Characteristics of steroid hormone receptors in cultured MC3T3-E1 osteoblastic cells and effect of steroid hormones on cell proliferation.

We examined the binding characteristics of three kinds of steroid hormones--estrogen, androgen, and glucocorticoid--in cultured MC3T3-E1 mouse osteoblastic cells by whole-cell binding assay. The binding studies revealed the presence of a single class of high-affinity binding sites for [3H]17 beta-estradiol, [3H]mibolerone (a synthetic androgen), and [3H]triamcinolone acetonide (a synthetic glucocorticoid). The numbers of binding sites for these steroid hormones were found to be 4534 +/- 819, 14312 +/- 1884, and 24898 +/- 655 sites/cell; and the Kd values were 8.57 +/- 0.62 x 10(-10) M, 1.12 +/

Modulation of steroid hormone receptor activity.

Classical steroid hormones (SHs) - estrogens, androgens, progestins, glucocorticoids and mineralocorticoids - play critical roles in the regulation of reproduction, metabolism and cancer. SHs act via their cognate steroid hormone receptors (SHRs) in multiple target tissues throughout the body, exerting their physiological effects through nuclear receptor (NR)-mediated gene transcription. Since SHRs are the mediators of steroid hormone signalling in cells, regulation of their expression and function is critical for appropriate physiological responses to SHs. Cells regulate SHRs by determining t

Neuroactive steroid actions at the GABAA receptor.

Neuroactive steroids are a new class of steroids that do not interact with any of the classical cytosolic hormonal steroid receptors. The most well-documented examples are those that interact with the gamma-aminobutyric acidA (GABAA) receptor/chloride channel complex in the central nervous system. The GABAA receptors are known to contain allosteric modulatory sites for therapeutically useful drugs such as benzodiazepines (BZs) and barbiturates. The interaction of neuroactive steroids with the GABAA receptor is specific to a site on the receptor complex distinct from the benzodiazepine and barb

Stabilization of androgen receptor by nonandrogenic steroids.

Binding of androgens including 5 alpha-dihydrotestosterone, testosterone and androstenedione stabilized the androgen receptor from rat submandibular gland with respect to its [3H]methyltrienolone binding activity. Nonandrogenic steroids, 0.5 microM of progesterone and estradiol-17 beta, or 5 microM of triamcinolone acetonide also satisfactorily stabilized the androgen receptor. These results suggest that the association of ligands, even if those ligands were not stereospecific for the androgen receptor, stabilizes the androgen receptor and prevents the loss of its binding capacity.

Effects of environmental steroid mixtures are regulated by individual steroid receptor signaling.

Fish are exposed to steroids of different classes in contaminated waters, but their effects are not sufficiently understood. Here we employed an anti-sense technique using morpholino oligonucleotides to knockdown the glucocorticoid receptors (GRs, GRα and GRβ) and androgen receptor (AR) to investigate their role in physiological and transcriptional responses. To this end, zebrafish embryos were exposed to clobetasol propionate (CLO), androstenedione (A4) and mixtures containing different classes of steroids. CLO caused a decrease of spontaneous muscle contraction and increase of heart rate, as

Steroid binding activity is retained in a 16-kDa fragment of the steroid binding domain of rat glucocorticoid receptors.

The steroid binding domain of the rat glucocorticoid receptor is considered as extending from amino acids 550 to 795. However, such a synthetic protein (i.e. amino acids 547-795; Mr approximately 31,000) has been reported to show very little affinity for the potent synthetic glucocorticoid dexamethasone. We now disclose that digestion of steroid-free rat glucocorticoid receptors with low concentrations of trypsin yields a single species, of Mr = 16,000, that is specifically labeled by dexamethasone 21-mesylate. This 16-kDa fragment retains high affinity binding for [3H]dexamethasone that is on

The role of adrenergic receptors and sex steroid hormones in takotsubo syndrome.

Takotsubo syndrome (also known as stress-induced cardiomyopathy or takotsubo cardiomyopathy) is an acute, reversible left ventricular dysfunction typically triggered by emotional or physical stress. TS is a rare but dangerous disease. In-hospital mortality in patients with TS is identical to mortality of patients with ST-segment elevation myocardial infarction. There is no obstructive coronary plaque or thrombosis in patients with TS, but there is injury of both cardiomyocytes and endothelial cells. The main manifestations of TS are apical akinesia and apical ballooning. The main cause of deat

Molecular determinants of the recognition of ulipristal acetate by oxo-steroid receptors.

The human progesterone receptor (PR) plays a key role in reproductive function in women. PR antagonists have numerous applications in female health care including regular and emergency contraception, and treatment of hormone-related pathological conditions such as breast cancer, endometriosis, and leiomyoma. The main factor limiting their long-term administration is the fact that they cross-bind to other oxo-steroid receptors. Ulipristal acetate (UPA), a highly potent PR antagonist, has recently come onto the market and is much more selective for PR than the other oxo-steroid receptors (androg

Tissue expression of steroid hormone receptors is associated with differential immune responsiveness.

