Engineering and Adapting Disulfide-Containing Proteins to Enable Intracellular Functionality.

Disulfide bonds are key structural components ensuring proper protein folding. Those bonds, however, do not form in the reducing intracellular environment, often resulting in misfolded, unstable proteins with subpar function. To enable intracellular functionality of such proteins, disulfide-free variants with restored stability and function should be engineered. Here, we present a method to substitute the cysteine residues with optimal amino acid combinations using site saturation mutagenesis and screening for improved disulfide-free variants using yeast surface display. We provide a detailed

Engineered Lysosome-Targeted Protein Degradation via Hijacking Macropinocytosis.

Recently, targeted protein degradation (TPD) strategies have emerged as an effective tool for addressing undruggable targets in both biomedical research and the pharmaceutical industry, selectively binding proteins of interest and targeting them to the intracellular degradation machinery for degradation. However, the targeting of extracellular proteins with current degradation tools requires a tedious, case-specific selection and design process based on lysosomal trafficking of cell surface receptors. Here, we introduce Macropinocytosis-Targeting Chimeras (MapTACs), a TPD platform that exploit

Alimentary 'green' proteins as electrospun scaffolds for skin regenerative engineering.

As a potential alternative to currently available skin substitutes and wound dressings, we explored the use of bioactive scaffolds made of plant-derived proteins. We hypothesized that 'green' materials, derived from renewable and biodegradable natural sources, may confer bioactive properties to enhance wound healing and tissue regeneration. We optimized and characterized fibrous scaffolds electrospun from soy protein isolate (SPI) with addition of 0.05% poly(ethylene oxide) (PEO) dissolved in 1,1,1,3,3,3-hexafluoro-2-propanol, and from corn zein dissolved in glacial acetic acid. Engineering of a polymeric bacterial protein as a scaffold for the multiple display of peptides.

Protein assemblies with a high degree of repetitiveness and organization are known to induce strong immune responses. For that reason they have been postulated for the design of subunit vaccines by means of protein engineering. The enzyme lumazine synthase from Brucella spp. (BLS) is highly immunogenic, presumably owing to its homodecameric arrangement and remarkable thermodynamic stability. Structural analysis has shown that it is possible to insert foreign peptides at the ten amino terminus of BLS without disrupting its general folding. These peptides would be displayed to the immune system

[Evaluation of penicilloyl proteins of allergic impurity in gene engineering drugs].

It is well known that penicillins are necessary for prevention of contamination during cell culture or fermentation in the production of gene engineering drugs. As penicillins are easily combined with proteins forming allergic impurities, the benzyl penicilloyl proteins in aqueous solution, an evaluation system should be established for control of the gene engineering drugs. An enzyme linked immunosorbent assay (ELISA) with high specificity and high sensitivity in vitro, by which the conjugated penicillins of 0.3 ppm in a sample could be detected and a passive cutaneous anaphylaxis (PCA) test,

Unleashing the potential of S-layer proteins for engineered living materials.

The field of engineered living materials (ELMs) aims to create self-regenerative, self-assembled, and multifunctional materials that mimic natural biomaterials. Novel ELMs can be produced by engineering biomolecules that are naturally secreted and displayed on bacterial cell surfaces. Surface-layer (S-layer) proteins are a class of proteins that form a two-dimensional paracrystalline lattice on the surface of many prokaryotes. These proteins provide a secretion, surface-anchoring, and high-density display platform that can be exploited for material formation. In this review, we discuss two str

Cell Engineering for Increasing Production of Recombinant Proteins in Mammalian Cells.

Mammalian cell lines are the preferred host cells for the biopharmaceutical industry. Chinese hamster ovary (CHO) and human embryonic kidney 293 (HEK293) cells are frequently utilized in the production of recombinant therapeutic proteins (RTPs) owing to their capacity to facilitate appropriate protein folding and perform accurate post-translational modifications (PTMs). However, there are still some bottlenecks in the biopharmaceutical process using mammalian cells, including lower productivity, higher production cost and increased risk of contamination compared with bacterial or yeast express

Prevention of influenza by targeting host receptors using engineered proteins.

There is a need for new approaches for the control of influenza given the burden caused by annual seasonal outbreaks, the emergence of viruses with pandemic potential, and the development of resistance to current antiviral drugs. We show that multivalent biologics, engineered using carbohydrate-binding modules specific for sialic acid, mask the cell-surface receptor recognized by the influenza virus and protect mice from a lethal challenge with 2009 pandemic H1N1 influenza virus. The most promising biologic protects mice when given as a single 1-μg intranasal dose 7 d in advance of viral chall

Genetically engineered protein in hydrogels tailors stimuli-responsive characteristics.

Certain proteins undergo a substantial conformational change in response to a given stimulus. This conformational change can manifest in different manners and result in an actuation, that is, catalytic or signalling event, movement, interaction with other proteins, and so on. In all cases, the sensing-actuation process of proteins is initiated by a recognition event that translates into a mechanical action. Thus, proteins are ideal components for designing new nanomaterials that are intelligent and can perform desired mechanical actions in response to target stimuli. A number of approaches hav

Developing Anti-EGFR/Anti-HER2 Bifunctional Antibody for Solid Tumors by Protein Engineering.

Overexpression of epidermal growth factor receptor (EGFR) and human epidermal growth factor receptor 2 (HER2) is common in solid tumors like breast, colorectal, and head and neck cancers, driving oncogenic signaling, therapy resistance, and poor prognosis. Monotherapies often fall short in fully suppressing tumor progression, particularly in cases with EGFR/HER2 co-amplification. Dual-targeting strategies offer enhanced efficacy by mitigating resistance and amplifying antitumor responses. Our study focused on developing and characterizing a bispecific anti-EGFR/HER2 antibody designed to simult

Protein scaffold engineering for immune checkpoint targeting, tumor microenvironment modulation, and Cancer immunotherapy.

