Speed breeding: protocols, application and achievements.

One of the limiting factors in breeding and genetic research is the time required to develop pure lines. This is due, on the one hand, to the prolonged vegetative period of a single generation and, on the other hand, to the specifics of inbreeding, which typically requires 4-6 consecutive generations of self-pollination in plant material. Researchers have always sought approaches that enable the rapid development of homozygous plant lines. Consequently, methods such as greenhouse cultivation during the autumn-winter period, single-seed descent, shuttle breeding, embryo culture, and doubled hap

Temporal transcriptional networks coordinate ABA-GA-JA crosstalk during cold-induced modulation of seed dormancy and germination in wheat (Triticum aestivum L.).

Using a combination of genome-wide transcriptome, targeted hormonome and pharmacological approaches, this study showed that temporally resolved transcriptional regulatory networks integrating hormonal crosstalk underlie cold-induced modulation of seed dormancy and germination in wheat. Gene homoeologues (GHs) differentially expressed between cold-treated and untreated dormant seeds imbibed at optimal temperature (OT, 22°C) as well as between the same seed samples imbibed in cold and at OT represent distinct biological processes (BPs), indicating the role of coordinated activation/repression sp

The MADS-box transcription factor VvSVP1 negatively regulates grapevine bud dormancy release.

Grape (Vitis vinifera L.), an economically important fruit tree grown worldwide, exhibits high sensitivity to environmental fluctuations during bud dormancy release. To address challenges such as warm winters and late-spring cold spells and to elucidate the molecular mechanisms underlying grapevine bud dormancy release, this study investigated the regulatory role of the MADS-box transcription factor VvSVP1 in this process. Transient silencing of VvSVP1 promoted bud break in grapevine, whereas its overexpression delayed bud break, providing clear evidence that VvSVP1 has a negative regulatory r

Aggregation and toxicity in bacteria of Rep-WH1 prion-like proteins from the phytopathogen Xylella fastidiosa.

Xylella fastidiosa is a global bacterial phytopathogen that colonizes the xylem vessels, leading to their occlusion and thus to plant death. The expression of the amyloidogenic, prion-like WH1 domain of the replication protein RepA from the Pseudomonas savastanoi plasmid pPS10 severely damages Escherichia coli cells. Here, we characterize the WH1 domains in the Rep proteins from the X. fastidiosa plasmids pXF51 and pXF64. The expression of these Rep-WH1 proteins, as happens with the archetypical RepA-WH1 protein from pPS10, decreased the culturability of E. coli in vitro, promoting dormancy. E

Role of starvation survival response mechanisms on ribosome integrity, antibiotic tolerances, and virulence of Pseudomonas aeruginosa biofilms.

Bacterial biofilms contain physiologically diverse subpopulations of cells, including cells that are nutrient stressed or dormant. We determined how two dormancy pathways, ribosome hibernation and the stringent response, contribute to the survival and antibiotic tolerance of Pseudomonas aeruginosa biofilms. Analyses of whole biofilms and single cells showed that these pathways have differing effects on biofilm cell physiology. Ribosome hibernation, mediated by hibernation promoting factor (HPF), is essential for optimal survival and resuscitation of starved biofilm cells. In the absence of HPF

Evaluating the Efficacy of Polyphenol-Enriched Plant Extracts and Pathway Modulators in the Bone Marrow to Suppress Breast Cancer Progression.

Disseminated tumor cells (DTCs) in the bone marrow can survive in a dormant state and later reawaken to drive metastatic relapse. Hypoxia-driven angiogenic signaling (HIF-1α → VEGF), matrix remodeling (MMP-2), and inflammation (COX-2, TNF-α) are central determinants of niche permissiveness for colonization. We investigated whether rational combinations of selected plant extracts with pathway-targeting small molecules can reprogram the bone marrow microenvironment, suppress pro-metastatic signaling, and limit tumor progression by promoting dormancy-like states. Complementary in vitro and in viv

Dynamics of alkaloid accumulation in Narcissus cv. Hawera: a source of Sceletium-type alkaloids.

