PLASTID ENVELOPE ION CHANNELS (PEC1/2) link Ca2+ and jasmonic acid signaling in plant cells.

Plants constantly encounter adverse environmental interactions. One organelle is particularly specialized in stress signaling and phytohormone synthesis: the plastid. Calcium (Ca2+), a key second messenger, is known to intersect with cellular phytohormone signaling networks. While cytosolic Ca2+ dynamics have been studied extensively, the physiological relevance of stromal Ca2+ transients and the identity of channels mediating rapid Ca2+ flux into plastids remain largely unexplored. In this study, we provide evidence for PLASTID ENVELOPE ION CHANNELS (PECs) as long-sought mediators of fast-act

Mitochondria and plastid response to genome duplication in diversified strategies to reconcile nucleo-cytoplasmic interaction in autotetraploid rice.

Whole-genome duplication is an important evolutionary mechanism in plants. Although numerous studies have morphologically characterized the polyploid plants, such as increased grain yield and enhanced resistance to pests and diseases, few studies are involved in the response of mitochondria and chloroplasts to the nuclear genome shock to reconcile the nucleo-cytoplasmic interactions in tetraploid rice. In this study, we employed the diploid rice cultivars Huang-Jin-Ke (HJK) and 032 and the artificially synthesized autotetraploid rice lines HJK-4n and 032-4n with normal fertility, to investigat

Impact of the Simultaneous Loss of the Plastid Envelope-Localized Homologous Pair Ycf10 and DLDG1 in Plant Growth and Photosynthetic Performance.

Ycf10 and DLDG1 are homologous proteins embedded in the chloroplast envelope membranes (EM) and encoded in the plastidial and nuclear genomes, respectively. We previously characterized Arabidopsis dldg1 and tobacco ycf10 mutants, and showed that these proteins mediate proton transport across the EM, thereby influencing ATP synthesis and the non-photochemical quenching (NPQ) induction. However, the consequences of the simultaneous loss of Ycf10/DLDG1 homologs have remained unclear. Here, we generated and analyzed Arabidopsis ycf10 and ycf10-dldg1 double mutants. These mutants exhibited a pale-g

Interplay between Heat Shock Protein 90 and HY5 controls PhANG expression in response to the GUN5 plastid signal.

The presence of genes encoding organellar proteins in different cellular compartments necessitates a tight coordination of expression by the different genomes of the eukaryotic cell. This coordination of gene expression is achieved by organelle-to-nucleus or retrograde communication. Stress-induced perturbations of the tetrapyrrole pathway trigger large changes in nuclear gene expression in plants. Recently, we identified HSP90 proteins as ligands of the putative plastid signal Mg-ProtoIX. In order to investigate whether the interaction between HSP90 and Mg-ProtoIX is biologically relevant, we

Genome-Wide and GWAS Dissection of Maize Fibrillin Genes Reveals Plastid Regulators of Drought and Salt Stress Tolerance.

Fibrillins (FBNs) are conserved plastid-associated proteins implicated in plant development and abiotic stress responses; however, their roles in maize remain unclear. In this study, through a genome-wide bioinformatic analysis, we identified 14 ZmFBN genes in the maize genome and characterized their phylogeny, chromosomal distribution, gene structure, conserved motifs, and promoter cis-elements. ZmFBN members were grouped into several subfamilies that all retain a conserved PAP_fibrillin domain, whereas the variation in exon-intron organization, motif composition, and regulatory elements sugg

Optimized Extraction Protocol of Chloroplast DNA from Chenopodium album (Amaranthaceae) Enabling Whole Plastid Genome Sequencing.

Chenopodium is one of the most complex genera in the family Amaranthaceae, exhibiting ploidy levels ranging from diploid to hexaploid. The lack of distinct morphological characters and phenotypic plasticity has complicated species identification in this genus. The advent of next-generation sequencing technology has enabled in-depth genomic studies of Chenopodium species. To date, the chloroplast genomes of six Chenopodium species-Chenopodium acuminatum, C. ficifolium, C. berlandieri, C. petiolare, C. quinoa, and C. album-have been reported using genomic DNA as templates. On the other hand, hig

Extensive reorganization of the plastid genome of Trifolium subterraneum (Fabaceae) is associated with numerous repeated sequences and novel DNA insertions.