Protein scaffolds are being developed as novel non-antibody-binding platforms to address significant challenges in traditional cancer immunotherapy, including high production costs, limited tumor penetration, immune-related toxicity, and therapeutic resistance. Scaffolds, which consist of Affibodies, DARPins, Anticalins, Monobodies (Adnectin), Knottins, and Avimers, are structurally stable, small in size, and serve as extensively modified proteins, offering higher solubility, rapid clearance from unwanted tissues, and effective recombinant production compared with monoclonal antibodies (mAbs).

Combination of Fc-Protein Engineering Strategies: Design and Evaluation of Antibody Effector Functions.

Monoclonal antibodies have revolutionized cancer therapy and are integral part of standard therapies. Despite their clinical efficacy, not all cancer patients benefit from established antibody therapies. Preclinical and clinical findings revealed that Fc-mediated effector functions play an essential role in antibody therapy. Therefore, current antibody-based immunotherapies can be optimized by antibody Fc engineering to generate next-generation monoclonal antibodies with tailor-made effector functions. The enhancement of Fc-mediated effector functions such as antibody-dependent cell-mediated c

Cell-free systems as complementary Test layers for protein engineering in biofoundry workflows.

Biofoundries have expanded the scale and design space of synthetic biology by combining laboratory automation with artificial intelligence-assisted protein engineering. As these workflows process larger numbers of variants, the Test phase becomes increasingly important: engineered proteins need to be characterized rapidly, quantitatively, and in formats that fit automated operation. Cell-free systems (CFSs) offer a practical way to meet these needs because they separate protein synthesis from cell growth and provide direct access to both the expressed proteins and the reaction environment. Protein Engineering of a Genetically Encodable Biosensor for Wastewater Detection of Profen NSAIDs.

Non-steroidal anti-inflammatory drugs (NSAIDs) are pervasive environmental contaminants due to their frequent and widespread use, multiple paths of release into surface and ground water supply, diversity of the chemical class, and toxicity to aquatic and other non-target species. In particular, the 2-arylpropionic acid ("profen") class of NSAIDs poses significant risks to aquatic ecosystems due to incomplete removal during wastewater treatment. Current monitoring precludes high-frequency testing at point sources. Here, we present the engineering and application of a genetically encodable, prot

Engineering a hybrid chitinase-sonorensin fusion protein for enhanced antibacterial and antifungal activity.

The main objective of the study was to attain improved effectiveness against various bacterial and fungal infections, tackling the widespread problem of multidrug resistance. The study presents the development and evaluation of a hybrid antimicrobial protein created by combining chitinase and sonorensin (HyChiSono). The hybrid protein HyChiSono, a 49.3 kDa chitinase-sonorensin fusion protein, was engineered, expressed in E. coli BL21(DE3), and purified to > 95 % homogeneity. Independent folding of each domain linked by a flexible (GGGGS) spacer was revealed by AlphaFold modelling; a highly ord

Robust protein-based engineering of hepatocyte-like cells from human mesenchymal stem cells.

BACKGROUND: Cells of interest can be prepared from somatic cells by forced expression of lineage-specific transcription factors, but it is required to establish a vector-free system for their clinical use. Here, we report a protein-based artificial transcription system for engineering hepatocyte-like cells from human umbilical cord-derived mesenchymal stem cells (MSCs). METHODS: MSCs were treated for 5 days with 4 artificial transcription factors (4F), which targeted hepatocyte nuclear factor (HNF)1α, HNF3γ, HNF4α, and GATA-binding protein 4 (GATA4). Then, engineered MSCs (4F-Heps) were subje

Enhanced Biomass and Protein Synthesis in Engineered Cyberlindnera jadinii Growing on Ethanol/Acetate: Metabolic Engineering and Transcriptomic Mechanism.

Producing single-cell protein (SCP) from syngas-derived ethanol and acetate offers a sustainable solution to global protein shortages, yet microbial utilization mechanisms for these mixtures remain underexplored. This study establishes a systematic bioconversion strategy using Cyberlindnera jadinii TU389. To mitigate acetaldehyde accumulation during ethanol metabolism, we engineered the strain TU546 to overexpress acylating acetaldehyde dehydrogenase (ADA6). TU546 achieved a maximum biomass of 46.7 g/L and a protein yield of 21.69 g/L, representing enhancements of 28.16% and 23.02% over the wi

Multilevel Metabolic Engineering of Cupriavidus necator for CO2-Derived Single-Cell Protein Production.

Enhancing nitrogen assimilation and coordinating carbon-energy metabolism in the autotrophic chassis Cupriavidus necator remain key challenges for maximizing single-cell protein (SCP) production from CO2. In this study, multilevel engineering was performed on C. necator. First, glnA was overexpressed to enhance nitrogen assimilation, which increased crude protein content to 72.75% in the best-performing strain, GS13. Preliminary physiological characterization suggested that this increase correlated with enhanced heme level. Second, glcR was disrupted to alter carbon allocation toward amino aci

Engineering quantum dot surfaces to preserve protein-DNA interactions for single-molecule visualization.

Quantum dots (QDs) are fluorescent nanoparticles widely used for single-molecule imaging because of their exceptional brightness and photostability. However, the impact of QD surface chemistry on biomolecular interactions has not been systematically investigated. Here, we report that commercial QDs unexpectedly destabilize protein-DNA complexes by inducing protein dissociation from DNA. Using the human nucleotide excision repair protein, xeroderma pigmentosum complementation group A (XPA) as a model system, we demonstrate that antibody-conjugated QDs promote dissociation of XPA from DNA substr
