BACKGROUND: Parkinson's disease (PD) is a multifactorial neurodegenerative disorder, and peripheral blood-based biomarkers may help capture disease-related molecular changes. Because protein prenylation is essential for Rab-mediated intracellular trafficking, we investigated whether genes encoding key prenylation enzymes are dysregulated in PD.
METHODS: Peripheral blood samples were collected from patients with PD and healthy controls. Quantitative real-time PCR was performed for FNTA, FNTB, RABGGTA, RABGGTB, and PGGT1B, using B2M as the endogenous reference gene. Expression was analyzed with

UNLABELLED: Leprosy is a chronic infectious disease caused by Mycobacterium leprae and M. lepromatosis. Brazil consistently ranks among the countries with the highest number of leprosy cases. Data from our group showed that M. leprae upregulates the mevalonate pathway (MP), contributing to the accumulation of cholesterol-ester-enriched lipid droplets in infected macrophages, and that the inhibition of this pathway by statins decreases bacterial intracellular viability. It has already been shown that part of the deleterious effect of statins on M. leprae survival is related to the reduced chole

Prenylation, which involves the addition of hydrophobic molecules, is considered to enhance the bioavailability and biological activity of flavonoids. However, the effect of prenylation on the estrogenic activity of flavonoids with different structures remains unclear. This study evaluated the estrogen receptor-α (ER-α) agonistic and antagonistic activities of estrogenic flavonoids in both unprenylated and prenylated forms using OECD standardized in vitro ER-α transactivation assay and in vivo uterine hypertrophy assay. A luciferase reporter assay using ER-α-HeLa-9903 cells revealed that twelv

Protein prenylation is an important protein modification that is responsible for diverse physiological activities in eukaryotic cells. This modification is generally catalyzed by three types of prenyl transferases, which include farnesyl transferase (FT), geranylgeranyl transferase (GGT-1) and Rab geranylgeranyl transferase (GGT-2). Studies in malaria parasites showed that these parasites contain prenylated proteins, which are proposed to play multiple functions in parasites. However, the prenyl transferases have not been functionally characterized in parasites of subphylum Apicomplexa. Here,

Prenylated isoflavonoids are phytochemicals with promising antifungal properties. Recently, it was shown that glabridin and wighteone disrupted the plasma membrane (PM) of the food spoilage yeast Zygosaccharomyces parabailii in distinct ways, which led us to investigate further their modes of action (MoA). Transcriptomic profiling with Z. parabailii showed that genes encoding transmembrane ATPase transporters, including Yor1, and genes homologous to the pleiotropic drug resistance (PDR) subfamily in Saccharomyces cerevisiae were upregulated in response to both compounds. Gene functions involve

Prenylated diresorcinols exhibit various bioactivities, including cytotoxic, antibacterial, and antifungal activities. Therefore, establishing facile and efficient synthetic routes for prenylated diresorcinols facilitates their development as chemical probes or drugs with a novel mode of action. In this study, microwave-assisted copper catalysis was explored as a cost-effective and environmentally friendly method for the cross-coupling of sterically hindered ortho-prenylated phenols and aryl halides to produce bioactive prenylated diresorcinols, diorcinol I and leotiomycene B. Notable advantag

Prenylated flavonoids are an important class of naturally occurring flavonoids with important biological activity, but their low abundance in nature limits their application in medicines. Here, we showed the hemisynthesis and the determination of various biological activities of seven prenylated flavonoids, named 7-13, with an emphasis on antimicrobial ones. Compounds 9, 11, and 12 showed inhibitory activity against human pathogenic fungi. Compounds 11, 12 (flavanones) and 13 (isoflavone) were the most active against clinical isolated Staphylococcus aureus MRSA, showing that structural require

Influenza virus neuraminidase (NA) is the primary target for influenza therapeutics. Severe complications are often related to secondary pneumonia caused by Streptococcus pneumoniae (pneumococci), which also express NAs. Recently, a NA-mediated lethal synergism between influenza A viruses and pneumococci was described. Therefore, dual inhibitors of both viral and bacterial NAs are expected to be advantageous for the treatment of influenza. We investigated the traditional Chinese herbal drug sāng bái pí (mulberry root bark) as source for anti-infectives. Two prenylated flavonoid derivatives, sa

BACKGROUND: Prenyltrasferases (PTases) are a class of enzymes known to be responsible for promoting posttranslational modification at the carboxyl terminus of proteins containing a so-called CaaX-motif. The process is responsible for proper membrane localization and the appropriate function of several intracellular signaling proteins. Current research demonstrating the pathomechanistic importance of prenylation in inflammatory illnesses emphasizes the requirement to ascertain the differential expression of PT genes under inflammatory settings, particularly in periodontal disease.
METHODS: Tel

Human WASP and N-WASP are homologous proteins that require the binding of multiple regulators, including the acidic lipid PIP2 and the small GTPase Cdc42, to relieve autoinhibition before they can stimulate the initiation of actin polymerization. Autoinhibition involves intramolecular binding of the C-terminal acidic and central motifs to an upstream basic region and GTPase binding domain. Little is known about how a single intrinsically disordered protein, WASP or N-WASP, binds multiple regulators to achieve full activation. Here we used molecular dynamics simulations to characterize the bind

Investigations of major mevalonate pathway enzymes have demonstrated the importance of local isoprenoid synthesis in cardiac homeostasis. Farnesyl diphosphate synthase (FPPS) synthesizes isoprenoid precursors needed for cholesterol biosynthesis and protein prenylation. Wang, Zhang, Chen et al, in a recently published article in The Journal of Pathology, elegantly elucidated the pathological outcomes of FPPS deficiency in cardiomyocytes, which paradoxically resulted in increased prenylation of the small GTPases Ras and Rheb. Cardiomyocyte FPPS depletion caused severe dilated cardiomyopathy that

Statins and various isoprenoids of dietary origins inhibit L-mevalonic acid synthesis, which in turn downregulates cholesterol and various other dependent substances, including farnesyl- and geranylgeranyl-conjugated proteins involved in cell signaling processes. Such signaling processes are stimulated by protease-activated receptor-1 (PAR-1), which upon activation, causes the expression of various substances including tissue factor (TF) and plasminogen activator inhibitor-1 (PAI-1). Tissue factor promotes thrombin generation, where thrombin stimulates a variety of cellular processes, as well

Prenylated phenols, the primary class of mushroom-derived meroterpenoids, exhibit significant structural diversity and biological activities. Two new prenylated phenols (1 and 2), along with four aromatic derivatives, were isolated from the wild mushroom Lentinula edodes for the first time. Their structures were established based on extensive spectroscopic data (UV, IR, MS, and NMR) and further corroborated by X-ray crystallographic analysis. Compounds 1 and 2 are unusual meroterpenoids characterized by 4-prenylated benzene 1,2,3-triols or 3-prenylated benzene 1,2-diols. Compound 2 showed mode

Kidney vetch (Anthyllis vulneraria L.) is an old medicinal plant mostly known for ethnomedicine. Nowadays, its herbal parts are only scarcely used in phytotherapy, whereas medicinal applications of roots have not been reported so far. In this study, we investigated ethyl acetate (EtOAc) extracts obtained by solvent partition of an acetone/water crude extract of A. vulneraria roots. Mainly prenylated chalcones, flavones, isoflavones, and cinnamic acid derivatives dominated the compound profile when the extracts were studied by HPLC-ESI-(HR)MSn and GC-MS analysis. Two main chalcones (23 and 24)
