Identification, Screening and Mechanism Analysis of Anti-Parkinson's Disease Peptides from Rapana venosa Protein Hydrolysates.

At present, there is still a lack of effective treatments to slow the progression of Parkinson's disease. Naturally derived active substances, valued for their safety and multi-target potential, have become an important direction in anti-PD drug development, with marine organisms representing a valuable source of bioactive peptides. This study aimed to isolate and identify anti-PD peptides from Rapana venosa protein hydrolysates. Through bioactivity-guided screening combined with an MPTP-induced zebrafish PD model, three novel active peptides-KSTELLI, FLVKLPMFM, and SDSLSEILIS-were successfull

Antioxidant and ACE-Inhibitory Activity of Protein Hydrolysates Produced from Atlantic Sea Cucumber (Cucumaria frondosa).

Atlantic sea cucumber is a benthic marine echinoderm found in Northwest Atlantic waters and is harvested mainly for its body wall. The body wall, along with internal organs and aquaphyrangeal bulb/flower, is a rich source of proteins, where the latter parts are often considered as processing discards. The objective of this research was to produce protein hydrolysates from sea cucumber tissues (body wall, flower, and internal organs) with bioactive properties associated with antioxidants, DNA and LDL cholesterol oxidation inhibition, and angiotensin-I-converting enzyme (ACE) inhibitory effects.

Blood glycemia-modulating effects of melanian snail protein hydrolysates in mice with type II diabetes.

Freshwater animal proteins have long been used as nutrient supplements. In this study, melanian snail (Semisulcospira libertina) protein hydrolysates (MPh) were found to exert anti-diabetic and protective effects against liver and kidney damage in mice with type II diabetes adapted to a 45% kcal high-fat diet (HFD). The hypoglycemic, hepatoprotective and nephroprotective effects of MPh were analyzed after 12 weeks of the continuous oral administration of MPh at 125, 250 and 500 mg/kg. Diabetic control mice exhibited an increase in body weight, and blood glucose and insulin levels, with a decre

Reduction of L-phenylalanine in protein hydrolysates using L-phenylalanine ammonia-lyase from Rhodosporidium toruloides.

L-Phenylalanine ammonia-lyase (PAL, EC 4.3.1.25) from Rhodosporidium toruloides was utilized to remove L-phenylalanine (L-Phe) from different commercial protein hydrolysates. A casein acid hydrolysate (CAH, L-Phe ~2.28 %) was employed as a model substrate. t-Cinnamic acid resulting from deamination of L-Phe was extracted, analyzed at λ = 290 nm, and used for PAL activity determination. Optimum reaction conditions, optimized using successive Doehlert design, were 35 mg mL(-1) of CAH and 800 mU mL(-1) of PAL, while temperature and pH were 42 °C and 8.7, respectively. Reaction kinetics of PAL wit

A Length-Aware C-Terminal Rule for Prioritizing Short ACE-Inhibitory Peptides from Food Protein Hydrolysates.

The discovery of angiotensin-converting enzyme (ACE)-inhibitory peptides from food protein hydrolysates is commonly guided by empirical fractionation or sequence-based prediction, but few screening rules have been evaluated at the hydrolysate level and independently benchmarked across peptide lengths. Here, we developed a length-aware C-terminal screening rule (Rule 5: P1' ∈ {W, Y, F, P} and P2' ∈ {L, I, V, K, R, H}) through an experimentally anchored framework. The rule was derived using 24 stratified protein-protease hydrolysates and showed the strongest associations with ACE inhibition (r =

Biochemical and functional benchmarking of plant protein hydrolysates in serum-reduced adherent CHO-K1 cell culture.

Chinese Hamster Ovary (CHO) cells are used for therapeutic protein production. Although industrial platforms rely on serum-free media, fetal bovine serum (FBS) remains common in research and development. Reducing serum while maintaining cell growth is of interest. In this study, CHO-K1 cells were adapted from 10% to 1% FBS in Ham's F-12 medium. Six plant protein hydrolysates (wheat, soy, pea, broad bean, rice, and a rice-pea blend) were biochemically characterized and evaluated for their effects on cell proliferation under serum-reduced conditions. CHO-K1 cells tolerated adaptation to 1% FBS,

Physicochemical and Antioxidative Properties of Protein Hydrolysates from Residual Goat Placenta Extract by Two Different Methods.

