Bone Morphogenic Protein 4-Smad-Induced Upregulation of Platelet-Derived Growth Factor AA Impairs Endothelial Function.

OBJECTIVE: Bone morphogenic protein 4 (BMP4) is an important mediator of endothelial dysfunction in cardio-metabolic diseases, whereas platelet-derived growth factors (PDGFs) are major angiogenic and proinflammatory mediator, although the functional link between these 2 factors is unknown. The present study investigated whether PDGF mediates BMP4-induced endothelial dysfunction in diabetes mellitus. APPROACH AND RESULTS: We generated Ad-Bmp4 to overexpress Bmp4 and Ad-Pdgfa-shRNA to knockdown Pdgfa in mice through tail intravenous injection. SMAD4-shRNA lentivirus, SMAD1-shRNA, and SMAD5 shRN

Measurement of beta-thromboglobulin and platelet factor 4 to follow up patients with artificial heart valves.

Efficient antithrombotic therapies are necessary for the prevention of thrombotic complications in patients with artificial heart valves. Antiplatelet drugs are commonly used since platelet activation is a feature frequently found in these pathologic conditions. The measurement of beta TG and PF4 is useful for the diagnosis of platelet activation, which may increase the thrombotic risk. Two commercially available ELISA kits for beta TG and PF4 were tested and compared with the corresponding RIA methods used previously. Coefficients of correlation were 0.976 for beta TG and 0.985 for PF4. Both

Effect of low molecular weight aliphatic alcohols and related compounds on platelet factor 4 subunit association.

Titration of platelet factor 4 (PF4) with increasing concentrations of various low molecular weight aliphatic alcohols disrupts dimer and tetramer quaternary structure while preserving tertiary structural integrity. Normally observed slow subunit exchange (1H NMR time scale) is shifted into the fast chemical exchange regime. The order of effectiveness is butanol > 2-propanol > propanol > ethanol > methanol. Fluorination of ethanol (trifluoroethanol) and 2-propanol (hexafluoro-2-propanol) increases effectiveness by approximately 2 orders of magnitude. Oxidation to 2-propanone or trifluoro-2-pro

Platelet factors 4 produces an antidepressant effect in mice via inhibition of neuroinflammation.

INTRODUCTION: Platelet factor 4 (PF4) is secreted by platelets and can cross blood-brain barrier (BBB) to affect brain function, including regulations of neuroinflammation and synaptic plasticity, which are involved in the pathophysiology of depression. Nevertheless, whether PF4 participates in the development of depression has yet to be determined. OBJECTIVES: The aim of this study was to investigate the role and the underlying mechanisms of PF4 in the pathophysiology of depression. METHODS: Mouse models of depression were established using chronic social defeat stress (CSDS) and lipopolysa

Platelet factor 4 modulates endothelial cell antimicrobial activity to enhance bacterial clearance and improve sepsis outcomes.

Sepsis is a life-threatening condition characterized by dysregulated host responses to infection. Here, we identify platelet factor 4 (PF4) as a key mediator of vascular antimicrobial defense. In vitro, PF4 enhanced endothelial cell internalization of Escherichia coli via interactions with the PF4 receptor CXCR3 and the endothelial glycocalyx, directing bacteria to clathrin-mediated endocytosis and lysosomal degradation. In vivo, PF4 administration improved survival and reduced sepsis severity, bacterial burden, inflammation, and thrombosis in wild-type (WT) and PF4 knockout (PF4italic-/-) mic

Platelet Factor 4 and Longevity of Patients with Essential Thromobocythemia: An Example of Antagonistic Pathogenic Pleiotropy.

This article presents the concept of Antagonistic Pathogenic Pleiotropy, in which an abnormality that causes a specific pathology can simultaneously reduce other morbidities through unrelated mechanisms, resulting in the pathology causing less morbidity or mortality than expected. The concept is illustrated by the case of essential thrombocythemia (ET). Patients with ET have substantially elevated platelets and are therefore expected to have increased thrombotic events leading to reduced life expectancy. However, patients with ET do not have reduced life expectancy. A possible explanation is t

Platelet factor 4 limits neutrophil extracellular trap- and cell-free DNA-induced thrombogenicity and endothelial injury.

