Characterization of in vitro digestion and fermentation of Polygonatum sibiricum ethanol extract and implications for antioxidant and lipid-lowering potential.

BACKGROUND: Polygonatum sibiricum is known to possess antioxidant and lipid-regulating properties, but due to the presence of various macromolecular active ingredients such as saponins, it is difficult to digest. The mechanism by which it is digested, absorbed in the gastrointestinal tract and exerts functional activity is still unclear. RESULTS: The results of in vitro digestion based on static INFOGEST showed that the content of saponin in P. sibiricum ethanol extract (PSE) gradually decreased during oral, gastric and intestinal digestion, with relatively stable degradation rate of saponin

Therapeutic potential of polygonatum polysaccharides in modulating gut microbiota and alleviating psoriasis-like inflammation in a mouse model.

Psoriasis is a chronic inflammatory skin disorder characterized by keratinocyte hyperproliferation and immune dysregulation. The gut microbiota has emerged as a significant factor influencing the pathogenesis of psoriasis, particularly through the gut-skin axis. This study investigates the therapeutic effects of Polygonatum polysaccharides (PSP) on imiquimod (IMQ)-induced psoriasis-like dermatitis in mice, focusing on gut microbiota modulation and systemic immune responses. Mice were treated with varying doses of PSP, and their effects were compared to methotrexate (MTX) as a positive control.

Polygonatum sibiricum polysaccharide inhibits osteoporosis by promoting osteoblast formation and blocking osteoclastogenesis through Wnt/β-catenin signalling pathway.

Bone homeostasis is maintained by a balance between bone formation by osteoblasts and bone resorption by osteoclasts. Osteoporosis occurs when osteoclast activity surpasses osteoblast activity. Our previous studies showed the plant-derived natural polysaccharide (Polygonatum sibiricum polysaccharide or PSP) had significant anti-ovariectomy (OVX)-induced osteoporosis effects in vivo, but the mechanisms of PSP's anti-osteoporosis effect remains unclear. In this study, we assessed PSP's effect on the generation of osteoblast and osteoclast in vitro. This study showed that PSP promoted the osteoge

Associations Between Gut Microbiota and Immune Profiles in Community-Dwelling Older Adults and the Regulatory Role of Polygonatum odoratum.

Aging is often associated with intestinal microbiota imbalance and immune system degeneration. The dysregulation and crosstalk of these two factors markedly elevate the elderly's susceptibility to infections and age-related diseases. Polygonatum odoratum, a traditional Chinese medicinal herb, is widely used in geriatric care for its lung-moistening and nourishing effects. However, its potential regulatory roles on the microbiota and immunity in the aging context remain unreported. A total of 140 independently living elderly individuals aged 60 and above were recruited for this study. The rando

A haplotype-resolved genome reveals intron expansion and enzyme innovation associated with the starch-to-fructan shift in Polygonatum cyrtonema.

Huangjing (Polygonati Rhizoma) is a traditional food-medicine substance in China, which can serve as a grain substitute historically. However, its material basis is not starch but agavin-type fructans with dual regulatory effects on energy and biological activity, which also constituted the staple diet component of prehistoric Mesoamerican humans. Yet, the formation mechanism of these compounds remains unclear. Here we presented a haplotype-resolved assembly of the Polygonatum cyrtonema giant genome, spanning two phased haplotypes of ∼7.18 Gb and 7.05 Gb respectively. Genomic analyses revealed

Exploring the Mechanism of Selenium-Biofortified Polygonatum Kingianum in Alzheimer's Disease: An Integrated Metabolomics and Network Pharmacology In Silico Study.

BACKGROUND: Alzheimer's disease [AD] involves multifactorial pathogenesis such as Aβ deposition and Tau hyperphosphorylation, yet effective multi-target therapies remain scarce. The mechanisms by which selenium-biofortified Polygonatum kingianum [Se-PK] modulates AD pathways are poorly understood, limiting its clinical translation. METHODS: An integrated in silico approach was employed: 1] UPLC-MS/MS metabolomics to identify differential metabolites in Se-PK [VIP > 1, fold change ≥2 or ≤0.5]; 2] network pharmacology to construct compound-target-pathway networks; and 3] molecular docking and d

Synthesis of RARα-PROTAC and its delivery via Polygonatum sibiricum exosome-like nanoparticles for the treatment of colorectal cancer.

Retinoic acid receptor α (RARα) is a promising yet challenging target in treating colorectal cancer (CRC). Traditional RARα-binding modulators often fail to fully inhibit downstream oncogenic pathways and cannot eliminate the pre-existing RARα protein, frequently resulting in drug resistance and treatment failure. To overcome these limitations, we designed a series of proteolysis-targeting chimeras (PROTACs) based on the ligand CA77.1, which are bifunctional molecules that recruit the ubiquitin-proteasome system for RARα degradation.Among these candidates, compound Z1 proved the most potent d
