Poly(3-hydroxybutyrate) production from swine wastewater and waste vegetable oil by Cupriavidus necator in nonsterile bioreactors.

Swine wastewater (SW), rich in nitrogen and phosphorus, and waste vegetable oil (WVO) are abundant feedstocks with potential for microbial biopolymers production. However, high polyhydroxyalkanoates (PHA) yields under nonsterile conditions remain challenging due to inhibitory compounds and microbial competition. Here, we cultivated Cupriavidus necator DSM545 under nonsterile conditions with varying SW dilutions combined with WVO for PHA synthesis. Batch bioreactors containing two-thirds (v/v) non-centrifuged wastewater yielded the highest polyhydroxybutyrate titers of 6.8 ± 0.1 gPHB/L, with C.

Kinetic and stoichiometric parameters in the fed-batch bioreactor production of poly(3-hydroxybutyrate) by Bacillus megaterium using different carbon sources.

This study investigates the effects of different strategies on poly(3-hydroxybutyrate)-P(3HB) production in a fed-batch bioreactor by Bacillus megaterium using candy industry effluent (CIE), sucrose, and rice parboiled water (RPW) as carbon sources. In biosynthesis, kinetic and stoichiometric parameters of substrate conversion into products and/or cells, productivity, instantaneous, and specific conversion rates were evaluated. The maximum concentration of P(3HB) was 4.00 g.L-1 (77% of the total dry mass) in 42 h of cultivation in minimal medium/RPW added with a carbon source based on CIE, dem

Tuning the biodegradation rate of poly(3-hydroxybutyrate) by incorporating acidified or oxidized biochar.

This study revealed that the acidification and oxidation of biochar retarded the biodegradation of poly(3-hydroxybutyrate) (PHB) films to a different extent. Herein, film samples were fabricated from neat poly(3-hydroxybutyrate) and PHB filled with various forms of biochar (1 wt%) by extrusion and thermocompression. The properties of the films were investigated by optical microscopy, scanning electron microscopy (SEM), atomic force microscopy (AFM), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA) and X-ray diffraction. The PHB films underwent a biodegradation test in

Determination of geometrical and viscoelastic properties of PLA/PHB samples made by additive manufacturing for urethral substitution.

Additive manufacturing has a great potential for creating hard tissue substitutes, such as bone and cartilage, or soft tissues, such as vascular and skin grafts. This study is a pilot study for 3D printing of a new material mixture potentially used as a tubular substitute for urethra replacement. This new mixture is a blend of polylactic acid (PLA) and polyhydroxybutyrate (PHB). The basic aspect that affects the 3D printing process is correct material preparation and setting of 3D printer parameters. Selection of material and printing parameters depend on printing technology. The printing tech

Statistical media optimization for growth and PHB production from methanol by a methylotrophic bacterium.

Media components were optimized by statistical design for cell growth and PHB production of Methylobacterium extorquens DSMZ 1340. Four important components of growth media were optimized by central composite design. The growth increased from an OD=1.35 for Choi medium as control to an OD=2.15 for optimal medium. Then media components for PHB production were optimized. Optimization of five important factors was conducted by response surface method. The optimal composition of PHB production medium was found to be at 7.8 (g/L) Na2HPO4 x 12H2O, and surprisingly at zero concentration of (NH4)2SO4,

Demonstration of bioplastic production from CO2 and formate using the reductive glycine pathway in E. coli.

There is a strong need to develop technologies that reduce anthropogenic pollution and the dependence on nonrenewable Earth resources. One way of doing so is by harnessing biological systems for replacing the production of fossil-fuel based goods with low-environmental-impact alternatives. Recently, progress was made in engineering the model organism E. coli to grow using CO2 and formate as its only carbon and energy sources using the reductive glycine pathway (rGlyP). Here, we use this engineered strain of E. coli as a host system for the production of polyhydroxybutyrate (PHB), a biologicall

[Effects of moist-heat sterilization on the structure of poly(3-hydroxybutyrate-co-4-hydroxybutyrate) microspheres and injectability of the composite hydrogels].

We assessed the effects of terminal moist-heat sterilization on the structure of poly(3-hydroxybutyrate-co-4-hydroxybutyrate) (P34HB) microspheres and the composite hydrogels with hyaluronic acid (HA), aiming to define an acceptable process window. According to pharmacopeial and sterilization standards, we selected 121 ℃ as the sterilization temperature and set holding time of 10, 20, and 30 min, with non-sterilized samples as controls. P34HB microspheres were first blended with HA and then subjected to moist-heat sterilization (co-sterilization). Gel permeation chromatography (GPC), different

Metabolomics Reveals Acetate-Formate compatibility for Dual-Carbon PHB biosynthesis in Cupriavidus necator.

The production of polyhydroxybutyrate (PHB) from organic substrates is often limited by energetic imbalance when single-carbon sources are used. Formate, while easily generated via electrochemical CO2 reduction, fails to sustain robust biomass and polymer yields when utilized as a primary carbon source. Similarly, acetate provides the necessary skeletal structures for PHB biosynthesis, but lacks the intrinsic energy to drive high-yield accumulation. To bridge this gap, we implemented a dual-carbon framework in wild-type Cupriavidus necator H16 that decouples energy supply from carbon assimilat

Biodegradability of commercial biopolyesters in marine sediments under anoxic conditions.

