Selective Chemogenetic Blockade of Cerebello-Rubral Pathway by Double-Virus Infection Method Using Lentivirus and Adeno-Associated Virus in Rats.

Double-virus infection method of lentivirus and adeno-associated virus was used to reveal the contribution of cerebellum in rehabilitative training after intracerebral hemorrhage (ICH) in rats. Chemogenetic blockade of cerebello-rubral pathway was achieved by the injection of a lentiviral vector pseudotyped with fusion glycoprotein type E and carrying Cre recombinase into the red nucleus and an adeno-associated viral vector encoding hM4D(Gi)-mCherry in the double-floxed, inverted orientation into the cerebellar lateral nucleus. This silencing system induced by clozapine-N-oxide significantly r

The neurotoxicity of valine deficiency in rats.

When valine, an essential amino acid, was withdrawn from the diet of weanling rats, the animals rapidly developed a unique pattern of neurological symptoms characterized by head retraction, staggering and aimless circling. At necropsy degenerative changes were most prominent in the neurons of the red nuclei, brain stem structures which modulate motor function. To explore the pathogenesis of the neurotoxicity associated with valine deficiency, we fed rats purified diets deficient in valine alone or in valine plus other branched chain and neutral amino acids, and we examined brain tissues by lig

Blepharoclonus: anatomical localization and etiological consideration.

Blepharoclonus refers to myoclonic rhythmic eyelid closure. This is an extremely rare abnormal movement of the eyelids. The symptom has an ill-defned anatomical localization and hypothesized etiologies are diverse. We describe a 42 year-old woman with known poorly controlled hypertension (HTN) who presented with a three-week history ofataxia, dysmetria, and uncontrolled eyelid twitching. The bilateral abnormal eyelid movement occurred during either eyelid closure or opening, and was compatible with blepharoclonus. MRI revealed multiple cerebral infarctions at red nucleus, dentate nucleus, and

Quantitative susceptibility mapping from basal ganglia and related structures: correlation with disease severity in progressive supranuclear palsy.

OBJECTIVE: We examined whether mean magnetic susceptibility values from deep gray matter structures in patients with progressive supranuclear palsy (PSP) differed from those in patients with Parkinson's disease (PD) and healthy volunteers, and correlated with the PSP rating scale. METHODS: Head of caudate nucleus, putamen, globus pallidus, substantia nigra and red nucleus were the regions of interest. Mean susceptibility values from these regions in PSP patients were estimated using quantitative susceptibility mapping. Correlations with clinical severity of disease as measured by the PSP rati

Concurrent excitatory and inhibitory effects of high frequency stimulation: an oculomotor study.

OBJECTIVE: To describe a reversible neurological condition resembling a crossed midbrain syndrome resulting from high frequency stimulation (HFS) in the midbrain. METHODS: Postoperative evaluation of quadripolar electrodes implanted in the area of the subthalamic nucleus of 25 patients with Parkinson's disease (PD) successfully treated by HFS. RESULTS: Four of the 25 patients experienced reversible acute diplopia, with dystonic posture and tremor in the contralateral upper limb when the white matter between the red nucleus and the substantia nigra was stimulated. The motor signs resembled th

Buyang Huanwu Decoction Promotes Recovery after Spinal Cord Injury by Regulating cAMP/PKA/NF-κB p65 Pathway.

OBJECTIVE: To investigate whether Buyang Huanwu Decoction (BYHWD) had a good curative effect on the neuroprotection of red nucleus neurons after spinal cord injury (SCI) and the possible molecular mechanism. METHODS: Ninety male Sprague-Dawley rats were divided into 5 groups (n=18 per group) according to a random number table, including the control, model, low- (12.78 g/kg, BL group), medium- (25.65 g/kg, BM group), and high-dose BYHWD groups (51.30 g/kg, BH group). A rubrospinal tract transection model in rats was established, and different doses of BYHWD were intragastrically administrated

Red nuclear and cerebellar but no olivary activation associated with essential tremor: a positron emission tomographic study.

There has been debate as to whether essential tremor has a central origin and over the possible role of the inferior olivary nucleus in its genesis. We used positron emission tomography with radioactive water (H2(15)O) to detect abnormal patterns of cerebral activity associated with this condition, at rest, without tremor, and on posture when the tremor was present. At rest, cerebellar blood flow was significantly increased bilaterally in the group with essential tremor (30-40%) but no increased olivary activity was evident. Essential tremor during arm extension was associated with further abn

Distribution of manganese-dependent superoxide dismutase in the human brain.

The distribution of manganese-dependent superoxide dismutase, an enzyme that transforms superoxide radicals into hydrogen peroxide, was studied in the human brain post mortem using a sheep polyclonal antiserum raised against the enzyme from liver mitochondria. One band, corresponding to a protein of 22,000 mol. wt was detected in the human brain by western blot analysis. At the light-microscopy level, a punctate immunostaining was observed in the neuropil and in some but not all, glial and neuronal cell bodies. Electron-microscopy revealed that the staining was exclusively confined to the inne

Enrichment and distribution of 3,6-dichlorocarbazole in red crucian carp (Carassius auratus) and its hepatotoxicity.

