The present study was performed to assess the effects exerted by the dopamine (DA) D2/D3 antagonist (-) eticlopride on rodent behavioral models considered to be predictive of antipsychotic activity, namely, antagonism toward DA agonist-induced stereotyped behavior (SB), and ketamine-and cocaine-induced hypermotility. (-) Eticlopride (10-50 micrograms/kg) dose-dependently inhibited SB elicited by SND 919 (10 mg/kg), CQP 201-403 (0.5 mg/kg), and 7-OH-DPAT (5 mg/kg); moreover, it significantly counteracted the hypermotility induced in rats and mice by ketamine (5 and 10 mg/kg). When (-) eticlopri

Behavioural and biochemical evidence for the existence of a powerful specific postsynaptic interaction between adenosine A1 and dopamine D1 receptors in the mammalian brain was found. Behavioural data showed that A1 receptor stimulation induced a decrease in the D1-induced motor activation in reserpinized mice and a decrease in the D1-dependent oral dyskinesia in rabbits. Biochemical data suggested that A1 receptor stimulation could produce a GTP-independent uncoupling of the rat striatal D1 receptor to the G protein. The A1-D1 receptor-receptor interaction might represent an important additio

Dopamine receptors were studied in post-mortem brains from control and Huntington's disease patients, using the specific binding of [3H]spiperone to dopamine D-2 receptors and [3H]piflutixol to dopamine D-1 receptors. Both [3H]spiperone binding and [3H]piflutixol binding were reduced by 45-50% in Huntington's disease putamen. The loss of [3H]spiperone and [3H]piflutixol binding sites correlated with decreased GABA concentrations observed in Huntington's disease putamen. A selective loss (48%) of [3H]piflutixol binding was observed in Huntington's disease substantia nigra pars reticulata, [3H]p

There are currently several studies in which individual dopamine receptor ligands have been reported to bind with relatively low affinity to opiate receptors. To extend these studies, and to examine the opiate receptor subclass selectivity of such agents, we have examined the ability of six dopamine receptor ligands (prochlorperazine, chlorpromazine, haloperidol, bromocriptine, pimozide and metoclopramide) to compete with four tritiated tracers - [3H]naloxone, [3H][D-Ala2,D-Leu5]enkephalin, [3H]morphine and [3H]ethylketocyclazocine - for binding to rat brain membrane opiate receptors. The dopa

Dopamine transporter knockout (DATk) mice are known to demonstrate profound hyperactivity concurrent with elevated (5-fold) extracellular dopamine in the basal ganglia. At the same time, heterozygous DAT mice (DATh) demonstrate a 2-fold increase in dopamine levels yet only a marginal elevation in locomotor activity level. Another model of dopaminergic hyperactivity is the D3 dopamine receptor knockout (D3k) mice, which present only a modest hyperactivity phenotype, predominately manifested as stereotypical behaviors. In the D3k mice, the hyperactivity is also correlated with elevated extracell

This review presents a systematic analysis of the cerebellar dopaminergic system and its role in regulating social behavior. It examines the mechanisms of dopamine synthesis and transmission, the main types and signaling pathways of dopamine receptors, and data on the localization and functional significance of these receptors in cerebellar cells, particularly D2 receptors. Special attention is given to experimental results demonstrating that modulation of D2 receptors in the cerebellum affects social preferences and memory in animals. The connections between the cerebellum and dopaminergic zo

The lateral epithelial cells of gill of Crassostrea virginica are innervated by dopamine and serotonin nerves that regulate the beating rate of their lateral cilia. Terminal release of dopamine slows down the beating rate of the cilia, while serotonin release increases the beating rate. Previously, we showed that the dopaminergic, but not the serotonergic, mechanism regulating the beating rate of the lateral cilia was disrupted by manganese treatments and that this disruption was occurring postsynaptically, at the level of the dopamine receptor or further downstream in the signal transduction

Earlier studies indicate that striatal dopamine D(2) receptors are involved in pain regulation in non-neuropathic conditions. We assessed whether striatal dopamine D(2) receptors contribute to pain regulation also in neuropathic conditions. The spared nerve injury model of neuropathy was induced by unilateral ligation of the tibial and common peroneal nerves in the rat. In awake nerve-injured animals, pain-related withdrawal responses to calibrated monofilaments or noxious heating were attenuated following striatal administration of a dopamine D(2) receptor agonist quinpirole. Pain-related res

Alterations in presynaptic and postsynaptic dopaminergic system and cerebral glucose metabolism in corticobasal degeneration (CBD) were assessed to evaluate the potential usefulness of different imaging methods for CBD. (123)I-FP-CIT/(123)I-beta-CIT SPECT and (123)I-IBZM SPECT as well as (18)F-FDG PET were performed in eight CBD patients. Decreased presynaptic dopamine transporter binding was found in all CBD patients while D2 receptor binding was reduced in only one patient. (18)F-FDG PET displayed a contralateral hypometabolism in cortical and subcortical areas in seven out of eight patients

