Four New Cadinane-Type Sesquiterpenoids From the Roots of Fissistigma tientangense.

Four new cadinane-type sesquiterpenoids, named fisscadinanes A-D (1-4), together with four known biogenetically related cadinane-type sesquiterpenoids (5-8) were isolated from the roots of Fissistigma tientangense. The structures of these sesquiterpenoids were determined by comprehensive spectroscopic analyses. Compound 1 is a 15-norcadinane sesquiterpene containing one p-π-π-system. Compounds 2-4 are cadinane sesquiterpenes. The absolute configurations of compounds 1-4 were determined via ECD calculations. The inhibitory activities of fisscadinanes A-D against the nitric oxide (NO) production

Functional characterization of PcMYB25, a candidate regulator of Patchouli alcohol biosynthesis in Pogostemon cablin, via exogenous hormone-elicited transcriptome analysis.

BACKGROUND: Plant hormones often cause changes in secondary metabolites. Patchouli alcohol (PA) is a bioactive compound of the medicinal plant Pogostemon cablin. Similar to other secondary metabolites, its synthesis is also regulated by transcription factors. While the PA biosynthetic pathway has been characterized, its regulatory mechanisms remain incompletely understood. RESULTS: Transcriptome analysis revealed that exogenous hormone application significantly altered the transcriptional landscape of Pogostemon cablin, particularly affecting genes involved in terpenoid metabolism. Through we

Indirect Defense Responses of Arabidopsis Thaliana To Insect Feeding Vibrations Utilizing a Miniature GC-FID.

The release of volatile organic compounds (VOCs) plays a crucial role in plant-to-plant and plant-to-arthropod communication. Each blend of VOCs released carries a specific meaning for the receiving party and the stimulus that triggers a response. One such stimulus, vibrations from insect feeding, has been shown to elicit chemical defense responses in rosettes of Arabidopsis thaliana. In this study, we measured herbivore-induced plant volatiles (HIPVs) in response to 4 h of insect feeding vibrations using solid-phase microextraction (SPME) fibers in small collection chambers and a miniature Ga

Root volatiles in plant-plant interactions II: Root volatiles alter root chemistry and plant-herbivore interactions of neighbouring plants.

Volatile organic compounds (VOCs) emitted by plant roots can influence the germination and growth of neighbouring plants. However, little is known about the effects of root VOCs on plant-herbivore interactions of neighbouring plants. The spotted knapweed (Centaurea stoebe) constitutively releases high amounts of sesquiterpenes into the rhizosphere. Here, we examine the impact of C. stoebe root VOCs on the primary and secondary metabolites of sympatric Taraxacum officinale plants and the resulting plant-mediated effects on a generalist root herbivore, the white grub Melolontha melolontha. We sh

Anticoccidial activities of Piper betle L essential oil on Eimeria tenella oocysts.

Coccidiosis poses a significant threat to the poultry industry, with synthetic antibiotics and disinfectants being the primary tools for control. This study investigated the potential of Piper betle L essential oil (PBEO) as a natural alternative against Eimeria tenella, one of the most pathogenic Eimeria species affecting poultry. Our findings revealed that PBEO exhibits significant anticoccidial effects through two primary mechanisms: (i) oocysticidal activity by disintegrating oocyst walls and (ii) inhibition of the sporulation process. PBEO demonstrated oocysticidal activities ranging from

The volatile sesquiterpene β-caryophyllene generated by the Phoebe bournei terpene synthase PbTPSa21 regulates seed germination by mediating ROS signaling.

Phoebe bournei is an evergreen tree with high economic and social value because of its fine timber and pleasant aroma, which is predominantly produced by terpenoids, including β-caryophyllene. However, the natural reproduction and artificial cultivation of P. bournei are severely hindered by long seed dormancy times and poor seed fragility. Terpenoids are secondary metabolites with vast commercial application potential in medicine, agriculture, and gardening, but their role in seed germination remains elusive. Here, we identified a novel P. bournei β-caryophyllene synthase gene, PbTPSa21, whos

The Potential Therapeutic Role of Beta-Caryophyllene as a Chemosensitizer and an Inhibitor of Angiogenesis in Cancer.

The natural, highly lipophilic bicyclic sesquiterpenes, Beta-Caryophyllene (BCP), was highlighted in several recent preclinical studies to enhance chemo-sensitization in chemo-resistant tumors and to efficiently inhibit angiogenesis and cancer cells' ability to invade and metastasize. Previous studies have researched the reasons for the synergistic effect of Beta-Caryophyllene in combination therapy and its role as a chemosensitizer and an inhibitor of angiogenesis through investigating the involved mechanisms and signaling molecules. These include the lipophilic nature of BCP, the selective i

Recruitment of entomopathogenic nematodes by insect-damaged maize roots.

Plants under attack by arthropod herbivores often emit volatile compounds from their leaves that attract natural enemies of the herbivores. Here we report the first identification of an insect-induced belowground plant signal, (E)-beta-caryophyllene, which strongly attracts an entomopathogenic nematode. Maize roots release this sesquiterpene in response to feeding by larvae of the beetle Diabrotica virgifera virgifera, a maize pest that is currently invading Europe. Most North American maize lines do not release (E)-beta-caryophyllene, whereas European lines and the wild maize ancestor, teosin

ZmTPS18-mediated β-caryophyllene biosynthesis contributes to resistance against southern corn rust in maize.

Southern corn rust (SCR), caused by the obligate biotrophic fungus Puccinia polysora Underwood, is a major foliar disease of maize associated with substantial yield losses in many production regions. Terpenoids are important components of plant defense, but the terpene synthases and upstream regulators involved in pathogen-induced terpenoid biosynthesis in maize are still poorly defined. In this study, volatile profiling showed that β-caryophyllene emission was induced in the SCR-resistant maize inbred line Yanzi F7016K after pathogen inoculation, reaching levels 3.18-fold higher than those in

Integrating Structural Bioinformatics and Functional Mechanisms of Sesquiterpene Synthases CARS and CADS in Lavandula angustifolia (Lavender).

Lavender species are economically valuable plants, widely cultivated for their essential oils (EOs), which include sesquiterpenes. The sesquiterpenes caryophyllene and cadinol are major constituents, contributing woody and balsamic notes. However, the specific enzymes catalyzing their formation in lavender have not been elucidated. This study reports the comprehensive functional and structural characterization of two pivotal sesquiterpene synthases from Lavandula angustifolia (lavender): caryophyllene synthase (CARS) and cadinol synthase (CADS). Mutation experiments were performed based on mol
