Epileptogenic activity induced by histamine H(1) antagonists in amygdala-kindled rats.

The epileptogenic activities induced by histamine H(1) antagonists in amygdala-kindled rats were studied in comparison with activities in nonkindled rats (sham rats). Intraperitoneal injection of pyrilamine, diphenhydramine and ketotifen resulted in behavioral and electroencephalogram (EEG)-detected seizures in amygdala-kindled rats at doses which caused no or negligible seizures in sham rats. On the other hand, loratadine and cetirizine caused no behavioral or EEG seizures in either amygdala-kindled or sham rats even at a dose of 40 mg/kg. In conclusion, first-generation H(1) antagonists like

In vitro antagonism of benzodiazepine binding to cerebral receptors by H1 and H2 antihistamines.

Antihistamines directed at either the H1 or H2 histamine receptor have the well-known side effect of sedation. The mechanism for the CNS depressant action of antihistamines is unknown. We examine here the possibility that the mechanism may involve modulation of the cerebral benzodiazepine receptor. We demonstrate that cimetidine and pyrilamine are competitive antagonists of 3H-benzodiazepine binding to human cerebral receptor in vitro. The inhibition of radioligand binding was not specific for benzodiazepine receptor, however, since antihistamines also antagonized binding to GABA, opiate, and

Histaminergic involvement in thyrotropin-releasing hormone stimulation of antral tissue in the rat.

The mechanism of action of thyrotropin-releasing hormone on antral motility was investigated in vitro. With the recent detection of thyrotropin-releasing hormone in gastrointestinal tissue, the possibility exists that the peptide may play a physiologic role in gastrointestinal motility. Results indicate that the hormone stimulates antral motility through a non-cholinergic excitatory pathway based on the inability of the muscarinic antagonist, atropine, to block the response. A histaminergic pathway is an attractive possibility to explain the excitatory response, because both pyrilamine and cim

The role of mast cells in citric acid-induced airway constriction and cough.

Inhalation of citric acid (CA) causes airway constriction and coughing. To investigate the role of mast cells in CA-induced airway constriction and cough, three experiments using guinea pigs were carried out. In the first experiment, we used compound 48/80 to deplete mast cells, cromolyn sodium to stabilize mast cells, MK-886 to inhibit synthesis of leukotrienes, pyrilamine to antagonize histamine H1 receptor, methysergide to antagonize serotonin receptor, and indomethacin to inhibit cyclooxygenase. In the second experiment, compound 48/80-pretreated animals were divided into 2 parts; the firs

Histamine: new thoughts about a familiar mediator.

Any health-care provider knows that the sneezing, wheezing, and itching that are commonplace most often involve a small molecule, namely, histamine. In addition to its inherent physiologic role, this seemingly small "actor" is of profound historical and fiscal significance. This is evidenced in part by the awarding of the 1936 Nobel Prize in physiology or Medicine to Sir Henry Hallett Dale and Dr Otto Loewi who discovered the actions of histamine and the 1957 Nobel Prize in physiology or medicine to pharmacologist Dr Daniel Bovet who discovered the first antihistamine, pyrilamine (neoantergan)

Determination of amine ingredients in cough-cold liquids by reversed-phase ion-pair high-performance liquid chromatography.

The chromatographic behavior of phenylephrine, codeine, pseudoephedrine, phenylpropanolamine, methoxyphenamine, pheniramine, pyrilamine, dextromethorphan, and chlorpheniramine was examined by reversed-phase, ion-pair high-performance liquid chromatography. An isocratic chromatographic system was devised for the analysis of cough-cold liquids containing these amine drugs by optimization of the mobile phase ionic strength, buffer pH, pairing ion concentration, and secondary ion concentration. Quantitative recovery and excellent precision were demonstrated for the simultaneous determination of ph

Behavioral effects of histamine H1 antagonists: comparison with other drugs and modification by haloperidol.

The behavioral effects of several histamine H1 antagonists were compared in groups of squirrel monkeys trained to respond under fixed-interval schedules involving response-produced shock or termination of a stimulus associated with shock. Low to intermediate doses of the conventional H1 antagonists chlorpheniramine (up to 3.0 mg/kg), diphenhydramine (up to 1.0 or 3.0 mg/kg), pyrilamine (up to 3.0 mg/kg) and tripelennamine (up to 0.3 or 1.0 mg/kg) produced dose-related increases in response rate under all conditions in which they were studied; higher doses either increased responding less or de

[Effect of antihistaminic preparations on gastric secretion in rats].

