Molecular characterization of Moniezia expansa (Rudolphi, 1810) (Cestoda: Anoplocephalidae) from a captive common eland (Tragelaphus oryx) in Taiwan.

The genus Moniezia comprises globally distributed parasitic cestodes that cause significant economic losses in domestic ruminants, yet its molecular characterization in wildlife remains limited. This study provides the first molecular confirmation of M. expansa in zoo animals in Taiwan. Specimens were obtained from a captive-born common eland (Tragelaphus oryx) and a domestic goat (Capra hircus); those from the eland underwent morphological examination of gravid proglottids, while all samples were analyzed genetically. Using the 18S ribosomal RNA, 28S ribosomal RNA, cytochrome c oxidase subuni

SARCOCYSTIS CANADENSIS N. SP. (PROTOZOA: SARCOCYSTIDAE) FROM LIMB MUSCLES OF THE CANADA LYNX (LYNX CANADENSIS).

Most species of endotherms and some ectotherms harbor muscle cysts (sarcocysts) of 1 or more species of apicomplexan parasites in the genus Sarcocystis. We report a novel species discovered in the muscles of a Canada lynx (Lynx canadensis). We found microscopic sarcocysts in limb muscles of 1 of 3 endangered Canada lynx from Minnesota. The sarcocyst wall was relatively thin (<2 µm) and had short, stubby protrusions with apical thickening. We characterized these further by sequencing the 18S, 28S, and ITS1 portions of nuclear ribosomal RNA genes. The sequences of each marker perfectly matched t

Discovery of new species of mesoparasitic pennellid (Copepoda: Siphonostomatoida) from the endemic mesopelagic lightfish Vinciguerria mabahiss in the Red Sea.

A new species of the genus Cardiodectes Wilson, 1917 (Siphonostomatoida: Pennellidae), Cardiodectes tofaili n. sp., is described based on 13 adult females from ten specimens of the endemic lightfish Vinciguerria mabahiss (Stomiiformes: Phosichthyidae). These hosts were inadvertently captured by a remotely operated vehicle at depths of 454-645 m in the pelagic waters of the Saudi Arabian Red Sea. The new species is placed under the "rubosus" group, characterized by possession of a trunk without a discrete abdomen. It is distinguished from its 12 congeners within this group by having a short nec

Genetic diversity and population structure of penis fish (Urechis unicinctus) based on mitochondrial and nuclear gene markers.

Urechis unicinctus is distributed only in Bohai Gulf of China and Korean and Japanese coast. The wild populations of this species have sharply declined in China and Japan. We collected 105 samples from six localities of Bohai Gulf and Korea coast, and investigated genetic diversity and population structure with mitochondrial COI, 16S-rRNA and nuclear 28S-rRNA gene fragments. Genetic diversity of U. unicinctus based on COI sequences was still high (Hd: 0.9595, π: 0.0101), however, 28S-rRNA gene sequences showed low level of genetic diversity (Hd: 0.4084, π: 0.0007). Based on COI sequences, FST

High prevalence of Ancylostoma caninum infection in black-eared opossums (Didelphis aurita) in an urban environment.

Marsupials of the genus Didelphis, such as black-eared opossums (Didelphis aurita), are common synanthropic animals in urban areas of Brazil. These marsupials are frequently parasitized by numerous helminth species, including ancylostomatid nematodes. This study aimed to report the occurrence of Ancylostoma caninum in black-eared opossums captured in an urban environment of Southeastern Brazil and discuss the potential impact of these findings for public health. From January to June 2019, we collected fecal samples from 49 restrained opossums and evaluated by a simple flotation method; Helmint

An unidentified species of Leucochloridium (Trematoda: Leucochloridiidae) found in an Amber snail (Succinea lauta) from Chiba Prefecture, Honshu, Japan.

Leucochloridium spp. are intriguing parasites due to their colorful, pulsating larval broodsacs in amber snails' eyestalks. The unusual appearance is believed to mimic caterpillars to attract insectivorous birds. Sporocysts of Leucochloridium sp. were discovered in an amber snail (Succinea lauta) in Chiba, located in the central region of Honshu, Japan. The broodsacs displayed a distinctive pattern, characterized by large dark brown spots, bands of the same color at the anterior end, and light brown vertical stripes running through them. Phylogenetic analyses were conducted based on DNA sequen

Extraction of total RNA from single-oocytes and single-cell mRNA sequencing of swine oocytes.

OBJECTIVE: Analyses of single oocytes are essential for a fine dissection of molecular features governing developmental competence. We adapted the phenol-chloroform procedure for the purification of total RNA from single oocytes. RESULTS: Key modifications include the use of Phasemaker™ tubes, a second chloroform wash of the aqueous phase, and the precipitation of the RNA with glyclogen in a 200 μl micro-centrifuge tube. Assessment of the RNA profile from single oocytes showed distinct peaks for 18S and 28S ribosomal subunits. This approach permitted the extraction of small RNAs from single o

Grillotia (Cestoda: Trypanorhyncha) plerocerci in an anglerfish (Lophius piscatorius) from the Tyrrhenian Sea.

