1A) agonist 8-OH-DPAT in the pigeon.

The involvement of adrenoreceptor blocking drugs with the 5-HT(1A)-mediated discriminative stimulus was studied in pigeons trained to discriminate the 5-HT(1A) receptor agonist 8-OH-DPAT (0.3mg/kg) from saline. Cumulative dose-response curves were determined for 8-OH-DPAT, the beta-adrenergic antagonist pindolol, the alpha(1)-antagonist prazosin, as well as for the 5-HT(1A) agonist buspirone, before, during and after (8-OH-DPAT and pindolol) chronic administration of 8-OH-DPAT (3.0mg/kg/day for 6 weeks). The dose-response curves for 8-OH-DPAT and buspirone, which substituted for 8-OH-DPAT, wer

Effects of a beta-blocker on the cardiovascular response to MDMA (Ecstasy).

BACKGROUND: MDMA (3,4-methylenedioxymethamphetamine, 'Ecstasy') produces tachycardia and hypertension and is rarely associated with cardiovascular and cerebrovascular complications. In clinical practice, beta-blockers are often withheld in patients with stimulant intoxication because they may increase hypertension and coronary artery vasospasm due to loss of beta(2)-mediated vasodilation and unopposed alpha-receptor activation. However, it is unknown whether beta-blockers affect the cardiovascular response to MDMA. METHODS: The effects of the non-selective beta-blocker pindolol (20 mg) on the

Treatment of chronic atrial fibrillation in the elderly, assessed by ambulatory electrocardiographic monitoring.

This study concerns 8 elderly patients with age-related chronic atrial fibrillation that required control of the ventricular response. The effects of oral administration of placebo, pindolol, verapamil, digoxin and digoxin + pindolol were assessed (each for a week) during three daily activity and sleep sessions, over a six-week period. Cardiac rhythm was monitored by taped electrocardiograms. Either digoxin or pindolol was effective, but digoxin was the best tolerated of all the drugs. A combination of pindolol and digoxin reduced the maximum ventricular rate without further depression of the

Studies on the stereoisomers of beta-adrenoceptor antagonists in conscious A-V blocked dogs.

Atrial and ventricular chronotropic effects of the individual stereoisomers of propranolol, pindolol, metoprolol and penbutolol were studied in conscious dogs with chronic atrio-ventricular (A-V) block. Ventricular beta-adrenoceptor blocking activity was assessed for all drugs against isoprenaline under the same experimental conditions. At low doses, the stereoisomers of propranolol and penbutolol decreased atrial rate, whereas those of pindolol and metoprolol produced an increase. At higher doses, all drugs increased atrial rate. All drugs decreased ventricular rate dose-dependently except (+

Beta-adrenoceptor blocking agents induce different counter-regulatory responses to insulin.

Simultaneous treatment with antidiabetic drugs and beta-adrenoceptor blocking agents is frequent on account of the high incidence of diabetes and cardiovascular disease. The effects of oral doses of pindolol, metoprolol, and propranolol on the response to insulin-induced hypoglycemia and an oral glucose load were investigated. During hypoglycemia metoprolol and propranolol inhibited the clearance of insulin (2 p less than 0.01) and caused a delay of glucose nadirs. This was not observed after pindolol. [Increase in beta-adrenergic receptors in rabbits in long-term local administration of beta-blockers].

Chronic application of beta-blockers often induces tachyphylaxia of unknown origin. After long-term topical pretreatment of pindolol and timolol in rabbit eyes (up to 12 weeks, twice a day) beta 2-adrenergic receptors were localized and quantified with autoradiographic methods. Frozen sections of albino rabbit eyes were labelled with [125I] cyanopindolol. Quantification revealed a significant increase in the density of beta 2-receptors after premedication with pindolol in the ciliary body and the corneal epithelium. This increase was detectable after 2 weeks of premedication and reached its ma

5-HT1A receptor blockade potentiates the subjective effects of DMT.

INTRODUCTION: Serotonergic psychedelics are being investigated in the treatment of various disorders and in the improvement of well-being. Evidence suggests that their subjective effects may play a role in long-term behavioral outcomes. The subjective effects are mediated by 5-HT2A receptor agonism, but the 5-HT1A receptor may also play a role in the subjective effects. This study elucidates the effects of 5-HT1A receptor blockade using pindolol pre-treatment on the subjective effects induced by N,N-dimethyltryptamine (DMT). METHODS: In a double-blind, randomized, placebo-controlled within-su

Interactions of some partial agonists with high and low affinity binding sites in beta-adrenoceptors.

Interactions of derivatives of befunolol (BFE-37, BFE-55, and BFE-61), carteolol, and pindolol with beta-adrenoceptors were tested in guinea pig isolated taenia caecum. All the drugs used acted as partial agonists on the beta-adrenoceptors when compared with isoprenaline, a full agonist. The pA2 values of BFE-61, carteolol, and pindolol were significantly larger than their pD2 values, while there was no significant difference between the pA2 and pD2 values for BFE-37 and BFE-55. The specific binding of [3H]befunolol to microsomal fractions from the guinea pig taenia caecum distinguished two bi

[High dose betablocker treatment in experimental renal hypertension (author's transl)].

Rats with experimental renovascular hypertension were treated with high doses of beta-blocking agents. Pindolol 10 mg/kg per day increased Goldblatt-type hypertension, whereas Propranolol 100 mg/kg per day showed an antihypertensive effect. A linear correlation between the weight of the left-heart ventricle and the systolic blood pressure was found in animals treated with Pindolol as well as in untreated Goldblatt rats. On the contrary, the weights of the left-heart ventricle were significantly higher in those animals treated with Propranolol than in the other groups. The results show that Pin

Endogenous and exogenous agonist regulation of responses to beta-adrenoceptor stimulation in patients with chronic autonomic failure.

1. Two patients with orthostatic hypotension due to peripheral autonomic neuropathy were studied. The diagnosis was based on severe hypotension during 60 degrees head-up tilting and absence of the systolic pressure overshoot in phase IV of the Valsalva response. Plasma noradrenaline levels were 50 and 90 pg/ml in the two patients. Noradrenaline was unresponsive to head-up tilting. Heart rate did not change after atropine, 1 mg i.v. Thus the patients had combined efferent sympathetic and parasympathetic lesions. 2. Infusion of isoprenaline in these patients showed supersensitivity for the chron

Involvement of beta-adrenoceptors in regulation of the yawning induced by neuropeptides, oxytocin and alpha-melanocyte-stimulating hormone, in rats.

The present study was undertaken to investigate whether beta-adrenoceptors are involved in regulation of yawning responses to oxytocin and alpha-melanocyte-stimulating hormone (alpha-MSH) in rats. Oxytocin administered intracerebroventricularly (ICV) at doses of 50 and 100 ng/rat elicited yawning. alpha-MSH (20 micrograms/rat, ICV) elicited not only yawning but also stretching and body shaking.
