Low phosphorus promotes NSP1-NSP2 heterodimerization to enhance strigolactone biosynthesis and regulate shoot and root architecture in rice.

Phosphorus is an essential macronutrient for plant development and metabolism, and plants have evolved ingenious mechanisms to overcome phosphate (Pi) starvation. However, the molecular mechanisms underlying the regulation of shoot and root architecture by low phosphorus conditions and the coordinated utilization of Pi and nitrogen remain largely unclear. Here, we show that Nodulation Signaling Pathway 1 (NSP1) and NSP2 regulate rice tiller number by promoting the biosynthesis of strigolactones (SLs), a class of phytohormones with fundamental effects on plant architecture and environmental res

Characterization of efficient plant-growth-promoting bacteria isolated from Sulla coronaria resistant to cadmium and to other heavy metals.

The inoculation of plants with plant-growth-promoting rhizobacteria has become a priority in the phytoremediation of heavy-metal-contaminated soils. A total of 82 bacteria were isolated from Sulla coronaria root nodules cultivated on four soil samples differently contaminated by heavy metals. The phenotypic characterization of these isolates demonstrated an increased tolerance to cadmium reaching 4.1mM, and to other metals, including Zn, Cu and Ni. Polymerase Chain Reaction/Restriction Fragment Length Polymorphism (PCR/RFLP) analysis showed a large diversity represented by genera related to Ag

A cytokinin-NIN module drives spontaneous root nodule formation in alfalfa.

Alfalfa can form root nodules even in the absence of rhizobial bacteria in a process controlled by cytokinin and the regulator NIN, and initiated by the plant itself; these observations provide a foundation for engineering nodule-like organs in other crops.

Roles of AGD2a in Plant Development and Microbial Interactions of Lotus japonicus.

Arabidopsis AGD2 (Aberrant Growth and Death2) and its close homolog ALD1 (AGD2-like defense response protein 1) have divergent roles in plant defense. We previously reported that modulation of salicylic acid (SA) contents by ALD1 affects numbers of nodules produced by Lotus japonicus, but AGD2's role in leguminous plants remains unclear. A combination of enzymatic analysis and biological characterization of genetic materials was used to study the function of AGD2 (LjAGD2a and LjAGD2b) in L. japonicus. Both LjAGD2a and LjAGD2b could complement dapD and dapE mutants of Escherichia coli and had a

Estimating Rhizobial Fitness During Legume Symbiosis: Enriching Viable Undifferentiated Bacteria from Root Nodules.

Advances in understanding the evolutionary ecology of the rhizobia-legume mutualism have been constrained by methodological limitations in efficiently measuring relative strain frequencies alongside measurements of absolute population sizes of rhizobia living in nodules. To examine strain competition in natural and agricultural ecosystems that harbor multiple strains of rhizobia, an increasing number of manipulative and observational studies have recently begun to examine dozens or hundreds of strains simultaneously. Assessing the competitive fitness of multiple strains in legume nodules requi

Research advances in the mechanisms of nitrogen-phosphorus synergy mediated by root exudates and mycorrhizal networks in cereal-legume intercropping systems.

The cereal-legume intercropping system achieves efficient utilization of nitrogen and phosphorus through interactions between roots and soil organisms, making it a key practice in sustainable agriculture. We systematically summarized the nitrogen and phosphorus mutual promotion mechanism mediated by root exudates and mycorrhizal networks in the cereal-legume intercropping system, and proposed future research directions. Cereal plants mobilize phosphorus in rhizosphere by secreting organic acids and phosphatases. The released phosphorus is then transferred to the rhizosphere of legumes via the

Medicago truncatula Molybdate Transporter type 1 (MtMOT1.3) is a plasma membrane molybdenum transporter required for nitrogenase activity in root nodules under molybdenum deficiency.