While the sea bottom is the major final sink for plastic items in the oceans, where they end up buried in sediments, the biodegradation of biopolymers has been studied so far under oxic conditions in the water column or the water-sediment interface. Here, the biodegradability of thin films of commercial biopolyesters, i.e., poly(butylene succinate) (PBS), poly(butylene succinate-co-adipate) (PBSA), poly(lactic acid) (PLA), poly(butylene adipate-co-terephthalate) (PBAT), and poly(hydroxybutyrate-co-hydroxyhexanoate) (PHBH), was investigated in microcosms of an anoxic marine sediment. Physiology-informed use of Cupriavidus necator in biomanufacturing: a review of advances and challenges.

Biomanufacturing offers a potentially sustainable alternative to deriving chemicals from fossil fuels. However, traditional biomanufacturing, which uses sugars as feedstocks, competes with food production and yields unfavourable land use changes, so more sustainable options are necessary. Cupriavidus necator is a chemolithoautotrophic bacterium capable of consuming carbon dioxide and hydrogen as sole carbon and energy sources, or formate as the source of both. This autotrophic metabolism potentially makes chemical production using C. necator sustainable and attractive for biomanufacturing. Add

Production of poly(3-hydroxybutyrate)/poly(lactic acid) from industrial wastewater by wild-type Cupriavidus necator H16.

The massive production of urban and industrial wastes has created a clear need for alternative waste management processes. One of the more promising strategies is to use waste as raw material for the production of biopolymers such as polyhydroxyalkanoates (PHAs). In this work, a lactate-enriched stream obtained by anaerobic digestion (AD) of wastewater (WW) from a candy production plant was used as a feedstock for PHA production in wild-type Cupriavidus necator H16. Unexpectedly, we observed the accumulation of poly(3-hydroxybutyrate)/poly(lactic acid) (P(3HB)/PLA), suggesting that the non-eng

Community composition and physiological plasticity control microbial carbon storage across natural and experimental soil fertility gradients.

Many microorganisms synthesise carbon (C)-rich compounds under resource deprivation. Such compounds likely serve as intracellular C-storage pools that sustain the activities of microorganisms growing on stoichiometrically imbalanced substrates, making them potentially vital to the function of ecosystems on infertile soils. We examined the dynamics and drivers of three putative C-storage compounds (neutral lipid fatty acids [NLFAs], polyhydroxybutyrate [PHB], and trehalose) across a natural gradient of soil fertility in eastern Australia. Together, NLFAs, PHB, and trehalose corresponded to 8.5-

Simple and rapid method for isolation and quantitation of polyhydroxyalkanoate by SDS-sonication treatment.

We developed a new method for isolation and quantitation of polyhydroxyalkanoate (PHA) from culture broth. In this method, the cells were sonicated in sodium dodecyl sulfate (SDS) solution and centrifuged to recover PHA. The recovered PHA was rinsed with deionized water and ethanol, and then weighed after drying. Hazardous chemicals such as chloroform, methanol, and sulfuric acid were not used, and no expensive analytical instruments were needed. We applied this method to Cupriavidus necator culture broths that included various amounts of poly(3-hydroxybutyrate) (PHB) or poly(3-hydroxybutyrate

ATMOSPHERIC NITROGEN FIXATION BY METHANE-OXIDIZING BACTERIA.

Bacteriol. 88:468-472. 1964.-Methane-oxidizing bacteria capable of fixing atmospheric nitrogen were isolated from garden soil, pond mud, oil field soil, and soil exposed to natural gas, indicating a rather wide prevalence in nature. This may explain the high concentration of organic nitrogen commonly found in soils exposed to gas leakage from pipelines or natural-gas seeps. Added molybdenum was a requirement for growth in a nitrogen-free minera

Vinasse: from a residue to a high added value biopolymer.

This work aimed to study the feasibility of using vinasse for polyhydroxybutyrate (PHB) production by Bacillus megaterium. To optimize the culture medium, a Box-Behnken design was employed considering carbon (C), nitrogen (N), and phosphorus (Ph) concentrations as independent variables and PHB productivity as the response variable. The productivity decreased when C or N were increased, probably due to the presence of phenolic compounds and the limitation of N for the production of PHB by Bacillus sp. bacteria. An additional experimental design to optimize the C/N ratio and growing conditions (

Effect of R-plasmid RP1 and nutrient depletion on the gross cellular composition of Escherichia coli and its resistance to some uncoupling phenols.

The resistance of Escherichia coli batch cultures depleted of carbon (C-dep), magnesium (Mg-dep), or phosphate (P-dep) against low concentrations of 3-chlorophenol, 4-chlorophenol, or 2-phenoxyethanol varied. C-dep cultures were always significantly more sensitive than Mg-dep or P-dep cultures. The presence of R-plasmid RP1 increased the sensitivity of C-dep cultures to 3- and 4-chlorophenol, yet had little effect on those cultured depleted in magnesium or phosphate ions. Cultures with R-plasmid RP1 had increased levels of beta-polyhydroxybutyrate irrespective of the nature of the depleting nu
