To reduce reliance on time consuming postharvest seed potato grow outs, a Whatman Flinders Technology Associates Plantsaver Cards (FTA)-based pathogen detection protocol based on dormant tubers was developed for four potato viruses. Viruses tested included potato virus Y (PVY), potato virus S (PVS), potato mop-top virus (PMTV), and tobacco rattle virus (TRV). Viruses were also detected from potato tubers, sprouts, and leaves using different experimental test methods and sample collection timings (harvest-FTA, storage-FTA, sprouting-FTA, sprout-enzyme-linked immunosorbent assay [ELISA], sprout-

The applications of mesoionic compounds and their analogues as agents against plant viruses remain unexplored. This was the first evaluation of the antiviral activities of mesoionic compounds on this issue. Our study involved the design and synthesis of a series of novel imidazo[1,2-a]pyridine mesoionic compounds containing a sulfonamide moiety and the assessment of their antiviral activities against potato virus Y (PVY). Compound A33 was assessed on the basis of three-dimensional quantitative structure-activity relationship (3D-QSAR) model analysis and displayed good curative, protective, and

The potential effect of genetic modification on nutritional properties of potatoes transformed to improve resistance to a necrotic strain of Potato virus Y was determined in a rat experiment. Autoclaved tubers from four transgenic lines were included to a diet in the amount of 40% and compared with the conventional cv. Irga. The experiment lasted 3 weeks and special attention was paid to nutritional properties of diets, caecal metabolism and serum indices. Genetic modification of potato had no negative effect on the chemical composition and nutritional properties of tubers, ecosystem of the ca

Cell-intrinsic restriction factors (CIRFs) negatively regulate plant virus infections and represent valuable resources for breeding virus-resistant crops. However, the potential for ribosomal proteins to function as CIRFs and the underlying mechanisms remains elusive. Our previous work demonstrated that the Nicotiana benthamiana chloroplastic ribosomal protein of the large subunit 1 (NbRPL1) promotes the infection of tobacco vein banding mosaic virus (TVBMV) by antagonizing the NbBeclin1-mediated degradation of the viral RNA-dependent RNA polymerase, NIb. Continuing this line of research, we e

BACKGROUND: Certification of seed potato as free of viruses is essential for stable potato production. Among more than 30 virus species infecting potato, potato leafroll virus (PLRV), potato virus S (PVS), potato virus X (PVX), and potato virus Y (PVY) predominate worldwide and should be the targets of a high-throughput detection protocol for seed potato quarantine.
RESULTS: We developed an assay based on one-step real-time multiplex reverse transcription-polymerase chain reaction (mRT-PCR) with melt curve analysis for the four viruses and one internal control, potato elongation factor 1 alph

Spodoptera frugiperda (fall armyworm) is a notorious pest that threatens maize production worldwide. Current control measures involve the use of chemical insecticides and transgenic maize expressing Bacillus thuringiensis (Bt) toxins. Although additional transgenes have confirmed insecticidal activity, limited research has been conducted in maize, at least partially due to the technical difficulty of maize transformation. Here, we describe implementation of a sugarcane mosaic virus (SCMV) vector for rapidly testing the efficacy of both endogenous maize genes and heterologous genes from other o

Plants employ diverse responses mediated by phytohormones to defend themselves against pathogens and herbivores. Adapted pathogens and herbivores often manipulate these responses to their benefit. Previously, we demonstrated that Turnip mosaic virus (TuMV) infection suppresses callose deposition, an important plant defense induced in response to feeding by its aphid vector, the green peach aphid (Myzus persicae), and increases aphid fecundity compared with uninfected control plants. Further, we determined that production of a single TuMV protein, Nuclear Inclusion a-Protease (NIa-Pro) domain,

The effect of abiotic stress responses on Potato virus A (PVA; genus Potyvirus) infection was studied. Salt, osmotic and wounding stress all increased PVA gene expression in infected Nicotiana benthamiana leaves. According to the literature, an early response to these stresses is an elevation in cytosolic Ca(2+) concentration. The infiltration of 0.1 m CaCl(2) into the infected leaf area enhanced the translation of PVA RNA, and this Ca(2+) -induced effect was more profound than that induced solely by osmotic stress. The inhibition of voltage-gated Ca(2+) channels within the plasma membrane abo

Plant viral diseases pose a significant threat to agricultural production, and the availability of effective drugs against viral diseases remains limited. In this study, we discovered that betulinic acid (BA), a pentacyclic triterpenoid derived from plants, delays infection by turnip mosaic virus (TuMV) in Nicotiana benthamiana. Transcriptomic analysis revealed that BA treatment specifically induced the expression of N. benthamiana phytosulfokine 3 (NbPSK3), a plant pentapeptide hormone with diverse functions, while TuMV infection suppressed its expression. Further study demonstrated that NbPS

Papaya ringspot virus (PRSV) limits papaya production worldwide. Previously, we generated transgenic lines of hybrid Tainung No.2 (TN-2) carrying the coat protein (CP) gene of PRSV with broad resistance to PRSV strains. Unfortunately, all of them were female, unacceptable for growers and consumers in practical applications. With our reported flanking sequences and the newly released papaya genomic information, the CP-transgene insert was identified at a non-coding region in chromosome 3 of the papaya genome, and the flanking sequences were verified and extended. The female transgenic line 16-0

