BACKGROUND: Conventional pesticide delivery systems often suffer from low deposition and insufficient targeted release in complex pathogenic microenvironments. Inspired by the pathogen infection process, this study aimed to develop a novel controlled-release platform capable of responding to specific infection microenvironments to achieve efficient and targeted pesticide delivery.
RESULTS: A dual-responsive nano-delivery platform (BZP) was successfully constructed by loading the fungicide boscalid (Bos) onto a zeolitic imidazolate framework-8 (ZIF-8) nanocarrier and utilizing pectin as a path

The objective of the current study was to investigate and compare the impact of enzymatic hydrolysis and pre-fermentation with probiotics on the biological activity and metabolic profile of melon juice. We found that fermentation with Lactiplantibacillus plantarum N13, not only decreased the juice pH, total soluble solids and reducing sugar, but also promoted the release of phenolic compounds, increased antioxidant activity and improved juice's aroma profile. Although pectinase and cellulase aided in the release of reducing sugar and total phenols in the juice, the heating process involved in

This study utilized 'Qinmi No.9' passion fruit to explore pre-harvest methyl jasmonate (MeJA) treatment effects on fruit quality and regulatory mechanisms via physiological assays, transcriptomics, and metabolomics. MeJA delayed peel yellowing, elevated total soluble solids, total flavonoid, soluble protein contents, and polygalacturonase, polyphenol oxidase, and lipoxygenase activities, but decreased titratable acidity, pectin content, and electrical conductivity, with MeJA_10 (1.0 mM MeJA) presenting optimal comprehensive performance. Transcriptomic analysis revealed that MeJA_5 (0.5 mM MeJA

We have used an hrp-positive strain of the soft rot pathogen Erwinia carotovora subsp. carotovora to elucidate plant responses to this bacterial necrotroph. Purified virulence determinants, harpin (HrpN) and polygalacturonase (PehA), were used as tools to facilitate this analysis. We show that HrpN elicits lesion formation in Arabidopsis and tobacco and triggers systemic resistance in Arabidopsis. Establishment of resistance is accompanied by the expression of salicylic acid (SA)-dependent, but also jasmonate/ethylene (JA/ET)-dependent, marker genes PR1 and PDF1.2, respectively, suggesting tha

When nematodes invade and subsequently migrate within plant roots, they generate cell wall fragments (in the form of oligogalacturonides; OGs) that can act as damage-associated molecular patterns and activate host defence responses. However, the molecular mechanisms mediating damage responses in plant-nematode interactions remain unexplored. Here, we characterized the role of a group of cell wall receptor proteins in Arabidopsis, designated as polygalacturonase-inhibiting proteins (PGIPs), during infection with the cyst nematode Heterodera schachtii and the root-knot nematode Meloidogyne incog

Suspension-cultured cells of Vitis vinifera cv Monastrell were used to investigate the effects of methyljasmonate, ethylene and salicylic acid separately or in combination with cyclodextrins on both trans-resveratrol production and the induction of defense responses. The results showed that the addition of methyljasmonate or ethylene to suspension-cultured cells jointly treated with cyclodextrins and salicylic acid provoked a decrease of trans-resveratrol levels suggesting that salicylic acid has a negative and antagonistic effect with methyljasmonate or ethylene on trans-resveratrol productio

To increase its operational stability and ongoing reusability, B. subtilis pectinase was immobilized on iron oxide nanocarrier. Through co-precipitation, magnetic iron oxide nanoparticles were synthesized. Scanning electron microscopy (SEM) and energy dispersive electron microscopy (EDEX) were used to analyze the nanoparticles. Pectinase was immobilized using glutaraldehyde as a crosslinking agent on iron oxide nanocarrier. In comparison to free pectinase, immobilized pectinase demonstrated higher enzymatic activity at a variety of temperatures and pH levels. Immobilization also boosted pectin

Chinese hickory (Carya cathayensis Sarg.) is an economically important tree species widely cultivated in southeastern China, where trunk canker disease caused by Botryosphaeria dothidea poses a serious threat to tree health and production. Pectin-degrading enzymes are important virulence-associated factors that facilitate fungal colonization and host tissue maceration, but their evolutionary diversification and functional roles in B. dothidea during woody host infection remain poorly understood. Comparative genomic analysis revealed lineage-specific variation in the GH28 glycoside hydrolase fa

Oligogalacturonides (OGs) released from plant cell walls by pathogen polygalacturonases induce a variety of host defense responses. Here we show that in Arabidopsis (Arabidopsis thaliana), OGs increase resistance to the necrotrophic fungal pathogen Botrytis cinerea independently of jasmonate (JA)-, salicylic acid (SA)-, and ethylene (ET)-mediated signaling. Microarray analysis showed that about 50% of the genes regulated by OGs, including genes encoding enzymes involved in secondary metabolism, show a similar change of expression during B. cinerea infection. In particular, expression of PHYTOA

Dietary polysaccharides have attracted considerable attention because of their species-specific chemical composition and diverse biological activities. Polysaccharides derived from Houttuynia cordata (HC) exhibit broad pharmacological activities, emphasizing their diverse roles in biological processes. This study investigated the effects of HC-derived polysaccharides on Peyer's patch (PP)-mediated intestinal immune response. The crude polysaccharide (HCP) isolated from HC extract (HCE) was identified as a pectin-type polysaccharide rich in the homogalacturonan (HG) domain, which was further hy

