Clinical phenomenology of episodic rage in children with Tourette syndrome.

OBJECTIVE: Episodic rage of unknown etiology causes significant morbidity in children with Tourette's syndrome (TS). Using modified Diagnostic and Statistical Manual of Mental Disorders (DSM-IV) criteria for intermittent explosive disorder (IED), we developed a screen and symptom questionnaire to explore rage attack phenomenology and to preliminarily investigate whether symptom clusters can identify clinical subgroups of TS children with rage attacks. METHODS: 48 children with TS between ages 7 and 17 years consecutively presenting with rage attacks completed the Rage Attacks Screen and Quest

Paroxetine treatment of episodic rages associated with Tourette's disorder.

BACKGROUND: Episodic rages have been estimated to occur in as many as 30% of patients with Tourette's syndrome (Tourette's disorder), but their treatment has never been systematically investigated. We report on the results of an open-label pilot study using paroxetine for the treatment of Tourette's disorder-associated rage episodes. METHOD: Forty-five Tourette's/rage patients (DSM-IV) were treated with paroxetine, specifically to control their rages. Other symptoms such as tics, attention-deficit/hyperactivity disorder (ADHD) and obsessive-compulsive disorder (OCD) were not targeted by this

Aging promotes a RAGE-dependent increase in breast cancer metastasis.

Breast cancer incidence and mortality increase with age, but the mechanisms linking aging to metastasis remain unclear. Most preclinical studies use young animal models, limiting insight into how the aged microenvironment influences cancer. Here, we show that aging markedly increases breast cancer metastasis in multiple mouse models, and this effect is dependent on host expression of the Receptor for Advanced Glycation End-products (RAGE). Aging increased tumor RAGE ligand levels, including S100A8/9 and AGEs, and induced RAGE-dependent changes in inflammatory, EMT, angiogenic, and extracellula

Rage gene -374T/A polymorphism in South Indian diabetic nephropathy.

Diabetic nephropathy (DN) is a major complication of type 2 diabetes mellitus (T2DM) and genetic variants like RAGE -374T/A may influence disease susceptibility. Therefore, it is of interest to analyze the association between RAGE -374T/A polymorphism and DN in T2DM patients from Kolar, Karnataka. Genotyping was done by PCR-RFLP and clinical parameters were compared. No significant association was found between the polymorphism and DN (p > 0.05), though DN patients showed elevated HbA1c, microalbuminuria and dyslipidemia. These findings suggest metabolic factors, rather than this specific RAGE

RAGE-mediated intestinal pro-inflammatory responses triggered by Giardia duodenalis.

Giardia duodenalis is a waterborne zoonotic protozoan that causes gastrointestinal inflammation. Giardiasis and metabolic illnesses share features such as chronic inflammation and intestinal symptoms. Receptor for advanced glycation end products (RAGE) signaling plays a role in metabolic illnesses and intestinal inflammatory responses. The presence of protozoan viruses can influence host immunological responses triggered by protozoa. However, these effects of G. duodenalis remain unknown. In this study, mice treated with the RAGE inhibitor FPS-ZM1 showed more severe intestinal damage, includin

Biofeedback-Based Videogame May Improve Rage Attacks in Tourette Syndrome.

Background: Approximately 20%-40% of individuals with Tourette syndrome (TS) have rage attacks (RAs), which are recurrent, explosive behavioral outbursts that can cause significant functional impairment. Despite the impact of RA in TS, there has been limited research on treatment, and most studies have focused on pharmacologic interventions. Nonpharmacologic interventions have the potential to improve symptoms with fewer side effects. Mightier, a video game-based biofeedback therapy, may help teach emotional regulation through heart rate control and has the potential to improve RA in youth wit

Metal-induced neurotoxicity in a RAGE-expressing C. elegans model.

Environmental and occupational metal exposure poses serious global concerns. Metal exposure have severally been associated with neurotoxicity and brain damage. Furthermore, receptor for advanced glycation end products (RAGE) is also implicated in neurological disorders, particularly those with altered glucose metabolism. Here, we examine potential compounding effect of metal exposure and RAGE expression on dopamine (DA) and serotonin (SER) neurons in C. elegans. In addition, we evaluate the effect of RAGE expression on DA and SER neurons in hyperglycemic conditions. Newly generated RAGE-expres

The RAGE/STAT5/autophagy axis regulates senescence in mesangial cells.

Renal aging and associated functional decline are associated with an increase in cellular senescence. Previous studies show a direct correlation between advanced glycation end products (AGEs) accumulation and renal aging, chronic kidney disease (CKD) and other nephropathies, although the underlying molecular mechanisms remain largely unclear. We found elevated levels of the receptor of advanced glycation end product (RAGE) as well as STAT5 in aged human kidneys, as well as in human mesangial cells aged artificially through AGEs. Furthermore, genetic and pharmacological ablation of STAT5 signif

RAGE signaling contributes to neuroinflammation in infantile neuronal ceroid lipofuscinosis.

