Functional Characterization of Odorant Binding Protein 27 (RproOBP27) From Rhodnius prolixus Antennae.

Olfactory proteins mediate a wide range of essential behaviors for insect survival. Odorant binding proteins (OBPs) are small soluble olfactory proteins involved in the transport of odor molecules (=odorants) through the sensillum lymph to odorant receptors, which are housed on the dendritic membrane of olfactory sensory neurons also known as olfactory receptor neurons. Thus, a better understanding of the role(s) of OBPs from Rhodnius prolixus, one of the main vectors of Chagas disease, may ultimately lead to new strategies for vector management. Here we aimed at functionally characterize OBPs

Social Control of Hypothalamus-Mediated Male Aggression.

How environmental and physiological signals interact to influence neural circuits underlying developmentally programmed social interactions such as male territorial aggression is poorly understood. We have tested the influence of sensory cues, social context, and sex hormones on progesterone receptor (PR)-expressing neurons in the ventromedial hypothalamus (VMH) that are critical for male territorial aggression. We find that these neurons can drive aggressive displays in solitary males independent of pheromonal input, gonadal hormones, opponents, or social context. By contrast, these neurons c

Odorant-binding proteins and chemosensory proteins in insects: structural insights, functional plasticity, and prospects for targeted pest management.

Odorant-binding proteins (OBPs) and chemosensory proteins (CSPs) are integral to the chemical communication systems of insects, mediating the detection, transport, and discrimination of hydrophobic ligands such as pheromones, odorants, and xenobiotics. While OBPs are largely confined to olfactory tissues and exhibit high structural specificity-with six conserved cysteines forming three disulfide bridges-CSPs are structurally simpler, featuring four conserved cysteines and two disulfide bridges, yet display remarkable conformational plasticity and functional versatility. Recent evidence undersc

Ionotropic glutamate receptors mediate inducible defense in the water flea Daphnia pulex.

Phenotypic plasticity is the ability held in many organisms to produce different phenotypes with a given genome in response to environmental stimuli, such as temperature, nutrition and various biological interactions. It seems likely that environmental signals induce a variety of mechanistic responses that influence ontogenetic processes. Inducible defenses, in which prey animals alter their morphology, behavior and/or other traits to help protect against direct or latent predation threats, are among the most striking examples of phenotypic plasticity. The freshwater microcrustacean Daphnia pu

MaltEms-MaltOBP9 axis mediates sexually dimorphic olfactory behavior in Monochamus alternatus via recognition of pheromone 2-methoxy-4-vinylphenol.

The olfactory system is essential for insect survival and behavior, with odorant-binding proteins (OBPs) playing a key role by binding and transporting chemical signals to olfactory receptors. Although MaltOBP9 from Monochamus alternatus, a primary vector of pine wilt disease, has been linked to olfactory functions in M. alternatus, its regulatory mechanisms and specific roles are not well understood. In this study, we show that MaltOBP9 is highly expressed in the antennae of both male and female adults after sexual maturation. Using fluorescence spectroscopy, molecular docking, and site-direc

The influence of steroids on vascular tension of isolated superficial veins of the nose and face during the estrous cycle of gilts.

The arrangement of the superficial facial veins enables blood flow from the nasal cavity into the peripheral circulation by two pathways: through the frontal vein into the cavernous sinus and through the facial vein into the external jugular vein. The current study was designed to determine whether estradiol and progesterone affect the vascular tone of the superficial veins of the nose and face in cycling gilts (Sus scrofa f. domestica) and to analyze the immunolocalization of progesterone receptors and estradiol receptors in these veins. The influence of hormones on vascular tension differed

Putative ligand binding sites of two functionally characterized bark beetle odorant receptors.

BACKGROUND: Bark beetles are major pests of conifer forests, and their behavior is primarily mediated via olfaction. Targeting the odorant receptors (ORs) may thus provide avenues towards improved pest control. Such an approach requires information on the function of ORs and their interactions with ligands, which is also essential for understanding the functional evolution of these receptors. Hence, we aimed to identify a high-quality complement of ORs from the destructive spruce bark beetle Ips typographus (Coleoptera, Curculionidae, Scolytinae) and analyze their antennal expression and phylo

Electrophysiological Responses of Eighteen Species of Insects to Fire Ant Alarm Pheromone.

