Long-term effects of prostaglandin analogues on the anterior chamber depth of patients with primary open-angle glaucoma.

OBJECTIVE: To evaluate the effect of long-term application of bimatoprost and latanoprost on the anterior chamber depth of primary open-angle glaucoma patients. METHODS: The anterior chamber depth (ACD) and axial length (AL) of patients using prostaglandin analogues for open-angle glaucoma (group I) and an age- and gender-adjusted control group (group II) were measured by ultrasonography. Patients using bimatoprost and those using latanoprost were also compared in terms of ACD and AL. RESULTS: The mean ACD of group I (27 eyes of 27 subjects) was significantly lower than that of group II (30

The role of prostaglandins in blastocyst implantation.

This review summarizes the information available on the involvement of prostaglandins in blastocyst implantation, and examines their interactions with three other inflammatory mediators, platelet-activating factor (PAF), interleukin (IL-1) and corticotrophin-releasing factor (CRF). Essential elements of this information, consistent with the assumption that prostaglandins play an important role in implantation, appear to be: (i) the burst of endometrial prostaglandin production, following the blastocyst signal(s) or an artificial stimulus; (ii) the main localization of this production at the lu

[Effects of acute changes in water balance on hydrosaluresis and on urinary excretion of prostaglandin E, in the presence and absence of a cyclooxygenase inhibitor].

Five healthy females were studied in order to evaluate the effects of a cyclo-oxygenase inhibitor agent (Indomethacin, I) on the hydro-saluresis and urinary excretion of prostaglandins (PG) of E series. Each subject was studied during both hypotonic polyuria (oral water load) and hypertonic olyguria (18 h water intake withheld). 1) In water diuresis I. treatment significantly (P less than 0.05) decreased urinary flow (36%) as well as sodium and potassium excretions (32%, 46%, respectively); the rise in urinary osmolarity (86%) and the fall in PGE excretion (63%) were statistically insignifican

Gastroprotections of escins Ia, Ib, IIa, and IIb on ethanol-induced gastric mucosal lesions in rats.

Effects of escins Ia, Ib, IIa, and IIb isolated from horse chestnuts on ethanol-induced gastric mucosal lesions and the roles of capsaicin-sensitive afferent neurons, endogenous nitric oxide (NO), sulfhydryls, prostaglandins, as well as gastric secretion and the sympathetic nervous system, were investigated in rats. Test samples were given orally to fasted rats 1 h before ethanol (1.5 ml/rat, p.o.) treatment or ligation of the pylorus. Escins Ia-IIb (10-50 mg/kg) potently inhibited ethanol-induced gastric mucosal lesions, whereas desacylescins I and II (50 mg/kg) showed no such effect. These a

Structural and functional characterization of striated ducts isolated from the rabbit mandibular salivary gland.

Ductal elements within salivary glands are responsible for modifying the electrolyte composition of primary saliva secreted by the acini. To study the mechanism and regulation of the transport processes involved requires a suitable preparation of functional ducts. To this end we have isolated intralobular ducts from rabbit mandibular salivary glands using the technique of tissue dissociation and microdissection. Light and electron microscopy demonstrated that the ducts corresponded ultrastructurally to striated intralobular ducts of the intact gland. Ducts could be maintained in tissue culture

Reversal by acetylsalicylic acid of the captopril-induced inhibition of angiotensin converting enzyme in the hindquarters of guinea-pig.

The conversion of angiotensin I to angiotensin II was studied in the isolated perfused hindquarters of guinea-pig. The relative enzyme activity was determined by vasoconstrictor response to the peptides and by the contraction of rat ascending colon superfused with the venous effluent. 45% of conversion of angiotensin I to angiotensin II was determined in this preparation as measured by vasoconstrictor responses. However, only 22% of conversion was detected in the venous return as measured in the rat colon. Captopril significantly inhibited angiotensin converting enzyme activity in this prepara

The role of anesthesia and catecholamines in the renal response to mild hemorrhage.

The role of the sympathoadrenal system in the renal response to mild hemorrhage was investigated in animals anesthetized with either sodium pentobarbital (Group I) or alpha-chloralose (Group II). Significantly greater decreases in renal blood flow (RBF) and increases in renal vascular resistance (RVR) were seen after hemorrhage in Group II than in Group I. Concomitant with these changes were increases both in circulating catecholamines and plasma renin activities. In Group II, however, the rise in plasma epinephrine was also significantly higher than in Group I. Additional studies were perform

Mechanisms of mucosal injury and healing: the role of non-steroidal anti-inflammatory drugs.

Biological insights into injurious effects of non-steroidal anti-inflammatory drugs (NSAIDs), including aspirin (ASA), on mucosal protection and repair, are largely from studies of acute injury. That chronic ulceration is similar is not established. NSAIDs directly injure tissues, including endothelia, and at the same time impair the operation of many of the processes that normally contribute to mucosal protection, whatever the injurious agent. Many protective processes are mediated through prostaglandins, whose synthesis is abolished by inhibition of the constitutive isoenzyme, cyclooxygenase

Des-aspartate-angiotensin I, a novel angiotensin AT(1) receptor drug.