Trypanorhynch cestodes are common parasites of marine fish with complicated life cycles which have been suggested as model taxa to study the evolution of marine helminth parasites and their life cycles. Among the Trypanorhyncha, the genus Grillotia includes 18 valid species, of which only four have been found in Mediterranean fish hosts. Morphological, histopathological, and molecular data are presented on a massive Grillotia plerocercus infection in an anglerfish (Lophius piscatorius) from the Tyrrhenian Sea. BLAST analysis of the 28S rDNA sequences revealed 99% similarity between specimens h

Description of Sarcocystis arvalis n. sp. from the Common Vole (Microtus arvalis) in Lithuania Using Morphological and Molecular Methods.

Records of Sarcocystis spp. diversity in rodents are relatively extensive; however, the increasing application of molecular approaches indicates that our current knowledge of these parasites remains incomplete. In the present study, morphological, genetic and phylogenetic data are provided on Sarcocystis arvalis n. sp. from the common vole (Microtus arvalis). Using light microscopy, the observed sarcocysts had a relatively thin (<1 μm) and smooth cyst wall. Via transmission electron microscopy, the sarcocyst wall thickness ranged from 0.7 to 1 μm, and the parasitophorous vacuolar membrane exhi

Morpho-molecular characterization and phylogenetic relationships of Encotyllabe percussa n. sp. (Monogenea: Capsalidae) from the spangled emperor Lethrinus nebulosus (Teleostei, Lethrinidae).

Encotyllabe percussa n. sp. is proposed based on morphology and DNA sequences analysis of ribosomal (18S, 28S) and mitochondrial (COI) gene fragments. Encotyllabe percussa n. sp. was found infecting the spangled emperor Lethrinus nebulosus (n = 75) with higher prevalence from Dibba, Musandam (Gulf of Oman) than in Dhofar Salalah (Indian Ocean), Oman (p = 0.03). The general morphology of E. percussa n. sp. resembles E. caballeroi, E. chironemi and E. spari, which exhibit pre-equatorial testes. However, E. percussa n. sp. shows unique morphological characteristics distinguishing from congeneric

The use of large and small subunits of ribosomal DNA in evaluating phylogenetic relationships between species of Cornudiscoides Kulkarni, 1969 (Monogenoidea: Dactylogyridae) from India.

Two partial regions of ribosomal DNA (28S and 18S) were used to evaluate genetic variations among the species of Cornudiscoides, viz. C. proximus, C. geminus and C. agarwali, all parasites of Mystus vittatus (Bagridae) from River Gomati, Ganges River basin, India. Our findings demonstrated that both the large and small ribosomal subunits are useful for species identification and genetic characterization of parasites, leading to resolution of inter/intra-relationships at generic and specific levels. The secondary structures of all three species for 28S and 18S rRNA genes contained exact pattern

The complete mitochondrial genome of 3 species of allocreadiids (Digenea, Allocreadiidae): characterization and phylogenetic position within the order Plagiorchiida.

Trematodes of the family Allocreadiidae are primarily found in the intestines of freshwater fishes around the world. The family includes 15 genera and c. 130 species. The last 2 decades have witnessed an increase in the genetic library of its species. Molecular data have been crucial for species delimitation and species description within Allocreadiidae and for understanding their evolutionary and biogeographical history and classification. Here, the mitogenomes of 3 species of allocreadiids were obtained using high throughput sequencing methods. Mitogenomes were compared with other members of

Uncovering further diversity of Ochoterenatrema Caballero, 1943 (Digenea: Lecithodendriidae) in South American bats.

Ochoterenatrema Caballero, 1943 is a genus of lecithodendriid digeneans that prior to this study included 8 species parasitic in bats in the Western Hemisphere. Species of Ochoterenatrema possess a unique morphological feature in form of the pseudogonotyl on the sinistral side of the ventral sucker. In this study, we describe 2 new species of Ochoterenatrema from bats in Ecuador. The new species are readily differentiated from their congeners by a combination of morphological characters, including the distribution of vitelline follicles, length of oesophagus, sucker ratio and the body shape, a

First report of white piedra caused by Cutaneotrichosporon debeurmannianum.

Piedras are small nodules found on the hair shafts. White piedra was initially described as caused by Trichosporon beigelii, which was later reclassified in several species. We describe the first case of white piedra caused by Cutaneotrichosporon debeurmannianum. Affected hairs were examined in natura with scanning electron microscopy, after gold metallization. The typical whitish cerebriform creamy colony was obtained in Sabouraud medium. Fungal genomic DNA extracted from cultures and locus was amplified with the NL1/NL4 primer pair from the D1/D2 region of the large ribosomal subunit (LSU) o

Stability of Nuclear and Mitochondrial Reference Genes in Selected Tissues of the Ambrosia Beetle Xylosandrus germanus.

The fungus-farming ambrosia beetle Xylosandrus germanus (Blandford) uses a pouch-like structure (i.e., mycangium) to transport spores of its nutritional fungal mutualist. Our current study sought to identify reference genes necessary for future transcriptome analyses aimed at characterizing gene expression within the mycangium. Complementary DNA was synthesized using selected tissue types from laboratory-reared and field-collected X. germanus consisting of the whole body, head + thorax, deflated or inflated mycangium + scutellum, inflated mycangium, and thorax + abdomen.
