Pueraria flavone (PF), the main component of Pueraria lobata, is a traditional Chinese medicine used for the treatment of cardiovascular and cerebrovascular diseases; however, it exhibits low oral bioavailability because of its poor membrane permeability. In this study, PF-loaded sodium deoxycholate-decorated liposomes (SDC-Lips) were prepared using the reverse-phase evaporation method and optimised using the Box-Behnken design method. The morphology, particle size, zeta potential, and entrapment efficiency of these PF-loaded SDC-Lips were evaluated. The release behaviours of PF-loaded SDC-Lip

Puerariae Radix, the root of Pueraria lobata (Willd.) Ohwi is primarily used as a traditional herbal medicine in East Asia. Although phytochemical and biological investigations of the roots have been well-reported, those of the leaves are limited. To explore the potential value of P. lobata leaves, this study aimed to evaluate their effect on melanin synthesis in B16-F1 murine melanoma cells and three-dimensional (3D) human skin equivalents and investigate the underlying molecular mechanism. The leaf extract increased the melanin content in B16-F1 cells, with stronger induction potency than th

Phytoestrogens have attracted attention for their potential in the prevention of postmenopausal osteoporosis. Recently, phytoestrogen-rich herb Pueraria mirifica has been demonstrated to possess an osteogenic effect on bone in ovariectomized rats, but its underlying cellular mechanism was not known. Here, we investigated the effects of P. mirifica extract and its major isoflavone compound, puerarin, on cell viability, cell proliferation and the expression of differentiation markers in rat osteoblast-like UMR106 cells. After exposure to 17β-estradiol (E2), genistein, P. mirifica extract and pue

Neuronal degeneration is known to be due to oxidative stress acting through a pathway involving the excessive activation of glutamate receptors. We studied the neuroprotection potential of an ethyl acetate-ethanol extract of Pueraria mirifica (P. candollei var. mirifica) root (PM extract). PM extract was evaluated for its antioxidant and neuroprotective activities against glutamate toxicity in mouse hippocampal HT22 neuronal cells. The extract at concentrations of 10 and 50 μg/ml exhibited considerable antioxidant activity with significant neuroprotection, based on the microscopic observations

Exessive drinking is commonly associated with a wide spectrum of liver injuries. The term alcoholic liver disease (ALD) is generally used to refer to this spectrum of hepatic abnormalities, and the term hepatic steatosis denotes early lesions. Puerariae Lobatae Radix (PLR) is a common traditional Chinese medicine and has been widely used as an efficient treatment for alcohol-induced damage. Flavonoids are the principal components of PLR that could potentially be responsible for the activation of alcohol metabolism and lipid-lowering effects. However, little is known about the mechanisms underl

Capsaicin is a bioactive substance with diverse health-promoting properties, but its intense pungency and irritant side effects limit oral application. This study developed a Pueraria lobata particle-based Pickering emulsion (PE) as an oral delivery platform for capsaicin. Modified Pueraria lobata particles (MP-ps) were prepared using a microwave-assisted enzymolysis technique (15% hydration, 252 J/g microwave energy and 8 h pullulanase hydrolysis). MP-ps exhibited surface cracks and pores, altered starch crystallinity, and enhanced water-holding capacity and swelling power. Compared with orig

Medicinal and edible plants are promising antidiabetic candidates due to their combined nutritional and pharmacological properties. However, the disconnection between bioactive substance extraction and efficacy validation hampers the development of integrated functional interventions. Here, a green and efficient subcritical ethanol-water extraction (SEWE) system was developed to obtain puerarin-enriched extracts (PEE) from Pueraria lobata. By introducing ethanol as a green polarity-modifying co-solvent, the SEWE system enabled polarity-tunable extraction under relatively mild subcritical condi

ETHNOPHARMACOLOGICAL RELEVANCE: Pueraria lobatae-Citrus reticulata Decoction (PLRY) is derived from a modified version of the "Jiawei Jiecheng Decoction" described in the renowned Qing Dynasty text Wai Ke Zheng Zhi Quan Shu. Traditionally, it has been used to treat symptoms such as "severe pain, diffuse joint swelling, and mild redness" associated with gout. Preclinical studies have shown that the combined use of Pueraria and Citrus possesses anti-hyperuricemic effects. However, the specific molecular mechanisms by which PLRY influences hyperuricemia have yet to be completely understood.

Pueraria lobata root and Glycine max (L.) Merrill are rich in phytoestrogens. However, these bioactive ingredients have limited bioavailability due to their high molecular weight. In this study, we extracted two natural products and fermented with Lactiplantibacillus plantarum before mixing the fermented extracts (FPE-FGE). To understand whether FPE-FGE could alleviate menopause with dyslipidemia, we examined their effects on ovariectomy (OVX)-induced dyslipidemia in mice. Oral administration of the FPE-FGE (1:9, 3:7, and 9:1) did not affect safety-related biomarkers, such as uterus index (%),
