A series of polymers of polyglutamic acid have been tested as plasma volume expanders. The results indicate that those polymers prepared from backbones of 40,000 molecular weight or over and side chains of 3,000 molecular weight or over, will have plasma volume retention half-life of 15 hours or longer. Measurements of the oncotic efficiency of these polymers in vivo indicated a blood stream fluid retention of approximately 51 ml./gm. of polymer. Similar measurements of polymer-serum albumin solutions in vitro showed a retention of 52 ml./gm. of polymer. The clinical response to infusion of so

Methylofuran (MYFR) is a formyl-carrying coenzyme essential for the oxidation of formaldehyde in most methylotrophic bacteria. In Methylorubrum extorquens, MYFR contains a large and branched polyglutamate side chain of up to 24 glutamates. These glutamates play an essential role in interfacing the coenzyme with the formyltransferase/hydrolase complex, an enzyme that generates formate. To date, MYFR has not been identified in other methylotrophs, and it is unknown whether its structural features are conserved. Here, we examined nine bacterial strains for the presence and structure of MYFR using

Volumetric absorptive microsamplers (VAMS) have been developed recently as a promising tool for clinical blood sampling. Compared to dried blood spot samples analyzed by accurate stable isotope dilution assays (SIDAs), the new technique could provide further substantial miniaturizing of folate assays by eliminating hematocrit effects and uneven analyte distribution within the sample. Herein, we present a miniaturized SIDA coupled with LC-MS/MS measurement of 5-methyltetrahydrofolic acid as main folate vitamer in whole blood (WB) using [13C5]-5-methyltetrahydrofolic acid as internal standard. The increasing demand for biopolymers has positioned poly-γ-glutamic acid (γ-PGA) as a promising alternative to fossil-based polymers due to its biodegradability and biocompatibility. γ-PGA biosynthesis in Bacillus subtilis is closely linked to intracellular glutamate availability, which is typically maintained via the supply of an exogenous glutamate source, a cost-intensive factor for industrial production. This study investigates the metabolic interplay between glycerol, citrate, and glutamate during γ-PGA synthesis, focusing on how cellular glutamate demand influences carbon source utiliza

Gout is a chronic and degenerative disease that affects the joints and soft tissues because of the crystalline deposit of monosodium urate. The interaction between monosodium urate crystals (MSU) and synoviocytes generates oxidative and inflammatory states. These physiological characteristics have promoted the study of poly-gallic acid (PGAL), a poly-oxidized form of gallic acid reported to be effective in in vitro models of inflammation. The effect of PGAL in an in vitro model of oxidation and synovial inflammation induced by MSU was evaluated after 24 h of stimulation through the morphologic

The causative agent of Huntington's disease, the poly-Q homo-repeat in the N-terminal region of huntingtin (httex1), is flanked by a 17-residue-long fragment (N17) and a proline-rich region (PRR), which promote and inhibit the aggregation propensity of the protein, respectively, by poorly understood mechanisms. Based on experimental data obtained from site-specifically labeled NMR samples, we derived an ensemble model of httex1 that identified both flanking regions as opposing poly-Q secondary structure promoters. While N17 triggers helicity through a promiscuous hydrogen bond network involvin

Bone sialoprotein (BSP) is a multi-functional extracellular matrix (ECM) protein associated with mineralized tissues, particularly bone and cementum. The amino acid sequence of BSP includes three evolutionarily conserved sequences which contribute to functions of the protein: an N-terminal collagen-binding domain, polyglutamic acid (polyE) sequences involved in hydroxyapatite nucleation and crystal growth, and a C-terminal arginine-glycine-aspartic acid (RGD) integrin-binding domain. BSP promotes attachment and differentiation of osteogenic and osteoclastic cells. Genetic ablation of BSP in mi

The accumulation of senescent cells can lead to tissue degeneration, chronic inflammatory disease and age-related tumorigenesis. Interventions such as senolytics are currently limited by off-target toxicity, which could be circumvented by instead enhancing immune-mediated senescent cell clearance; however, immune surveillance of senescent cells is often impeded by immunosuppressive factors in the inflammatory microenvironment. Here, we employ a chimeric peptide as a 'matchmaker' to bind to the urokinase-type plasminogen activator receptor, a cell surface marker of senescent cells. This peptide

A CCRF-CEM mutant, CEM-p, has been shown to exhibit resistance to methotrexate due to decreased methotrexate polyglutamate accumulation. To ascertain the mechanism(s) responsible for this phenotype, we analyzed FPGS and SLC19A1 mRNA expression, examined FPGS promoter activity, and determined nucleotide sequence of the FPGS promoter and full length cDNA from CCRF-CEM and CEM-p cells. We identified in FPGS from CEM-p cells three amino acid substitutions that altered the ATP binding P-loop, glutamate/folate binding, and a conserved domain located at the carboxyl-terminal. Our data demonstrated fo

Novel foods are food products that were not in circulation for consumption to any significant extent in the European Union before 1997. The introduction of novel foods harbors the risk of allergic reactions to new allergens or new cross-reactions that need to be taken into account. The aim of the study is to highlight the allergenic potential of selected novel foods. Allergens in edible insects in particular harbor the risk of anaphylactic reactions. In western industrial nations cross-sensitivity to crustaceans and shellfish are likely to play an important role in the future. Chia seeds, whic

