Heavy metal-induced cytotoxicity to cultured human epidermal keratinocytes and effects of antioxidants.

Human epidermal keratinocytes which have been cultured were treated with the heavy metal ions of cadmium, mercury, copper and zinc. Cytotoxicity was measured either by protein estimation or by using the neutral red assay. Antioxidants were added in order to find out whether heavy metal-induced cytotoxicity is related to oxidative stress. All metals used showed considerable cytotoxic effects within 24 h in moderate concentrations. None of the antioxidants vitamin E (alpha-tocopherol), pyrogallol, propyl gallate, BHT or ebselen showed any protective or preventive effect. This indicates that oxid

β-cyclodextrin inclusion complexes with short-chain phenolipids: An effective formulation for the dual sustained-release of phenolic compounds.

The β-cyclodextrin and short-chain alkyl gallates (A-GAs), which are representative of phenolipids, such as butyl, propyl, ethyl, and methyl gallates, were chosen to form inclusion complexes by the use of the freeze-drying process. In the everted rat gut sac model, HPLC-UV analysis demonstrated that the released A-GAs from inclusion complexes were degraded to yield free gallic acid (GA) (sustained-release function 1). The small intestine membrane may be crossed by both the GA and the A-GAs. A-GAs may also undergo hydrolysis to provide GA (sustained-release function 2) following transmembrane t

Free radicals and lipid peroxidation in ethanol- or aspirin-induced gastric mucosal injury.

In this study the role of free radicals and lipid peroxidation as mediators of chemically induced mucosal damage was investigated. Two enzymatic antioxidants, superoxide dismutase or catalase injected intravenously, reduced mucosal damage either by ethanol or aspirin. Of six nonenzymatic antioxidants, given in a wide dose range subcutaneously 30 min before intragastric administration of absolute ethanol, only propyl gallate decreased mucosal damage, while four of the antioxidants tested against aspirin were protective. These nonenzymatic antioxidants were antisecretory in the pylorus-ligated r

Ascorbate-induced lipid peroxidation and the binding of [3H]dihydroalprenolol.

Sodium ascorbate addition caused a modest loss in the specific binding of [3H]dihydroalprenolol ([3H]DHA) in a rat cortical membrane preparation. Sodium ascorbate also caused a modest increase in lipid peroxide formation in the membrane preparation. Both the lipid peroxidation and the loss in [3H]DHA binding were greatly potentiated by the addition of low levels of iron (Fe2+). A kinetic analysis indicated that the iron/ascorbate caused a loss in the number of [3H]DHA binding sites but had no effect on the affinity of [3H]DHA for the binding site. Both the lipid peroxidation and the loss of [3

A novel differential ratiometric molecularly imprinted electrochemical sensor for determination of sulfadiazine in food samples.

Herein, we report a novel differential ratiometric molecularly imprinted polymer (MIP) electrochemical sensor for sulfadiazine (SDZ). An MIP membrane with double templates, SDZ and propyl gallate (PG), was fabricated on glassy carbon electrode (GCE) modified by CuInS2/ZnS nanocomposites. After adding PG in the samples as the reference, the current differences between MIP@CuInS2/ZnS/GCE and non-imprinted polymer@CuInS2/ZnS/GCE at the potentials of 0.18 V (ΔIPG) and 0.92 V (ΔISDZ) were measured. The ratio of ΔISDZ/ΔIPG was used for SDZ determination in the differential and ratiometric dual-mode.

Methylmercury-induced reactive oxygen species formation in neonatal cerebral astrocytic cultures is attenuated by antioxidants.

Excessive generation of reactive oxygen species (ROS) has been suggested as a causal factor in various neurodegenerative disorders, such as Parkinson's disease and Alzheimer's disease [Brain Res. 830 (1999) 10-15; Biochem. J. 310 (1995) 83-90; Free Radic. Biol. Med. 27 (1999) 612-616]. The present work examined the role of ROS in the neurotoxicity of methylmercury (MeHg). ROS formation in primary astrocytic cultures of neonatal rat cerebral cortex was monitored by 2',7'-dichlorodihydrofluorescein diacetate (H(2)DCF-DA) fluorescence. MeHg, at 10 and 20 microM caused a significant increase in RO

Development of a sonodynamic approach for Listeria monocytogenes inactivation and quality preservation in strawberries.

This study developed an ultrasonically activated sonosensitizer composite, curcumin-lecithin-propyl gallate-zein (CLPGZ) nanoparticles, evaluated its bactericidal mechanism of Listeria monocytogenes and its potential in food preservation applications. The synthesized CLPGZ nanoparticles were characterized for their physicochemical properties, including particle-size distribution, storage stability, and antioxidant activity. In conjunction with low-frequency ultrasound treatment, CLPGZ effectively induced the generation of reactive oxygen species (ROS) via sonodynamic effects, leading to bacter

Involvement of cyanide-resistant respiration in cell-type proportioning during Dictyostelium development.

Involvement of cyanide (CN)-resistant respiration in cell-type proportioning was analyzed using the developmental system of Dictyostelium discoideum. When migrating slugs were vitally stained with rhodamine 123, which is known to stain actively respirating mitochondria coupled with an elevated electronic potential of the inner membrane, the posterior prespore region was stained more strongly than the anterior prestalk region. Application of benzohydroxamic acid (BHAM) and propyl gallate, specific inhibitors of CN-resistant respiration, to starved Dictyostelium cells induced formation of unique

Modulatory effect of glutathione status and antioxidants on methylmercury-induced free radical formation in primary cultures of cerebral astrocytes.

