The ferroxidases LPR1 and LPR2 control iron translocation in the xylem of Arabidopsis plants.

Iron (Fe) deficiency is common in agricultural crops and affects millions of people worldwide. Translocation of Fe in the xylem is a key step for Fe distribution in plants. The mechanism controlling this process remains largely unknown. Here, we report that two Arabidopsis ferroxidases, LPR1 and LPR2, play a crucial and redundant role in controlling Fe translocation in the xylem. LPR1 and LPR2 are mainly localized in the cell walls of xylem vessels and the surrounding cells in roots, leaves, and stems. Knockout of both LPR1 and LPR2 increased the proportion of Fe(II) in the xylem sap, and caus

OsDRP1C mediates root development by modulating OsPIN1 cycling in rice.

Root length plays a crucial role in plants' ability to absorb water and nutrients from the soil. However, the molecular mechanisms underlying root elongation in rice (Oryza sativa) remain unclear. Here, we identified a short root mutant, short root-31 (sr31), and cloned the causal gene by map-based cloning, which carries a point mutation in the coding sequence of DYNAMIN-RELATED PROTEIN 1C (OsDRP1C). Genetic complementation analyses demonstrated that OsDRP1C is responsible for the phenotypic defect of sr31. OsDRP1C is widely expressed in roots, with higher expression in root meristem, epiderma

Flexible Drinking Straws of the Cretaceous: A Novel Type of Labium in Mesozoic Auchenorrhyncha and Its Functional Implications.

The evolution of novel feeding structures played a major role in the diversification of Hemiptera. Here, we investigate the morphology and inferred function of a highly specialized labium in the extinct Cretaceous planthopper families Neazoniidae and Mimarachnidae. These insects possessed a feeding apparatus unparalleled among extant Hemiptera that is characterized by an extremely elongated labium with a densely corrugated apical segment formed by sinusoidal cuticular folds. Comparative analysis, aided by micro-computed tomography, indicates that the cuticular corrugations were a mechanical ad

MtGA2ox10 encoding C20-GA2-oxidase regulates rhizobial infection and nodule development in Medicago truncatula.

Gibberellin (GA) plays a controversial role in the legume-rhizobium symbiosis. Recent studies have shown that the GA level in legumes must be precisely controlled for successful rhizobial infection and nodule organogenesis. However, regulation of the GA level via catabolism in legume roots has not been reported to date. Here, we investigate a novel GA inactivating C20-GA2-oxidase gene MtGA2ox10 in Medicago truncatula. RNA sequencing analysis and quantitative polymerase chain reaction revealed that MtGA2ox10 was induced as early as 6 h post-inoculation (hpi) of rhizobia and reached peak transcr

VpSTS29/STS2 enhances fungal tolerance in grapevine through a positive feedback loop.

Accumulation of stilbene phytoalexins stimulates resistance mechanisms against the grapevine fungus Uncinula necator. However, the defensive mechanisms triggered by stilbene synthase (STS) genes, remain largely unknown. Here, we report the function and molecular mechanism of the stilbene synthase gene VpSTS29/STS2 from Vitis pseudoreticulata in the regulation of plant responses to powdery mildew. Stilbene synthesis occurred mainly in root tips and mesophyll cells of transgenic grapevines via transport through the vascular bundles. Overexpression of VpSTS29/STS2 in Vitis vinifera increased the

The Arabidopsis COPT6 transport protein functions in copper distribution under copper-deficient conditions.

Copper (Cu), an essential redox active cofactor, participates in fundamental biological processes, but it becomes highly cytotoxic when present in excess. Therefore, living organisms have established suitable mechanisms to balance cellular and systemic Cu levels. An important strategy to maintain Cu homeostasis consists of regulating uptake and mobilization via the conserved family of CTR/COPT Cu transport proteins. In the model plant Arabidopsis thaliana, COPT1 protein mediates root Cu acquisition, whereas COPT5 protein functions in Cu mobilization from intracellular storage organelles. The f

Fluorescence Detection of Pyrene-Stained Bacillus amyloliquefaciens MHR24 in Tomato (Solanum lycopersicum L.) Stem Tissues.

The PGPR strain of Bacillus amyloliquefaciens MHR24 (MHR24) was recently reported as a strong biocontrol strain. In this study, MHR24 was used to investigate phyllosphere effects during inoculations of tomato leaves (Solanum lycopersicum L.). When MHR24 was inoculated on foliar tissue, it caused apical chlorosis symptoms at 3-6 days after infiltration or submersion, which suggests that the bacterium may adopt a potentially pathogenic lifestyle in the phyllosphere. In order to detect the MHR24 interaction with the plant, it was stained with the commercial fluorophore 8-hydroxypyrene-1,3,6-trisu

AtCRCT1 and AtCRCT2, homologs of CO2 responsive CCT protein in Arabidopsis thaliana, are involved in the regulation of starch synthesis-related gene expression.

The CO2-responsive CCT protein (CRCT) is a transcription factor that regulates the expression of starch synthesis-related genes, and consequently starch content in the vegetative organs of rice. Two CRCT homologs, AtCRCT1 and AtCRCT2, were identified in the genome of Arabidopsis thaliana. AtCRCT1 and AtCRCT2 were expressed in most organs of the plant, and AtCRCT2 in particular was expressed in their vascular bundles. The expression of these genes was markedly upregulated by sucrose treatment and tended to increase under elevated CO2 conditions. The overexpression or knockout of these genes did

Functional analysis of the cotton CLE polypeptide signaling gene family in plant growth and development.