Phaseolus vulgaris is subjected to serious post-harvest diseases such as grey mold and cottony rot diseases caused by Botrytis cinerea and Pythium aphanidermatum, respectively. In current study, potassium silicate (KSi), potassium thiosulfate (KTS) and potassium sulfate (KS) suppressed moderately the growth of B. cinerea and P. aphanidermatum in vitro. The applied treatments significantly suppressed grey mold and cottony rot of Xera and Valentino snap beans varieties' pods stored at 7 ± 1°C and 90-95% RH for 20 days. Take-all (caused by the fungal pathogen Gaeumannomyces graminis var. tritici, Ggt) and common root rot (caused by Bipolaris sorokiniana) are devastating root diseases of wheat (Triticum aestivum L.). Development of resistant wheat cultivars has been a challenge since no resistant wheat accession is available. GmPGIP3, one member of polygalacturonase-inhibiting protein (PGIP) family in soybean (Glycine max), exhibited inhibition activity against fungal endopolygalacturonases (PGs) in vitro. In this study, the GmPGIP3 transgenic wheat plants were generated and used to assess the effectiveness of

Plant mitogen-activated protein kinases (MPKs) transduce signals required for the induction of immunity triggered by host recognition of pathogen-associated molecular patterns. We isolated a full-length cDNA of a group B MPK (PgMPK4) from pearl millet. Autophosphorylation assay of recombinant PgMPK4 produced in Escherichia coli confirmed it as a kinase. Differential accumulation of PgMPK4 mRNA and kinase activity was observed between pearl millet cultivars 852B and IP18292 in response to inoculation with the downy mildew oomycete pathogen Sclerospora graminicola. This increased accumulation of

Pomegranate peel, a major by-product of processing, is rich in water-soluble pectin made of D-galacturonic acid units with α (1-4) linkage. Efficiently converting pomegranate peel into safe bioactive principles minimizes waste in the fruit industry and opens new avenues in functional foods. In this work, pomegranate pectin was enzymatically depolymerized with extracellular pectinases from Streptomyces hydrogenans YAM1 to generate pectic oligosaccharides (POS). Products were characterized by TLC and LC-ESI-MS, which indicated unsaturated mono-, di-, and trigalacturonic acids among the main spec

Fruit softening is a critical physiological process that occurs during fruit ripening and senescence. Nitric oxide (NO) and NAM-ATAF1/2-CUC2 (NAC) transcription factors (TFs) have been implicated in regulating fruit softening. However, the functions of NO and NAC-like, activated by apetala3/pistillata (NAP) TFs and their interaction in fruit softening are not well understood. Here, we found that NO might suppress cell wall-related enzyme [polygalacturonase (PG), pectin methyl esterase (PME)] activity, thereby effectively postponing tomato fruit softening. Rhizopus delemar, an opportunistic fungal pathogen, causes a highly fatal disease, mucormycosis. Spore germination is a crucial mechanism for disease pathogenesis. Thus, exploring the molecular mechanisms of fungal germination would underpin our knowledge of such transformation and, in turn, help control mucormycosis. To gain insight into the developmental process particularly associated with cell wall modification and synthesis, weighted gene co-expression network analysis (WGCNA) was performed including both coding and non-coding transcripts identified in the current study, to find out the m

Mature plant cell walls lose their ability to expand and become unresponsive to expansin. This phenomenon is believed to be due to cross-linking of hemicellulose, pectin, or phenolic groups in the wall. By screening various hydrolytic enzymes, we found that pretreatment of nongrowing, heat-inactivated, basal cucumber (Cucumis sativus) hypocotyls with pectin lyase (Pel1) from Aspergillus japonicus could restore reconstituted exogenous expansin-induced extension in mature cell walls in vitro. Recombinant pectate lyase A (PelA) and polygalacturonase (PG) from Aspergillus spp. exhibited similar ca

Plant pollens are airborne allergens, as their inhalation causes immune activation, leading to rhinitis, conjunctivitis, sinusitis and oral allergy syndrome. A myriad of pollen proteins belonging to profilin, expansin, polygalacturonase, glucan endoglucosidase, pectin esterase, and lipid transfer protein class have been identified. In the present in silico study, the protein domains of fifteen pollen sequences were extracted from the UniProt database and submitted to the interactive web tool SMART (Simple Modular Architecture Research Tool), for finding the protein domain profiles. Activities of twelve hydrolytic enzymes in the digestive tract of young rabbits before weaning (4 weeks old) and adult rabbits (3 months old) were measured. The principal digestive enzymes in both groups of rabbits appeared to be amylase (EC 3.2.1.1), maltase (EC 3.2.1.20), pectinase (EC 3.2.1.15) and proteinases. The stomach of young rabbits contained most of the lipolytic activity and 45.7% of the total proteolytic activity of the digestive tract. The highest specific activities (per g digesta) of amylase, maltase and proteinase in young rabbits were found in the small intestine. Total activ