The review describes DAA-I (des-aspartate-angiotensin-I) as a prototype of a novel class of drugs that acts as agonists on the angiotensin AT1 receptor or ARAs (angiotensin receptor agonists). DAA-I is a component of the renin angiotensin system. Earlier studies showed that it was rapidly metabolized to angiotensin III. However, when administered at doses below the Km of enzymes, DAA-I produces specific actions that antagonize the deleterious actions of angiotensin II. DAA-I exerts protective actions in animal models of eight human pathologies in which angiotensin II is implicated. The patholo

Neuroprotective effects of indomethacin and aminoguanidine in the newborn rats with hypoxic-ischemic cerebral injury.

Nitric oxide (NO) and prostaglandins (PG) play important roles in delayed mechanisms of brain injury. While NO disrupts oxidative metabolism, prostaglandins are responsible for free radical attack in reperfusion interval. Relatively little is known about neuroprotection exerted at this level in perinatal models. The aim of this study was to investigate the effect of indomethacin and aminoguanidine on endogenous inducible nitric oxide synthase (iNOS) biosynthesis and neuroprotection in the newborn rats with hypoxic ischemic cerebral injury.Seven-day old rat pups with model of hypoxic-ischemic c

[Prostaglandin analogues and central corneal thickness].

UNLABELLED: Prostaglandin analogs reduce intraocular pressure (IOP) by activation of matrixmetalloproteinase enzymes (MMP) and increasing uveoscleral outflow. MMP's were found in cornea and their activation could change the central corneal thickness (CCT). PURPOSE: To compare the effects of different prostaglandin analogues on central corneal thickness (CCT). METHODS: Clinical, observational, prospectively and randomised study during 3 month on 26 patients (52 eyes) newly diagnosed with primary open angle glaucoma or ocular hypertension. The patients were distributed in two groups concerning

Role of prostaglandins in androgenetic alopecia.

Androgenetic alopecia (AGA) is the most prevalent progressive hair loss disorder worldwide, and its pathogenesis is closely linked to dysregulated androgen metabolism. Conventional therapies for AGA have limited efficacy and adverse reactions. Prostaglandins (PGs), a class of lipid signaling molecules, have recently been implicated in AGA development. This review systematically examines the association of PGs with key pathogenic mechanisms of AGA, including androgen pathway dysregulation, immune response, inflammation, vascular microenvironment disturbances, and lipid metabolism disorders. Effect of latanoprost on the extracellular matrix of the ciliary muscle. A study on cultured cells and tissue sections.

Prostaglandin F2alpha and its analogue latanoprost, both prostanoid FP receptor agonists, reduce the intraocular pressure mainly by enhancing uveoscleral outflow. Changes in the extracellular matrix of the ciliary muscle may be involved in the increased outflow. The effect of latanoprost and prostaglandin F2alpha on the extracellular matrix of the ciliary muscle was investigated. Cell cultures of human ciliary muscle were treated with latanoprost acid or prostaglandin F2alpha for 1-2 days and were immunostained against various extracellular matrix components and metalloproteinases. Dengue virus co-opts innate type 2 pathways to escape early control of viral replication.

Mast cell products and high levels of type 2 cytokines are associated with severe dengue disease. Group 2 innate lymphoid cells (ILC2) are type-2 cytokine-producing cells that are activated by epithelial cytokines and mast cell-derived lipid mediators. Through ex vivo RNAseq analysis, we observed that ILC2 are activated during acute dengue viral infection, and show an impaired type I-IFN signature in severe disease. We observed that circulating ILC2 are permissive for dengue virus infection in vivo and in vitro, particularly when activated through prostaglandin D2 (PGD2). ILC2 underwent produc

PGE-metabolite levels in CSF correlate to HIE score and outcome after perinatal asphyxia.

AIM: Acute anoxic exposure rapidly increases prostaglandin E2 (PGE2 ) production and release in neonatal mice brains. We hypothesize that PGE2 is released in human cerebrospinal fluid (CSF) during perinatal asphyxia and that it might be used as a biomarker for perinatal asphyxia. METHODS: In full-term infants with lumbar puncture performed within 72 h of birth (n = 35), CSF was analysed for prostaglandin E2 metabolite (PGEM) using an enzyme immunoassay. Term infants with suspected but unverified infections were used as controls (n = 11). Hypoxic-ischaemic encephalopathy (HIE) was classified a

Oxidants, nitric oxide and prostanoids in the developing ocular vasculature: a basis for ischemic retinopathy.

The choroid is the main source of oxygen to the retina. In contrast to the adult, the absence of autoregulation of choroidal blood flow in the newborn leads to hyperoxygenation of the retina. In the immature retina which contains relatively low levels of antioxidants this hyperoxygenation favors peroxidation including the generation of biologically active isoprostanes, and results in vasoconstriction and vascular cytotoxicity leading to ischemia, which predisposes to the development of a vasoproliferative retinopathy, commonly termed retinopathy of prematurity. During frequently encountered ox

The cAMP-dependent kinase pathway does not sensitize the cloned vanilloid receptor type 1 expressed in xenopus oocytes or Aplysia neurons.

Capsaicin-activated channels present in sensory neurons are ligand-gated cation channels that largely account for mediating some types of pain. The cAMP-dependent protein kinase (PKA) signal pathway was suggested to mediate the prostaglandin-induced enhancement of capsaicin-evoked inward current (I(CAP)) in rat sensory neurons. It is not clear, however, whether PKA acts directly on the capsaicin-sensitive channel that is responsible for I(CAP). To address this issue, we overexpressed the cloned capsaicin receptor, VR1, in heterologous expression systems such as Xenopus oocytes or Aplysia R2 ne