The Sceletium-type alkaloids, known for their anxiolytic and antidepressant activities, have been recently found to be biosynthesized in Narcissus cv. Hawera, which is largely used as an ornamental plant. An alkaloid fraction enriched with Sceletium-type alkaloids from the plant has shown promising antidepressant and anxiolytic activities. In the present study, qualitative and quantitative analyses of the alkaloids in the plant organs were performed during one vegetation season by GC-MS. The alkaloid pattern and total alkaloid content was found to depend strongly on the stage of development an

S-nitrosoglutathione Reductase-Mediated Nitric Oxide Affects Axillary Buds Outgrowth of Solanum lycopersicum L. by Regulating Auxin and Cytokinin Signaling.

Auxin and cytokinin are two kinds of important phytohormones that mediate outgrowth of axillary buds in plants. How nitric oxide and its regulator of S-nitrosoglutathione reductase (GSNOR) take part in auxin and cytokinin signaling for controlling axillary buds outgrowth remains elusive. We investigated the roles of GSNOR during tomato axillary bud outgrowth by using physiological, biochemical and genetic approaches. GSNOR negatively regulated NO homeostasis. Suppression of GSNOR promoted axillary bud outgrowth by inhibiting the expression of FZY in both apical and axillary buds. Meanwhile, AU

Exploring the Role of Carbon Monoxide in Seed Physiology: Implications for Stress Tolerance and Practical Uses.

Carbon monoxide (CO) is recognized as a signaling molecule in plants, inducing various physiological responses. This article briefly examines the physiological functions of CO in seed biology and seedlings' responses to environmental stresses. The activity of heme oxygenase (HO), the main enzyme responsible for CO synthesis, is a key factor controlling CO levels in plant cells. CO can influence seed germination by regulating seed dormancy through interactions with genes and hormones. Additionally, CO positively affects seedling growth by enhancing the antioxidant system, thereby increasing res

Signaling by hydrogen sulfide and cyanide through post-translational modification.

Two cysteine metabolism-related molecules, hydrogen sulfide and hydrogen cyanide, which are considered toxic, have now been considered as signaling molecules. Hydrogen sulfide is produced in chloroplasts through the activity of sulfite reductase and in the cytosol and mitochondria by the action of sulfide-generating enzymes, and regulates/affects essential plant processes such as plant adaptation, development, photosynthesis, autophagy, and stomatal movement, where interplay with other signaling molecules occurs. The mechanism of action of sulfide, which modifies protein cysteine thiols to for

Induction of Autopolyploidy and Preliminary Investigation of the Dwarfing Mechanism in Hedychium coccineum.

In this study, Hedychium coccineum tetraploid plants and octaploid plants induced by colchicine were used as materials. The ploidy levels were precisely identified by combining root tip squash and flow cytometry analyses, and the differences between plants of different ploidy levels were systematically investigated at cytological, morphological, and molecular levels. The results showed that the highest polyploid induction efficiency was achieved when callus tissues were treated with 0.1 g/L colchicine for 4 days. The fluorescence peak value of the induced plants was twice that of the tetraploi

Integrating Physiology, Transcriptome, and Metabolomics Reveals the Potential Mechanism of Nitric Oxide Concentration-Dependent Regulation of Embryo Germination in Sorbus pohuashanensis.

Nitric oxide (NO) breaks a seed's dormancy and stimulates germination by signaling. However, the key physiological metabolic pathways and molecular regulatory mechanisms are still unclear. Therefore, this study used physiological, transcriptomic, and metabolomics methods to analyze the key genes and metabolites involved in the NO regulation of plant embryo germination and their potential regulatory mechanisms. The physiological analysis results indicate that the appropriate concentration of NO increased the content of NO and hydrogen peroxide (H2O2) in cells, stimulated the synthesis of ethyle

Warming, Snow Exclusion, and Soil Type Alter the Timing of Plant and Soil Activity and Associated Nutrient Losses.

In seasonally snow-covered ecosystems, changing temperatures and snowpack dynamics under climate change have increased the occurrence and duration of soil temperatures that support microbial activity during plant dormancy. During these periods of microbial activity without plant activity (i.e., plant-microbe asynchronies), soil nutrients that build up are vulnerable to leaching loss, with potentially important consequences for ecosystem productivity. Furthermore, asynchronies likely do not occur uniformly in space; rather, their occurrence may be modulated by subsurface characteristics. Soil t

Coordinated Transport of Nitrate, Potassium, and Sodium.