Palmitoyl-protein thioesterase-1 (PPT1) deficiency causes infantile neuronal ceroid lipofuscinosis (INCL), a devastating childhood neurodegenerative storage disorder. We previously reported that neuronal apoptosis in INCL is mediated by endoplasmic reticulum-stress. ER-stress disrupts Ca(2+)-homeostasis and stimulates the expression of Ca(2+)-binding proteins. We report here that in the PPT1-deficient human and mouse brain the levels of S100B, a Ca(2+)-binding protein, and its receptor, RAGE (receptor for advanced glycation end-products) are elevated. We further demonstrate that activation of

Beyond "toxicity:" Rethinking rage, rivalry, and regulation in online games.

The nomenclature used to describe harmful behavior in gaming remains vague and inconsistently applied. "Toxicity" has become the dominant label, yet it functions as an umbrella term encompassing heterogeneous behaviors that differ by context, intent, and cultural norms. This conceptual ambiguity obscures underlying psychological mechanisms that are critical for designing effective interventions. This article looks beyond descriptors to focus on psychological dynamics and how they can inform effective prevention efforts. We conducted a conceptual analysis integrating mimetic theory with existin

RAGE contributes to persistent sepsis-induced muscle and mitochondrial alterations.

A majority of patients surviving sepsis develop muscle weakness. However, the underlying cellular and molecular pathways remain largely unexplored. To determine whether sepsis leads to long-term persistent muscular consequences and to identify the underlying mechanisms, we used a murine model of reanimated sepsis induced by intraperitoneal injection of a heterologous stool slurry. Muscles were analyzed 3 months later. The oxidative muscle exhibited reduced fatigue resistance and decreased mitochondrial respiration, without a corresponding reduction in mitochondrial OXPHOS proteins. Glycolytic

Increase of Soluble RAGE in Cerebrospinal Fluid following Subarachnoid Haemorrhage.

Receptors for advanced glycation end-products (RAGE) mediate the inflammatory reaction that follows aneurysmal subarachnoid haemorrhage. Soluble RAGE (sRAGE) may function as a decoy receptor. The significance of this endogenous anti-inflammatory mechanism in subarachnoid haemorrhage (SAH) remains unknown. The present study aims to analyse sRAGE levels in the cerebrospinal fluid (CSF) of SAH patients. sRAGE levels were assayed by ELISA kit in 47 CSF samples collected on post-SAH days 0-3, 5-7, and 10-14 from 27 SAH patients with acute hydrocephalus. CSF levels of sRAGE were compared with a cont

RAGE blockade and hepatic microcirculation in experimental endotoxaemic liver failure.

BACKGROUND: Activation of the receptor for advanced glycation endproducts (RAGE) causes sustained activation of multiple inflammatory pathways. Therefore, RAGE has potential as a new therapeutic target in sepsis. The aim of this study was to analyse whether RAGE blockade in vivo prevents microcirculatory dysfunction and subsequent tissue injury in endotoxaemic liver failure. METHODS: The hepatic microcirculation was analysed using intravital fluorescence microscopy in murine livers exposed to galactosamine/lipopolysaccharide (G/L) and treated with an anti-RAGE antibody (abRAGE) either 12 h be

Effects of specific genes activating RAGE on polycystic kidney disease.

BACKGROUND: Autosomal-dominant polycystic kidney disease (ADPKD) is characterized by progressive cyst formation and secretion of fluid and is associated with interstitial inflammation and fibrosis, resulting in the loss of renal function. We previously generated mice overexpressing PKD2, causing progressive cyst development with an inflammatory and fibrotic phenotype in the kidneys. METHODS: To profile the gene expression related to inflammation and cystogenesis, microarray analysis was performed with kidney tissue from 6-, 12- and 18-month-old mice. Subsequently, levels and related mechanism

Role of the RAGE signaling pathway in myocardial ischemia/reperfusion injury.

The receptor for advanced glycation end products (RAGE), a multi-ligand receptor of the immunoglobulin superfamily, plays a pivotal regulatory role in myocardial ischemia/reperfusion (I/R) injury. This review summarizes the regulatory mechanisms governing RAGE expression; the dynamic alterations of its major ligands-advanced glycation end products (AGEs), high mobility group box 1 (HMGB1), and the S100 protein family-during myocardial I/R; and the multifaceted pathological mechanisms by which RAGE-mediated downstream signaling pathways, including activation of the MAPK and JAK/STAT cascades an

"It Was Like They'd Lit a Fuse": A Mixed-Methods Investigation Into Rage.

Impulsive acts of violence are perpetrated by a small proportion of society, which anecdotal evidence suggests often occurs in a rage state. Rage has generally been conceptualized as extreme anger, but it may be qualitatively different from extreme anger. Is rage best considered as part of the anger continuum or is it a different phenomenon altogether? The present research addressed this question in a large sample of university students (N = 545) who completed qualitative and quantitative measures of rage attacks, anger, emotional dysregulation, trait aggression and displaced aggression, angry

The anatomy of rage: conceptualization, operationalization, and validation of a new measurement scale.

INTRODUCTION: Despite its prevalence among gamers, rage remains an understudied phenomenon in digital gaming. The present study aimed to conceptualize rage as an emotional response to in-game failure or frustration and to develop a reliable and valid self-report instrument to measure this experience. METHODS: The study was conducted in two phases. In the first, theoretical and qualitative phase, a focus group and expert assessment procedure were used to define the construct of rage in gaming and develop initial items. In the second, empirical phase, an international sample of 267 participants