Plasma cell-free DNA (cfDNA), a marker of disease severity in sepsis, is a recognized driver of thromboinflammation and a potential therapeutic target. In sepsis, plasma cfDNA is mostly derived from neutrophil extracellular trap (NET) degradation. Proposed NET-directed therapeutic strategies include preventing NET formation or accelerating NET degradation. However, NET digestion liberates pathogens and releases cfDNA that promote thrombosis and endothelial cell injury. We propose an alternative strategy of cfDNA and NET stabilization with chemokine platelet factor 4 (PF4, CXCL4). We previously

Frequency and Clinical Impact of Platelet Factor 4-Specific Antibodies in Patients Undergoing Extracorporeal Membrane Oxygenation.

INTRODUCTION/OBJECTIVES: Extracorporeal membrane oxygenation (ECMO) provides circulatory support in patients with severe heart failure, but the frequent use of unfractionated heparin exposes patients to high risk of heparin-induced thrombocytopenia (HIT). We prospectively evaluated the development and clinical impact of platelet factor 4 (PF4)-specific antibodies (Abs) during ECMO and whether specific biological characteristics could predict HIT. MATERIALS AND METHODS: From 2014 to 2018, we studied 57 adults who underwent an ECMO for at least 5 days. The plasma samples collected daily were te

Studies of thromboxane B2, platelet factor 4, and fibrinopeptide A in bleeding-time blood of patients deficient in von Willebrand factor, platelet glycoproteins Ib and IIb-IIIa, and storage granules.

The blood volumes and concentrations of thromboxane B2 (TxB2), platelet factor 4 (PF4), and fibrinopeptide A (FPA) were measured every 30 seconds in bleeding-time blood in normal subjects and in patients with idiopathic thrombocytopenic purpura (ITP), delta and alpha delta storage pool deficiency (SPD), Bernard-Soulier Syndrome (BSS), thrombasthenia (TSA), and von Willebrand's disease (vWD). Data were fitted to second-order (TxB2, PF4, and FPA) or third-order (volumes) polynomials. Average values for various parameters over fixed-time intervals were determined by numerical methods. Persistence of anti-platelet factor 4 antibodies in vaccine-induced immune thrombocytopenia and thrombosis for 3 years.

BACKGROUND: Vaccine-induced immune thrombocytopenia and thrombosis (VITT) is a rare and potentially life-threatening complication of adenoviral vector-based vaccines against SARS-CoV-2. In VITT, antibodies against platelet factor 4 (PF4; ie, CXCL4) cause platelet activation, which then leads to thrombocytopenia and thrombosis. VITT resembles the immune-mediated drug reaction of heparin-induced thrombocytopenia; however, unlike heparin-induced thrombocytopenia antibodies, which have been shown to be transient, VITT antibodies appear to persist for much longer. OBJECTIVE(S): To identify the cli

[Heparin-induced thrombocytopenia and vaccine-induced immune thrombotic thrombocytopenia: novel insights on anti-platelet factor 4 antibodies].

The levels of anti-platelet factor 4 (PF4) antibodies, also known as anti-PF4 or heparin complex antibodies, are used to diagnose heparin-induced thrombocytopenia (HIT). In HIT, anti-PF4 antibodies induced by heparin exposure cause thrombocytopenia and thrombosis. However, anti-PF4 antibodies were recently reported to be associated with the development of fatal vaccine-induced immune thrombotic thrombocytopenia (VITT) after adenoviral vector vaccination for coronavirus disease 2019. HIT and VITT are caused by anti-PF4 antibodies and have similar pathological conditions. However, the severity o

Unique features of vaccine-induced immune thrombotic thrombocytopenia; a new anti-platelet factor 4 antibody-mediated disorder.

Vaccine-induced immune thrombotic thrombocytopenia (VITT) is a highly prothrombotic disorder caused by anti-PF4 antibodies that activate platelets and neutrophils, leading to thrombosis. Heparin-induced thrombocytopenia (HIT) is a related anti-PF4 mediated disorder, with similar pathophysiology and clinical manifestations but different triggers (i.e., heparin vs adenoviral vector vaccine). Clinically, both HIT and VITT typically present with thrombocytopenia and thrombosis, although the risk of thrombosis is significantly higher in VITT, and the thromboses occur in unusual anatomical sites (e.

Characterization of bonds formed between platelet factor 4 and negatively charged drugs using single molecule force spectroscopy.

Immunogenicity (i.e., the ability to initiate immune reactions) is one of the major challenges for the development of new drugs, as it may turn the developed drug therapeutically ineffective or cause severe immune-related effects. Using single molecule force spectroscopy, we study rupture forces between the positively charged, endogenous protein platelet factor 4 (PF4; also known as CXC chemokine ligand 4, CXCL4) and the antithrombotic drug heparin and other negatively charged glycosaminoglycans (GAGs), which are known to form immunogenic PF4/GAG-complexes (e.g., heparin and dextran sulfate) a

Fluorinated benzyloxyphenyl piperidine-4-carboxamides with dual function against thrombosis: inhibitors of factor Xa and platelet aggregation.