Olfaction plays a dominant role in insect communication. Alarm pheromones, which alert other insects of the same species of impending danger, are a major class of releaser pheromones. The major components of alarm pheromones in red imported fire ants, honeybees and aphids have been identified as 2-ethyl-3,6-dimethylpyrazine (2E-3,6-DP), isopentyl acetate (IPA), and E-β-farnesene (EβF), respectively. In this study, electroantennography (EAG) responses to EDP (a mixture of 2-ethyl-3,6-dimethylpyrazine and 2-ethyl-3,5-dimethylpyrazine), IPA and EβF were investigated in a wide range of insect spec

Activation of family C G-protein-coupled receptors by the tripeptide glutathione.

The Family C G-protein-coupled receptors include the metabotropic glutamate receptors, the gamma-aminobutyric acid, type B (GABAB) receptor, the calcium-sensing receptor (CaSR), which participates in the regulation of calcium homeostasis in the body, and a diverse group of sensory receptors that encompass the amino acid-activated fish 5.24 chemosensory receptor, the mammalian T1R taste receptors, and the V2R pheromone receptors. A common feature of Family C receptors is the presence of an amino acid binding site. In this study, a preliminary in silico analysis of the size and shape of the amin

Transcriptome Analysis and Identification of Chemosensory Genes in Leguminivora glycinivorella.

The soybean pod borer, Leguminivora glycinivorella, is a monophagous pest that threatens soybean production. Its larvae feed concealed within pods, which limits the efficacy of conventional insecticides. Elucidating its chemosensory system is therefore essential for developing green, behavior-based management strategies. Reference-based transcriptomics across multiple tissues of L. glycinivorella identified a comprehensive repertoire of chemosensory genes, including 76 odorant receptors (ORs), 15 gustatory receptors (GRs), 18 ionotropic receptors (IRs), 52 odorant-binding proteins (OBPs), 18 c

Cryptococcal titan cell formation is regulated by G-protein signaling in response to multiple stimuli.

The titan cell is a recently described morphological form of the pathogenic fungus Cryptococcus neoformans. Occurring during the earliest stages of lung infection, titan cells are 5 to 10 times larger than the normal yeast-like cells, thereby resisting engulfment by lung phagocytes and favoring the persistence of infection. These enlarged cells exhibit an altered capsule structure, a thickened cell wall, increased ploidy, and resistance to nitrosative and oxidative stresses. We demonstrate that two G-protein-coupled receptors are important for induction of the titan cell phenotype: the Ste3a p

Pheromone-based communication influences the production of somatic extracellular vesicles in C. elegans.

Extracellular vesicles (EVs) are integral to numerous biological processes, yet it is unclear how environmental factors or interactions among individuals within a population affect EV-regulated systems. In Caenorhabditis elegans, the evolutionarily conserved large EVs, known as exophers, are part of a maternal somatic tissue resource management system. Consequently, the offspring of individuals exhibiting active exopher biogenesis (exophergenesis) develop faster. Our research focuses on unraveling the complex inter-tissue and social dynamics that govern exophergenesis. We found that ascr#10, t

Immunolocalization of c-Fos, ELOVL5 and oestradiol in the ewe vulva in relation to oestrus behaviour after treatment with lipopolysaccharide.

Sudden activation of the stress axis by a lipopolysaccharide endotoxin (LPS) significantly reduces ewes' sexual attractivity to rams by delaying all signs of oestrous behaviour. To understand mechanisms involved in attracting male interest, we examined c-Fos (nuclear activation), ELOVL5 (enzyme involved in pheromone synthesis) and oestradiol receptors (ER) using immunohistochemistry on ewe vulval tissue at 0, 31 and 40 hr in the ovarian follicular phase with or without exposure to LPS at 28 hr (5 groups of 4 ewes per group). While there was intense staining for immunoreactive (IR)-c-Fos and IR

Effects of pesticide-adjuvant combinations used in almond orchards on olfactory responses to social signals in honey bees (Apis mellifera).

