The RLK protein TaCRK10 activates wheat high-temperature seedling-plant resistance to stripe rust through interacting with TaH2A.1.

Plants sense various pathogens and activate immunity responses through receptor-like kinases (RLKs). Cysteine-rich receptor-like kinases (CRKs) are involved in massive transduction pathways upon perception of a pathogen. However, the roles of CRKs in response to stripe rust are unclear. In the present study, we identified a CRK gene (designated TaCRK10) from wheat variety Xiaoyan 6 (XY6) that harbors high-temperature seedling-plant (HTSP) resistance to stripe rust caused by fungal pathogen Puccinia striiformis f. sp. tritici (Pst). The expression level of TaCRK10 was induced by Pst inoculation

Comparative Transcriptomics Reveals Constitutive Immunity and Divergent Defence Strategies in Wheat against Leaf Rust.

Wheat leaf rust, caused by Puccinia triticina, poses a continuous threat to global food security. To dissect durable resistance mechanisms, we compared early infection responses between the near-immune cultivar 'Yunmai 34' (YM) and the susceptible 'Chinese Spring' (CS). While CS exhibited massive transcriptional reprogramming upon infection, YM displayed a relatively narrow scale-changes yet could arrested the pathogen prior to haustorium formation. This resistance might be associated with a constitutive immune state, evidenced by elevated basal expression of pathogenesis-related (PR) genes an

The development and validation of a genotyping-by-target sequencing chip for fungal population genetic analysis.

Advanced genotyping technologies for understanding the genetic intricacies of fungal pathogens have broad applications in crop protection. Here, we introduce a novel genotyping-by-target sequencing (GBTS) chip, a versatile tool designed for comprehensive genetic analysis of fungal populations. This technology overcomes key limitations of traditional molecular marker-based approaches by providing a more efficient, economic, and streamlined solution while bypassing the need for labor-intensive pathogen culturing. We demonstrate its utility by applying it to profile Puccinia striiformis f. sp. tr

A PR1-Like Protein Stabilizes a Barwin-Like Protein to Enhance Wheat Resistance to Leaf Rust.

Pathogenesis-related 1 (PR1) proteins contribute to plant immunity through antimicrobial activity and the regulation of target protein stability and condensation. In wheat, TaPR1-2 has been widely used as a molecular marker of immune responses to rust fungi. However, it remains unclear whether other members of the PR1 family are involved in wheat resistance to leaf rust. Here, we show that the transcript levels of TaPR1-11 are induced following inoculation with Puccinia triticina (Pt). Overexpression of TaPR1-11 significantly enhanced wheat resistance to Pt, accompanied by reduced fungal growt

Mimicry in plant-parasitic fungi.

Mimicry is the close resemblance of one living organism (the mimic) to another (the model), leading to misidentification by a third organism (the operator). Similar to other organism groups, certain species of plant-parasitic fungi are known to engage in mimetic relationships, thereby increasing their fitness. In some cases, fungal infection can lead to the formation of flower mimics (pseudo flowers) that attract insect pollinators via visual and/or olfactory cues; these insects then either transmit fungal gametes to accomplish outcrossing (e.g. in some heterothallic rust fungi belonging to th

TaCAP1 Interacts with TaLHCB1s and Positively Regulates Wheat Resistance Against Stripe Rust.

Actin filaments and their associated actin-binding proteins play key roles in plant innate immune signaling. CAP1, or cyclase-associated protein 1, is an important regulatory factor of the actin cytoskeleton-associated signaling network and was hypothesized here to be involved in resistance against wheat stripe rust because TaCAP1 expression was upregulated in response to Puccinia striiformis f. sp. tritici (Pst). Downregulation of TaCAP1 expression led to decreased resistance against Pst, in contrast to increased resistance upon TaCAP1 overexpressing, as demonstrated by the changes of phenoty

Neighbour Effects and Dynamics of Plant-Associated Microbiomes in a Strip-Crop System.

