Systematic Review of Vaccine Strategies Against Tritrichomonas foetus Infection in Cattle: Insights, Challenges, and Prospects.

Tritrichomonas foetus is a protozoan parasite that causes bovine trichomonosis (also referred to as trichomoniasis) resulting in substantial economic loss in extensive grazing systems. The parasite colonises the reproductive tracts of both male and female cattle, being asymptomatic in males but causing early reproductive failure in infected females. This systematic review aimed to examine research manuscripts describing the development of T. foetus vaccines, the strategies employed, and the immune response associated with T. foetus infection. A systematic review was conducted using indexed sou

[Research Advance on Toxoplasma gondii Rhoptry Protein 2 Family as Candidate Vaccine Molecules].

Toxoplasma gondii rhoptry protein 2 family (ROP2 family), secreted by the rhoptry, plays an important role in T. gondii invasion of host cells and its virulence. The ROP2 family members include ROP2, ROP4, ROP5, ROP8, ROP13, ROP16, ROP17, and ROP18. Recent studies have found that these members are potential vaccine candidates against toxoplasmosis, and can induce the protective immunity of the host. This paper reviews the research advance on the ROP2 family members as DNA or protein vaccines against toxoplasmosis.

Immunoproteomic analyses identify broadly cross-reactive sporozoite immunogens of Eimeria maxima recognized by antisera from chickens infected with E. maxima, E. necatrix, E. tenella or E. acervulina.

Coccidiosis is a widespread, protozoan disease that continues to impose a high risk to the global poultry industry despite various control measures, including live, attenuated vaccines. To set the stage for next-generation vaccines that are broadly protective, our study aimed to identify sporozoite immunogens that are cross-reactive with hyper-immune sera against Eimeria maxima, E. tenella, E. necatrix and E.acervulina. In all, 2D electrophoresis, immunoblots and MALDI-TOF-MS/MS revealed 12 immunogenic proteins of interest, including 11 that have amino acid sequences matching those of non-redu

Strategies for developing multi-epitope subunit vaccines against tick-borne protozoa and bacteria.

The emergence of tick-borne protozoa and bacteria poses significant threats to both livestock and human health, underscoring the urgent need for effective vaccination strategies. Multi-epitope subunit vaccines offer a promising solution, as they can elicit robust humoral and cellular immune responses. This review synthesizes current knowledge on the development of multi-epitope subunit vaccines against tick-borne protozoan and bacterial pathogens. Advances in immunoinformatics, bioinformatics, molecular biology, and proteomics have enabled the identification of key immunogenic proteins and epi

Immunological evaluation of a novel multi-antigenic DNA vaccine encoding SAG1, SAG3, MIC4, GRA5, GRA7, AMA1and BAG1 against Toxoplasma gondii in BALB/c mice.

Many recent studies have been conducted to find new DNA vaccines based on Toxoplasma gondii antigens. DNA vaccines encoding complex of different antigens showed better immune responses compared to single antigen vaccine. In this study, we constructed a DNA vaccine encoding SAG1, SAG3, MIC4, GRA5, GRA7, AMA1 and BAG1 against T. gondii, and evaluated the immune response it induced in BALB/c mice. For this purposes, thirty BALB/c mice were randomly divided into three groups containing tenmice each. There were two negative control groups (PBSand pVAX1 vector) and one vaccination group (pVAX1-MAF,

Proof-of-concept trial in mature bulls prophylactically and therapeutically vaccinated with an experimental whole-cell killed Tritrichomonas foetus vaccine.

Tritrichomonas foetus causes bovine trichomonosis, a venereal disease that reduces productivity in naturally mated cattle. Its high prevalence in Northern Australian herds underscores the need for a locally made strain-specific vaccine. This study developed and tested a whole-cell killed T. foetus vaccine using the Queensland isolate TfOz5 (vaccine strain) and TfOz-N36 (Northern Territory isolate) as the challenge strain. The heat-inactivated vaccine, adjuvanted with Montanide ISA 61 VG, was administered subcutaneously in 2 doses (5 × 10⁷ cells/dose) at a 1-month interval to mature bulls (n =

Protective mucosal and systemic immunity induced by virus-like particles expressing Toxoplasma gondii cyst wall protein.

Toxoplasma gondii host cellular invasion factors such as the rhoptry proteins, micronemal antigens, or other subcellular compartment proteins have shown limited vaccine efficacies. T. gondii cyst wall protein (CST1) as a cyst persistence factor is critical for cyst wall integrity and bradyzoite persistence. Here, we generated influenza virus-like particles (VLPs) expressing the T. gondii CST1 and evaluated the mucosal as well as systemic immunities induced by VLPs. Intranasal immunization with the VLPs induced parasite-specific IgG and IgA antibody responses in sera and intestines. VLP immuniz

Advancement in the development of DNA vaccines against Trypanosoma brucei and future perspective.

Trypanosomes are the extracellular protozoan parasites that cause human African trypanosomiasis disease in humans and nagana disease in animals. Tsetse flies act as a vector for the transmission of the disease in African countries. Animals infected with these parasites become useless or workless, and if not treated, disease can be fatal. There are many side effects associated with old treatments and some of them result in death in 5% of cases. There is a major surface glycoprotein in the parasite known as variant surface glycoprotein. The immune system of the host develops antibodies against t

Protective immunity against congenital toxoplasmosis with recombinant SAG1 protein in a guinea pig model.