Potassium (K+) and nitrogen (N) are essential nutrients, and their absorption and distribution within the plant must be coordinated for optimal growth and development. Potassium is involved in charge balance of inorganic and organic anions and macromolecules, control of membrane electrical potential, pH homeostasis and the regulation of cell osmotic pressure, whereas nitrogen is an essential component of amino acids, proteins, and nucleic acids. Nitrate (NO3 -) is often the primary nitrogen source, but it also serves as a signaling molecule to the plant. Nitrate regulates root architecture, st

AHG1-AFP interaction as a regulatory node in the ABA response during seed germination.

Seed dormancy and germination are tightly regulated by complex signaling networks that integrate internal and external cues, including the endogenous phytohormone abscisic acid (ABA). ABA HYPERSENSITIVE GERMINATION 1 (AHG1), a group A type 2C protein phosphatase (PP2C), is thought to modulate the activity of transcription factors such as ABA INSENSITIVE 5 (ABI5) in seeds and during germination. AHG1 is regulated by DELAY OF GERMINATION 1 (DOG1), a key regulator of seed dormancy, through physical interaction. We previously reported that AHG1 also interacts with ABI FIVE BINDING PROTEIN 2 (AFP2)

Molecular aspects of the antagonistic interaction of smoke-derived butenolides on the germination process of Grand Rapids lettuce (Lactuca sativa) achenes.

Smoke-derived compounds provide a strong chemical signal to seeds in the soil seed bank, allowing them to take advantage of the germination niche created by the occurrence of fire. The germination stimulatory activity of smoke can largely be attributed to karrikinolide (KAR(1) ), while a related compound, trimethylbutenolide (TMB), has been shown to have an inhibitory effect on germination. The aim of this study was to characterize the interaction of these potent fire-generated compounds. Dose-response analysis, leaching tests and a detailed transcriptome study were performed using highly KAR(

Role of reactive oxygen species in the regulation of Arabidopsis seed dormancy.

Freshly harvested seeds of Arabidopsis thaliana, Columbia (Col) accession were dormant when imbibed at 25°C in the dark. Their dormancy was alleviated by continuous light during imbibition or by 5 weeks of storage at 20°C (after-ripening). We investigated the possible role of reactive oxygen species (ROS) in the regulation of Col seed dormancy. After 24 h of imbibition at 25°C, non-dormant seeds produced more ROS than dormant seeds, and their catalase activity was lower. In situ ROS localization revealed that germination was associated with an accumulation of superoxide and hydrogen peroxide i

Phytochrome-interacting factors integrate environmental signals to regulate tomato growth and development.

Plants respond to environmental cues, such as light and temperature, which regulate their growth and development. In the model plant Arabidopsis (Arabidopsis thaliana), PHYTOCHROME-INTERACTING FACTORS (PIFs) are central regulators of both shade avoidance and thermomorphogenesis. However, their functional roles in crop species are less well known. Here, we generated a tomato (Solanum lycopersicum) mutant lacking all 8 known PIF genes (slpifo), to investigate their roles under controlled and field conditions. We showed that SlPIFs are essential for shade responses, while thermomorphogenesis-indu

Genome-wide association analysis identifies novel genetic loci and candidate genes associated with seed dormancy in cultivated peanut.

Two significant regions on chromosomes A03 and A04, along with the candidate gene Arahy.5SR8FF, were identified to be associated with seed dormancy in peanut through GWAS. Two KASP markers were developed and validated, demonstrating their potential for use in marker-assisted breeding. Seed dormancy is a crucial trait that can influence both peanut yield and quality. In this study, a genome-wide association analysis (GWAS) was conducted using 353 peanut accessions with diverse genetic backgrounds from various global regions. The germination index was evaluated across multiple years and environm

AI-Augmented Multi-Omics for Abiotic Stress Responses: A New Frontier in Plant Hormone Systems Biology.

Plant phytohormone networks control growth, dormancy, and stress responses throughout plants' long lifespans and heterogeneous tissues. While multi-omics approaches have advanced the understanding of these regulatory systems, their application remains constrained by low spatial resolution, static sampling strategies, and limited capacity to capture nonlinear, context-dependent interactions. These limitations are especially evident in forest tree species, where hormone gradients change dynamically across developmental stages, seasons, and environmental conditions. The emergence of Artificial In
