Qualitative and quantitative analysis of muscarinic acetylcholine receptors in the piebald lethal mouse model of Hirschsprung's disease.

Cholinergic innervation in the aganglionic bowel of the piebald lethal mouse model of Hirschsprung's disease was investigated by analysis of muscarinic acetylcholine receptors before and after administration of hexamethonium. After hexamethonium administration in the normal rectum, the maximum specific binding (Bmax) of [3H]quinuclidinyl benzilate increased from 196.6 to 346.2 fmol/mg protein without affecting the dissociation constant. This increase of muscarinic acetylcholine receptors was associated with a decrease in the 50% effective dose (ED50) of contractile response to oxotremorine fro

Evidence of muscarinic acetylcholine receptors and GTP-binding proteins in peritoneal mesothelial cells in vitro.

Preliminary evidence of muscarinic acetylcholine receptors (AchRs) in rabbit and human peritoneal mesothelial cells grown in tissue culture is reported. Atropine displaceable binding of L-quinuclidinyl[phenyl-4-3H]-benzilate (QNB), an antagonist that binds to all AchR subtypes, and [N-methyl-3H]-4-diphenylacetoxy-N-methyl-piperidine methiodide (4-DAMP), an antagonist specific for AchR subtypes 1 and 3, to homogenates of mesothelial cells was approximately 400 and 120 fmol ligand bound/milligram cell protein, respectively. A similar value for specific [3H]-QNB binding was observed in living cel

Muscarinic cholinergic and histamine H1 receptor binding of phenothiazine drug metabolites.

In vitro binding affinities of chlorpromazine, fluphenazine, levomepromazine, perphenazine and some of their metabolites for dopamine D2 receptors, alpha 1- and alpha 2 adrenoceptors in rat brain were previously reported from our laboratories. The present study reports the in vitro binding affinities of the same compounds for muscarinic cholinergic receptors and for histamine H1 receptors in rat brain, using 3H-quinuclidinyl benzilate and 3H-mepyramine as radioligands. Chlorpromazine, levomepromazine, and their metabolites had 5-30 times higher binding affinities for muscarinic cholinergic rec

Decreased binding of the muscarinic antagonist [3H]N-methylscopolamine in mouse brain following acute treatment with an organophosphate.

The effect of the irreversible acetylcholinesterase inhibitor diisopropylfluorophosphate (DFP) on mouse brain muscarinic acetylcholine receptors was assessed using the muscarinic antagonists [3H]N-methylscopolamine [( 3H]NMS) and [3H]quinuclidinyl benzilate [( 3H]QNB). No alteration in the maximal binding capacity (Bmax) or equilibrium dissociation constant (KD) was observed in brain homogenates obtained from mice 12 h after a single injection of DFP when [3H]NMS was employed as the ligand. However, one administration of DFP produced a 24 and 33% decrease in Bmax as measured by [3H]NMS binding

Urinary and brain beta-carboline-3-carboxylates as potent inhibitors of brain benzodiazepine receptors.

Benzodiazepines probably exert their anxiolytic, hypnotic, and anticonvulsant effects by interacting with brain-specific high-affinity benzodiazepine receptors. In searching for possible endogenous ligands for these receptors we have purified a compound 10(7)-fold from human urine by extractions, treatment with hot ethanol, and column chromatography. The compound was identified as beta-carboline-3-carboxylic acid ethyl ester (IIc) by mass spectrometry, NMR spectrometry, and synthesis; IIc was also isolated from brain tissues (20 ng/g) by similar procedures. Very small concentrations of IIc dis

[Progress in the study of muscarinic acetylcholine receptors; establishment of the subtypes or subgroups].

