Regulation of feeding-associated peptides and receptors by nicotine.

Although numerous epidemiological studies have provided convincing evidence for the inverse association between tobacco smoking and body weight, the molecular mechanisms underlying this relationship are not well-understood. Nicotine, as a potent secretagogue, could be expected to influence the levels and expression of many classes of neurotransmitters, as well as of cell-membrane constituents linked to neurotransmission, including signal transducers and related effectors. A potentially major group of candidate molecules that could be involved in feeding-related actions of nicotine are the nume

Receptors for opioid peptides in the guinea-pig ileum.

Cryptic receptor sites in the guinea-pig ileum preparation have been uncovered by the treatment of the preparation with the highly selective, irreversible mu opioid receptor antagonist, beta-funaltrexamine. These beta-funaltrexamine-insensitive sites appear to interact only with opioid peptides ([D-Ala2, D-Leu5]enkephalin, [D-Ala2, Met5]enkephalinamide, Tyr-D-Ser-Gly-Phe-Leu-Thr and [D-Ala2, MePhe4, Gly-ol5]enkephalin) but not with nonpeptide agonists. These new sites could not be protected by either mu-selective (morphiceptin and [D-Ala2, MePhe4, Gly-ol5]enkephalin) or delta-selective ([D-Ala

Opioid Receptor Independent Effects of Opioid Peptides.

Almost immediately after the discovery of opioid peptides and opioid receptors (ORs), reports were published that some effects of opioid peptides are independent of the activation of ORs. It was demonstrated that Des-Tyr1-fragments of opioid peptides did not interact with ORs but can exert different effects both in vivo and in vivo. They modify the functional state of the brain, the heart, the trachea, the aorta, arteries, natural killers, neutrophils, lymphocytes, and adrenal glands. It is quite obvious that non-ORs of Des-Tyr1-enkephalins, Des-Tyr1-dynorphins A, β-endorphin (2- 17), and β-en

Pheochromocytomatosis Treated With Peptide Receptor Radionuclide Therapy.

Pheochromocytomatosis refers to pheochromocytoma tumorlets developed as a result of seeding of tumor cells around the surgical bed due to intraoperative tumor capsule rupture and tumor cell spillage. As pheochromocytomatosis is relatively rare, optimal management is not clear. We describe a 42-year-old man with progressive pheochromocytomatosis despite surgical debulking. He did not have a family history of pheochromocytoma or harbor mutations in pheochromocytoma-predisposing genes. The pheochromocytomatosis tumorlets exhibited uptake on DOTATATE PET. He underwent PRRT (peptide receptor radion

Efficacy of Peptide Receptor Radionuclide Therapy for Esthesioneuroblastoma.

Esthesioneuroblastoma is rare, with limited therapeutic options when unresectable or metastatic; however, expression of somatostatin receptors qualifies it for peptide receptor radionuclide therapy (PRRT). We report outcomes of PRRT in esthesioneuroblastoma from 2 referral centers. Methods: Using PRRT databases at 2 European Neuroendocrine Tumor Society Centers of Excellence, cases were sought between 2004 and 2018 of patients who had PRRT with recurrent or metastatic esthesioneuroblastoma deemed unsuitable for further conventional therapies. Evaluations of survival and of response using a com

Natriuretic peptide receptor A as a novel target for cancer.

The receptor for the cardiac hormone atrial natriuretic peptide (ANP), natriuretic peptide receptor A (NPR-A), has been reported to be expressed in lung cancer, prostate cancer and ovarian cancer. NPR-A expression and signaling is important for tumor growth; its deficiency protects C57BL/6 mice from lung, skin and ovarian cancers. This suggests that NPR-A is a new marker and a new target for cancer therapy. Recently, NPR-A has been demonstrated to be expressed in pre-implantation embryos and in embryonic stem cells, which has a novel role in the maintenance of self-renewal and pluripotency of

Adrenomedullin and related peptides: receptors and accessory proteins.

