Hierarchical Mesoscale Trapping Controls Pathway Selection in Supramolecular Polymerization.

Supramolecular polymerization often proceeds through competing kinetic and thermodynamic pathways, giving rise to pathway complexity that governs the structure and function of supramolecular materials. While pathway selection is typically interpreted in terms of molecular packing, the potential regulatory role of organization beyond the molecular scale remains largely unexplored. Here we demonstrate that hierarchical mesoscale aggregation can control pathway selection during supramolecular polymerization. A donor-acceptor dicyanostilbene derivative forms two primary aggregates that further ass

Beyond geometric constraint: dynamic fluid-lattice-activated topochemical polymerization.

Topochemical polymerization affords lattice-directed polymers with high structural fidelity, but its scope is limited by the need for precise, static preorganization of reactive groups-an intrinsically fragile outcome of subtle intermolecular packing. Here we introduce dynamical fluid lattices into topochemical design to circumvent these geometric constraints, achieving near-quantitative monomer conversion even without ideal preorganization. Employing core-shell columnar liquid crystals, rapid molecular fluctuations within the fluid lattices enable temporary proximity of reactive sites that ac

Chain Length Dependence of Chemically Controlled Reactions in Polymerization.

Chemically controlled reactions in polymerization processes are traditionally assumed to have chain-length-independent rate coefficients. However, this work challenges that assumption by using multiscale modeling to demonstrate that chemically controlled polymer-polymer reactions can exhibit significant chain length effects. Taking reversible addition-fragmentation chain transfer (RAFT) polymerization as a case study, we compare the chain length convergence of the reaction entropy for two processes: the addition of a growing polymer radical to a simultaneously growing polymeric RAFT agent, and

Sonochemical Nitroxide-Mediated Polymerization: Harnessing Sonochemistry for Polymer Synthesis.

In polymer science, mechanochemistry is emerging as a powerful tool for materials science and molecular synthesis, offering novel avenues for controlled polymerization and post-synthetic modification. Building upon the previous research, nitroxide-mediated polymerization (NMP) is merged with mechanochemistry through the design of nitroxide-based mechanophore macroinitiators, pioneering the first instance of a sonochemical nitroxide-mediated-type polymerization. As NMP usually requires high temperatures, this study demonstrates that a sonochemical NMP-type process allows polymerization under re

Taste Detection of Maltooligosaccharides with Varying Degrees of Polymerization.

Previous studies have shown that humans can taste maltooligosaccharides [MOS; degree of polymerization (DP) of 3-20] but not maltopolysaccharides (MPS; DP of >20) and that their taste detection is independent of the canonical sweet taste receptor. The objectives of this study were to determine the DP ranges of target stimuli that are tasted and further to investigate the impact of DP on taste detectability. To achieve this goal, we prepared three food-grade MOS samples with narrow DP ranges using flash chromatography: low (4-6), medium (7-12), and high (14-21) DP samples. Following sample prep

Mineralization versus polymerization pathways in heterogeneous Fenton-like reactions.

Fenton reaction has been widespread application in water purification due to the excellent oxidation performances. However, the poor cycle efficiency of Fe(III)/Fe(II) is one of the biggest bottlenecks. In this study, graphite (GP) was used as a green carbon catalyst to accelerate Fenton-like (H2O2/Fe3+ and persulfate/Fe3+) reactions by promoting ferric ion reduction and intensifying diverse peroxide activation pathways. Significantly, the carboxyl group on GP anchors iron ions to form GP-COOFe(III) which promote persulfate adsorption to form surface complexes and induce an electron transfer p

Nanoconfined polymerization limits crack propagation in hysteresis-free gels.

Consecutive mechanical loading cycles cause irreversible fatigue damage and residual strain in gels, affecting their service life and application scope. Hysteresis-free hydrogels within a limited deformation range have been created by various strategies. However, large deformation and high elasticity are inherently contradictory attributes. Here we present a nanoconfined polymerization strategy for producing tough and near-zero-hysteresis gels under a large range of deformations. Gels are prepared through in situ polymerization within nanochannels of covalent organic frameworks or molecular si

Macrochain Transfer Agents for Catalytic Ring-Opening Metathesis Polymerization.

A monosubstituted 1,3-diene derivative attached to a polymer is demonstrated to act as a macrochain transfer agent in catalytic ring-opening metathesis polymerization. PEG- and PLA-based macrochain transfer agents were synthesized in a few steps and were characterized using NMR spectroscopy, size exclusion chromatography (SEC) and matrix-assisted laser desorption/ionization-time-of-flight (MALDI-ToF) mass spectrometry. Poly(l-lactide) based diblock copolymer, poly(ethylene glycol)-based diblock, and triblock (ABA type) copolymers of varied chain lengths were prepared catalytically in a one-pot

Electrochemical Investigation of Iron-Catalyzed Atom Transfer Radical Polymerization.

Use of iron-based catalysts in atom transfer radical polymerization (ATRP) is very interesting because of the abundance of the metal and its biocompatibility. Although the mechanism of action is not well understood yet, iron halide salts are usually used as catalysts, often in the presence of nitrogen or phosphorous ligands (L). In this study, electrochemically mediated ATRP (eATRP) of methyl methacrylate (MMA) catalyzed by FeCl3, both in the absence and presence of additional ligands, was investigated in dimethylformamide. The electrochemical behavior of FeCl3 and FeCl3/L was deeply investiga

Multicomponent Polymerization toward Cationic Polymers for Efficient Gene Delivery.

