Hardwired survival algorithms: RelBE1 and MadR1 couple carbon state to metabolic persistence in Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb) survives host-imposed stress through dynamic metabolic adaptation and growth modulation. Toxin-antitoxin (TA) systems and cell cycle regulators, including RelBE1 and MadR1, are increasingly recognized as contributors to these processes, yet their physiological roles remain unclear. In this perspective, we propose that these systems function as genomically encoded metabolic control modules that couple carbon source availability to persistence. The relBE1 loci, encoded adjacent to the α-ketoglutarate decarboxylase gene (kgd), and the cell division regulator, madR

Physiological and Molecular Aspects of Two Thymus Species Differently Sensitive to Drought Stress.

Drought is one of the most important threats to plants and agriculture. Here, the effects of four drought levels (90%, 55%, 40%, and 25% field capacity) on the relative water content (RWC), chlorophyll and carotenoids levels, and mRNA gene expression of metabolic enzymes in Thymus vulgaris (as sensitive to drought) and Thymus kotschyanus (as a drought-tolerant species) were evaluated. The physiological results showed that the treatment predominantly affected the RWC, chlorophyll, and carotenoids content. The gene expression analysis demonstrated that moderate and severe drought stress had grea

Antioxidant Therapy Significantly Attenuates Hepatotoxicity following Low Dose Exposure to Microcystin-LR in a Murine Model of Diet-Induced Non-Alcoholic Fatty Liver Disease.

We have previously shown in a murine model of Non-alcoholic Fatty Liver Disease (NAFLD) that chronic, low-dose exposure to the Harmful Algal Bloom cyanotoxin microcystin-LR (MC-LR), resulted in significant hepatotoxicity including micro-vesicular lipid accumulation, impaired toxin metabolism as well as dysregulation of the key signaling pathways involved in inflammation, immune response and oxidative stress. On this background we hypothesized that augmentation of hepatic drug metabolism pathways with targeted antioxidant therapies would improve MC-LR metabolism and reduce hepatic injury in NAF

Phenylacetylcarbinol biotransformation by disrupted yeast cells using ultrasonic treatment in conjunction with a dipropylene glycol mediated biphasic emulsion system.

Biotransformation of a pharmaceutical precursor, phenylacetylcarbinol (PAC), could be achieved by frozen-thawed Candida tropicalis whole cells (FT-WHC). The treatment of FT-WHC, which contained intracellular pyruvate decarboxylase (PDC) enzyme, using high-power ultrasonication with varying amplitudes were compared with glass bead attrition (GBA) as well as control for the release of PDC. Ultrasonication at 20% amplitude (Ult20) proved to be the most effective, resulting in the highest volumetric and specific PDC activities of 0.210 ± 0.004 U/mL and 0.335 ± 0.033 U/mg protein, respectively. A case of pyruvate carboxylase deficiency with later prenatal diagnosis of an unaffected sibling.

A severely mentally retarded infant with congenital lactic acidosis due to pyruvate carboxylase deficiency is reported. The patient suffered from vomiting and convulsions soon after birth and developed severe mental and motor retardation at 3 months of age. The persistent elevation of pyruvate and lactate in both blood and cerebrospinal fluid and hyperalanaemia suggested an impairment of pyruvate oxidation. The enzyme activities of pyruvate carboxylase in both liver tissues and cultured skin fibroblasts of the patient revealed values of about 5% of controls. However, pyruvate dehydrogenase and

Hypoglycemic effect of deoxynojirimycin-polysaccharide on high fat diet and streptozotocin-induced diabetic mice via regulation of hepatic glucose metabolism.

