Effect of a nuclease-resistant derivative of polyriboinosinic-polyribocytidylic acid complex on yellow fever in rhesus monkeys (Macaca mulatta).

Rhesus monkeys (Macaca mulatta) treated with a newly developed nuclease-resistant complex of polyriboinosinic-polyribocytidylic acid, poly-L-lysine, and carboxymethylcellulose [poly (ICLC)] did not die after challenge with virulent Asibi strain yellow fever (YF) virus. The strain of virus is sensitive to the effects of interferon in vitro and is lethal for rhesus monkeys four to six days after subcutaneous administration of 1,000 plaque-forming units of the virus. The mortality rate was reduced in monkeys initially treated 8 hr before or after inoculation of virus but was unchanged in monkeys

TLR3 activation enhances antitumor effects of sorafenib in hepatocellular carcinoma by activating NK cell functions through ERK and NF-κB pathways.

Background Sorafenib is a standard therapeutic agent for advanced hepatocellular carcinoma (HCC). However, its efficacy is moderate, as the survival of patients is prolonged for only a few months, and the response rate is low. The mechanism of low efficacy remains unclear. In this study, we investigated the effect of Toll-like receptor 3 (TLR3) on the effects of sorafenib on HCC. Methods Polyinosinic-polycytidylic acid [poly(I: C)] was used as a double-stranded RNA analog and TLR3 agonist in subsequent experiments. After orthotopic implantation of HCC tumors in BALBc nu/nu or C57BL/6 mice, sur

Protective role of Toll-like Receptor 3-induced type I interferon in murine coronavirus infection of macrophages.

Toll-like Receptors (TLRs) sense viral infections and induce production of type I interferons (IFNs), other cytokines, and chemokines. Viral recognition by TLRs and other pattern recognition receptors (PRRs) has been proven to be cell-type specific. Triggering of TLRs with selected ligands can be beneficial against some viral infections. Macrophages are antigen-presenting cells that express TLRs and have a key role in the innate and adaptive immunity against viruses. Coronaviruses (CoVs) are single-stranded, positive-sense RNA viruses that cause acute and chronic infections and can productivel

Induction of natural killer cell activity by the antitumour compound flavone acetic acid (NSC 347 512).

Flavone acetic acid (FAA), an antitumour drug at present undergoing clinical trial for cancer treatment, has been found to activate natural killer cell activity in spleen cells of mice following in vivo treatment at doses of 45-330 mg/kg. The activity of FAA was measured using a 51Cr-release assay, employing the YAC-1 lymphoma line as a target cell population, and was found to be comparable to that of the interferon inducer polyinosinic acid-polycytidylic acid. The induction of activity was blocked by anti-asialo GM1 antibody. The time course of induction of activity by FAA was similar to that

Solvent-Free Processing of i-P3HB Blends: Enhancing Processability and Mechanical Properties for Sustainable Applications.

Poly(3-hydroxybutyrate) is a biobased and biodegradable polymer, produced via bacterial fermentation and characterized by an isotactic structure and mechanical properties similar to those of polyethylene and polypropylene. However, its brittleness-due to high crystallinity (~70%) and thermal degradation, starting at a temperature range of 180-190 °C near its melting point (175 °C)-makes its processing difficult and limits its applications. Most recent studies on modifying P3HB involved solution casting, typically using chloroform, which raises sustainability concerns. In this study blends of i

Crosstalk between Toll-like receptor 3 and Notch signaling contributes to CD14+ monocytes activity in enterovirus 71 infected hand, foot, and mouth disease.

Interaction between Toll-like receptor (TLR) and Notch signaling contributes to inflammatory response in nephropathy and fungicidal infection, however, the role of this crosstalk remains not fully elucidated in enterovirus 71 (EV71)-induced hand, foot, and mouth disease (HFMD). The aim of this study was to investigate the crosstalk between TLR and Notch in inflammatory regulation in EV71 infection. Thirty-seven EV-71-indcued HFMD (16 mild and 21 severe cases) and eleven normal control (NC) were enrolled. CD14+ monocytes were purified, and were stimulated with either TLR3/4 agonists [poly(I: C)

Functional characterization of specialized immune cells in a cnidarian reveals an ancestral antiviral program.

Examining early-branching animal phyla can help reconstruct the evolutionary origins of immune cells. Here, we characterize immune-related cell programs in embryos of the sea anemone Nematostella vectensis, a model of Cnidaria, which diverged ~600 million years ago from other animals. Using a transgenic Nematostella reporter line expressing mCherry under the RLRb antiviral promoter, we identify a morphologically and transcriptomically distinct cell population activated by the viral mimic poly(I:C). These cells upregulate immune effector and regulator genes and show increased phagocytic activit

Generation of the Early-Gestational Maternal Immune Activation Mouse Model to Assess Prenatal Inflammation on Neurodevelopment.

Neurodevelopmental disorders (NDDs) arise from complex interactions between genetic and environmental risk factors, including prenatal immune activation. Maternal immune activation (MIA) in mice is a widely used model to study environmental contributions to NDDs; variability in experimental outcomes limits reproducibility. Here, we present a standardized protocol for inducing MIA during early gestation at embryonic day 9.5 (E9.5) in mice, incorporating multiple quality control metrics to assess immune response and predict pregnancy outcomes. Following administration of polyinosinic:polycytidyl

SIFD-associated TRNT1 deficiency unveils importance of TSPO during macrophage antibacterial and antiviral responses.