Whereas the activation of resistance (R) proteins has been intensively studied, the downstream signaling mechanisms leading to the restriction of the pathogen remain mostly unknown. We studied the immunity network response conditioned by the potato Ny-1 gene against potato virus Y. We analyzed the processes in the cell death zone and surrounding tissue on the biochemical and gene expression levels in order to reveal the spatiotemporal regulation of the immune response. We show that the transcriptional response in the cell death zone and surrounding tissue is dependent on salicylic acid (SA). F

Epigenetic modifications play pivotal roles in regulating plant adaptive responses to viral infection and various other stresses. However, how viral infection rewires and shapes chromatin-based epigenetic regulatory networks in crops with contrasting resistance remains unclear. To this end, we investigated the consequences of epigenetic variations in resistant and susceptible soybean cultivars following soybean mosaic virus (SMV) infection. SMV infection mediates the depletion of 24-nucleotide small interfering RNAs (24-nt siRNAs) in susceptible cultivars and induces the accumulation of 24-nt

Cymbidium mosaic virus (CymMV) and Odontoglossum ringspot virus (ORSV) are among the world's most serious and widespread orchid viruses; they often infect orchids, causing devastating losses to the orchid industry. Therefore, it is critical to establish a method that can rapidly and accurately detect viruses in the field using simple instruments, which will largely reduce the further spread of viruses and improve the quality of the orchid industry and is suitable for mass promotion and application at grassroots agrotechnical service points. In this investigation, we established a rapid amplifi

Recent studies have documented effects of plant viruses on host plants that appear to enhance transmission by insect vectors. But, almost no empirical work has explored the implications of such apparent manipulation for interactions among co-infecting pathogens. We examined single and mixed infections of two potyviruses, watermelon mosaic virus (WMV) and zucchini yellow mosaic virus (ZYMV), that frequently co-occur in cucurbitaceae populations and share the same aphid vectors. We found that ZYMV isolates replicated at similar rates in single and mixed infections, whereas WMV strains accumulate

BACKGROUND: Soybean mosaic virus (SMV) causes substantial yield losses of soybean worldwide. Although Pseudomonas chlororaphis IRHB3 is known to promote soybean growth and suppress fungal diseases, however, its efficacy against SMV has remained unclear. This study evaluated IRHB3 impacts on SMV infection and explored the underlying physiological and molecular mechanism.
RESULTS: Foliar application of IRHB3 significantly reduced SMV disease index, with control efficacy of 52.69%, and decreased viral accumulation in inoculated and systemic leaves. IRHB3 also alleviated SMV-induced growth inhibi

BACKGROUND: Sustainable nanotechnology, particularly chitosan (CS)-based biodegradable nanoparticles (NPs), offers an eco-friendly delivery system compared to conventional techniques. Potato virus Y (PVY) causes severe potato yield losses and reduces tuber quality. Its control is challenging owing to nonpersistent aphid transmission, especially by Myzus persicae, the most efficient vector. Chemical insecticides lead to environmental pollution, health risks, and resistance, highlighting the need for alternatives. This study assessed pure CS, thyme essential oil (TEO)-loaded CS, CS-lecithin (LH)

Potato virus Y (PVY) causes significant damage to crops worldwide. Nucleoside analogs and their derivative drugs are currently a research hot spot in antiviral medications. We designed and synthesized a nucleoside guanidine compound FluoroGuanidoNucleosin (FGN) with anti-plant virus effects by structural modification of the nucleoside scaffold. The structure of compound FGN was characterized by 1H NMR, 13C NMR, and HRMS, and is named (R)-N-(1-((2R,3R,4S,5R)-3,4-dihydroxy-5-(hydroxymethyl)tetrahydrofuran-2-group)-2-oxo-1,2-dihydropyrimidine-4-group)-3-guanidine-4-(2,4,5-trifluorophenyl)butanami

Tomato-infecting viruses have been considered as a serious threat to tomato crops in Poland. Therefore, during 2014-2021, 234 tomato samples delivered directly by greenhouse tomato growers to Plant Disease Clinic of IPP-NRI were tested. Eight virus species: pepino mosaic virus (PepMV), tomato yellow ring orthotospovirus (TYRV), tomato spotted wilt orthotospovirus (TSWV), potato virus Y (PVY), cucumber mosaic virus (CMV), tomato black ring virus (TBRV) and tomato mosaic virus (ToMV) were detected in single or mixed infection in 89 samples. The presence of TYRV was established for the first time

Sugarcane mosaic virus (SCMV) is a pathogen of worldwide importance that causes dwarf mosaic disease on maize (Zea mays). Until now, few maize genes/proteins have been shown to be involved in resistance to SCMV. In this study, we characterized the role of maize phenylalanine ammonia-lyases (ZmPALs) in accumulation of the defence signal salicylic acid (SA) and in resistance to virus infection. SCMV infection significantly increased SA accumulation and expression of SA-responsive pathogenesis-related protein genes (PRs). Interestingly, exogenous SA treatment decreased SCMV accumulation and enhan

Watermelon mosaic virus (WMV) is a major constraint in squash production, necessitating sustainable antiviral strategies. This study evaluated the efficacy of aqueous extracts from six medicinal plants: Artemisia sp., Matricaria chamomilla, Allium cepa, Allium sativum, Moringa oleifera, and Citrullus colocynthis in promoting plant growth and mitigating WMV infection. To the best of our knowledge, this is the first report demonstrating the efficacy of the combined application of Artemisia extract and B. mycoides revealed a synergistic effect, significantly enhancing physiological recovery and r