Oppositely charged polyelectrolytes are known to form polyelectrolyte complexes. Polyelectrolyte complexes are typically structurally disordered. In recent years, complexes of oppositely charged polypeptides have received increasing attention. Polypeptides provide additional interaction motifs such as hydrogen bonds and salt bridges that could stabilize ordered assemblies. Under specific conditions, the formation of secondary structures has been reported. So far, it has been difficult to obtain long-range ordered assemblies of polypeptide complexes. Here we show that liquid-solid phase separat

Chitosan/calcium carbonate core-shell type composite microparticles were fabricated via a biomineralization-inspired process with chitosan particles as starting materials. The obtained composite particles were spherical, and the diameter of the composite particles was larger than that of the chitosan particles. XRD measurement indicated that the shell of the composite particle had a crystalline structure. The shell fabrication process was carried out with polyacrylic acid (PAA) or polyglutamic acid (PGlu) as binder molecules between calcium carbonate and chitosan. The crystal form of the shell

Microbes across diverse species and environments form biofilms, living materials composed of cells and extracellular polymers. Biofilm-dwelling cells benefit from emergent soft matter physics, which sculpts three-dimensional morphologies and facilitates osmotic nutrient uptake. Although biofilms are modeled as viscoelastic gels, the physical origins of the phase transition underlying their conversion from groups of cells to living gels have not been systematically investigated. Here, we show that Bacillus subtilis biofilms use polymer composition to tune their physical properties and drive gel

Radiotherapy (RT) is an essential treatment modality for several types of cancer. Despite its therapeutic potential, RT is frequently insufficient to overcome the immunosuppressive nature of the tumor microenvironment, failing to control tumor metastases. Innovative immunomodulatory strategies, like immunostimulatory biomaterials could be used to boost the immunogenic effects of RT. Herein, we addressed the synergistic potential of immunostimulatory chitosan/poly(γ-glutamic acid) nanoparticles (Ch/γ-PGA NPs) combined with RT to induce antitumor immunity in the 4T1 orthotopic breast tumor mouse

The rheological behaviors of a polyanionic collagen, fabricated using poly(γ-glutamic acid)-N-hydroxysuccinimide (γ-PGA-NHS) as a novel modifier, were investigated in this study. It was found that both of the native and modified collagen solutions were pseudoplastic fluids, as shown from the steady-shear tests. While the storage modulus and loss modulus of collagen increased with increasing the amount of γ-PGA-NHS, or with increasing the degree of esterification of γ-PGA-NHS; meanwhile, the dynamic denaturation temperature determined by dynamic temperature sweep was also increased, indicating

In order to reveal mechanisms for the electrospinning of proteins, this study focuses on the polymer chain conformation, which is considered to be a critical factor for successful electrospinning. Poly(gamma-benzyl-L-glutamate) (PBLG) is employed and the relationships between the chain conformations of the pre-spun PBLG molecules and the morphologies of the post-spun PBLG fibers are investigated. By combining viscosity measurements, and circular dichroism and FT-IR spectroscopies, chain conformations of the pre- and post-spun PBLG are characterized. The chain conformations of the pre-spun PBLG

Plant growth-promoting bacteria and biochar have been widely used as immobilizers to remediate heavy metal contaminated soil. However, few studies have unraveled the effect and synergistic mechanism of combined application of plant growth-promoting bacteria and biochar on in situ heavy metal contaminated soil remediation and plant yield and quality improvement under heavy metal pollution stress. In this study, the effects of biochar, γ-PGA-producing bacteria (Bacillus amyloliquefaciens strain W25) and their combined application on Cd and Pb immobilization, γ-PGA production in soil filtrate, th

The purpose of this study was to evaluate poly(L-glutamic acid)-benzyl-DTPA-Gd (PG-Gd), a new biodegradable macromolecular magnetic resonance imaging contrast agent, for its pharmacokinetics and MRI enhancement in nonhuman primates. Studies were performed in rhesus monkeys at intravenous doses of 0.01, 0.02 and 0.08 mmol Gd/kg. T(1)-weighted MR images were acquired at 1.5 T using fast spoiled gradient recalled echo and fast spin echo imaging protocols. The small-molecule contrast agent Magnevist was used as a control. PG-Gd in the monkey showed a bi-exponential disposition. The initial blood c

Drug overdose remains a major public health challenge, particularly for non-opioid substances that lack effective antidotes and are often treated through gastrointestinal decontamination with activated charcoal. While activated charcoal is broadly effective, its use is associated with aspiration risks and gastrointestinal complications. Here, we report a class of synthetic poly(glutamic acid-block-tyrosine) (EY) peptide adsorbent materials designed as biocompatible, tuneable alternatives for gastrointestinal drug adsorption. EY polypeptides were synthesized via N-carboxyanhydride ring-opening

Gambogic acid (GA) exhibits potent antitumor effects through multiple synergistic mechanisms, making it a promising candidate for adjuvant therapy in refractory cancers. However, its clinical translation has been hindered by poor aqueous solubility, low bioavailability, and systemic toxicity. In this study, we develop a dual-responsive biomimetic nano-prodrug (CM@pGGH) that addresses these limitations and facilitates effective, tumor-targeted therapy. CM@pGGH is synthesized by conjugating GA and hydroxychloroquine (HCQ)-an FDA-approved antimalarial and immunomodulator-to polyglutamic acid (PGA
