Altered Brain Structure and Functional Connectivity of Primary Visual Cortex in Optic Neuritis.

Previous studies have revealed brain adaptations to injury that occurs in optic neuritis (ON); however, the mechanisms underlying the functional connectivity (FC) and gray matter volume (GMV) changes in ON have not been clarified. Here, 51 single attack ON patients and 45 recurrent attacks ON patients were examined using structural MRI and resting-state functional MRI (RS-fMRI), and compared to 49 age- and gender-matched healthy controls (HC). FC analysis with a seed in primary visual cortex (V1 area) was used to assess the differences among three groups. Whole brain GMV was assessed using vox

Unilateral peripheral vestibular dysfunction disrupts motion-evoked neuronal responses in primary visual cortex.

Acute unilateral peripheral vestibular destruction (AUPVD) causes dizziness, characterized by both visual and motion-related disturbances. Although it is well established that the vestibular nuclei receive auditory and visual sensory inputs and play a critical role in maintaining balance and gaze, the extent to which vestibular activity influences the primary visual cortex (V1) remains unclear. In this study, we observed that V1 neurons failed to generate appropriate responses to motion stimuli using chronic two-photon microscopy in awake AUPVD mice, despite responding normally to static visua

Context-dependent orientation discontinuity encoding by gamma rhythms in mouse primary visual cortex.

Through the modulation of its surround, an identical visual stimulus can be perceived as more or less salient, allowing it to either stand out or seamlessly integrate with the rest of the visual scene. Gamma rhythms are associated with processing stimulus features across extensive areas of the visual field. Consistent with this concept, the magnitude of visually induced gamma rhythm depends on how well stimulus features aligned both within and outside the classical receptive field (CRF) at the recording site. However, there still exists some uncertainty regarding the encoding of context-modula

Irreversible Primary Visual Cortex Impairment in a Mouse Model of High-Risk Schizophrenia.

PURPOSE: Although visual deficits can be observed at any stage of schizophrenia, few studies have focused on visual cortex alterations in individuals at high risk of schizophrenia. This study aimed to investigate the pathological changes of the primary visual cortex in a prenatal mouse model of MK801-induced high-risk schizophrenia. METHODS: The high-risk schizophrenia model was generated by MK801 injection into pregnant mice. The male offspring without schizophrenia-like behaviors in early adulthood were defined as the high-risk mouse model of schizophrenia (HRMMS) and divided into two group

Cortical thickness of primary visual cortex correlates with motion deficits in periventricular leukomalacia.

Impairments of visual motion perception and, in particular, of flow motion have been consistently observed in premature and very low birth weight subjects during infancy. Flow motion information is analyzed at various cortical levels along the dorsal pathways, with information mainly provided by primary and early visual cortex (V1, V2 and V3). We investigated the cortical stage of the visual processing that underlies these motion impairments, measuring Grey Matter Volume and Cortical Thickness in 13 children with Periventricular Leukomalacia (PVL). The cortical thickness, but not the grey matt

Progressive thinning of visual cortex in primary open-angle glaucoma of varying severity.

The aim of this study was to investigate possible changes of cortical thickness in the visual cortex in primary open-angle glaucoma (POAG) of varying severity. Twenty normal controls (NC), 20 mild (MP) and 17 severe (SP) POAG patients were recruited and scanned using magnetic resonance imaging. Cortical thickness analyses with regions of interest (V1, V2, ventral V3, V4 and V5/MT+) were used to assess the cortical changes among the three groups. Furthermore, the associations of cortical thickness with retinal nerve fiber layer (RNFL) thickness and mean deviation of visual field were analyzed.

Visual Recognition Is Heralded by Shifts in Local Field Potential Oscillations and Inhibitory Networks in Primary Visual Cortex.

Learning to recognize and filter familiar, irrelevant sensory stimuli eases the computational burden on the cerebral cortex. Inhibition is a candidate mechanism in this filtration process, and oscillations in the cortical local field potential (LFP) serve as markers of the engagement of different inhibitory neurons. We show here that LFP oscillatory activity in visual cortex is profoundly altered as male and female mice learn to recognize an oriented grating stimulus-low-frequency (∼15 Hz peak) power sharply increases, whereas high-frequency (∼65 Hz peak) power decreases. These changes report

Dendritic Cell Factor 1-Knockout Results in Visual Deficit Through the GABA System in Mouse Primary Visual Cortex.

The visual system plays an important role in our daily life. In this study, we found that loss of dendritic cell factor 1 (DCF1) in the primary visual cortex (V1) caused a sight deficit in mice and induced an abnormal increase in glutamic acid decarboxylase 67, an enzyme that catalyzes the decarboxylation of glutamate to gamma aminobutyric acid and CO2, particularly in layer 5. In vivo electrophysiological recordings confirmed a decrease in delta, theta, and beta oscillation power in DCF1-knockout mice. This study presents a previously unknown function of DCF1 in V1, suggests an unknown contac

Retinotopic organization of primary visual cortex in glaucoma: Comparing fMRI measurements of cortical function with visual field loss.

Primary open angle glaucoma (POAG) is a progressive optic neuropathy characterized by retinal ganglion cell loss. Experimental primate glaucoma indicates neuronal degeneration of the lateral geniculate nucleus (LGN) and activity changes in the visual cortex (V1). Neuronal degeneration has also been shown in a post-mortem human study of the optic nerve, LGN and visual cortex. Functional magnetic resonance imaging (fMRI), a non-invasive means of inferring function-specific neuronal activity, provides an opportunity to evaluate glaucomatous changes in neuronal activity throughout the visual pathw

Cross-modal activation of the primary visual cortex by auditory stimulation in RCS rats: considerations in visual prosthesis.