Since acetylcholine (ACh) was identified as a neurotransmitter at parasympathetic nerve terminals by pioneering pharmacologists such as O. Schmiedeberg, R. Hunt, O. Loewi and H.H. Dale, muscarinic acetylcholine receptors (mACh-R) serving as a transducer of muscarinic action have been assumed to exist. After many tries to identify the mACh-R, it's existence was established by the group of S.H. Snyder, who employed binding assays with the radioligand 3H-QNB. The presence of a neuronal (M1) and a peripheral (M2) mACh-R was suggested from the action of an M1-specific agonist, McN-A-343; and this o

The change in the threshold for short-term desensitization in isolated smooth muscle cells showing an all-or-none response to acetylcholine.

1. Isolated smooth muscle cells from guinea-pig taenia caecum were prepared by collagenase digestion. The cells showed an all-or-none response to acetylcholine (ACh) under our experimental conditions. 2. Desensitized cells showed an all-or-none response but required a higher concentration of ACh for induction of contraction, indicating that the desensitization was due to a change in the threshold concentration. 3. In [3H]-quinuclidinyl benzilate ([3H]-QNB) binding to the desensitized cells, KD and Bmax were not significantly different from those estimated in the control cells. The competitive

Imprinting. A study of cholinergic receptor sites in parts of the chick brain.

The distribution of cholinergic receptor binding sites was studied in certain areas of the chick brain, especially that part (IMHV) of hyperstriatum ventrale which has previously been implicated in the imprinting process. There was relatively little binding of [125I]-alpha-bungarotoxin in this region, as shown using an autoradiographic technique. In contrast muscarinic receptor sites, with an affinity for [3H]-quinuclidinyl benzilate (QNB), are present in relatively high concentration in a medial forebrain sample containing IMHV and also in a visual projection area, the hyperstriatum accessori

Antagonism by antimuscarinic and neuroleptic compounds at the five cloned human muscarinic cholinergic receptors expressed in Chinese hamster ovary cells.

We determined the affinity and selectivity of binding for 24 compounds: nine antimuscarinics (including some antiparkinson drugs) and 15 neuroleptics (including the atypical compounds clozapine, fluperlapine, melperone, rilapine, risperidone, tenilapine, tiosperone and zotepine) at the five human muscarinic receptor subtypes expressed in Chinese hamster ovary cells. Equilibrium dissociation constants (Kd) were obtained from competitive radioligand binding studies with [3H]quinuclidinyl benzilate and membranal preparations of these cells. As expected, QNB had the highest affinity of the compoun

Muscarinic binding and choline acetyltransferase in postmortem brains of demented patients.

Postmortem human brain samples were taken from non-neurological controls as well as demented subjects who died with Alzheimer's disease (AD), multi-infarct dementia (MID), or a combination of AD and MID dementia (MIXED). Choline acetyltransferase (ChAT) activity was measured radiometrically using [1-14C]acetyl-coenzyme A as the substrate, muscarinic binding was assayed with [3H]quinuclidinyl benzilate, and the proportion of binding associated with high affinity agonist sites was measured by carbamylcholine displacement of the radioligand. Effect of estrogen on muscarinic acetylcholine receptor expression in rat myometrium.

We report the effect of acute estrogen treatment in the expression of muscarinic acetylcholine receptors (mAChRs) in myometrium. Strips were obtained from rats in estrus (control) and treated with estrogen, 24h before the experiments. Reverse transcriptase-polymerase chain reaction (RT-PCR) was performed and m2, m3 and m5 mAChR mRNA subtypes were detected in myometrium from both groups. [(3)H]Quinuclidinyl benzilate [(3)HQNB] binding studies indicated that estrogen treatment did not change the affinity and density of mAChRs in myometrial membranes. Distribution of cholinergic and dopaminergic receptors in rainbow trout pineal gland.

The involvement of multiple receptors in modulating the function of the pineal gland was investigated by searching for dopaminergic and cholinergic receptors in trout pineal gland. Dopamine D1 and D2 receptors were measured using [3H]SCH23390 and [3H]spiperone, respectively. Muscarinic and nicotinic cholinergic receptors were measured using quinuclidinyl benzilate ([3H]QNB) and [3H]methylcarbamyl choline, respectively. High-affinity choline uptake sites were measured using [3H]hemicholinium-3. The distribution of dopaminergic receptors varied throughout the pineal gland in that the density of

Autoradiographic localization of muscarinic cholinergic receptors in rat brainstem.