Adrenomedullin (AM), alpha- and beta-calcitonin gene-related peptide (CGRP), amylin and calcitonin (CT) are structurally and functionally related peptides. The structure of a receptor for CT (CTR) was elucidated in 1991 through molecular cloning, but the structures of the receptors for the other three peptides had yet to be elucidated. The discovery of receptor-activity-modifying proteins (RAMP) 1 and -2 and their co-expression with an orphan receptor, calcitonin receptor-like receptor (CRLR) has led to the elucidation of functional CGRP and AM receptors, respectively. RAMP1 and -3 which are c

Receptor biology of the melanocortins, a family of neuroimmunomodulatory peptides.

Melanocortins, melanocyte-stimulating hormones (MSH) and adrenocorticotropic hormone (ACTH) are homologous natural peptides derived from pro-opiomelanocortin (POMC). Recent breakthroughs in melanocortin receptor (MCR) biology are relevant to neuroimmunomodulation because melanocortins are known to modulate fever, inflammation and immunity, by acting both on peripheral targets and within the brain. During fever, endogenous melanocortins exert antipyretic effects by acting on MCR located within the brain, suggesting a protective counterregulatory role of the central melanocortin system. MCR are

Differential mechanisms of activation of the Ang peptide receptors AT1, AT2, and MAS: using in silico techniques to differentiate the three receptors.

The renin-angiotensin system is involved in multiple conditions ranging from cardiovascular disorders to cancer. Components of the pathway, including ACE, renin and angiotensin receptors are targets for disease treatment. This study addresses three receptors of the pathway: AT1, AT2, and MAS and how the receptors are similar and differ in activation by angiotensin peptides. Combining biochemical and amino acid variation data with multiple species sequence alignments, structural models, and docking site predictions allows for visualization of how angiotensin peptides may bind and activate the r

Heterobivalent Dual-Target Peptide for Integrin-αvβ3 and Neuropeptide Y Receptors on Breast Tumor.

Background/Objectives: Heterodimer peptides targeting more than one receptor can be advantageous, as tumors can simultaneously express more than one receptor type. For human breast cancer, a promising biological target is tumor angiogenesis through αvβ3 integrin expression. Another promising target is Neuropeptide Y receptors, considering Y1R is overexpressed in 90% of human breast tumors. This article details the development and preclinical evaluation, both in vitro and in vivo, of a novel heterodimer peptide dual-receptor-targeting probe, [99mTc]HYNIC-cRGDfk-NPY, designed for imaging breast

Role of nociceptin/orphanin FQ and nociceptin opioid peptide receptor in depression and antidepressant effects of nociceptin opioid peptide receptor antagonists.

Nociceptin/orphanin FQ (N/OFQ) and its receptor, nociceptin opioid peptide (NOP) receptor, are localized in brain areas implicated in depression including the amygdala, bed nucleus of the stria terminalis, habenula, and monoaminergic nuclei in the brain stem. N/OFQ inhibits neuronal excitability of monoaminergic neurons and monoamine release from their terminals by activation of G protein-coupled inwardly rectifying K+ channels and inhibition of voltage sensitive calcium channels, respectively. Therefore, NOP receptor antagonists have been proposed as a potential antidepressant. Indeed, mounti

Natriuretic Peptide Receptors (NPRs) as a Potential Target for the Treatment of Heart Failure.

PURPOSE OF REVIEW: Heart failure is defined as a complex clinical syndrome that results from any structural or functional impairment of ventricular filling or ejection of blood. The natriuretic peptide is known to exert its biological action on the kidney, heart, blood vessels, renin-angiotensin system, autonomous nervous system, and central nervous system. The natriuretic peptide-natriuretic receptor system plays an important role in the regulation of blood pressure and body fluid volume through its pleiotropic effects. RECENT FINDINGS: The clinical and animal studies suggest that natriureti

Coagonist of glucagon-like peptide-1 and glucagon receptors ameliorates nonalcoholic fatty liver disease.

Nonalcoholic fatty liver disease (NAFLD) is often associated with obesity and type 2 diabetes. Coagonists of glucagon-like peptide-1 receptor (GLP-1R) and glucagon receptor (GCGR) are under clinical investigation for the treatment of obesity and type 2 diabetes. In this study, we have demonstrated the effect of a balanced coagonist in the treatment of NAFLD using mouse models. GLP-1R agonist exendin-4, glucagon, and coagonist (Aib2 C24 chimera2) were administered to C57BL6/J mice, in which NAFLD was induced by carbon tetrachloride (CCl4) treatment after high-fat diet (HFD) feeding, and choline

Characterization of specific calcitonin gene-related peptide receptors present in hamster pancreatic beta cells.

