Carbonyl stress, characterized by pathological accumulation of reactive dicarbonyl species such as methylglyoxal (MGO), disrupts mitochondrial function and drives formation of advanced glycation end-products (AGEs), and is implicated in chronic neurobehavioral and metabolic disorders. Here, we established a strain-resolved activity metabolomics framework to rationally select postbiotic extracts with MGO-AGEs breaking activity. We screened a postbiotic library of 177 intracellular metabolite extracts from 113 bacterial strains by untargeted metabolite profiling and in vitro MGO-AGEs breaking ac

People living with HIV (PLWH) develop cardiovascular diseases (CVDs) about a decade earlier and at rates 2-3 times higher than the general population. At present, pharmacological strategies to delay the onset of CVDs in PLWH are unavailable, in part because of an incomplete understanding of its molecular causes. We and others recently uncovered elevated levels of the toxic glycolysis and inflammation-induced byproduct methylglyoxal (MG) in plasma from PLWH and from HIV-infected humanized mice (Hu-mice). We also found a reduction in expression of the primary MG-degrading enzyme glyoxalase I (Gl

During long-term peritoneal dialysis, peritoneal fibrosis (PF) often happens and results in ultrafiltration failure, which directly leads to the termination of dialysis. The accumulation of extracellular matrix produced from an increasing number of myofibroblasts was a hallmark characteristic of PF. To date, glucose degradation products (GDPs, i.e., methylglyoxal (MGO)) that appeared during the heating and storage of the dialysate are considered to be key components to initiating PF, but how GDPs lead to the activation of myofibroblast in fibrotic peritoneum has not yet been fully elucidated.

Intracellular pathogens neutralize and evade macrophage-intrinsic host defenses. In this issue of Cell Host & Microbe, Anaya-Sanchez et al. show that methylglyoxal, a metabolic byproduct of glycolysis, is part of the macrophage arsenal limiting L. monocytogenes and M. tuberculosis infections but is countered by pathogen expression of methylglyoxal detoxification enzymes.

BACKGROUND: Dihydroxyacetone (DHA) stands as a crucial chemical material extensively utilized in the cosmetics industry. DHA production through the dephosphorylation of dihydroxyacetone phosphate, an intermediate product of the glycolysis pathway in Escherichia coli, presents a prospective alternative for industrial production. However, insights into the pivotal enzyme, dihydroxyacetone phosphate dephosphorylase (HdpA), remain limited for informed engineering. Consequently, the development of an efficient tool for high-throughput screening of HdpA hypermutants becomes imperative.
RESULTS: Thi

BACKGROUND: Lotus rhizome juice (LRJ) is susceptible to the Maillard reaction (MR) and caramelization, which tend to cause a reduction in quality and lower consumer acceptance of the product. 1,2-Dicarbonyl compounds (DCs) and heterocyclic compounds have attracted increasing attention as key intermediates responsible for the formation of brown pigments during MR and caramelization. However, little is known about the effects of these two types of compounds on brown pigments in LRJ during sterilization. This study quantified the changes in brown intensity (A420), DCs, and heterocyclic compounds

Taurine is a recently recognized plant growth regulator under abiotic stress. However, the information on taurine-mediated plant defense responses is scarce, particularly on taurine-mediated regulation of the glyoxalase system. There is currently no report available on the use of taurine as seed priming under stress. Chromium (Cr) toxicity considerably subsided growth characteristics, photosynthetic pigments, and relative water content. Furthermore, plants encountered intensified oxidative injury due to a significant increase in relative membrane permeability, H2O2, O2•‒, and MDA production. T

Type 2 diabetes (T2D) is associated with elevated frequencies of micronuclei (MNi) and other DNA damage biomarkers. Interestingly, individuals with T2D are more likely to be deficient in micronutrients (folic acid, pyridoxal-phosphate, cobalamin) that play key roles in one-carbon metabolism and maintaining genomic integrity. Furthermore, it has recently been shown that deficiencies in these nutrients, in particular folic acid leaves cells susceptible to glucose-induced DNA damage. Therefore, we sought to investigate if the B lymphoblastoid WIL2-NS cell line cultured under folic acid-deficient

Similar to their mammalian counterpart, protozoan parasites including Leishmania donovani detoxify methylglyoxal (MG),(1) a toxic ubiquitous product generated in glycolysis pathway. However, it differs in one or more way(s) from the humans. It is known that MG is eliminated either through glyoxalase (GLO)(2) pathway and/or excreted across the cell membrane. This toxic metabolic by-product is known to be detoxified predominantly by the GLO pathway and excretion across the cell membrane is never considered to be a significant pathway for its detoxification. We have tried to modulate these pathwa

Recently, several types of foods and drinks, including coffee, cream, and cake, have been found to result in high methylglyoxal (MG) levels in the plasma, thus causing both nutritional and health concerns. MG can be metabolized by phase-II enzymes in liver through the positive regulation of nuclear factor-erythroid 2-related factor 2 (Nrf2). In this study, we investigated the ability of scopoletin (SP) to protect against MG-induced hyperglycemia and insulin resistance. Recently, SP was shown to be a peroxisome proliferator-activated receptor-γ activator to elevate insulin sensitivity. We inves