Experiments were made to study the influence of a new antihistaminic drug phencarol [(quinuclidyl-3) diphenylcarbinol hydrochloride] on gastric secretion in rats. Unlike the blockers of H1-histamine receptors (diphenhydramine, omeril, pyrilamine, and cyproheptadine), phencarol reduces secretion and the content of free hydrochloric acid in the gastric contents. The antacid activity of the drug is similar to that of the H2-receptor blocker, cimetidine. However, phencarol differs from the latter drug in the mechanism of action. The antacid action of phencarol is likely to be the result of its act

Blockade of brain histamine metabolism alters methamphetamine-induced expression pattern of stereotypy in mice via histamine H1 receptors.

The administration of methamphetamine (METH, 10 mg/kg, i.p.) to male ICR mice induced stereotyped behavior consisting of nail and/or wood chip biting (86.0%), continuous sniffing (12.0%), head bobbing (1.1%), and circling (1.0%) during the observation period of 1 h. Pretreatment of the mice with metoprine (2, 10, and 20 mg/kg, i.p.), a selective inhibitor of histamine N-methyltransferase (HMT), which metabolizes histamine in the brain, significantly increased and decreased METH-induced continuous sniffing (20.5, 51.3, and 80.3%) and nail and/or wood chip biting (77.4, 45.3, and 14.2%), respect

The anorectic effect of neurotensin is mediated via a histamine H1 receptor in mice.

Neurotensin (NT), a tridecapeptide found in the mammalian brain and peripheral tissues, induces a decrease in food intake after central administration. In this investigation, we examine whether the histaminergic system is involved in NT-induced suppression of feeding. Intracerebroventricular injection of NT (0.1-1 nmol/mouse) led to dose-dependent inhibition of food intake in fasted ddY mice. The anorectic effect induced by NT (0.1 nmol/mouse) was ameliorated upon co-administration of pyrilamine (3 nmol/mouse), an antagonist for histomine H1 receptor. The NT-induced anorectic effect was partia

Histamine H1 receptors in the ventromedial hypothalamus mediate the anorectic action of the pancreatic hormone amylin.

In the present study we investigated the role of hypothalamic histamine H(1) receptors in the mediation of peripheral amylin's anorectic effect. Rats with chronically implanted bilateral cannulas were infused into the ventromedial hypothalamus (VMH) with the specific histamine H(1) receptor antagonists pyrilamine (PYR, 104 nmol/rat) or chlorpheniramine (CPA, 52 nmol/rat), respectively, combined with an intraperitoneal (IP) injection of amylin (5 microg/kg). Amylin's inhibitory effect on food intake (i.e. 50% reduction in cumulative food intake 30 min after ingestion) was markedly reduced by CP

Pharmacological characterization of opossum distal colonic muscularis mucosae in vitro.

In muscularis mucosae from the opossum distal colon, both tone and spontaneous contractions were highly dependent on the available oxygen. Acetylcholine and histamine caused, respectively, atropine- and pyrilamine-sensitive contractions. Norepinephrine relaxed the tissue, an effect abolished by propranolol. Under these conditions norepinephrine failed to elicit contractions and at higher concentrations again caused relaxations. The tissue gave concentration-dependent relaxations to ATP but not to ADP, AMP, or adenosine. Electrical field stimulation (20-30 Hz, 1-2 ms, 120 mA) revealed a choline

Negligible potency of acetylcholine in increasing vascular permeability.

As acetylcholine (ACh) is claimed to be a possible mediator of cholinergic urticaria, the potency of acetylcholine (ACh) in increasing vascular permeability was tested by skin perfusion technique in rats and by dye extraction method from the skin of rabbits. The potency of vascular permeability increase of ACh was much lower than that of histamine (Hist) and bradykinin (BK) in rats and rabbits. The higher dose of ACh (10 mg in 0.1 ml of saline), however, produced slight increase of vascular permeability in rabbits, which was inhibited by the antihistamine, pyrilamine maleate. Furthermore, ACh

NTP Hepatotoxicity Studies of the Liver Carcinogen Methapyrilene Hydrochloride (CAS No. 135-23-9) Administered in Feed to Male F344/N Rats.