Calcitonin gene-related peptide (CGRP) shares about 46% and 20% amino acid sequence homology with islet amyloid polypeptide (IAPP) and salmon calcitonin (sCT). We investigated whether these related peptides could cross-react with the specific binding of 125I-[His]hCGRP I to the CGRP receptor in hamster insulinoma cell membranes. A rapid dissociation of membrane bound 125I-[His]hCGRP I could be induced in the presence of 1 microM chicken CGRP (cCGRP). The specific 125I-[His]hCGRP I binding was inhibited by the related peptides and their half-maximal inhibitory concentrations (IC50) were: cCGRP

Amylin increases cyclic AMP formation in L6 myocytes through calcitonin gene-related peptide receptors.

The cellular function of amylin is investigated in L6 myocytes, a rat skeletal muscle cell line. Both rat amylin and human amylin-amide acutely cause a dose-dependent increase in cyclic AMP formation in L6 myocytes. 100 nM amylin stimulates intracellular cyclic AMP concentrations 12-fold, whereas human amylin-amide at this concentration causes only a 2-fold increase. Up to 10 mM human amylin has no effect on cyclic AMP levels. Rat calcitonin gene-related peptide (CGRP) is more potent than amylin, causing a 60-fold increase over basal at 1 nM, with an EC50 value of 0.2 nM. The CGRP receptor ant

Receptors for peptide hormones. New insights into the pathophysiology of disease states in man.

The first step in the action of polypeptide hormones and many neurotransmitters is binding to receptor sites on the plasma membrane of the cell. These receptors are usually complex, high molecular weight proteins. Using a variety of receptor preparations and radioactively labeled hormones, radioreceptor assays for several hormones have been developed. These have allowed for assay of hormones for which no immunoassay exists. Such assays have shown increased levels of NSILA-s (an insulin-like peptide) in patients with nonpancreatic tumors and hypoglycemia. In disease states, the number or affini

Ryanoids and imperatoxin affect the modulation of cardiac ryanodine receptors by dihydropyridine receptor Peptide A.

Ca(2+)-entry via L-type Ca(2+) channels (DHPR) is known to trigger ryanodine receptor (RyR)-mediated Ca(2+)-release from sarcoplasmic reticulum (SR). The mechanism that terminates SR Ca(2+) release is still unknown. Previous reports showed evidence of Ca(2+)-entry independent inhibition of Ca(2+) sparks by DHPR in cardiomyocytes. A peptide from the DHPR loop II-III (PepA) was reported to modulate isolated RyRs. We found that PepA induced voltage-dependent "flicker block" and transition to substates of fully-activated cardiac RyRs in planar bilayers. Substates had less voltage-dependence than b

Human neutrophils recognize group B streptococci via formylated peptide receptors and toll-like receptor 8.

INTRODUCTION: Neutrophils, the most abundant leukocyte population, play a central role in host defense against group B Streptococcus (GBS), an encapsulated bacterium that frequently causes severe infections in humans, including sepsis and meningitis. However, the molecular pathways by which human neutrophils detect and respond to GBS remain incompletely defined. METHODS: Neutrophils were isolated from the peripheral blood of healthy donors and stimulated with live and killed GBS or their nucleic acids, in the presence of specific inhibitors of immune recognition receptors. Cytokine responses

Zeta Inhibitory Peptide attenuates learning and memory by inducing NO-mediated downregulation of AMPA receptors.

Zeta inhibitory peptide (ZIP), a PKMζ inhibitor, is widely used to interfere with the maintenance of acquired memories. ZIP is able to erase memory even in the absence of PKMζ, via an unknown mechanism. We found that ZIP induces redistribution of the AMPARGluA1 in HEK293 cells and primary cortical neurons, and decreases AMPAR-mediated currents in the nucleus accumbens (NAc). These effects were mimicked by free arginine or by a modified ZIP in which all but the arginine residues were replaced by alanine. Redistribution was blocked by a peptidase-resistant version of ZIP and by treatment with th