Diethylpropion (DEP) is an amphetamine-like agent used as an anorectic drug. Abuse of DEP has been reported and some restrictions of its use have been recently imposed. The conditioning place preference (CPP) paradigm was used to evaluate the reinforcing properties of DEP in adult male Wistar rats. After initial preferences were determined, animals weighing 250-300 g (N = 7 per group) were conditioned with DEP (10, 15 or 20 mg/kg). Only the dose of 15 mg/kg produced a significant place preference (358 +/- 39 vs 565 +/- 48 s). Pretreatment with the D1 antagonist SCH 23,390 (0.05 mg/kg, s.c.) 10

In order to determine whether the D2 and D3 dopamine receptors may have a role in prenatal development, we have studied the mRNA expression and distribution of these receptors during murine embryonic development. Using RT-PCR on RNA from embryos taken at progressive stages of development, we have shown that the D3 receptor is expressed significantly earlier than the D2 receptor, being detectable at day 9.5 post-conception (p.c.) compared with day 13.5 p.c. for the D2 subtype. We have also examined the mRNA distribution of the two receptors by whole mount in situ hybridisation. In agreement wit

Fencamfamine (FCF) is a psychostimulant classified as an indirect dopamine agonist. The conditioning place preference (CPP) paradigm was used to investigate the reinforcing properties of FCF. After initial preferences had been determined, animals were conditioned with FCF (1.75, 3.5, or 7.0 mg/kg; IP). Only at the dose of 3.5 mg/kg FCF produced a significant place preference. Pretreatment with SCH23390 (0.05 mg/kg; SC) or naloxone (1.0 mg/kg; SC) 10 min before FCF (3.5 mg/kg, IP) blocked both FCF-induced hyperactivity and CPP. Pretreatment with metoclopramide (10.0 mg/kg; IP) or pimozide (1.0

Dopamine receptors in glomeruli and renal cortical tubules were characterized using radioligand binding and adenylate cyclase studies. The binding of [3H]haloperidol to glomeruli and tubules was rapid, saturable with time and ligand concentration, reversible, of high affinity, and demonstrated stereoselectivity and antagonist and agonist rank potency for binding to dopamine receptors. Analysis of kinetic data and Rosenthal plots in glomeruli revealed a single class of [3H]haloperidol binding sites with an apparent dissociation constant (Kd) of 6 nM and maximum receptor density (Bmax) of 0.42 p

Dopamine receptors were studied in striatal synaptosomes prepared from rat brain with hepatic encephalopathy induced by galactosamine-HCl and documented by visual evoked potential recordings. In order to further characterize the model, plasma amino acid levels and striatal catecholamines and octopamine levels were assayed. In agreement with previous reports in animal and in man, plasma amino acids were increased both in mild and severe stage of this pathology. Striatal levels of norepinephrine and dopamine fell during the development of coma while octopamine rose. Dopamine binding studies show

In the rat, elevating dopamine content in corpus striatum with electrical stimulation of substantia nigra or direct administration of apomorphine (50-200 micrograms) into the lateral cerebral ventricle or apomorphine (2-10 microgram) into the caudate-putamen complex decreased pain sensitivity (as shown by an increase in the latency to hind-paw lick in the hot plate test). Furthermore, the decreased pain sensitivity after the central administration of apomorphine was antagonized by pretreatment with haloperidol (a dopamine antagonist). On the other hand, lowering dopamine content in corpus stri

The poor norepinephrine innervation and high density of Gi/o-coupled α2A- and α2C-adrenoceptors in the striatum and the dense striatal dopamine innervation have prompted the possibility that dopamine could be an effective adrenoceptor ligand. Nevertheless, the reported adrenoceptor agonistic properties of dopamine are still inconclusive. In this study, we analyzed the binding of norepinephrine, dopamine, and several compounds reported as selective dopamine D2-like receptor ligands, such as the D3 receptor agonist 7-OH-PIPAT and the D4 receptor agonist RO-105824, to α2-adrenoceptors in cortical

Positron emission tomographic(PET) study using 18F-6-fluoro-L-dopa (18FDOPA) can provide efficient information on the pre-synaptic function of nigrostriatal dopaminergic neurons. In juvenile parkinsonism(JP), the accumulation of 18FDOPA is markedly decreased in the caudate nucleus and putamen on both hemispheres. This finding is different from those in dystonia syndromes such as dopa-responsive dystonia (DRD) and hereditary progressive dystonia with marked diurnal fluctuation(HPD), and it is rather similar to late onset of Parkinson's disease. Furthermore, we studied dopamine D2 receptor bindi