Polyhalogenated carbazoles (PHCZs) are a class of organohalogen compounds where the hydrogen atom on the carbazole ring is replaced by a halogen atom. In recent years, PHCZs have drawn increasing concern due to their persistence, dioxin-like toxicity, bioaccumulation, potential ecological hazards and widespread occurrence in the environment. Current research on the enrichment and depuration of PHCZs in biological tissues and organs is insufficient, and the liver toxicity is unclear. Herein, to understand the enrichment and elimination of 3,6-DCCZ in fish tissues and organs as well as the hepat

Acute Thermal Elevation Alters Cellular Metabolic Activity, Redox Balance, and Iron Homeostasis in Butterfly Lizards (Leiolepis belliana belliana).

Global warming is increasing the frequency of acute temperature fluctuations that may threaten ectothermic reptiles, but the underlying physiological and cellular mechanisms remain poorly understood. This study investigated physiological changes, oxidative imbalance, antioxidant systems, nitric oxide, iron metabolism, and red blood cell morphology in butterfly lizards (Leiolepis belliana belliana) under acute temperature elevation. Physiological responses were evaluated in vivo in butterfly lizards exposed to temperatures of 27-37 °C, whereas biochemical responses in red blood cells were exami

Deep brain stimulation in a dentatorubral-pallidoluyisian atrophy patient with myoclonic dystonia.

We describe a patient with myoclonic dystonia caused by dentatorubral-pallidoluyisian atrophy (DRPLA), which was successfully controlled with bilateral deep brain stimulation (DBS) of the globus pallidus internus (GPi). DRPLA is a rare disease which can progressively cause a loss or degeneration of neurons in the globus pallidus, dentate nucleus, subthalamic nucleus, and red nucleus. This observation is another example of secondary dystonia which can be controlled by GPi-DBS in carefully selected patients.

Acceleration of Brain Susceptibility-Weighted Imaging with Compressed Sensitivity Encoding: A Prospective Multicenter Study.

BACKGROUND AND PURPOSE: While three-dimensional susceptibility-weighted imaging has been widely suggested for intracranial vessel imaging, hemorrhage detection, and other neuro-diseases, its relatively long scan time has necessitated the clinical verification of recent progresses of fast imaging techniques. Our aim was to evaluate the effectiveness of brain SWI accelerated by compressed sensitivity encoding to identify the optimal acceleration factors for clinical practice. MATERIALS AND METHODS: Ninety-nine subjects, prospectively enrolled from 5 centers, underwent 8 brain SWI sequences: 5 d

Differential participation of the corticospinal and corticorubral neurons during motor execution in the rat.

The sensorimotor cortex is crucial for learning and executing new movements with precision. It selectively modulates sensory information flow and represents motor information in a spatially organized manner. The pyramidal system is made up of layer 5 pyramidal tract neurons (PTNs), which are organized into populations with distinct morphological, genetic and functional properties. These subpopulations project to different subcortical structures in a segregated manner. To understand whether PTNs projecting to different structures play distinct functional roles in motor control, we characterized

[Relations of aqueduct with some structures of mesencephalon].

INTRODUCTION: Aqueductus mesencephali is the biggest part of the ventricular system and that is why it is the most common place of intraventricular obstruction of cerebrospinal fluid. This study was done in order to study topographic characteristics of aqueduct more thoroughly. MATERIALS AND METHODS: Transversal sections of mesencephalon were made in three levels. The first section was made caudally immediately from the posterior commissure. The second section was made in the middle part of the superior colliculi, and the third section was made in the rostral parts of the caudal sections of t

Failed DBS for palliation of visual problems in a case of oculopalatal tremor.

OBJECTIVE: To report the results of attempted bilateral red nucleus (RN) deep brain stimulation (DBS) for the palliative treatment of visual problems associated with oculopalatal tremor (OPT). BACKGROUND: It is hypothesized that OPT results from a defect in the Guillain-Mollaret triangle, a circuit that includes connections with the dentate nucleus, the contralateral red nucleus, and the inferior olive. We present a high functioning patient (an accountant) who underwent a palliative trial of RN region DBS in an approach targeted through the subthalamic nucleus region. The aim was to reduce ey

[Benedikt Syndrome Associated with Neck Clipping of Ruptured Basilar-Superior Cerebellar Artery Aneurysm:A Case Report].

Benedikt syndrome is caused by a rare type of brain damage to the red nucleus and nearby locations. Clinically, it presents as ipsilateral oculomotor palsy and specific contralateral involuntary movement. These involuntary movements may affect activities of daily living(ADLs)in affected patients. We treated a case of Benedikt syndrome associated with neck clipping of a ruptured basilar-superior cerebellar artery aneurysm. A 66-year-old female patient was admitted to our department with headache. Initial computed tomography imaging and angiography showed subarachnoid hemorrhage and left basilar