The CLAVATA3 (CLV3)/EMBRYO SURROUNDING REGION (ESR)-RELATED (CLE) gene family encodes a large number of polypeptide signaling molecules involved in the regulation of shoot apical meristem division and root and vascular bundle development in a variety of plants. CLE family genes encode important short peptide hormones; however, the functions of these signaling polypeptides in cotton remain largely unknown. In the current work, we studied the effects of the CLE family genes on growth and development in cotton. Based on the presence of a conserved CLE motif of 13 amino acids, 93 genes were charac

A high affinity pyruvate decarboxylase is present in cottonwood leaf veins and petioles: a second source of leaf acetaldehyde emission?

Considerable evidence indicates that acetaldehyde is released from the leaves of a variety of plants. The conventional explanation for this is that ethanol formed in the roots is transported to the leaves where it is converted to acetaldehyde by the alcohol dehydrogenase (ADH) found in the leaves. It is possible that acetaldehyde could also be formed in leaves by action of pyruvate decarboxylase (PDC), an enzyme with an uncertain metabolic role, which has been detected, but not characterized, in cottonwood leaves. We have found that leaf PDC is present in leaf veins and petioles, as well as in

Arabidopsis dynamin-related proteins, DRP2A and DRP2B, function coordinately in post-Golgi trafficking.

Dynamin-related proteins (DRPs) are large GTPases involved in a wide range of cellular membrane remodeling processes. In Arabidopsis thaliana, two paralogous land plant-specific type DRPs, DRP2A and DRP2B, are thought to participate in the regulation of post-Golgi trafficking. Here, we examined their molecular properties and functional relationships. qRT-PCR and GUS assays showed that DRP2A and DRP2B were expressed ubiquitously, although their expressions were strongest around root apical meristems and vascular bundles. Yeast two-hybrid, bi-molecular fluorescent complementation, and co-immunop

A mixed microscopic method for differentiating seven species of "Bixie"-related Chinese Materia Medica.

Confusion in the species associated with the name of "Bixie" in Chinese Materia Medica began centuries ago. In recent decades, diverse medicinal plants from the genera Dioscorea and Smilax, and even minor species from the genus Heterosmilax, have been documented under the name Bixie or a very similar name as folk medicines in different areas of China. However, the traditional efficacies and chemical profiles of these herbs are not exactly the same and even vary wildly. Comprehensive authentication of multiple Bixie herbs has not yet been attempted. To differentiate and ensure the correct use o

Knockdown of Fa-miR159/160 reprograms plant development and improves heat-stress resistance in tall fescue.

MicroRNA159/160 (miR159/160) has been implicated in plant development and stress resistance in annual crops and Arabidopsis, yet their functional roles in perennial cool-season forage and turf grass remain elusive. Tall fescue (Festuca arundinacea Schreb.) is one main perennial cool-season forage and turfgrass species worldwide. Here, we found that Fa-miR159/160 has 7/4 copies in tall fescue, respectively. Knockdown of both Fa-miR159 and Fa-miR160 (STTM159/160) using the short tandem target mimics (STTM) system resulted in a dwarf phenotype with higher tiller density, lower growth rate levels,

Spatiotemporal metabolomics of tobacco leaves in response to Pseudomonas syringae infection.

Tobacco (Nicotiana tabacum L.) is a major economic crop and a model for plant-pathogen interactions, yet the spatiotemporal dynamics of defense metabolism during infection remain poorly characterized. Here, we used MALDI-MSI-based spatial metabolomics to systematically profile tobacco leaves during Pseudomonas syringae infection. Multidimensional analysis of 1,399 annotated metabolites revealed distinct spatiotemporal regulation patterns. Temporally, early infection (12 h postinfection (hpi)) was characterized by increased organic acids and terpenoids, followed by a mid-stage shift toward phen

Silicon nutrition improves growth of salt-stressed wheat by modulating flows and partitioning of Na+, Cl- and mineral ions.

Silicon (Si) is reported to improve salt stress tolerance of cereals, but little is known about the effects of Si on flows and partitioning of sodium (Na+), chloride (Cl-), and essential mineral ions at the tissue and cellular level. Wheat (Triticum aestivum L.) was exposed to 200 mM NaCl for 30 d in hydroponics, with or without 2 mM Si. X-ray microanalysis coupled with scanning electron microscopy (SEM) was used to quantify the cell-specific ion profiles across root and leaf cells, paralleled by measurements of wheat growth and physiological responses. Under salt stress, higher Na+ and Cl- co

Genome-wide identification of the ECERIFERUM (CER) gene family in cabbage and critical role of BoCER4.1 in wax biosynthesis.

The ECERIFERUM (CER) gene family is essential for the biosynthesis of plant cuticular wax. In this study, 32 BoCER genes were identified in cabbage through genome-wide analysis. We found that the BoCER genes are highly conserved with their homologous counterparts in Arabidopsis thaliana. However, there was a significant divergence in the expression pattern among the BoCER paralogs, which suggests the occurrence of functional specialization during evolution. The expression analysis also showed that most of the BoCER genes are expressed in the aboveground part. Cis-regulatory element analysis su

Individual architecture and photosynthetic performance of the submerged form of Drosera intermedia Hayne.

Drosera intermedia grows in acidic bogs in parts of valleys that are flooded in winter, and that often dry out in summer. It is also described as the sundew of the most heavily hydrated habitats in peatlands, and it is often found in water and even underwater. This sundew is the only one that can tolerate long periods of submersion, and more importantly produces a typical submerged form that can live in such conditions for many years. Submerged habitats are occupied by D. intermedia relatively frequently. The aim of the study was to determine the environmental conditions and architecture of in
