Construction of a DNA vaccine and its protective effect on largemouth bass (Micropterus salmoides) challenged with largemouth bass virus (LMBV).

Largemouth bass virus (LMBV) is the causative agent of a disease causing high mortality rates in largemouth bass during summer. However, there is little information available about the development of vaccines for LMBV disease. Hence, a DNA vaccine, named pCDNA3.1(+)-MCP-Flag, was constructed by inserting the cloned LMBV major capsid protein (MCP) gene into the pCDNA3.1(+)-Flag plasmid. The expression of the recombinant plasmid was confirmed by Western blot (WB) and RT-PCR. The WB result revealed that the MCP protein produced a band of approximately 53 kDa, consistent with the expected result.

Transcriptome analysis reveals the host immune response upon LMBV infection in largemouth bass (Micropterus salmoides).

Largemouth bass (Micropterus salmoides) is one of the important economical freshwater aquaculture species in China. However, the outbreak of viral diseases always caused great economic losses in the largemouth bass aquaculture industry. Largemouth bass virus (LMBV), a double-stranded DNA (dsDNA) virus belonging to genus Ranavirus, family Iridoviridae causes high mortality in cultivated largemouth bass. However, host responses, especially the molecular events involved in LMBV infection still remained largely uncertain. Here, we established an in vivo model of LMBV infection, and systematically

Antiviral activity of marine fungal polysaccharide AVP141-A against Singapore grouper iridovirus (SGIV) in vitro and in vivo.

Singapore grouper iridovirus (SGIV) represents a major viral pathogen responsible for severe economic losses in the grouper aquaculture industry, emphasizing the urgent need for effective antiviral agents. In the present study, the antiviral activity of the natural compound AVP141-A against SGIV was systematically evaluated in vitro and in vivo. In vitro, AVP141-A showed no cytotoxicity to GS cells at concentrations up to 5 mg/mL, and significantly inhibited SGIV replication in a concentration-dependent manner. This inhibitory effect was reflected by attenuated cytopathic effects, reduced tran

Lactate facilitates largemouth bass virus replication by promoting cholesterol biosynthesis.

Cellular metabolism has been hijacked by viruses for efficient replication due to their regulatory roles on both innate and adaptive immunity. Largemouth bass virus (LMBV), a member of Ranavirus, causes high mortality and severe economic losses in several fish species, seriously threatening the sustainable development of aquaculture industry. However, the roles of the metabolic events in LMBV replication remained largely uncertain. Here, our results showed that LMBV boosted aerobic glycolysis both in vitro and in vivo. Supplement with glucose significantly enhanced LMBV replication in vitro. C

Computational and experimental mapping of antigenic epitopes on iridovirus major capsid protein for precision vaccine design.

The major capsid protein (MCP), owing to its strong immunogenicity, has emerged as a critical target for largemouth bass ranavirus (LMBV) vaccine development. However, its key antigenic epitopes remain uncharacterized. This study integrated phage display technology with bioinformatics approaches to design a multiepitope vaccine against LMBV. By constructing a phage Ab library derived from LMBV-infected largemouth bass spleen tissues, we obtained high-affinity Abs (Ab-S1 and Ab-S2) capable of specifically binding LMBV. Through molecular docking and alanine scanning mutagenesis of MCP-Ab complex

The absence of zoonotic agents in invasive bullfrogs (Lithobates catesbeianus) in Belgium and The Netherlands.

Exotic invasive bullfrogs (Lithobates catesbeianus) are considered to exert a considerable negative impact on native amphibian communities. This can be due to competition and predation, but they are also a notorious source of the infectious diseases chytridiomycosis and ranavirosis, affecting amphibian populations globally. Little is known regarding their carriage of other microbial agents that might be transferred to humans or other animals. In this study we determined the occurrence of the amphibian pathogens Ranavirus and Batrachochytrium dendrobatidis and of the zoonotic agents Coxiella bu

Investigating the Antiviral Mechanism of Kaempferol Against Grouper Iridovirus: Combined Evaluation of In Vitro Activity and In Vivo Efficacy.

Singapore grouper iridovirus (SGIV) is one of the major pathogens that pose significant threats to the aquaculture industry. Kaempferol is a flavonoid compound extracted from Illicium verum Hook.f. (star anise), a material that is both edible and used in traditional medicine. In this study, we evaluated the antiviral effects of kaempferol against SGIV in vitro and in vivo, and elucidated the antiviral mechanism. Our study found that kaempferol could effectively resist SGIV infection in vitro and in vivo. Kaempferol might destroy the SGIV envelope and interfere with the attachment, invasion, an

Pathology, Tissue Distribution, and Phylogenetic Characterization of Largemouth Bass Virus Isolated from a Wild Smallmouth Bass (Micropterus dolomieu).

We performed a diagnostic disease investigation on a wild smallmouth bass (Micropterus dolomieu) with skin ulcers that was collected from Lake Oahe, South Dakota, following reports from anglers of multiple fish with similar lesions. Gross and histologic lesions of ulcerative dermatitis, myositis, and lymphocytolysis within the spleen and kidneys were consistent with largemouth bass virus (LMBV) infection. LMBV was detected by conventional PCR in samples of a skin ulcer, and the complete genome sequence of the LMBV (99,184 bp) was determined from a virus isolate obtained from a homogenized skin

Modulatory effects of curcumin on Singapore grouper iridovirus infection-associated apoptosis and autophagy in vitro.

Singapore grouper iridovirus (SGIV) with high pathogenicity can cause great economic losses to aquaculture industry. Thus, it is of urgency to find effective antiviral strategies to combat SGIV. Curcumin has been demonstrated effective antiviral activity on SGIV infection. However, the molecular mechanism behind this action needs to be further explanations. In view of the fact that apoptosis (type I programmed cell death) and autophagy (type II programmed cell death) were key regulators during SGIV infection, we aimed to investigate the relevance between antiviral activity of curcumin and SGIV
