Neuronanomedicine harnesses nanoparticle technology for the treatment of neurological disorders. An unavoidable consequence of nanoparticle delivery to biological systems is the formation of a protein corona on the nanoparticle surface. Despite the well-established influence of the protein corona on nanoparticle behavior and fate, as well as FDA approval of neuro-targeted nanotherapeutics, the effect of a physiologically relevant protein corona on nanoparticle-brain cell interactions is insufficiently explored. Indeed, less than 1% of protein corona studies have investigated protein coronas fo

The formation of a Protein Corona (PC) on the surface of nanoparticles (NPs) is a critical event that shapes their biological identity and governs interactions with the immune system. In this study, we investigated the composition of the PC formed on mixtures of PLGA and PEG-PLGA NPs, aiming to elucidate the link between NPsurface chemistry, proteomic fingerprint in cell culture medium, and uptake by bone marrow-derived macrophages (BMDMs). NPs showed different sizes but comparable actual PEG amount exposed on the surface, which is significantly lower than the theoretical values. The PC, isola

The protein corona is a layer of biomolecules-primarily proteins-that adsorbs to nanoparticle (NP) surfaces in biological fluids. If the purpose of the NP is therapeutic, this can have a profound effect on its biological activity and function in vivo. Protein corona formation can also be exploited for diagnostic purposes and to differentially enrich proteins for biomarker discovery. For all of these applications, it is useful to determine which proteins, and which specific proteoforms, bind to different types of NP. The traditional mass spectrometry (MS)-based bottom-up proteomics does not acc

Cholesterol is a key determinant of membrane stability and fluidity in both natural and synthetic lipid systems. Although widely used, its content varies greatly among commercial liposomes, and its regulatory effects on biological interactions remain insufficiently understood. Here, we reveal that cholesterol content critically governs the protein corona composition and binding affinity of liposomes, thereby dictating their cellular interactions and in vivo fate. Complement proteins exhibit a biphasic adsorption pattern that modulates monocyte and Kupffer cell uptake and systemic clearance. Hi

DNA nanomachines have significant potential for biosensing because of their high programmability, signal amplification ability, and excellent biocompatibility. However, under physiological conditions, their performance is often hindered by insufficient endogenous driving forces, nuclease degradation, and lysosomal entrapment. In this study, we develop a protein corona (PC)-armored DNA nanomachine with spatiotemporal control for multiple intracellular biomarker imaging. By pre-assembling a UV-responsive PC shell, the DNA nanomachine forms a physical barrier that protects nucleic acids from nucl

Most nanoparticles approved for medical applications are liposome formulations. In the environment of biological fluids, the corona protein was found to alter both the surface of the liposome and its size and zeta potential. As a result, it can also affect the stability of the carrier and its ability to hold a drug and release it sustainably into the cell. The key element of novelty of this research is the introduction of the Langmuir-Blodgett technique together with imaging microscopy to study the interactions between the monolayer corresponding to one liposome leaflet and the corona protein.

The dynamic protein corona formed on nanocarriers has been revealed to strongly affect their in vivo behaviors. Precisely manipulating the formation of protein corona on nanocarriers may provide an alternative impetus for specific drug delivery. Herein, we explore the role of glycosylated polyhydroxy polymer-modified nanovesicles (CP-LVs) with different amino/hydroxyl ratios in protein corona formation and evolution. CP-LVs with an amino/hydroxyl ratio of approximately 0.4 (CP1-LVs) are found to efficiently suppress immunoglobulin adsorption in blood and livers, resulting in prolonged circulat

Oral delivery of peptide/protein drugs has attracted worldwide attention due to its good patient compliance and convenience of administration. Orally administered nanocarriers always encounter the rigorous defenses of the gastrointestinal tract, which mainly consist of mucus and epithelium barriers. However, diametrically opposite surface properties of nanocarriers are required for good mucus penetration and high epithelial uptake. Here, bovine serum albumin (BSA) is adsorbed to cationic liposomes (CLs) to form protein corona liposomes (PcCLs). The aim of using PcCLs is to conquer the mucus an

Nanoparticles (NPs) have been widely investigated for enhancing the stability, bioavailability, and functionality of bioactive compounds, as well as for broadening their application in biomedical and food-related fields. Upon exposure to biological environments, proteins spontaneously adsorb onto the surface of NPs and form dynamic protein corona (PC), which determine the interfacial properties and influence the biological behavior and fate of NPs. This review summarizes recent advances in the formation mechanisms, influencing factors, characterization strategies, and biological functions of P

Lipid nanoparticles (LNPs) preferentially accumulate in the liver, but hepatic tropism alone does not ensure productive RNA interference. Here, we screened a library of LNPs in which lipid composition was varied to generate distinct protein corona identities and identify formulations capable of combining liver accumulation with functional siRNA delivery. LNP1 emerged as the lead formulation, showing favorable physicochemical properties, high transfection efficiency, and enhanced hepatic accumulation. Proteomic analysis associated this behavior with a distinct multicomponent corona signature. W