The plastid genome of Trifolium subterraneum is 144,763 bp, about 20 kb longer than those of closely related legumes, which also lost one copy of the large inverted repeat (IR). The genome has undergone extensive genomic reconfiguration, including the loss of six genes (accD, infA, rpl22, rps16, rps18, and ycf1) and two introns (clpP and rps12) and numerous gene order changes, attributable to 14-18 inversions. All endpoints of rearranged gene clusters are flanked by repeated sequences, tRNAs, or pseudogenes. One unusual feature of the Trifolium subterraneum genome is the large number of disper

The Complete Chloroplast Genome of Ficus gasparriniana var. laceratifolia Reveals Discordance Between Morphology-Based Classification and Plastid Phylogeny.

Background/Objectives:Ficus gasparriniana var. laceratifolia (H. Lév. & Vaniot) Corner is treated as a variety of F. gasparriniana and placed in Ficus subg. Ficus on morphological grounds, but complete plastome evidence for its plastid phylogenetic placement has been lacking. We assembled and analyzed its chloroplast genome to evaluate this morphology-based placement using plastid genomic evidence and to expand genomic resources for the genus. Methods: The plastome was assembled from paired-end reads using GetOrganelle and subsequently annotated. We characterized its genome architecture, simpl

Robust Phylogeny of Tetrastigma (Vitaceae) Based on Ten Plastid DNA Regions: Implications for Infrageneric Classification and Seed Character Evolution.

Tetrastigma (Miq.) Planch. is one of the most species-rich genera of the economically and agronomically important grape family Vitaceae. It includes ca. 95 species widely distributed in the tropics and subtropics of Asia and Australia. Species of Tetrastigma exhibit great diversity in both vegetative and reproductive characters. Here we inferred a well-supported phylogeny of Tetrastigma based on ten chloroplast DNA regions with an expanded taxon sampling of 72 species and two varieties. Our molecular results support six major clades within Tetrastigma and the relationships among these clades w

Identification and Characterization of a Plastidic Adenine Nucleotide Uniporter (OsBT1-3) Required for Chloroplast Development in the Early Leaf Stage of Rice.

Chloroplast development is an important subject in botany. In this study, a rice (Oryza sativa) mutant exhibiting impairment in early chloroplast development (seedling leaf albino (sla)) was isolated from a filial generation via hybridization breeding. The sla mutant seedlings have an aberrant form of chloroplasts, which resulted in albinism at the first and second leaves; however, the leaf sheath was green. The mutant gradually turned green after the two-leaf stage, and the third leaf was a normal shade of green. Map-based cloning indicated that the gene OsBT1-3, which belongs to the mitochon

How sequence variants of a plastid-replicating viroid with one single nucleotide change initiate disease in its natural host.

Understanding how viruses and subviral agents initiate disease is central to plant pathology. Whether RNA silencing mediates the primary lesion triggered by viroids (small non-protein-coding RNAs), or just intermediate-late steps of a signaling cascade, remains unsolved. While most variants of the plastid-replicating peach latent mosaic viroid (PLMVd) are asymptomatic, some incite peach mosaics or albinism (peach calico, PC). We have previously shown that two 21-nt small RNAs (PLMVd-sRNAs) containing a 12-13-nt PC-associated insertion guide cleavage, via RNA silencing, of the mRNA encoding a h

EMB2738, which encodes a putative plastid-targeted GTP-binding protein, is essential for embryogenesis and chloroplast development in higher plants.

In higher plants, chloroplasts carry out many important functions, and normal chloroplast development is required for embryogenesis. Numerous chloroplast-targeted proteins involved in embryogenesis have been identified. Nevertheless, their functions remain unclear. In this study, a chloroplast-localized protein, EMB2738, was reported to be involved in Arabidopsis embryogenesis. EMB2738 knockout led to defective embryos, and the embryo development in emb2738 was interrupted after the globular stage. Complementation experiments identified the AT3G12080 locus as EMB2738. Cellular observation indi

Elevated ROS-scavenging enzymes contribute to acclimation to UV-B exposure in transplastomic tobacco plants, reducing the role of plastid peroxidases.