Type 2 diabetes mellitus (T2DM) is currently considered a worldwide epidemic and finding effective therapeutic strategies against this disease is highly important. A deoxynojirimycin-polysaccharide mixture (DPM) has previously been shown to exert hypoglycemic effects on alloxan- or streptozotocin (STZ)-induced diabetic mice. The purpose of the present study was to evaluate the therapeutic effects and underlying mechanism(s) of DPM on T2DM induced by high fat diet following low-dose STZ treatment in mice. After daily oral treatment of diabetic mice with DPM (150 mg/kg b.w.) for 90 d, significan

Increased hydrogen and ethanol production in transformants of the filamentous cyanobacterium Phormidium lacuna.

Research on biofuel production by cyanobacteria has largely focused on unicellular species. To date, there are no reports on engineered filamentous cyanobacteria for bioethanol or biohydrogen production. However, biofilm formation in filamentous cyanobacteria could provide an advantage for biotechnological applications. We used natural transformation and homologous recombination to establish recombinant expression at the psaD locus of the filamentous cyanobacterium Phormidium lacuna. A similar approach has already been successful in the unicellular cyanobacterium Synechocystis sp. PCC 6803 (Ap

Enhanced Biomass and Protein Synthesis in Engineered Cyberlindnera jadinii Growing on Ethanol/Acetate: Metabolic Engineering and Transcriptomic Mechanism.

Producing single-cell protein (SCP) from syngas-derived ethanol and acetate offers a sustainable solution to global protein shortages, yet microbial utilization mechanisms for these mixtures remain underexplored. This study establishes a systematic bioconversion strategy using Cyberlindnera jadinii TU389. To mitigate acetaldehyde accumulation during ethanol metabolism, we engineered the strain TU546 to overexpress acylating acetaldehyde dehydrogenase (ADA6). TU546 achieved a maximum biomass of 46.7 g/L and a protein yield of 21.69 g/L, representing enhancements of 28.16% and 23.02% over the wi

Carbon dioxide enhances Akkermansia muciniphila fitness and anti-obesity efficacy in high-fat diet mice.

Numerous studies and clinical applications have underscored the therapeutic potential of the indigenous gut bacterium Akkermansia muciniphila in various diseases. However, our understanding of how Akkermansia muciniphila senses and responds to host gastrointestinal signals remains limited. Here, we demonstrate that A. muciniphila exhibits rapid growth, facilitated by its self-produced carbon dioxide (CO₂), with key enzymes such as glutamate decarboxylase, carbonic anhydrase, and pyruvate ferredoxin oxidoreductase playing pivotal roles. Additionally, we design a novel delivery system, comprisin

Intrahepatic glucose: a requirement for neonatal ODC induction by specific hormones.

We have shown previously that short-term nutritional deprivation causes a tissue-specific loss of liver ornithine decarboxylase (ODC) induction after isoproterenol, phenylephrine, or glucagon administration in rat pups. To examine the role of nutrition in the regulation of hepatic ODC, we tested the ability of intragastric nutrient administration to reverse nutritionally related deficits in the ODC response to hormonal challenge. Intragastric whole milk was effective in restoring ODC induction and accumulation of its immediate product, putrescine, in response to isoproterenol administration. G

Metabolic rewiring promotes anti-inflammatory effects of glucocorticoids.

Glucocorticoids represent the mainstay of therapy for a broad spectrum of immune-mediated inflammatory diseases. However, the molecular mechanisms underlying their anti-inflammatory mode of action have remained incompletely understood1. Here we show that the anti-inflammatory properties of glucocorticoids involve reprogramming of the mitochondrial metabolism of macrophages, resulting in increased and sustained production of the anti-inflammatory metabolite itaconate and consequent inhibition of the inflammatory response. The glucocorticoid receptor interacts with parts of the pyruvate dehydrog

Engineering Clostridium ljungdahlii for direct acetoin production from one-carbon gases.