An important brain re-wiring, the so-called cross-modal plasticity, occurs during progression of retinal degenerative diseases to compensate for lack of visual input. The visual cortex does not go 'unused', instead it is devoted to processing other sensory modalities. In this study we recorded, in the visual cortex, visual- and auditory-evoked potentials in an anesthetized murine model of retinal degeneration. The latency to the first peak of the recorded local field potentials was used to assess the speed of the response. Visual responses occurred significantly faster in the control group. Co

Population receptive field analysis of the primary visual cortex complements perimetry in patients with homonymous visual field defects.

Injury to the primary visual cortex (V1) typically leads to loss of conscious vision in the corresponding, homonymous region of the contralateral visual hemifield (scotoma). Several studies suggest that V1 is highly plastic after injury to the visual pathways, whereas others have called this conclusion into question. We used functional magnetic resonance imaging (fMRI) to measure area V1 population receptive field (pRF) properties in five patients with partial or complete quadrantic visual field loss as a result of partial V1+ or optic radiation lesions. Comparisons were made with healthy cont

Disruptions in primary visual cortex physiology and function in a mouse model of Timothy syndrome.

Timothy syndrome (TS) is a rare genetic disorder caused by mutations in the CACNA1C gene, which encodes the L-type calcium channel α1 CaV1.2 subunit. While it is expressed throughout the body, the most serious symptoms are cardiac and neurological. Classical TS type 1 (TS1) and TS type 2 (TS2) mutations cause prolonged action potentials (APs) in cardiomyocytes and in induced neurons derived from pluripotent stem cells taken from TS patients, but the effects of TS mutations on neuronal function in vivo are not fully understood. TS is frequently associated with autistic traits, which in turn hav

Structural changes to primary visual cortex in the congenital absence of cone input in achromatopsia.

Autosomal recessive Achromatopsia (ACHM) is a rare inherited disorder associated with dysfunctional cone photoreceptors resulting in a congenital absence of cone input to visual cortex. This might lead to distinct changes in cortical architecture with a negative impact on the success of gene augmentation therapies. To investigate the status of the visual cortex in these patients, we performed a multi-centre study focusing on the cortical structure of regions that normally receive predominantly cone input. Using high-resolution T1-weighted MRI scans and surface-based morphometry, we compared co

Topographic organization of human visual areas in the absence of input from primary cortex.

Recently, there has been evidence for considerable plasticity in primary sensory areas of adult cortex. In this study, we asked to what extent topographical maps in human extrastriate areas reorganize after damage to a portion of primary visual (striate) cortex, V1. Functional magnetic resonance imaging signals were measured in a subject (G.Y.) with a large calcarine lesion that includes most of primary visual cortex but spares the foveal representation. When foveal stimulation was present, intact cortex in the lesioned occipital lobe exhibited conventional retinotopic organization. Several vi

Functional effects of unilateral open-angle glaucoma on the primary and extrastriate visual cortex.

The purpose of this study was to use functional magnetic resonance imaging (fMRI) to investigate the response of the visual cortex to unilateral primary open-angle glaucoma (POAG). Specifically, we assessed whether regions of V1 and V2 with lost input from the glaucomatous eye had a greater response to input from the nonaffected fellow eye. Nine participants with unilateral POAG causing paracentral visual field defects and four controls participated in the study. We found no evidence for an increased response to the fellow eye in glaucoma-affected regions of the visual cortex; however, in agre

Global motion perception deficits in autism are reflected as early as primary visual cortex.

Individuals with autism are often characterized as 'seeing the trees, but not the forest'-attuned to individual details in the visual world at the expense of the global percept they compose. Here, we tested the extent to which global processing deficits in autism reflect impairments in (i) primary visual processing; or (ii) decision-formation, using an archetypal example of global perception, coherent motion perception. In an event-related functional MRI experiment, 43 intelligence quotient and age-matched male participants (21 with autism, age range 15-27 years) performed a series of coherent

Cortical and subcortical projections to primary visual cortex in anophthalmic, enucleated and sighted mice.

The purpose of this study was to identify and compare the afferent projections to the primary visual cortex in intact and enucleated C57BL/6 mice and in ZRDCT/An anophthalmic mice. Early loss of sensory-driven activity in blind subjects can lead to activations of the primary visual cortex by haptic or auditory stimuli. This intermodal activation following the onset of blindness is believed to arise through either unmasking of already present cortical connections, sprouting of novel cortical connections or enhancement of intermodal cortical connections. Studies in humans have similarly demonstr

Expression of inflammatory genes in the primary visual cortex of late-stage Alzheimer's disease.

Alzheimer's disease is associated with progressively dysfunctional gene expression in the limbic system of the brain. The thalamus and primary visual cortex are thought to be initially spared of Alzheimer-type changes that ravage the association neocortex. In this study, using DNA arrays and Western immunoassay, gene expression patterns were examined in the thalamus and primary visual cortex of moderate-stage and late-stage Alzheimer's disease and age-matched controls using a set of proinflammatory genes known to be upregulated in the temporal lobe neocortex and hippocampus of moderate-stage A