Molybdenum, as a component of the iron-molybdenum cofactor of nitrogenase, is essential for symbiotic nitrogen fixation. This nutrient has to be provided by the host plant through molybdate transporters. Members of the molybdate transporter family Molybdate Transporter type 1 (MOT1) were identified in the model legume Medicago truncatula and their expression in nodules was determined. Yeast toxicity assays, confocal microscopy, and phenotypical characterization of a Transposable Element from Nicotiana tabacum (Tnt1) insertional mutant line were carried out in the one M. truncatula MOT1 family

Systemic proteome remodeling in soybean during early nodulation: distinct responses in roots and leaves.

Here, we aimed to deepen our understanding of the integration between root and shoot signaling during early nodulation by jointly analyzing proteome-level dynamics in roots and leaves of the wild type (WT) and the hypernodulating autoregulation-of-nodulation (AON) mutant, nark. To this end, we performed a time-course shotgun proteomics experiment in soybean roots and leaves of the same plants inoculated with Bradyrhizobium diazoefficiens. In WT roots, we observed proteome changes linked to calcium-dependent signaling, cellular reorganization, and nutrient allocation. In the leaves, isoflavonoi

Enhanced biological nitrogen fixation and nodulation in alfalfa through the synergistic interactions between Sinorhizobium meliloti and Priestia aryabhattai.

Nitrogen fertilizer is crucial for agricultural output. However, prolonged overuse has resulted in nitrate leaching and potential soil acidification. Research on microbial fertilizer has become essential to enhance soil conditions and minimize nitrogen fertilizer usage. In alfalfa cultivation, research on efficient compound microbial agents remains limited, therefore this study concentrates on the investigation of dual microbial combinations. In the screening process, black soil was utilized with alfalfa plants as samples to identify a strain of rhizobacteria, Sinorhizobium meliloti LMGL3-1, e

Natural genetic variation in rhizobia and alfalfa influence nodule shape and symbiotic performance.

In many host-microbe symbioses, hosts develop specialized organs that house microbial symbionts. In legume-rhizobium symbiosis, these organs are called root nodules. While many studies have focused on host traits like plant growth, nodule number, and their structural changes, nodule morphology may influence rhizobial benefits (e.g., bacterial population size). Here, we investigated how variation in alfalfa nodule morphologies influences the fitness outcome of both partners. We used 117 strains and 3 alfalfa varieties to test how genetic variation influenced 1) nodule branch number, area, and r

Effect of seed priming with polyethylene glycol, distilled water, and sorbitol on physical, chemical quality parameters, and nodule microbiota of lentil.

The aim of this study was to investigate the effects of different seed priming solutions on physical and chemical quality parameters of lentils as well as nodule bacterial diversity before sowing. Therefore, lentil seeds were treated with polyethylene glycol (PEG 6000) (15%), sorbitol (6%), and distilled water, and none pretreated lentils (Lens culinaris) were used as control. The seeds were kept in these solutions for 24 h, then dried on toweling paper for 24 h, and used for the experiment. For nodule microbiota analysis, the plant root was divided into two equal parts, upper and lower, accor

Root nodules are distinct organs assembled from ancient developmental components. A new study shows that rhizobial Nod factors activate a conserved auxin module controlling lateral root formation, revealing how symbiotic signals can recruit pre-existing programmes without reproducing lateral root development.

Influence of Soil and Nonsoil Environments on Nodulation by Rhizobium trifolii.

Indigenous serotypes 1-01 and 2-02 of Rhizobium trifolii occupied similar percentages (18 to 23%) of root nodules on soil-grown subclover (Trifolium subterraneum L.) and were virtually absent (4.5%) from nodules of soil-grown white clover (Trifolium repens L.). In contrast (with the exception of one dilution [10]), serotype 1-01 occupied a substantial portion of nodules (16 to 40%) on white clover seedlings grown on mineral salts agar and exposed to samples of the same soil in the form of a 10-fold dilution series (10 to 10). Under the latter conditions, occupancy of subclover nodules by 1-01

Iron Oxide and Titanium Dioxide Nanoparticle Effects on Plant Performance and Root Associated Microbes.