A series of benzyloxy anilides of nipecotic (5, 6) and isonipecotic (7, 8) acids were synthesized and assayed in vitro as inhibitors of ADP-induced platelet aggregation and the blood coagulation enzymes factor Xa (FXa) and thrombin (FIIa). An exploration of effects of the amidine group attached at the piperidine nitrogen, position and substitution (F, phenyl) of the benzyloxy group, and addition of fluorine/s on the second (distal) phenyl ring, led us to single out some promising isonipecotamide derivatives 7. Addition of meta-F and para-CF(3) on the distal phenyl ring resulted in a 6-to-18-fo

Platelet-activating factor activates HIV promoter in transfected SH-SY5Y neuroblastoma cells and MOLT-4 T lymphocytes.

Transfected gene constructs comprising the long terminal repeat (LTR) sequence of the human immunodeficiency virus (HIV) genome spliced to an assayable reporter gene have made possible the evaluation of a lipid mediator, platelet-activating factor (PAF), as a potential HIV transcriptional regulatory molecule. We assessed the activation of the HIV LTR promoter sequence linked to the chloramphenicol acetyltransferase (CAT) reporter gene (HIV-CAT) by PAF in both a human neural (SH-SY5Y neuroblastoma) and a human leukocytic (MOLT-4 T-lymphocyte) cell line. Platelet factor 4 prevents neuroinflammation and neurodegeneration in Parkinson's disease model via regulating protocadherin gamma/Pyk2 pathway.

Recent studies have reported that platelet factor 4 (PF4), also known as platelet-derived exokine CXCL4, is a critical factor in rejuvenating brain ageing and cognitive impairment by attenuating neuroinflammation. Our transcriptomic analysis indicated that 3 protocadherin gamma (Pcdhg) members (Pcdhga10, Pcdhgb4, and Pcdhgb5) were prominently upregulated by PF4 overexpression. This study aims to investigate whether PF4 plays a protective role in the pathogenesis of Parkinson's disease models and explore its underlying molecular mechanism by regulating Pcdhg and its downstream Pyk2. Pentosan Polysulfate and Heparin Exhibit Comparable Interactions with Platelet Factor 4, Suggesting a Potential Risk of Thrombocytopenia.

Pentosan polysulfate (PPS) is an approved drug for the treatment of interstitial cystitis in humans and osteoarthritis in animals. This semisynthetic highly sulfated polysaccharide shares structural similarities with heparin and also interacts with platelet factor 4 (PF4), the key protein implicated in thrombocytopenia, a serious side effect of heparin administration. Thrombocytopenia arises from an immune response to structural features of multimeric complexes of heparin and PF4, although the prediction of disease progression in patients is complicated by the variable polyclonal and polyspeci

Antigenic assessment for the β2-glycoprotein I/Platelet factor 4 complex in thrombotic patients with antiphospholipid syndrome.

OBJECTIVE: Antiphospholipid syndrome (APS) is an autoimmune condition characterized by recurrent thrombosis, pregnancy-related complications and circulating antiphospholipid antibodies, including anti-β2-glycoprotein I (β2-GPI). Platelet factor 4 (PF4) is a pro-coagulant protein expressed by activated platelets. It is considered a potential platelet ligand for oxidized β2-GPI, and this interaction may play a role in the thrombotic manifestations of APS. This study aims to assess β2-GPI/PF4 complex autoantibodies in sera of thrombotic patients with APS and their potential functional role in pla

Anti-platelet factor 4 antibodies causing VITT do not cross-react with SARS-CoV-2 spike protein.

Vaccine-induced immune thrombotic thrombocytopenia (VITT) is a severe adverse effect of ChAdOx1 nCoV-19 COVID-19 vaccine (Vaxzevria) and Janssen Ad26.COV2.S COVID-19 vaccine, and it is associated with unusual thrombosis. VITT is caused by anti-platelet factor 4 (PF4) antibodies activating platelets through their FcγRIIa receptors. Antibodies that activate platelets through FcγRIIa receptors have also been identified in patients with COVID-19. These findings raise concern that vaccination-induced antibodies against anti-SARS-CoV-2 spike protein cause thrombosis by cross-reacting with PF4. Immun