Excessive free radical formation has been implicated as one of the causative factors in neurotoxic damage associated with variety of metals, including methylmercury (MeHg). Although the mechanism(s) associated with MeHg-dependent neurotoxicity remains far from clear, overwhelming data give credence to a mediatory role for astrocytes, a major cell type that preferentially accumulates MeHg. To extend our recent findings of MeHg-induced increase in ROS formation (G. Shanker, J.L. Aschner, T. Acute toxicities to typical aquatic organisms of synthetic phenolic antioxidants and QSAR study: The vital role of water solubility.

In this paper, the acute toxicities to Scenedesmus obliquus and Danio rerio of 22 Synthetic phenolic antioxidants (SPAs) were investigated in detail. The half-effective concentrations (EC50) of 22 SPAs and half-lethal concentrations (LC50) of 14 SPAs were calculated by the corresponding toxicity effect curves. Results showed that except for 2,4-di-tert-butyl-1,4-benzoquinone (BHT-Q), an important intermediate of 2,6-di-tert-butyl-4-methylphenol (BHT), 4-tert-octylphenol (4-tOP) and dodecyl gallate (DG) with the relative EC50 values of 2.35 and 2.53 mg/L were the most toxic among 22 SPAs. 4-tOP

Combinations of Terminalia bellirica (Gaertn.) Roxb. and Terminalia chebula Retz. Extracts with Selected Antibiotics Against Antibiotic-Resistant Bacteria: Bioactivity and Phytochemistry.

Antimicrobial resistance (AMR) has arisen due to antibiotic overuse and misuse. Antibiotic resistance renders standard treatments less effective, making it difficult to control some infections, thereby increasing morbidity and mortality. Medicinal plants are attracting increased interest as antibiotics lose efficacy. This study evaluates the antibacterial activity of solvent extracts prepared using Terminalia bellirica and Terminalia chebula fruit against six bacterial pathogens using disc diffusion and broth microdilution assays. The aqueous and methanol extracts of T. bellirica and T. chebul

Distinct characteristic compounds and preventive effects on ulcerative colitis of different grades of Keemun black tea.

Keemun black tea (KBT), a classic Chinese black tea with exquisite quality, was traditionally classified into seven grades. However, the chemical profiles and characteristic metabolites between the grades have not been investigated, and the grades-associated health benefits remain unexplored. In this study, metabolomics data revealed that premium grades P1 and P2, high grades T1, T2 and T3 exhibited very similar chemical profiles and clustered together, respectively. Low grades T4 and T5 formed a separate cluster. Therefore, P1 and P2 can merge into a new Premium grade; T1, T2, and T3 combine

[Possible clinical application of SOD and free radical scavengers].

Ischemia-reperfusion injury in various organs has been discussed in connection with reactive oxygen species (ROS). Xanthine oxidase (XOD) has been believed to be the source of O2- to produce this injury of dogs and rats but XOD is not detected in hearts of men, pigs or rabbits. This suggests the importance of O2- produced by leukocyte NAD(P)H oxidase. We demonstrated in 1976 that 3 injections of Cu, Zn-SOD (superoxide dismutase) (i.v.) suppressed rat carrageenan paw edema. McCord succeeded with a single injection in the same model using polyethyleneglycol (PEG-SOD) of long retention time in th

Effects of Supplemental Antioxidative Substances on Micronutrient Retention and Antioxidative Capacity in Rapeseed Oil During Low-Temperature Ethanol Steam Deodorization.

This study investigated the effects of four antioxidative substances, α-tocopherol (α-TP), phytosterols (PS), squalene (SQ), and propyl gallate (PG), on micronutrient retention and antioxidative capacity of rapeseed oil during ethanol steam deodorization (ESD, 140-220 °C). Results demonstrated that supplementation with these antioxidants increased the retention rates (percentage relative to initial content) of tocopherols, phenols, carotenoids, phytosterols, and squalene by up to 2.92%, 46.25%, 25.82%, 1.03%, and 60.15%, respectively. These improvements are attributed to the protective effects

Studies on survival of bacteria: rhythmic response of microorganisms to freeze-drying additives.

Various materials were mixed with suspensions of Serratia marcescens and other organisms. Samples were removed and frozen at intervals after mixing; the number of cells that survived both freeze-drying and exposure to air varied rhythmically as a function of time between mixing and freezing. When assayed before or immediately after drying there were essentially no fluctuations. The response was evident only when these dried samples were exposed to air. In a typical experiment, the number of cells surviving in the sample frozen 30 sec after adding propyl gallate was at least 10 times that in sa

Development of an orally delivered GLP-1 receptor agonist through peptide engineering and drug delivery to treat chronic disease.

Peptide therapeutics are increasingly used in the treatment of disease, but their administration by injection reduces patient compliance and convenience, especially for chronic diseases. Thus, oral administration of a peptide therapeutic represents a significant advance in medicine, but is challenged by gastrointestinal instability and ineffective uptake into the circulation. Here, we have used glucagon-like peptide-1 (GLP-1) as a model peptide therapeutic for treating obesity-linked type 2 diabetes, a common chronic disease. We describe a comprehensive multidisciplinary approach leading to th

Characterisation of QoI-resistant field isolates of Botrytis cinerea from citrus and strawberry.

BACKGROUND: In 2004, field isolates of Botrytis cinerea Pers. ex Fr., resistant to strobilurin fungicides (QoIs), were first found in commercial citrus orchards in Wakayama Prefecture, Japan. Subsequently, QoI-resistant isolates of this fungus were also detected in plastic strawberry greenhouses in Saga, Ibaraki and Chiba prefectures, Japan. Biological and molecular characterisation of resistant isolates was conducted in this study. RESULTS: QoI-resistant isolates of B. cinerea grew well on PDA plates containing kresoxim-methyl or azoxystrobin at 1 mg L(-1), supplemented with 1 mM of n-propyl