Primary infection with Toxoplasma gondii during pregnancy can induce fetal pathology and abortion in both humans and animals. The present study describes the development of an experimental model of congenital toxoplasmosis in the guinea pig. In this animal model, we evaluated the protective effect of vaccination with a recombinant form of SAG1 against maternofetal transmission of tachyzoites. The presence of parasites in fetuses was determined by nested PCRs and by an in vivo readout after fetal brain homogenate injections in mice. The absence of parasites was demonstrated in 66 to 86% of fetu

Comparison of Babesia rossi and Babesia canis isolates with emphasis on effects of vaccination with soluble parasite antigens: a review.

Babesia canis and B. rossi are large Babesia species that infect dogs and cause clinical disease. The spectrum of disease is highly diverse with either parasite, but upon evaluation of field cases it has been suggested that in general B. rossi is more virulent than B. canis. This difference was also found in experimental infections using B. canis and B. rossi isolates and appeared to be related to a difference in parasitaemia. Whether this difference reflects the essential difference between B. canis and B. rossi species in general, or merely reflects the variability in virulence of individual

Leishmania mediated regulation of host metabolism: impact on host immunity.

Leishmaniasis, a vector-borne disease affecting millions worldwide, is caused by protozoan parasite Leishmania. In mammalian hosts, Leishmania survives in the hostile environment of macrophages by exploiting key metabolic pathways to evade their destruction and subvert the host immune responses. Understanding of how Leishmania alters host immune cell metabolism could reveal novel targets for vaccines and therapeutics for effective control of Leishmaniasis. This review focuses on Leishmania-induced modulation of host immune response, with particular focus on reprograming of key metabolic pathwa

The African Green Monkey model for cutaneous and visceral leishmaniasis.

Non-human primates are valuable models for biomedical research because of their similarities to human anatomy, immunology and physiology. Leishmaniasis, a disease caused by protozoan parasites, has a worldwide distribution and results in high morbidity and mortality. Availability of a non-human primate model of leishmaniasis would facilitate the study of different aspects of this disease and would accelerate the development of vaccines and new drugs. In this article, some interesting features of the vervet monkey (African Green monkey) model of human cutaneous and visceral leishmaniasis are di

Comparative analysis of egg adapted vaccines and salinomycin against coccidiosis in chicks.

Avian coccidiosis is an intestinal protozoan parasitic disease of genus Eimeria. Wide use of anticoccidial drugs has resulted in development of drug resistant strains. Current study is based upon the development of egg adapted vaccines; gametocytes, formalin inactivated and formalin inactivated sonicated gametocytes against coccidiosis. Day old chicks (n = 90) were divided into six groups (A-F) with triplicate. On 5th day of age, three groups were orally given developed vaccines (0.2 ml) while on 21st day groups (A-E) were challenged orally with mixed Eimeria spp (60-70,000 oocysts/chick). The

Genetically modified organisms and visceral leishmaniasis.

Vaccination is the most effective method of preventing infectious diseases. Since the eradication of small pox in 1976, many other potentially life compromising if not threatening diseases have been dealt with subsequently. This event was a major leap not only in the scientific world already burdened with many diseases but also in the mindset of the common man who became more receptive to novel treatment options. Among the many protozoan diseases, the leishmaniases have emerged as one of the largest parasite killers of the world, second only to malaria. There are three types of leishmaniasis n

A dual-antigen malaria vaccine targeting Pb22 and Pbg37 was able to induce robust transmission-blocking activity.

BACKGROUND: Despite years of effort to develop an effective vaccine against malaria infection, a vaccine that provides individuals with sufficient protection against malaria illness and death in endemic areas is not yet available. The development of transmission-blocking vaccines (TBVs) is a promising strategy for malaria control. A dual-antigen malaria vaccine targeting both pre- and post-fertilization antigens could effectively improve the transmission-blocking activity of vaccines against the sexual stages of the parasite. METHODS: A chimeric recombinant protein Pb22-Pbg37 (Plasmodium berg

Cellular and humoral immune responses induced in cattle by vaccination with Babesia divergens culture-derived exoantigens correlate with protection.

Previous results with the Babesia divergens gerbil vaccination model were extended in studies with cattle. Two calves were vaccinated with culture-derived B. divergens exoantigens, and two others were treated with control supernatant; both preparations were adjuvanted with Quil-A saponin. A parasite-specific humoral response was observed after the first vaccine injection and was boosted by two succeeding vaccine injections. Sera from the two vaccinated calves immunoprecipitated eight major parasitic proteins (with molecular masses ranging between 17 and 110 kDa) whose patterns were close to th

Multi-epitope mRNA vaccine and protein vaccine protect mice against Toxoplasma gondii.

BACKGROUND: Toxoplasma gondii, an obligate intracellular protozoan parasite, infects almost one-third of the world's population and all warm-blooded animals, posing a substantial threat to public health. Accordingly, the development of effective vaccines against T. gondii has become an urgent priority. In this study, we constructed a multi-epitope chimeric antigen T-SGR targeting three key protective antigens of T. gondii (SAG1, GRA7, and ROP16), and developed both a messenger RNA (mRNA) lipid nanoparticle (LNP) vaccine and a recombinant protein vaccine based on T-SGR. Understanding Cell Communication Among Protozoan Parasites.

Infections caused by single-celled protozoan parasites affect countless individuals around the world, particularly in low- and middle-income countries. Despite the significant impact on global health, available therapeutic intervention strategies are still limited. Gaining insight into the mechanisms of infection is essential for developing new vaccines and drug therapies. While host-parasite interactions are well-recognized as critical factors in disease, recent evidence highlights the growing importance of communication among parasites. Various studies indicate that protozoans can interact a