We have localized muscarinic cholinergic receptors in the rat brainstem by a light microscopic autoradiographic method. Initially, we examined the kinetics and specificity of the binding of radiolabeled quinuclidinyl benzilate ([3H]QNB) in slide-mounted, frozen tissue sections to determine that we were labeling the muscarinic cholinergic receptors as they had been described previously in biochemical pharmacological studies. Subsequently, using optimal binding parameters to obtain high specific to nonspecific ratios, we labeled tissue sections and generated autoradiograms by the apposition of e

Muscarinic receptors mediating contraction of female mouse urinary bladder: effects of oestrogen.

Muscarinic receptors mediating contraction of bladder detrusor muscle from female mice were examined. Mice were untreated (A) or treated with oestradiol cypionate (200 microg/kg) 24 h (B) or 96 h (C) before experimentation, or were pregnant (day 17) (D). Saturation radioligand binding experiments using [(3)H]quinuclidinyl benzilate ([(3)H] QNB) indicated similar muscarinic receptor densities and affinities in bladders from groups A and B. Neither oestrogen treatment nor pregnancy altered pD(2) estimates for methacholine. Maximum responses to methacholine and high-K(+) physiological salt soluti

Muscarinic receptor regulation of Ca2+ mobilization in a human salivary cell line.

We have studied receptor-mediated Ca2+ mobilization in an established exocrine epithelial cell line (HSG-PA) derived from a human submandibular gland. These cells possess a single class of high-affinity muscarinic cholinergic receptors identified using [3H]-quinuclidinyl-benzilate (Kd = 0.17 +/- 0.07 nmol/l; Bmax = 37 +/- 2 fmol/mg protein; n = 3). The muscarinic agonist carbachol elicits a concentration dependent increase of [3H]-inositol trisphosphate in HSG-PA cells (100 mumol/l; greater than 2 fold by 30 s). Carbachol also results in a rapid, approximately 5-fold increase in cytosolic [Ca2

Changes in muscarinic receptors and amylase secretion in regenerating acinar cells of rat pancreas.

Atrophic pancreatic acinar cells of copper-deficient rats recovered after copper supplementation. To clarify whether the recovered cells are regenerative and amylase secretion via muscarinic receptors alters, [3H]-thymidine uptake, [3H]-quinuclidinyl benzilate binding and amylase release were studied in these rats and compared with the values of 5-day-old and 21-week-old rats. The [3H]-thymidine uptake study indicated that the recovered acinar cells were regenerative. The number of muscarinic receptors in regenerating cells was equal to that in mature cells and was one third in cells of 5-day-

Copper-induced alteration of muscarinic binding in bovine adrenal medulla.

In microsomes of bovine adrenal medulla, there were two affinity binding sites for carbamylcholine with a high and a low affinity. Copper (1-10 microM) largely enhanced the affinity of carbamylcholine at the low affinity binding site, with a slight increase in the affinity at the high affinity binding site. On the other hand, copper slightly decreased the binding affinity of pirenzepine and atropine. Thus, low concentrations of copper modulate the muscarinic receptors in the adrenal medulla by selectively increasing agonist affinity.

Characterization and gene organization of Taiwan banded krait (Bungarus multicinctus) gamma-bungarotoxin.

gamma-Bungarotoxin was isolated from Bungarus multicinctus (Taiwan banded krait) venom using a combination of chromatography on a SP-Sephadex C-25 column and a reverse-phase high-performance liquid chromatography column. Circular dichroism (CD) measurement revealed that its secondary structure was dominant with beta-sheet structure as is that of snake venom alpha-neurotoxins and cardiotoxins. gamma-Bungarotoxin exhibits activity on inhibiting the binding of [3H]quinuclidinyl benzilate to the M2 muscarinic acetylcholine receptor subtype, and competes weakly with radioiodinated alpha-bungarotoxi