A new class of cationic polymers containing tertiary amine, thioether, and hydroxyl groups are prepared via a catalyst-free, multicomponent polymerization method using dithiol, formaldehyde, and di-sec-amine with a ratio of 1:2:1, to access a library of water-soluble polymers with well-defined structures and suitable molecular weights (Mw ranging from 5000 to 8000 Da) in high yields (up to 90%). Such polycations are demonstrated to be promising nonviral gene delivery vectors with high transfection efficiency (up to 3.5-fold of PEI25k) and low toxicity with multiple functionalities: 1) efficien

Stoichiometry of Nck-dependent actin polymerization in living cells.

Regulation of actin dynamics through the Nck/N-WASp (neural Wiskott-Aldrich syndrome protein)/Arp2/3 pathway is essential for organogenesis, cell invasiveness, and pathogen infection. Although many of the proteins involved in this pathway are known, the detailed mechanism by which it functions remains undetermined. To examine the signaling mechanism, we used a two-pronged strategy involving computational modeling and quantitative experimentation. We developed predictions for Nck-dependent actin polymerization using the Virtual Cell software system. In addition, we used antibody-induced aggrega

RNA induced polymerization of the Borna disease virus nucleoprotein.

The Borna disease virus (BDV) nucleoprotein (N) monomer resembles the nucleoprotein structures from rabies virus (RABV) and vesicular stomatitis virus (VSV). We show that BDV N assembles into ring- and string-like structures in the presence of 5' genomic BDV RNA. RNA induced polymerization is partly RNA-specific since polymerization is inefficient in the presence of 3' genomic BDV RNA or E. coli RNA. Mutagenesis of basic residues located in the cleft made up by the N- and C-terminal domains of N abrogate RNA-induced polymerization indicating that BDV N binds RNA similarly as observed in case o

YopR impacts type III needle polymerization in Yersinia species.

A hallmark of Yersinia type III machines is the presence of needles extending from the bacterial surface. Needles perform two functions, serving as the conduits for the transport of effectors into immune cells but also acting as a sensor. The polymerized needle protein, YscF, is thought to perceive threshold levels of environmental calcium ions to trigger secretion. yopR (yscH) is a gene downstream of yscEFG, encoding the chaperones and principal building blocks of the needle. Here we investigated the contribution of YopR towards type III secretion and pathogenesis. Yersinia pestis KIM D27 mut

ARF1-mediated actin polymerization produces movement of artificial vesicles.

Vesicular trafficking and actin dynamics on Golgi membranes are both regulated by ADP-ribosylation factor 1 (ARF1) through the recruitment of various effectors, including vesicular coats. Actin assembly on Golgi membranes contributes to the architecture of the Golgi complex, vesicle formation, and trafficking and is mediated by ARF1 through a cascade that leads to Arp2/3 complex activation. Here we addressed the role of Golgi actin downstream of ARF1 by using a biomimetic assay consisting of liposomes of defined lipid composition, carrying an activated form of ARF1 incubated in cytosolic cell

Light-induced endocapsular polymerization of injectable lens refilling materials.

The use of advanced phacoemulsification techniques enables the surgeon to remove opacified lens material while preserving a nearly complete capsular bag. For this reason, several groups are presently working on lens refilling techniques. We have developed a method for the injection of a liquid monomer into the lens capsule. The material can be polymerized by exposure to cold light (wavelength, 400-500 nm) within the capsular bag. Using cell-culture experiments, we selected an optically clear material that has cytotoxic properties when placed in direct contact with cells but does not release cy

Polymeric immunosensors for tumor detection.

Cancer is a broad-spectrum disease which is spread globally, having high mortality rates. This results from genetic, epigenetic and molecular abnormalities caused by various mutations. The main reason behind this critical problem lies in its diagnostics, the late detection of the disease is the root cause of all this. This can be managed well by the timely diagnosis of cancer by means of the tumor biomarkers present in the body fluids such as serum, blood, and urine. These tumor biomarkers are present in normal conditions as well, but their concentrations are altered in the presence of a malig

Polymeric Nanocarriers for the Delivery of Antimalarials.

Malaria is an infectious disease caused by a protozoan parasite which is transmitted by female Anopheles mosquitoes around tropical and sub-tropical regions. Half of the world's population is at risk of being infected by malaria. This mainly includes children, pregnant women and people living with chronic diseases. The main factor that has contributed to the spread of this disease is the increase in the number of drug-resistant parasites. To overcome drug resistance, researchers have developed drug delivery systems from biodegradable polymers for the loading of antimalarials. The drug delivery

Vat Polymerization 3D Printing of Hydrogels With Tunable Porosity.

Three-dimensional, interconnected hydrogel networks are central to tissue engineering and disease modeling, where tailored pore architecture and mechanical robustness are essential for supporting cellular functions. However, the limited ability to engineer microstructural features in vat polymerization 3D-printed natural hydrogels often compromises scaffold performance, as oversized pores reduce cell attachment and cell-cell interactions while smooth pore walls lack essential topographical cues. Here, we report an emulsion-based ink for vat polymerization 3D printing that enables the fabricati