Glucocorticoids have been used as treatments against a number of diseases, especially autoimmune/inflammatory conditions in which the immune system is overactive. These treatments have varying degrees of responsiveness among individuals and in different tissues (including brain); therefore, it is important to determine what could account for these differences. In this study, we evaluated expression of stress hormone receptors in immune cells from lymphoid and non-lymphoid tissues (including brain) as a possible explanation. We analyzed leukocytes (CD45(+)) in kidney, liver, spleen, and thymus

Evolution of a new function by degenerative mutation in cephalochordate steroid receptors.

Gene duplication is the predominant mechanism for the evolution of new genes. Major existing models of this process assume that duplicate genes are redundant; degenerative mutations in one copy can therefore accumulate close to neutrally, usually leading to loss from the genome. When gene products dimerize or interact with other molecules for their functions, however, degenerative mutations in one copy may produce repressor alleles that inhibit the function of the other and are therefore exposed to selection. Here, we describe the evolution of a duplicate repressor by simple degenerative mutat

Immunohistochemical localization of gonadotropin and gonadal steroid receptors in human pineal glands.

Recently, we demonstrated that melatonin secretion was increased in male patients with GnRH deficiency and decreased to normal levels during testosterone treatment. These data suggested that gonadal steroids modulate melatonin secretion, probably by activating specific receptors in the pineal gland. We used immunohistochemistry to localize gonadotropin (LH and FSH) and gonadal steroid (androgens and estrogens) receptors in human pineal glands. Tissues were obtained at autopsy from 25 males, aged 19-87 yr, and five prepubertal children, aged 0.2-10 yr. Positive staining for all four types of re

Hormonal steroid receptors in intracranial tumours and their relevance in hormone therapy.

Determination of cytosolic estradiol and progesterone receptors was carried out in 30 intracranial tumors: 12 meningiomas, 13 metastases, two angioreticulomas, two gliomas and one sarcoma. The hormonal fraction found with the receptor and the dissociation constant (Kd) were determined by the Scatchard method. Values higher than 10 fmol/mg of protein were considered as positive. Ten of the 12 meningiomas (83%) showed progesterone receptors (RP), while estrogen receptors (RE) were not found in any of the cases; six of the 13 metastases showed the two types of receptors; the other tumors did not

Plasma steroid hormones, cytosol receptors, and thymidine incorporation rate in endometrial carcinoma.

Histopathologically evaluated endometrial carcinomas were analyzed for the presence of estradiol and progesterone receptors and for tumor thymidine incorporation rate. Plasma estradiol and progesterone concentrations were also measured. With the use of an improved assay, all endometrial carcinomas were found to contain estradiol receptors; 81 of 92 (88%) contained measurable concentrations of progesterone receptors. The concentrations of both receptors were positively correlated to the degree of differentiation. The moderately and poorly differentiated tumours could be further divided into sub

Steroid receptor phylogeny and vertebrate origins.

Vertebrates appear about 500 million years ago in the fossil record. This is only 25-50 million years after the great explosion of multicellular invertebrate body plans in the early Cambrian. On a geological time scale, this interval is a 'blink of an eye', suggesting that the evolution of regulatory genes is likely to be important in the origins of vertebrates. Here we present evidence for a role of steroid receptors in this process based on a phylogenetic analysis suggesting that receptors for androgens, glucocorticoids, mineralocorticoids, and progesterone evolved from an ancestral steroid

Sex steroid hormone receptors of telocytes - potential key role in leiomyoma development.

BACKGROUND: Uterine leiomyoma is the most widespread benign tumor affecting women of childbearing age. There are still gaps in the understanding of its pathogenesis. Telocytes are unique cells found in more than 50 different locations inside the human body. The functional relationship between cells could clarify the pathogenesis of leiomyomata. Examination of membrane receptors on telocytes could explain their role in fibrosis, oxidative stress, and myometrial contractility. AIM: This research was conducted to assess the density of telocytes in terms of their putative role in leiomyoma format

Low expression of glucocorticoid receptors in children with steroid-resistant nephrotic syndrome.

BACKGROUND: About 10-20 % of children with idiopathic nephrotic syndrome (NS) are steroid-resistant (SR). Low expression of glucocorticoid receptors (GRs) has been associated with poor response to steroids in a variety of autoimmune diseases. This study was done to assess the expression of cytoplasmic GRs for CD3 and CD14 in children with NS. METHODS: Expression of cytoplasmic GRs in lymphocytes (CD3(+)/GR) and monocytes (CD14(+)/GR) in the peripheral blood were assessed in 51 children with NS before the start of therapy by flow cytometry. Patients were divided into two groups: 30 children wh
