A spontaneous true breeding homeotic gene mutant Pps-1 with distinct partial petaloid sepals was detected in the population of downy mildew (DM)-resistant elite accession I-14 during our studies for the identification of disease resistance sources in opium poppy. The trait was found to be stable and inherited truly in the subsequent generations. Genetic studies were carried out through systematic reciprocal crosses with the parental wild-type genotype I-14, and segregation pattern of phenotypic characteristics in F(1) and F(2) populations clearly indicated single recessive nuclear gene control

BLADE-ON-PETIOLE 2 (BOP2) plays a pivotal role in leaf morphogenesis. Liriodendron tulipifera is a suitable model for exploring the molecular mechanisms underlying leaf serration formation, which are largely unknown. Here, we isolated the full-length LtuBOP2 gene and its promoter from L. tulipifera and characterized its function in leaf morphogenesis through multidimensional approaches. The spatiotemporal expression pattern of LtuBOP2 indicated the high expression of LtuBOP2 in stems and leaf buds. We constructed LtuBOP2 promoter, fused the promoter sequences to the β-glucuronidase (GUS) gene,

This study overcomes a key technical barrier by establishing transformation and enabling first CRISPR/Cas9 editing in Oxalis, providing a platform for functional genomics and breeding. The lack of an efficient genetic transformation system has considerably hindered functional genomics studies in Oxalis vulcanicola 'Sunset Velvet'. Here, we established a stable and efficient Agrobacterium-mediated transformation system using stem segments as explants. Key parameters, including pre-culture duration, infection time, Agrobacterium cell density, acetosyringone (AS) concentration, and co-cultivation

Environmental factors that damage DNA have various lengths of exposure and intensity levels. Although the results of increasing the intensity of a DNA damaging agent is often predictable, it is not clear whether the stage during development when the exposure is received has any influence on the amount of DNA damage. In this paper we analyzed the influence of UVB on the stability of Arabidopsis thaliana and the Nicotiana tabacum genomes. Our experiments showed that the acute exposure to UVB produces a significantly greater increase in homologous recombination frequency (HRF) and recombination r

Transgenes promise to reduce insecticide and fungicide use but relatively little has been done to significantly reduce herbicide use through genetic engineering. Recently, three strategies for transgene utilization have been developed that have the potential to change this. These are the improvement of weed-specific biocontrol agents, enhancement of crop competition or allelopathic traits, and production of cover crops that will self-destruct near the time of planting. Failsafe risk mitigation technologies are needed for most of these strategies.

Ionizing radiation (IR) is a known mutagen responsible for causing DNA strand breaks in all living organisms. Strand breaks thus created can be repaired by different mechanisms, including homologous recombination (HR), one of the key mechanisms maintaining genome stability [A. Britt, DNA damage and repair in plants, Annu. Rev. Plant. Phys. Plant Mol. Biol., 45 (1996) 75-100; H. Puchta, B. Hohn, From centiMorgans to basepairs: homologous recombination in plants, Trends Plant Sci., 1 (1996) 340-348.]. Acute or chronic exposure to IR may have different influences on the genome integrity. Recent understanding of plant metabolism has made it possible to increase the iron, zinc and beta-carotene (provitamin A) content in staple foods by both conventional plant breeding and genetic engineering. Improving the micronutrient composition of plant foods may become a sustainable strategy to combat deficiencies in human populations, replacing or complementing other strategies such as food fortification or nutrient supplementation.

Meiosis is a key process in sexual reproduction, orchestrated by numerous proteins that differ in their spatiotemporal expression level and dynamics. Employing meiotic gene promoters typically leads to varying levels of transgene expression within meiotic and non-meiotic cells in plants. Hence, their application for protein expression during meiosis is limited. Here, we present a tool for meiosis-specific protein co-expression in Arabidopsis thaliana. By utilizing ribosome-skipping peptides, we leverage the distinct expression patterns of the two meiotic genes ASY1 and MTOPVIB to co-express va

Our review illustrates how the dexamethasone-inducible LhGR/pOp system has been used across numerous tissues/organs and plant species, and summarizes the resources and inducer application procedures established to date. The establishment of groundbreaking molecular biology tools has enabled the rapid advancement of research based on the model plant Arabidopsis, which currently strongly benefits non-model but economically important species. Spatiotemporal control of transgene expression via the chemically inducible system GR-LhG4/pOp has proven to be a particularly powerful, universal and non-i

Lettuce (Lactuca sativa) is a popular leafy vegetable with global production of ~28 million Mt, cultivated >1 million hectares, with a market value of US$ 4 billion in 2022. However, lettuce is highly susceptible to fungal pathogens that drastically reduce biomass and quality due to spoilage/rot. Therefore, in this study, we investigated the expression of chitinase genes via the lettuce chloroplast genome to enhance biomass and disease resistance. Site-specific integration of the expression cassette into chloroplast genomes was confirmed using two sets of PCR primers. Homoplasmy in transplasto

