Teratogenic viruses have increased public health importance with the emergence of Zika virus and a recent decline in rubella virus vaccination. Of the seven viruses known to cause birth defects in humans, three are mosquito-borne pathogens. Ethical oversight, compliance, rising costs, and the need for specialized training slow the pace of study of these human pathogens compared to study of similar teratogenic viruses in plants. Plant viruses have served as models for human viruses which can be applied to animal systems. This review describes the similar features of plant and animal teratogenic

Long-distance molecular transport contributes to coordination between locally stressed tissues and distal organs, but assigning causal systemic-signaling functions to mobile RNAs and extracellular vesicle (EV)-associated molecules remains experimentally challenging. This review critically evaluates the evidence required to distinguish molecular association, source assignment, physical movement, recipient activity, persistence, and causal function in sequential-stress systems. We assess source-resolved RNA tracking, graft-based mobility assays, sequence-assignment and contamination errors, mole

BACKGROUND: miRNAs are a type of conserved, small RNA molecule that regulate gene expression and play an important role in the growth and development of plants. miRNAs are involved in seed germination, root development, shoot apical meristem maintenance, leaf development, and flower development by regulating various target genes. However, the role of miRNAs in the mechanism of tea plant flower sterility remains unclear. Therefore, we performed miRNA sequencing on the flowers of fertile male parents, female parents, and sterile offspring.
RESULTS: A total of 55 known miRNAs and 90 unknown miRN

Kiwifruit is a dioecious woody liana fruit tree, and the non-fruitfulness of male plants leads to a great deal of blindness in the selection of male plants in crossbreeding. In this study, we induced the development of male plant ovary by externally applying plant growth regulators (PGRs) and performed histological observation, phytohormone content determination and transcriptome analysis on the abortive ovary of the male kiwifruit (Con), the ovary of the female kiwifruit (Fem) and the PGR-induced developing ovary of the male kiwifruit (PT). Histological analysis showed that the Con ovary was

RNA editing is a post-transcriptional modification process, the chloroplast genes of which are involved in the process of chloroplast development in plant. However, the RNA editing sites of chloroplast genes remains unknown. In this study, we identified 39 RNA editing sites in 18 chloroplast genes from chloroplast genome of C. sinensis. Furthermore, the feature, structures and specificity of RNA editing sites were systematic analyzed. The differential editing efficiency were examined at 11 RNA editing sites among C. sinensis var. sinensis 'Huabai 1', 'Baiye 1' and 'Longjing 43'. Meanwhile, we

The leaf-mining insect Tuta absoluta is a highly destructive pest that deploys candidate effector proteins to modulate host defences; however, the molecular mechanisms underlying this manipulation remain poorly understood. In this study, we characterise TaCYPE1, a predicted secreted cyclophilin-like effector, and demonstrate its role in promoting insect virulence. Transient expression of TaCYPE1 in Nicotiana benthamiana suppresses pattern-triggered immunity (PTI), including INF1-induced cell death, reactive oxygen species (ROS) burst, and defence-related gene expression. Notably, this immune s

As the most prevalent RNA modification in eukaryotes, N6-methyladenosine (m6A) plays a crucial role in regulating various biological processes in plants, including embryonic development and flowering. However, the function of m6A RNA methyltransferase in moso bamboo remains poorly understood. In this study, we identified two m6A methyltransferases in moso bamboo, PheMTA1 and PheMTA2. Overexpression of PheMTA1 and PheMTA2 significantly promoted root development and enhanced salt tolerance in rice. Using the HyperTRIBE method, we fused PheMTA1 and PheMTA2 with ADARcdE488Q and introduced them int

Plants respond to abiotic stress stimuli, such as water deprivation, through a hierarchical cascade that includes detection and signaling to mediate transcriptional and physiological changes. The phytohormone abscisic acid (ABA) is well-characterized for its regulatory role in these processes in response to specific environmental cues. ABA-mediated changes in gene expression have been demonstrated to be temporally-dependent, however, the genome-wide timing of these responses are not well-characterized in the agronomically important crop plant Zea mays (maize). ABA-mediated responses are synerg

Plant pathogenesis-related 1 (PR1) proteins are members of the CAP protein superfamily and are widely used immune markers. The precise function and mode of action of CAP proteins, however, remains poorly understood. In Cytospora chrysosperma, the causative agent of poplar canker disease, three CAP family members, CcCAP1-3, have been identified. CcCAP1 is required for fungal virulence and suppresses the plant immune response to facilitate fungal colonisation, in a nuclear-localisation dependent way. In this study, we functionally characterised the other two members of the C. chrysosperma CAP fa

Owing to their safety, plant-derived nanovesicles (PDNVs), particularly those derived from edible plants, are expected to be natural delivery carriers for microRNAs (miRNAs). Among PDNVs, Citrus limon-derived extracellular vesicle-like nanovesicles have been shown to exert antitumor effects. We previously reported that PDNVs derived from C. limon L. exert selective inhibitory effects on the proliferation of p53-inactivated colon cancer cells. However, miRNAs in C. limon L.-derived nanovesicles have not yet been identified, and it remains unclear which miRNAs are contained in these PDNVs. We he