In this study, we investigated the effect of positively and negatively charged Fe₃O₄ and TiO₂ nanoparticles (NPs) on the growth of soybean plants (Glycine max.) and their root associated soil microbes. Soybean plants were grown in a greenhouse for six weeks after application of different amounts of NPs, and plant growth and nutrient content were examined. Roots were analyzed for colonization by arbuscular mycorrhizal (AM) fungi and nodule-forming nitrogen fixing bacteria using DNA-based techniques. We found that plant growth was significantly lower with the application of TiO₂ as compared to F

Engineering nitrogen-fixing symbiosis: bridging signaling and intracellular entry.

Nitrogen availability is a major constraint on plant growth and agricultural productivity. Legumes and several other plant lineages circumvent this limitation by establishing symbiotic associations with nitrogen-fixing microorganisms, which convert atmospheric dinitrogen into ammonia that the host assimilates. Since root nodules were first recognized as sites of biological nitrogen fixation in the late 19th century, engineering this capacity in nonlegume crops has remained a long-standing goal. However, two key challenges remain: enabling intracellular rhizobial infection and establishing a mo

CPOP1 is a key enzyme required for nodule microenvironment control and successful symbiotic nitrogen fixation in Lotus japonicus.

Symbiotic nitrogen fixation in legumes requires the exquisite regulation of the environment within the infected region of the nodule. The microaerobic environment critical for nitrogenase activity is maintained through the physical oxygen diffusion barrier of the cortex and locally the oxygen-binding protein leghemoglobin (Lb). Leghemoglobin binds and releases oxygen with heme moiety to maintain oxygen gradients inside the infected cell (IC) during nitrogen fixation. Heme binds to diverse proteins and plays critical roles in different redox reactions. However, the role and regulation of host-c

Molecular insights into bacteroid development during Rhizobium-legume symbiosis.

Rhizobial soil bacteria can form a symbiosis with legumes in which the bacteria fix atmospheric nitrogen into ammonia that can be utilized by the host. The plant, in turn, supplies the rhizobia with a carbon source. After infecting the host cell, the bacteria differentiate into a distinct bacteroid form, which is able to fix nitrogen. The bacterial BacA protein is essential for bacteroid differentiation in legumes producing nodule-specific cysteine-rich peptides (NCRs), which induce the terminal differentiation of the bacteria into bacteroids. NCRs are antimicrobial peptides similar to mammali

The Role of Hydrogen Sulfide in Plant Roots during Development and in Response to Abiotic Stress.

Hydrogen sulfide (H2S) is regarded as a "New Warrior" for managing plant stress. It also plays an important role in plant growth and development. The regulation of root system architecture (RSA) by H2S has been widely recognized. Plants are dependent on the RSA to meet their water and nutritional requirements. They are also partially dependent on the RSA for adapting to environment change. Therefore, a good understanding of how H2S affects the RSA could lead to improvements in both crop function and resistance to environmental change. In this review, we summarized the regulating effects of H2S

Dryas as a Model for Studying the Root Symbioses of the Rosaceae.

The nitrogen-fixing root nodule symbiosis is restricted to four plant orders: Fabales (legumes), Fagales, Cucurbitales and Rosales (Elaeagnaceae, Rhamnaceae, and Rosaceae). Interestingly all of the Rosaceae genera confirmed to contain nodulating species (i.e., Cercocarpus, Chamaebatia, Dryas, and Purshia) belong to a single subfamily, the Dryadoideae. The Dryas genus is particularly interesting from an evolutionary perspective because it contains closely related nodulating (Dryas drummondii) and non-nodulating species (Dryas octopetala). The close phylogenetic relationship between these two sp

Managing Friends and Foes: Sanctioning Mutualists in Mixed-Infection Nodules Trades off With Defense Against Antagonists.

Successful plant growth requires plants to minimize harm from antagonists and maximize benefit from mutualists. However, these outcomes may be difficult to achieve simultaneously, since plant defenses activated in response to antagonists can compromise mutualism function, and plant resources allocated to defense may trade off with resources allocated to managing mutualists. Here, we investigate how antagonist attack affects plant ability to manage mutualists with sanctions, in which a plant rewards cooperative mutualists and/or punishes uncooperative mutualists. We studied interactions among w