Brassinosteroids (BRs) are steroid phytohormones which regulate various developmental and physiological processes throughout plant life cycle, from seed development and germination, up to modulation of reproduction and senescence. Importantly, mutants defective in the BR biosynthesis or response show various degree of plant height reduction (dwarfism or semi-dwarfism). This agronomic trait is of particular importance considering that in contrast to tall cereal varieties, semi-dwarf cereal plants are more tolerant to lodging which occurs during unfavorable weather conditions and constitutes a s

In plant immunity, the mutually antagonistic hormones salicylic acid (SA) and jasmonic acid (JA) are implicated in resistance to biotrophic and necrotrophic pathogens, respectively. Promoters that can respond to both SA and JA signals are urgently needed to engineer plants with enhanced resistance to a broad spectrum of pathogens. However, few natural pathogen-inducible promoters are available for this purpose. To address this problem, we have developed a strategy to synthesize dual SA- and JA-responsive promoters by combining SA- and JA-responsive cis elements based on the interaction between

Site-directed nucleases (SDNs) used for targeted genome editing are powerful new tools to introduce precise genetic changes into plants. Like traditional approaches, such as conventional crossing and induced mutagenesis, genome editing aims to improve crop yield and nutrition. Next-generation sequencing studies demonstrate that across their genomes, populations of crop species typically carry millions of single nucleotide polymorphisms and many copy number and structural variants. Spontaneous mutations occur at rates of ∼10-8 to 10-9 per site per generation, while variation induced by chemical

Artemisinin, the endoperoxide sesquiterpene lactone, is an effective antimalarial drug isolated from the Chinese medicinal plant Artemisia annua L. Due to its effectiveness against multi-drug-resistant cerebral malaria, it becomes the essential components of the artemisinin-based combination therapies which are recommended by the World Health Organization as the preferred choice for malaria tropica treatments. To date, plant A. annua is still the main commercial source of artemisinin. Although semi-synthesis of artemisinin via artemisinic acid in yeast is feasible at present, another promising

Plants engineered with synthetic genetic programs can transform how we monitor and manage the extension of crop pests and diseases. Here, we establish a bioluminescent platform in Nicotiana benthamiana for autonomous viral sensing based on the fungal bioluminescence pathway (FBP). We first demonstrate that recombinant viruses can deliver missing pathway components, establishing a methodological framework for spatially resolved tracking of infection dynamics. Leveraging this starting point, we develop a dual-output sentinel circuit that uses a protease-responsive bioluminescence resonance energ

Phlorizin has antioxidant and anti-aging pharmacological activities, which render it a highly promising candidate for applications in functional foods and the pharmaceutical industry. However, the incomplete elucidation of phlorizin biosynthesis in plants has constrained its large-scale production and application. Lithocarpus litseifolius, which contains a notably higher level of phlorizin than most plant species, serves as an ideal model for identifying key enzymes involved in phlorizin biosynthesis. The genes MdDBR, MdCHS, and MdMYB10 from Malus domestica Borkh. and LlP2'GT from L. litseifol

Transgenic Arabidopsis thaliana expressing an anti-rabies monoclonal antibody (mAb), SO57, was obtained using Agrobacterium-mediated floral dip transformation. The endoplasmic reticulum (ER) retention signal Lys-Asp-Glu-Leu (KDEL) was tagged to the C-terminus of the anti-rabies mAb heavy chain to localize the mAb to the ER and enhance its accumulation. When the inaccurately folded proteins accumulated in the ER exceed its storage capacity, it results in stress that can affect plant development and growth. We generated T1 transformants and obtained homozygous T3 seeds from transgenic Arabidopsi

Antibiotic and herbicide resistance genes are currently the most frequently used selectable marker genes for plant research and crop development. However, the use of antibiotics and herbicides must be carefully controlled because the degree of susceptibility to these compounds varies widely among plant species and because they can also affect plant regeneration. Therefore, new selectable marker systems that are effective for a broad range of plant species are still needed. Here, we report a simple and inexpensive system based on providing transgenic plant cells the capacity to convert a nonmet

Expansins are cell wall loosening proteins that appear to permit the microfibril matrix network to slide in growing plant cell walls, thereby enabling the wall to expand. To scrutinize possible impacts on plant growth and development when expansins are over-expressed, we characterized phenotypic alterations of the transgenic plants that constitutively expressed AtEXP3 or AtEXP-beta1 under control of 35S-CaMV promoter. Our results suggest that both AtEXP3-OX and AtXPbeta1-OX are very sensitive to salt stress. However, the mechanisms underlying their enhanced salt sensitivity appear to be differ

Soybean is one of the major sources of high-quality protein and oil and plays a crucial role in ensuring nutritional and economic security. The rapidly increasing population, soybean supply-demand imbalance, and yield losses due to soil salinity collectively necessitate the development of salt-tolerant cultivars. Perennial wild Glycine species harbor-rich genetic diversity and constitute a reservoir of stress-tolerant genes. This study systematically characterized the COBL gene family in the salt-resilient Glycine tabacina (Labill.) Benth., a wild soybean that tolerates 500 mM NaCl with 100% s