Autotrophic gas-fermenting Clostridium species are recognized as key chassis for constructing C1 (CO/CO2) cell factories. However, their limited natural product spectrum restricts broader industrial applications, necessitating an expansion of their product repertoire. Acetoin is widely used in the food, pharmaceutical, and chemical synthesis industries, but its conventional sugar‑based production route is cost-prohibitive. Thus, low-cost alternative carbon sources are needed, and C1 gases offer a promising solution. In this study, using the gas‑fermenting bacterium Clostridium ljungdahlii as t

Glucosamine Yield Improvement in Engineered Saccharomyces cerevisiae with Ethanol Yield Reduction by Carbon Flux Redistribution.

Glucosamine (GlcN) is an essential amino monosaccharide widely used in pharmaceuticals, nutraceuticals, and cosmetics. Microbial fermentation presents a sustainable alternative to its traditional chemical production. However, in Saccharomyces cerevisiae, competitive carbon flux towards ethanol significantly limits GlcN yields. In this study, an S. cerevisiae strain for GlcN biosynthesis was engineered by integrating heterologous GlmD (glucosamine-6-phosphate deaminase) and GlmP (glucosamine-6-phosphate phosphatase) genes. To redirect carbon flux, the pyruvate decarboxylase genes pdc1, pdc5, an

Thiamine antivitamins--an opportunity of therapy of fungal infections caused by Malassezia pachydermatis and Candida albicans.

Severe skin diseases and systemic fungaemia are caused by Malassezia pachydermatis and Candida albicans respectively. Antifungal therapies are less effective because of chronic character of infections and high percentage of relapses. Therefore, there is a great need to develop new strategies of antifungal therapies. We previously found that oxythiamine decreases proliferation of yeast (Saccharomyces cerevisiae), therefore we suggest that thiamine antivitamins can be considered as antifungal agents. The aim of this study was the comparison of thiamine antivitamins (oxythiamine, amprolium, thioc

The m6A Reader MhYTP2 Regulates MdERF54 mRNA Stability and Contributes to Hypoxia Tolerance in Apple (Malus domestica).

Hypoxia is one of the main challenges in apple (Malus domestica) cultivation. However, breeding hypoxia-tolerant cultivars demands a thorough understanding of the responses of apple trees to low oxygen supply. Studies have indicated that N6-methyladenosine (m6A) reader regulates plant stress response by binding to their corresponding mRNA targets with m6A modification. The present study investigated the function and mechanism of apple m6A reader MhYTP2 under hypoxia stress. Here, we found that the overexpression of MhYTP2 improved hypoxia resistance in apple. Human Chorionic Gonadotropin-Stimulated Interleukin-4-Induced-1 (IL4I1) Promotes Human Decidualization via Aryl Hydrocarbon Receptor.

Decidualization is necessary for the successful establishment of early pregnancy in rodents and humans. Disturbed decidualization results in recurrent implantation failure, recurrent spontaneous abortion, and preeclampsia. Tryptophan (Trp), one of the essential amino acids in humans, has a positive effect on mammalian pregnancy. Interleukin 4-induced gene 1 (IL4I1) is a recently identified enzyme that can metabolize L-Trp to activate aryl hydrocarbon receptor (AHR). Although IDO1-catalyzed kynurenine (Kyn) from Trp has been shown to enhance human in vitro decidualization via activating AHR, wh

Isolation and sequencing of a cDNA encoding the decarboxylase (E1)alpha precursor of bovine branched-chain alpha-keto acid dehydrogenase complex. Expression of E1 alpha mRNA and subunit in maple-syrup-urine-disease and 3T3-L1 cells.

A cDNA clone encoding the entire decarboxylase (E1)alpha precursor of the bovine branched-chain alpha-keto acid dehydrogenase complex has been isolated from a lambda ZAP library prepared from bovine liver poly(A)+ RNA. Nucleotide sequencing indicates that this E1 alpha cDNA clone is 1821 base pairs (bp) in length with an open reading frame of 1365 bp and a 3'-untranslated region of 356 bp. A polyadenylation signal of the type AATAAA is located 27 bp upstream of the start of a poly(A)+ tail. There is a pair of identical 32-bp direct repeats of unknown function at the 5'-end of the cDNA.