Leaf peroxidases play a key role in the successful acclimation of plants to low UV-B doses. The aim of the present study was to examine whether selective enhancement of alternative chloroplast antioxidant pathways achieved by chloroplast transformation affected the need for peroxidase defense. Transplastomic tobacco lines expressing glutathione reductase in combination with either dehydroascorbate reductase or glutathione-S-transferase in their plastids exhibited better tolerance to supplemental UV-B than wild type plants. After 10days UV treatment, both the maximum and effective quantum yield

Barley anther culture: assessment of carbohydrate effects on embryo yield, green plant production and differential plastid development in relation with albinism.

Sugars and polyols were tested at different steps of anther culture in barley (Hordeum vulgare L.) to elucidate their influence on both the overall yield of androgenesis and the structure of plastids in relation to albinism. During the pretreatment period, the osmotic regulation in the medium was beneficial to microspore embryogenesis regardless of the type and concentration of the tested osmoticum. The use of mannitol (300 mOsm/kg), sorbitol (180 mOsm/kg), PEG (240 mOsm/kg) and sucrose (180 mOsm/kg) gave the best results in terms of green plant production, although the influence of each subst

Characterization of the complete plastid genome and 45S nrDNA sequences of Murdannia medica (Lour.) D.Y. Hong (Commelinaceae) and its phylogenetic analysis.

Murdannia medica is a Vietnamese traditional medicinal herb. Here, we report DNA super‑barcodes comprising the complete plastid genome (plastome, 177,455 bp) and 45S nuclear ribosomal DNA (45S nrDNA, 5,741 bp) of M. medica. The plastome has a large single‑copy (LSC, 100,132 bp), a small single‑copy (SSC, 21,125 bp), and a pair of inverted repeats (IRs) of 28,099 bp each. It contains 115 annotated genes and has an overall A/T content of 68.2%. Plastome‑based phylogenetic analyses show that M. medica sister with M. edulis. These new genomic resources provide a valuable foundation for future cult

Disrupting the plastidic iron-sulfur cluster biogenesis pathway in Toxoplasma gondii has pleiotropic effects irreversibly impacting parasite viability.

Like many other apicomplexan parasites, Toxoplasma gondii contains a plastid harboring key metabolic pathways, including the sulfur utilization factor (SUF) pathway that is involved in the biosynthesis of iron-sulfur clusters. These cofactors are crucial for a variety of proteins involved in important metabolic reactions, potentially including plastidic pathways for the synthesis of isoprenoid and fatty acids. It was shown previously that impairing the NFS2 cysteine desulfurase, involved in the first step of the SUF pathway, leads to an irreversible killing of intracellular parasites. However,

Plastid-mediated feedback regulation of Arabidopsis LYCOPENE EPSILON CYCLASE is modulated by the promoter and a 5'UTR structural variant harbouring a conserved IRES.

BACKGROUND: Perturbations in flux through the carotenoid pathway can signal control over nuclear gene expression to maintain metabolic homeostasis in plastids; however, the genetic targets and mechanisms are rarely characterised in plants. RESULTS: Utilising mutants, over- and exogenous expression lines, and chemical inhibitors of carotenogenesis, we demonstrate that Arabidopsis LYCOPENE EPSILON CYCLASE (εLCY) mRNA levels correlate with changes in the accumulation of cyclic carotenoids generated by the β-branch in the metabolic pathway. Cell-specific CO2 fixation rates of two distinct groups of plastidic protists in the Atlantic Ocean remain unchanged after nutrient addition.

To assess the role of open-ocean ecosystems in global CO2 fixation, we investigated how picophytoplankton, which dominate primary production, responded to episodic increases in nutrient availability. Previous experiments have shown nitrogen alone, or in combination with phosphorus or iron, to be the proximate limiting nutrient(s) for total phytoplankton grown over several days. Much less is known about how nutrient upshift affects picophytoplankton CO2 fixation over the duration of the light period. To address this issue, we performed a series of small volume (8-60 ml) - short term (10-11 h) n

Plastid phylogenomics of the parietal clade of Malpighiales: ancestral genome synteny and congruent signal across exons, introns and spacers in support of Salicaceae relationships.

BACKGROUND AND AIMS: The parietal clade is one of the major lineages of the angiosperm order Malpighiales. Several nodes in the phylogenetic trees inferred from concatenated chloroplast genes exhibited low support, and various Malpighiales genera were found to have highly altered plastid genomes. METHODS: Here we newly generated seven complete plastid genomes to represent the major lineages of the pantropical genus Casearia using genome skimming. Together with published genome sequences of various sources, we comparatively analysed the genome structure of all major lineages of the Salicaceae