INTRODUCTION: Mitochondria support cellular biosynthetic and bioenergetic demands and mediate cell signaling. Their dysfunction is implicated in a wide range of diseases, including congenital disorders. One such disorder, sideroblastic anemia with B-cell immunodeficiency, periodic fevers, and developmental delay (SIFD), is caused by mutations in the tRNA-nucleotidyltransferase enzyme TRNT1. While SIFD is known to affect immune function, the role of macrophages-key mediators between innate and adaptive immunity-remains underexplored. METHODS: To investigate the impact of TRNT1 deficiency on ma

GSTP1 is a negative regulator of MAVS in the antiviral signaling against SVCV infection.

Glutathione S-transferase P1 (GSTP1), the most ubiquitous member of the GST superfamily, plays vital roles in the detoxification, antioxidant defense, and modulation of inflammatory responses. However, limited studies have been conducted on the function of GSTP1 in antiviral innate immunity. In this study, we have cloned the homolog of GSTP1 in triploid hybrid crucian carp (3nGSTP1) and investigated its regulatory role in the interferon signaling pathway. The open reading frame of 3nGSTP1 is composed of 627 nucleotides, encoding 209 amino acids. In response to spring viremia of carp virus (SVC

Trial watch: TLR3 agonists in cancer therapy.

UNLABELLED: Toll-like receptor 3 (TLR3) is a pattern recognition receptor that senses exogenous (viral) as well as endogenous (mammalian) double-stranded RNA in endosomes. On activation, TLR3 initiates a signal transduction pathway that culminates with the secretion of pro-inflammatory cytokines including type I interferon (IFN). The latter is essential not only for innate immune responses to infection but also for the initiation of antigen-specific immunity against viruses and malignant cells. These aspects of TLR3 biology have supported the development of various agonists for use as stand-al

Poly ICLC in the treatment of postinfectious demyelinating encephalomyelitis.

Three weeks after a mild and presumably infectious illness, a 21-year-old man developed a CNS disorder characterized by involvement of the cerebellum, cerebrum, and brainstem. It progressed, sometimes stepwise, without remission, over five months to being bedfast with total spastic paraplegia, severe ataxia, and unintelligible dysarthric speech. The CSF showed increased levels of protein, IgG, and myelin basic protein, as well as five oligoclonal bands. Because of failure of the patient's condition to respond to prolonged prednisone therapy, poly ICLC was given intravenously weekly for 20 week

Immunochemistry, physical chemistry and biology of 2',5'-oligoadenylates.

Monoclonal antibodies directed against 2-5A were developed and characterized. 2',5'-Oligoadenylate-protein complexes possess at least three distinct antigenic surfaces defined primarily by the ribose-phosphate backbone. A schematic model for the three epitopes is presented. Antibodies directed against 2-5A, in conjunction with other techniques, were employed to quantify 2-5A in various tissues of pathogen-free mice. Levels of 2-5A were in the range of 400-800 fmole/gm. Mice injected with poly(I).poly(c) or encephalomyocarditis virus (EMCV) showed elevated levels of 2-5A. Administration of poly

Mismatched double-stranded RNA, Ampligen (poly(I): poly(C12U), demonstrates antiviral and immunostimulatory activities in HIV disease.

Mismatched double-stranded RNA (Ampligen) has broad spectrum antiviral and immunomodulatory activities. These activities generate stabilization or improvement in three important surrogate markers of HIV disease progression. Patients with HIV disease treated with Ampligen do not become positive for p24 antigen, in contrast to patients treated with AZT or placebo. Viral burden can also be decreased in patients receiving Ampligen/AZT therapy. In vitro studies indicate that both AZT sensitive and AZT resistant viruses can be inhibited by Ampligen alone and are synergistically inhibited by Ampligen

The relationship between serum interferon and an inhibitor of mouse haemopoietic colonies in vitro.

When mice are inoculated with Poly I—Poly C or influenza virus an inhibitor of haemopoietic colony forming cells appears in the serum. The colony inhibiting activity of whole serum is shown to be directly proportional to the interferon titre. Interferon and colony inhibitor are also shown to have the same properties with regard to molecular size and sensitivity to heat, pH and trypsin.

Role of interferons in the control of Lassa virus replication in human dendritic cells and macrophages.

Lassa fever is a hemorrhagic fever caused by Lassa virus (LV), which primarily targets human dendritic cells (DC) and macrophages (MP). Massive numbers of viral particles are released with no effect on the viability, activation or maturation of these cells. LV does not inhibit the activation of cells induced by sCD40L or LPS. We report here the consequences of exogenous activation of LV-infected human DC and MP for viral replication. The activation of cells with lipopolysaccharide or exogenous poly(I-C) and the transfection of cells with poly(I-C) strongly inhibited LV replication, at least pa

Integrated bioinformatics and experimental analysis of mitochondrial-associated membrane function and mechanism in acute respiratory distress syndrome.

Acute respiratory distress syndrome (ARDS) is a life-threatening lung condition characterized by severe inflammation, immune dysregulation, and oxidative stress, leading to high mortality (30-40%). This study explores the involvement of MAM-related genes in ARDS pathogenesis through bioinformatics and experimental validation. Publicly available RNA-sequencing data from ARDS and control samples were analyzed to identify differentially expressed genes (DEGs). Functional enrichment, gene set variation analysis (GSVA), and weighted gene co-expression network analysis (WGCNA) were performed to expl

Synthesis and Post-Polymerization Modification of Defined Functional Poly(vinyl ether)s.

Living cationic polymerization is known for a good control over chain growth yielding polymers with well-defined molar mass distributions and low dispersities. However, the practical challenges involved in the synthesis of poly(vinyl ether)s limited suitable post-polymerization modifications (PPM) via chemoselective click reactions. Herein the successful controlled cationic polymerization of vinyl ethers bearing pendant CC double and C≡C triple bonds using a single-component initiation under ambient conditions is reported. Furthermore, the PPM via thiol-ene/-yne and copper(I)-catalyzed alkyne