Patients with irritable bowel syndrome (IBS) show a wide range of symptoms including diarrhea, abdominal pain, changes in bowel habits, nausea, vomiting, headache, anxiety, depression and cognitive impairment. Methylglyoxal has been proved to be a potential toxic metabolite produced by intestinal bacteria. The present study was aimed at investigating the correlation between methylglyoxal and irritable bowel syndrome. Rats were treated with an enema infusion of methylglyoxal. Fecal water content, visceral sensitivity, behavioral tests and serum 5-hydroxytryptamine (5-HT) were assessed after met

Triosephosphate isomerase (TIM) is one of the most efficient enzymes known, yet its catalytic precision is not fully explained by classical biochemistry. Here we propose that TIM operates as a quantum logic gate, in which the proton transfer between dihydroxyacetone phosphate and glyceraldehyde-3-phosphate arises from quantum tunneling within a reversible two-state system. We extend this catalysis model by introducing a non-unitary decay channel that quantitatively describes the loss of quantum coherence (decoherence) at the level of the enediol intermediate. In this framework, the formation o

Reactive α-oxoaldehydes, such as glyoxal (GO) and methylglyoxal (MGO), are cytotoxic glycolysis byproducts that induce cellular dysfunction by nonenzymatically modifying diverse macromolecules. Although DJ-1 (PARK7), a protein linked to Parkinson's disease, acts as a type-III glyoxalase/methylglyoxalase, the physiological function(s) of its prokaryotic homologs remain poorly understood. Here, we investigated the enzymatic and physiological properties of YhbO, one of four DJ-1 homologs in Escherichia coli. We demonstrate that YhbO has higher catalytic efficiency than DJ-1 and exhibits broad-spe

The amino groups of lysine and arginine residues in proteins react with reducing sugars and carbonyl compounds to form advanced glycation endproducts (AGEs). Several AGEs have been detected in human lenses, and their levels have been shown to increase with age and cataract formation. AGEs can lead to structural changes, yellow pigmentation, and cross-linking of lens proteins. Studies have shown a positive correlation between AGEs levels and lens age, stiffness, and cataracts. Recent research suggests that DJ-1 can inhibit the accumulation of AGEs in cellular proteins by reversing the early gly

During Mycobacterium tuberculosis infections bacteria are engulfed by macrophages, a main line of defense against invading pathogens. Upon activation, macrophages increase glycolysis, producing the antibacterial aldehyde methylglyoxal. To test whether bacterial methylglyoxal resistance is required for robust infections, we sought to identify M. tuberculosis defense mechanisms against methylglyoxal. We identified phoP mutants were among the most highly sensitive strains to methylglyoxal in vitro. phoP mutants are highly attenuated in mice but a phoP mutant was even more attenuated in mice that

Ultrafast affinity extraction (UAE) and affinity microcolumns containing immobilized human serum albumin (HSA) were employed to evaluate the effect of advanced stage glycation on HSA and its binding to warfarin, a common site-specific probe for Sudlow site I of this protein. The modification of HSA by glyoxal (GO) and methylglyoxal (MGO) was considered, where GO and MGO are known to be important in the formation of many types of advanced glycation end products. Free drug fractions were measured by UAE for warfarin in solutions containing normal HSA or HSA that had been modified by GO or MGO at

Trypanosomatid parasites are the infectious agents causing Chagas disease, visceral and cutaneous leishmaniasis and human African trypanosomiasis. Recent work of others has implicated an aldo-keto reductase (AKR) in the susceptibility and resistance of Trypanosoma cruzi to benznidazole, a drug used to treat Chagas disease. Here, we show that TcAKR and homologues in the related parasites Trypanosoma brucei and Leishmania donovani do not reductively activate monocyclic (benznidazole, nifurtimox and fexinidazole) or bicyclic nitro-drugs such as PA-824. Rather, these enzymes metabolise a variety o

Citrus are mainly grown in low pH soils with high active aluminum (Al). 'Xuegan' (Citrus sinensis (L.) Osbeck) and 'Shatian pummelo' (Citrus grandis (L.) Osbeck) seedlings were fertilized for 18 weeks with nutrient solution containing either 0 mM (control) or 1 mM (Al toxicity) AlCl3·6H2O. Aluminum induced decreases of biomass, leaf photosynthesis, relative water content and total soluble protein levels, and increases of methylglyoxal levels only occurred in C. grandis roots and leaves. Besides, the Al-induced decreases of pigments and alterations of chlorophyll a fluorescence transients and f

Two evaluations were carried out in this study. The first was a comparison of the standard tube test with the automated microtitration test for the detection of antibodies to Pseudomonas pseudomallei by the indirect hemagglutination method. Data from this comparison indicated that the tests were equivalent. The second evaluation consisted of reproducibility studies on two lots of pyruvic aldehyde-stabilized sensitized erythrocytes in comparison with freshly prepared sensitized erythrocytes in the automated microtitration test. The influence of different types of the microtitration plates used

AlphaB-crystallin is a small heat shock protein that is found in the eye lens, retina, heart, brain, skin, and skeletal muscle. Mutations of αB-crystallin lead to the development of cataracts, human muscle and heart disorders. The role of non-enzymatic glycation of αB-crystallin in the development of pathological processes has been little studied. The aim of this study was to evaluate the influence of enhanced methylglyoxal (MGO) level on the functions of αB-crystallin. We modified recombinant αB-crystallin (aB-Cr) by MGO and investigated the effect of this modification on different functions