Protein hydrolysates from the goat placenta provide multiple benefits, such as immune system enhancement, antioxidant activities, and reductions in uric acid levels. Despite these benefits, their industrial applications have been underexplored. This study aimed to prepare extract protein hydrolysates (GPERPs) from residual goat placenta extract (GPER) and assess their functional properties, focusing on how different drying methods influence these properties. The essential amino acid contents were 30.94% for the GPER and 34.11% for the GPERPs. Moreover, all the essential amino acids were presen

Preparation and antioxidant properties of crayfish (Procambarus clarkii) By-products protein hydrolysates and ultra filtration fractions.

Protein isolate from crayfish by-products (CBPI) were hydrolyzed using Alcalase, neutrase, pancreatin and bromelain. Hydrolysis by Alcalase had more remarkable digesting efficiency on crayfish by-products protein than that by the other enzymes. Therefore, protein hydrolysate from Alcalase digestion (CBPHa) was selected to be fractionated by ultrafiltration according to molecular weight into three fractions F1 (MW <1kDa), F2 (MW 1-3kDa) and F3 (MW 3-10kDa). The amino acid determination revealed that CBPI had essential amino acid (EAA) close to that required for human protein synthesis. In vitro

Production of antioxidant and ACE-inhibitory peptides from Kluyveromyces marxianus protein hydrolysates: Purification and molecular docking.

Kluyveromyces marxianus protein hydrolysates were prepared by two different sonicated-enzymatic (trypsin and chymotrypsin) hydrolysis treatments to obtain antioxidant and ACE-inhibitory peptides. Trypsin and chymotrypsin hydrolysates obtained by 5 h, exhibited the highest antioxidant and ACE-inhibitory activities. After fractionation using ultrafiltration and reverse phase high performance liquid chromatography (RP-HPLC) techniques, two new peptides were identified. One fragment (LL-9, MW = 1180 Da) with the amino acid sequence of Leu-Pro-Glu-Ser-Val-His-Leu-Asp-Lys showed significant ACE inhi

Mutagenicity of glycerol chlorohydrines and of their esters with higher fatty acids present in protein hydrolysates.

3-chloro-1,2-propanediol and 1,3-dichloro-2-propanol caused base substitutions in Salmonella typhimurium TA1535 both with and without metabolic activation. Metabolic activation seemed to act mainly by decreasing the toxicity of these compounds. A difference in the growth of the wild-type and repair-deficient strains of Escherichia coli was observed only for 1,3-dichloro-2-propanol with S9 mix. Esters of both chlorohydrines with fatty acids has smaller mutagenic effects than unesterified compounds.

An integrated Peptidomics approach to analysing the formation mechanism of bitter taste in walnut protein hydrolysates.

Enzymatic hydrolysis enhances the functionality of walnut protein; however, the presence of bitter peptides limits its application. This study compared the hydrolytic efficiency, peptide patterns and bitterness of eight food-grade proteases, and revealed the mechanism behind the bitter taste of walnuts. Neutral protease performed best (60% degree of hydrolysis, 58% yield), producing mainly <3 kDa peptides. Trypsin yielded the most <0.5 kDa fragments, while acidic protease/trypsin released the most free amino acids. Papain, bromelain, and pepsin showed low activity, producing >3 kDa products. E

Malignant transformation of mouse M2-fibroblasts by glycerol chlorohydrines contained in protein hydrolysates and commercial food.

Glycerol chlorohydrines, such as 3-chloro-1,2-propanediol and 1,3-dichloro-2-propanol, are present in commercial protein hydrolysates used for human nutrition. These compounds are genotoxic and 1,3-dichloro-2-propanol induced tumors in rats. Now it is reported that both compounds are active at inducing malignant transformation of mouse fibroblasts. Therefore, the carcinogenic risk to humans by exposure to these compounds contained in food is of concern. The investigation of the in vivo carcinogenic potential of 3-chloro-1,2-propanediol is urgently required to further evaluate the carcinogenic

The Regulatory Effects of Pea Protein Hydrolysate on Lipid Metabolism in Gestational Diabetic Mice.

Gestational diabetes mellitus (GDM) poses significant risks to maternal health and adverse metabolic outcomes in offspring. Pea protein hydrolysate (PPH), a kind of bioactive peptide with anti-diabetic properties, was evaluated for its therapeutic potential in the GDM mouse model. While PPH exhibited mild glucose-lowering effects, high-dose PPH (1000 mg/kg/day) significantly ameliorated insulin resistance and systemic dyslipidemia, including reductions in serum triglycerides, total cholesterol, low-density lipoprotein cholesterol, and hepatic lipid accumulation. In addition, the PPH treatment