While multi-cropping systems, such as strip-cropping, assemble complex microbial communities that contribute to ecosystem resilience, we have limited understanding of their assembly and functions at field scale. Here, we examined the dynamics and site-specific assembly of soil and phyllosphere microbiomes in a rotational faba bean (Vicia faba) and spring barley (Hordeum vulgare) strip-system during two seasons. Using bacterial 16S and fungal ITS amplicon sequencing, we analysed microbial communities across gradients from the centre towards strip edges. We found that phyllosphere microbiomes ar

Effects of Rust Fungus and Aphid Infestation on Plant Height and Seed Production of Impatiens parviflora DC.

The effects of the rust fungus Puccinia komarovii Tranzschel and the aphid Impatientinum asiaticum Nevsky 1929 on plant height, seed number per capsule, and seed mass were investigated in eight small balsam (Impatiens parviflora DC.) stands in Hungary and one in Poland. Two stands were infected by rust, five by aphids, and two were healthy. The lowest average plant height was 37.5 cm and the highest 94.7 cm, both measured in aphid-infested stands. Examining the stands separately, no relationship appeared with the type of damage. For data pooled across stands, differences were significant: rust

Novel histone deacetylase-inhibiting 5-(trifluoromethyl)-1,2,4-oxadiazole derivatives with exceptional efficacy against Phakopsora pachyrhizi.

BACKGROUND: Asian soybean rust caused by Phakopsora pachyrhizi (P. pachyrhizi) poses a serious threat to global soybean production, and resistance to conventional fungicides is increasing. Therefore, new fungicides with novel modes of action are urgently needed. This study aimed to design and optimize potent 5-(trifluoromethyl)-1,2,4-oxadiazole (TFMO) derivatives inspired by fungal histone deacetylase inhibition. RESULTS: Forty-two TFMO derivatives were synthesized through bioisosteric modification and linker optimization. Among them, FM-17888 showed the highest activity against P. pachyrhizi

The TaRNS4-TaPP2C10 module regulates wheat resistance to stripe rust through RNA degradation and MAPK cascades.

RNase is a kind of RNA hydrolase that widely exists in organisms. RNase T2 in plants functions by hydrolyzing and processing RNA in the cell. However, the mechanism by which RNase T2 acts on intercellular RNA to exert the resistance function has not been studied. Here, we identified the S-like RNase TaRNS4 that positively regulates wheat resistance to stripe rust by degrading the RNA of Puccinia striiformis f. sp. tritici (Pst), the causative agent of stripe rust. Importantly, TaRNS4 phosphorylation mediated by receptor-like kinase TaRLK5 contributes to its degradation of Pst RNA and enhances

Proline Dehydrogenase Gene TaProDH1 Positively Regulates Wheat Resistance to Stripe Rust Disease.

Proline dehydrogenase (ProDH) is a rate-limiting enzyme in the proline metabolism cycle that plays an important role in plant stress response and disease resistance. However, the function and mechanism of ProDH1 in the wheat-Puccinia striiformis f. sp. tritici (Pst) interaction remain poorly understood. In this study, a wheat proline dehydrogenase gene TaProDH1 was induced by stripe rust. qRT-PCR analyses showed that the transcript levels of TaProDH1 were highly increased during early infection. The transient expression of TaProDH1 in Nicotiana benthamiana leaves revealed that TaProDH1 increas

A LysM receptor-like kinase TaLYK5 positively regulates wheat resistance to leaf rust.

Wheat leaf rust, caused by the biotrophic fungus Puccinia triticina (Pt), is one of the most destructive diseases threatening global wheat production. Lysin motif (LysM)-containing receptors play central roles in plant recognition of fungal chitin and activation of pattern-triggered immunity (PTI), but the function of the LysM receptor-like kinase TaLYK5 in wheat resistance to leaf rust remains largely unknown. Here, we show that TaLYK5 lacks conserved kinase motifs essential for catalytic activity and is predicted to be a pseudokinase, suggesting that it may function through a co-receptor to

Cytological and molecular characterization of non-host resistance in Arabidopsis thaliana against wheat stripe rust.

Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most destructive diseases of wheat worldwide. We report the use of the non-host plant Arabidopsis thaliana to identify the basis of resistance to Pst at the cytological and molecular levels. No visible symptoms were observed on Arabidopsis leaves inoculated with Pst. Microscopic observations showed that significantly reduced numbers of Pst urediospores had successfully achieved penetration in Arabidopsis compared with those in wheat. There were significant differences in the frequency of stomatal penetration

Comparative genomics reveals lineage-associated structural variation and diversification in a barley fungal pathogen.

Leaf rust, caused by Puccinia hordei, is a major barley disease worldwide. Despite repeated shifts in virulence, contrasting reproductive histories, and emerging fungicide insensitivity, the genomic basis of its diversification and adaptation remains poorly understood. In this study, we generated haplotype-resolved, chromosome-level genome assemblies for two isolates with contrasting virulence and analyzed 41 Australian isolates collected over 54 yr (1966-2020), integrating comparative and population genomics, mating-type gene phylogenies, chromosome-specific k-mer profiling, genome-wide copy-

Salicylic Acid Mediates Chitosan-Induced Immune Responses and Growth Enhancement in Barley.

Chitosan (CS), derived from the partial deacetylation and hydrolysis of chitin, varies in the degree of deacetylation, molecular weight, and origin, influencing its biological effects, including antifungal properties. In plants, CS triggers immune responses and stimulates biomass growth. Previously, we found that the antifungal activity of CS was strongly dependent on its physicochemical properties. This study revealed that the chitosan batch CS_10 with the strongest antifungal activity also effectively activated plant immune responses and promoted biomass growth. Barley treated with CS_10 exh

Standardized Workflow for Long Noncoding RNA Prediction and Expression Profiling in Wheat Under Stem Rust Stress.

Long noncoding RNAs (lncRNAs), typically over 200 nucleotides in length, with low or no coding capacity, possess significant regulatory roles in plant growth, development, and stress responses by interacting with DNAs, proteins, and other RNAs. This chapter outlines a comprehensive protocol for identifying and functionally characterizing wheat lncRNAs under stem rust infection. The workflow includes plant growth, pathogen inoculation, RNA isolation, and whole-transcriptome sequencing of wheat. In-silico approaches for data preprocessing, transcriptome assembly and subsequent characterization,

Comparative multi-omics uncover conserved and lineage-specific defense strategies to southern corn rust in temperate and tropical maize.

Temperate and tropical maize inbred lines achieve high-level resistance to southern corn rust through distinct ETI associated and PTI associated signaling networks, with the peroxidase gene PER1 functioning as a common positive regulator across both resistance types. Southern corn rust (SCR), caused by the fungus Puccinia polysora, is a devastating fungal disease that leads to significant yield losses in maize worldwide. This study employs an integrated multi-omics approach, combining phenotyping, transcriptomics, metabolomics, weighted gene co-expression network analysis (WGCNA), and gene sil

First Description of the Causal Agent of Fairy Ring of Cranberry and Its Novel Dispersal Method.

Fairy ring is an economically important disease of cranberry in the northeastern United States, yet its causal agent has remained unresolved for more than a century. Field sampling, targeted isolation, morphological and molecular characterization, pathogenicity tests, and population analyses consistently associated the disease with the genus Helicobasidium (Pucciniomycetes), a known teleomorph of the violet root rot fungus. Internal transcribed spacer and large subunit rDNA sequences placed all isolates from New Jersey and Massachusetts in a distinct lineage within H. longisporum, designated h

Proposal for a new standard area diagram to quantify the severity of orange rust in sugarcane.

Orange rust (Puccinia kuehnii), a fungal disease of major concern in the sugarcane industry, can reduce the productivity of susceptible sugarcane varieties by up to 50% in many sugarcane-producing countries. The use of a standard area diagram (SAD) enhances the assessment of rust severity, improving the accuracy and consistency of disease quantification by evaluators. The present study aimed to develop and validate a new SAD for assessing the severity of orange rust in sugarcane. The SAD was constructed from symptomatic leaves collected in the field, with severity levels of 0, 1.5, 3.8, 7.3, 1