Methapyrilene hydrochloride is a histamine H(1)-receptor antagonist that was an active ingredient in many over-the-counter cold and allergy medications. In the mid- to late 1970s, studies in rats suggested that methapyrilene hydrochloride was a hepatocarcinogen, and the drug was removed from these preparations. In most cases, methapyrilene hydrochloride was replaced by pyrilamine maleate, a structurally similar analogue. As part of a program to investigate mechanisms of toxicity whereby structurally similar chemicals produce different toxicities, these chemicals were studied for induction of c

Histamine-induced, central nervous system-mediated hyperglycemia is suppressed by atropine in the brain.

We investigated the relationship between histamine and muscarinic cholinergic neurons in central nervous system (CNS)-mediated glucose regulation in anesthetized fed rats. The injection of pyrilamine (5 x 10(-7) mol) into the third cerebral ventricle suppressed the hyperglycemia induced by intraventricular injection of histamine (5 x 10(-7) mol). Ranitidine (5 x 10(-7) mol), however, did not suppress this hyperglycemia. The injection of atropine (5 x 10(-9)-5 x 10(-7) mol) into the third cerebral ventricle suppressed the histamine-induced hyperglycemia in a dose-dependent manner. These finding

Effects of a histamine synthesis inhibitor and antihistamines on the sexual behavior of female rats.

Intraventricular administration of alpha-hydrazinohistidine, a histamine synthesis inhibitor, at different doses and times before testing produced a significant decrease of lordotic responses and sexual receptivity in ovariectomized estrogen plus progesterone-primed female rats. The H1-antihistamines pyrilamine and chlorfeniramine and the H2-antihistamine metiamide, injected in the lateral ventricle, significantly decreased the lordosis quotient but did not modify receptivity; antihistamine-injected rats showed no soliciting behavior. Exploratory activity was decreased by both alpha-hydrazinoh

Characterization of the rabbit detrusor response to histamine through pharmacologic antagonism.

The various effects of histamine on smooth muscles and other tissues are thought to be mediated by two pharmacologically distinct receptors, H1 and H2 types. These receptors are defined in terms of thier susceptibility to blockade by pyrilamine (type H1) or burimamide (type H2). In this study selective antagonism of the spasmodic effects of histamine on isolated strips of rabbit detrusor has shown that the response is mediated by H1 receptors, with no evidence of H2 activity. The p-A2 of the pyrilaminehistamine antagonism is 9.3. The histamine response is susceptible to muscarinic blockade (at

The effects of H1- and H2-receptor blocking agents on postsynaptic potentials recorded from the bull-frog sympathetic ganglion.

The effects of H1-blockers (pyrilamine, diphenhydramine and promethazine) and H2-blockers (cimetidine, ranitidine) on the postsynaptic potentials of the bull-frog sympathetic ganglion were studied by means of the sucrose gap method. All the H1-blockers tested, at concentrations of 10(-6)-10(-4) M, depressed the amplitudes of both action and slow postsynaptic potentials in positive and negative directions in a concentration-dependent manner; these drugs had more potent inhibitory effects on postsynaptic potentials. In the case of H2-blockers, action and postsynaptic potentials were slightly dep

Pharmacological Characterization of the Zebrafish (Danio Rerio) Histamine H1 Receptor Reveals the Involvement of the Second Extracellular Loop in the Binding of Histamine.

The zebrafish (Danio rerio) histamine H1 receptor gene (zfH1R) was cloned in 2007 and reported to be involved in fish locomotion. Yet, no detailed characterization of its pharmacology and signaling properties have so far been reported. In this study, we pharmacologically characterized the zfH1R expressed in HEK-293T cells by means of [3H]-mepyramine binding and G protein-signaling assays. The zfH1R [dissociation constant (KD), 0.7 nM] displayed similar affinity for the antagonist [3H]-mepyramine as the human histamine H1 receptor (hH1R) (KD, 1.5 nM), whereas the affinity for histamine is 100-f

Cholinergic control of smooth muscle peristalsis in the cat esophagus.

The aim of this study was to examine in detail the effects of selective cholinergic and other pharmacological antagonists on primary and secondary peristalsis in the smooth muscle of the cat esophagus in order to fully characterize the cholinergic contribution to peristalsis in this species. Primary and secondary peristalsis in the smooth muscle part of the feline esophagus was completely abolished by atropine, 4-dephenylacetoxy-N-methylpiperidine methiodide (4-DAMP) (a selective M2 muscarinic antagonist), hexamethonium, and high doses of nicotine.