Cardiovascular diseases (CVDs), the primary cause of global mortality, face persistent therapeutic challenges due to poor drug targeting and systemic side effects. While innovative nanotherapeutic approaches offer improved drug delivery through targeting and stimulus-responsive release mechanisms, clinical translation is limited by poor tissue specificity and manufacturing limitations. We present a universal therapeutic hypothermia (TH, i.e., 32 to 35 °C)-enhanced strategy that amplifies nanocarrier targeting across multiple CVD models. Both hard and soft nanocarriers showed 1.6- to 3-fold inc

Nanocarriers entering the body are usually coated by plasma protein, leading to a protein "corona" easily recognized by tissues and cells. Adjusting the composition of protein coronas may be an efficient way to change the properties and behavior of nanoparticles in vivo. In this study, we modified doxorubicin-loaded liposomes (Lip/DOX) with an albumin-binding domain (ABD) to prepare nanoparticles (ABD-Lip/DOX) that can specifically bind to albumin and form albumin-based protein coronas in vivo for targeted tumor therapy. The prepared liposomes were spherical with a particle size of about 100 n

Chitosan is a polymer that is extensively used to prepare nanoparticles (NPs) with tailored properties for applications in many fields of human activities. Among them, targeted drug delivery, especially when cancer therapy is the main interest, is a major application of chitosan-based NPs. Due to its positive charges, chitosan is used to produce the core of the NPs or to cover NPs made from other types of polymers, both strategies aiming to protect the carried drug until NPs reach the target sites and to facilitate the uptake and drug delivery into these cells. A major challenge in the design

Signature-based protein detection coupled with machine learning algorithms has revolutionized traditional sensing methods, providing rapid, inexpensive, and selectivity-driven detection without the use of specialized equipment. This strategy leverages selective interactions with a sensor array to create a global signature pattern using machine learning. The reference pattern provides an effective tool to stratify the analytes, identify blinded unknowns, and predict altered signatures of the analyte that could be missed by the common specificity-based sensors. Protein corona adsorbed on a nanop

Plasma proteins (human serum albumin, immunoglobulin gamma-1, complement C3, and fibrinogen) are simulated with 26 and 36 nm-sized cationic, anionic, and zwitterionic liposomes in water using coarse-grained models. Proteins, initially placed randomly around a liposome, adsorb more significantly onto charged liposomes than onto zwitterionic liposomes through electrostatic interactions with lipid headgroups and hydrophobic interactions with lipid tails, with the greatest adsorption observed on cationic liposomes, in agreement with experimental observations. For zwitterionic liposomes, adsorption

After intravenous administration of nanocarriers, plasma proteins may rapidly adsorb onto their surfaces. This process hampers the prediction of the nanocarriers' pharmacokinetics as it determines their physiological identity in a complex biological environment. Toward clinical translation it is therefore an essential prerequisite to investigate the nanocarriers' interaction with plasma proteins. Here, this work evaluates a highly "PEGylated" squaric ester-based nanogel with inherent prolonged blood circulation properties. After incubation with human blood plasma, the nanogels are isolated by

Cardiovascular disease continues to be a leading cause of mortality worldwide, but the mechanistic interplay between hypercholesterolemia and environmental pollutants in the progression of atherosclerosis remains poorly understood. This study investigated the role of silver nanoparticles (AgNPs), a typical particulate contaminant, in promoting atherogenic risk under dyslipidemic conditions. Elevated serum cholesterol levels significantly promoted the adsorption of low-molecular-weight hydrophobic proteins to AgNPs. The hyperlipidemia-specific PC facilitated macrophage uptake of AgNPs through l

Protein corona formation is widely viewed as an inevitable but poorly controlled event when nanoparticles encounter biological environments. Converting this interfacial layer into a programmable functional entityremains a major challenge. Here, we report an adaptive protein corona nanoassembly strategy that integrates cytokine signaling with endogenous antigen transport. Coordination-engineered manganese nanoscaffolds were assembled with interleukin-12 (IL-12) and protected by an enzyme-responsive hyaluronic acid shell to form nanoshuttles (NSs) capable of dynamic protein recruitment in the tu

Lipid nanoparticles (Lips) are studied as drug delivery systems for superior biocompatibility, low toxicity, and high encapsulation efficiency. Their size, charge, and hydrophilicity are tunable via material composition and functional groups for tailored tissue distribution. In biological environments, Lip adsorbs proteins to form a "protein corona (PC)," which alters its physicochemical property and biological interaction. Nevertheless, the mechanisms of PC regulating Lip-tissue targeting remain unclear, with limitations of inaccurate in vivo fluorescence quantification and poor stability. To