Gene targeting (GT) is a useful technology for accurate genome engineering in plants. A reproducible approach based on a positive-negative selection system using hygromycin resistance and the diphtheria toxin A subunit gene as positive and negative selection markers, respectively, is now available. However, to date, this selection system has been applied exclusively in rice (Oryza sativa). To establish a universally applicable positive-negative GT system in plants, we designed a selection system using a combination of neomycin phosphotransferaseII (nptII) and an antisense nptII construct. The

Wastewater-based epidemiology (WBE) is a powerful method that allows community surveillance to identify diseases/pandemic dynamics in a city, especially in metropolitan areas with high overpopulation. This study investigated the detection and quantification of SARS-CoV-2 RNA in wastewater and sewage sludge in two different wastewater treatment plants (WWTPs) in Istanbul, the 5th largest city in the world, during the COVID-19 pandemic. Statistical analysis was performed to examine the relationships between SARS-CoV-2 concentrations and COVID-19 case numbers, seasonal variations, and key WWTP pa

High concentrations of sulfur dioxide (SO(2) ) as an air pollutant, and its derivative sulfite, cause abiotic stress that can lead to cell death. It is currently unknown to what extent plant fumigation triggers specific transcriptional responses. To address this question, and to test the hypothesis that sulfite oxidase (SO) is acting in SO(2) detoxification, we compared Arabidopsis wildtype (WT) and SO knockout lines (SO-KO) facing the impact of 600 nl l(-1) SO(2) , using RNAseq to quantify absolute transcript abundances. These transcriptome data were correlated to sulfur metabolism-related en

RNA silencing plays a crucial role in defending against viral infections in diverse eukaryotic hosts. Despite extensive studies on core components of the antiviral RNAi pathway such as DCLs, AGOs and RDRs proteins, host factors involved in antiviral RNAi remain incompletely understood. In this study, we employed the proximity labelling approach to identify the host factors required for antiviral RNAi in Nicotiana benthamiana. Using the barley stripe mosaic virus (BSMV)-encoded γb, a viral suppressor of RNA silencing (VSR), as the bait protein, we identified the DEAD-box RNA helicase RH20, a br

Sensitive quantification of Tobacco Mosaic Virus RNA (TRNA) is imperative for preventing large-scale agricultural losses. In this study, we engineered a sophisticated quenching-based electrochemiluminescence (ECL) biosensor by integrating a high-performance CdTe@ZnS nanoscaffold with a target-triggered tripedal DNA walker. To ensure a robust and stable initial luminophore density, multi-walled carbon nanotubes (MWCNTs) were utilized to anchor CdTe@ZnS core-shell quantum dots, where the ZnS shell effectively passivates surface defects and prevents cadmium leakage. Upon recognition of TRNA, a ca

Deep RNA-Seq profiling, a revolutionary method used for quantifying transcriptional levels, often includes non-specific transcripts from other co-existing organisms in spite of stringent protocols. Using the recently published walnut genome sequence as a filter, we present a broad analysis of the RNA-Seq derived transcriptome profiles obtained from twenty different tissues to extract the biodiversity and possible plant-microbe interactions in the walnut ecosystem in California. Since the residual nature of the transcripts being analyzed does not provide sufficient information to identify the e

Chromatin-associated RNAs (caRNAs) serve as central hubs for regulation within the cell nucleus and are involved in key biological processes such as transcriptional homeostasis, chromatin remodeling, and transgenerational inheritance. In recent years, the development of various genome-wide RNA-chromatin interaction capture technologies has made it possible to resolve the spatial interaction networks between caRNAs and chromatin with high precision. This paper systematically reviews the concepts, functions, and interaction patterns of caRNA, and provides a comprehensive overview of the principl

BACKGROUND: Plants attenuate their responses to a variety of bacterial and fungal pathogens, leading to higher incidences of pathogen infection at night. However, little is known about the molecular mechanism responsible for the light-induced defence response; transcriptome data would likely facilitate the elucidation of this mechanism.
RESULTS: In this study, we observed diurnal changes in tomato resistance to Pseudomonas syringae pv. tomato DC3000 (Pto DC3000), with the greatest susceptibility before midnight. Nightly light treatment, particularly red light treatment, significantly enhanced

Strawberry plant tissues and particularly fruit material are rich in polysaccharides and polyphenolic compounds, thus rendering the isolation of nucleic acids a difficult task. This work describes the successful modification of a total RNA extraction protocol, which enables the isolation of high quantity and quality of total RNA from small amounts of strawberry leaf, root and fruit tissues. Reverse-transcription polymerase chain reaction (RT-PCR) amplification of GAPDH housekeeping gene from isolated RNA further supports the proposed protocol efficiency and its use for downstream molecular app

The young leaves and shoots of albino tea cultivars are usually characterized as having a yellow or pale color, high amino acid, and low catechin. Increasing attention has been paid to albino tea cultivars in recent years because their tea generally shows high umami and reduced astringency. However, the genetic mechanism of yellow-leaf variation in albino tea cultivar has not been elucidated clearly. In this study, bulked segregant RNA-seq (BSR-seq) was performed on bulked yellow- and green-leaf hybrid progenies from a leaf color variation population. A total of 359 and 1134 differentially exp