Exposure to agrochemical sprays containing pesticides and tank-mix adjuvants has been implicated in post-bloom mortality, particularly of brood, in honey bee colonies brought into California almond orchards for pollination. Although adjuvants are generally considered to be biologically inert, some adjuvants have exhibited toxicity and sublethal effects, including decreasing survival rates of next-generation queens. Honey bees have a highly developed olfactory system to detect and discriminate among social signals. To investigate the impact of pesticide-adjuvant combinations on honey bee signal

From sexual attraction to maternal aggression: when pheromones change their behavioural significance.

This article is part of a Special Issue "Chemosignals and Reproduction". This paper reviews the role of chemosignals in the socio-sexual interactions of female mice, and reports two experiments testing the role of pup-derived chemosignals and the male sexual pheromone darcin in inducing and promoting maternal aggression. Female mice are attracted to urine-borne male pheromones. Volatile and non-volatile urine fractions have been proposed to contain olfactory and vomeronasal pheromones. In particular, the male-specific major urinary protein (MUP) MUP20, darcin, has been shown to be rewarding an

The sensory circuitry for sexual attraction in C. elegans males.

BACKGROUND: Why do males and females behave differently? Sexually dimorphic behaviors could arise from sex-specific neurons or by the modification of circuits present in both sexes. C. elegans males exhibit different behaviors than hermaphrodites. Although there is a single class of sex-specific sensory neurons in the head of males, most of their neurons are part of a core nervous system also present in hermaphrodites. Are the behavioral differences due to sex-specific or core neurons? RESULTS: We demonstrate that C. elegans males chemotax to a source of hermaphrodite pheromones. This sexual-

DREADD-Induced Silencing of the Medial Olfactory Tubercle Disrupts the Preference of Female Mice for Opposite-Sex Chemosignals(1,2,3).

Attraction to opposite-sex pheromones during rodent courtship involves a pathway that includes inputs to the medial amygdala (Me) from the main and accessory olfactory bulbs, and projections from the Me to nuclei in the medial hypothalamus that control reproduction. However, the consideration of circuitry that attributes hedonic properties to opposite-sex odors has been lacking. The medial olfactory tubercle (mOT) has been implicated in the reinforcing effects of natural stimuli and drugs of abuse. We performed a tract-tracing study wherein estrous female mice that had received injections of t

DREADD-induced silencing of the medial amygdala reduces the preference for male pheromones and the expression of lordosis in estrous female mice.

Sexually naïve estrous female mice seek out male urinary pheromones; however, they initially display little receptive (lordosis) behavior in response to male mounts. Vomeronasal-accessory olfactory bulb inputs to the medial amygdala (Me) regulate courtship in female rodents. We used a reversible inhibitory chemogenetic technique (Designer Receptors Exclusively Activated by Designer Drugs; DREADDs) to assess the contribution of Me signaling to females' preference for male pheromones and improvement in receptivity normally seen with repeated testing. Sexually naïve females received bilateral Me

The Drosophila pheromone cVA activates a sexually dimorphic neural circuit.

Courtship is an innate sexually dimorphic behaviour that can be observed in naive animals without previous learning or experience, suggesting that the neural circuits that mediate this behaviour are developmentally programmed. In Drosophila, courtship involves a complex yet stereotyped array of dimorphic behaviours that are regulated by Fru(M), a male-specific isoform of the fruitless gene. Fru(M) is expressed in about 2,000 neurons in the fly brain, including three subpopulations of olfactory sensory neurons and projection neurons (PNs). One set of Fru(+) olfactory neurons expresses the odora

Deconstructing pheromone-mediated behavior one layer at a time.

The vomeronasal organ, a sensory structure within the nasal cavity of most tetrapods, detects pheromones that influence socio-sexual behavior. It has two neuronal layers, each patterned by distinct receptor sub-families coupled to different G-proteins. Work recently published in this journal found female mice with one layer genetically inactivated are deficient in a surprisingly wide range of reproductive behaviors, providing new insights into how the nose can influence the brain.
