Estimate of the Deterministic Neutron RBE for Radiation-induced Pseudo-Pelger Huët Cell Formation.

Using archival peripheral blood slides from radiation accident patients, we have recently described the pseudo-Pelger Huët anomaly (PPHA) in neutrophils as a new radiation-induced biomarker, useful for dosimetry not only immediately after a radiation incident but also potentially helpful as a tool in retrospective dosimetry. In conjunction with the Radiation Accident Registry at the Radiation Emergency Assistance Center/Training Site (REAC/TS), the frequency of PPHA cells has been compared from selected patients in the Y-12 criticality accident in Oak Ridge, TN, in 1958 and from the patient in

Proton and photon absorbed-dose conversion coefficients for embryo and foetus from top-down irradiation geometry.

Absorbed-dose conversion coefficients are calculated for the embryo of 8 weeks and the foetus of 3, 6 or 9 months when the mother's body is exposed to protons and photons from top-down (TOP) direction. It provides data sets in addition to other standard irradiation geometries published previously. The TOP-irradiation geometry is considered here, because high-energy particles are often peaked from the TOP direction onboard aircrafts. The results show that absorbed-doses from high-energy particles could be underestimated significantly if isotropic (ISO) irradiation geometry is assumed. For proto

Dose-effect relationships for recurrence of keloid and pterygium after surgery and radiotherapy.

PURPOSE: To show radiation dose-response relationships for recurrence of keloid and pterygium after radiotherapy following surgery. METHODS AND MATERIALS: Using PubMed, we performed a retrospective review of articles reporting incidences and/or dose-response relationships for recurrence of keloid and pterygium after radiotherapy following surgery. The irradiation regimens identified were normalized by use of the linear-quadratic model; biologically effective doses (BEDs) were calculated. RESULTS: For keloid recurrence after radiotherapy following keloid removal, with either teletherapy or br

Chromosome aberration frequencies in human lymphocytes irradiated in a multi-layer array by protons with different LET.

PURPOSE: To provide data on the dose-and linear energy transfer (LET)-dependence of the production of dicentrics in human lymphocytes. MATERIALS AND METHODS: Track segment irradiation with 16.5 MeV protons was performed in a multi-layer array allowing the simultaneous exposure of human peripheral lymphocytes in three successive samples. Within these samples the dose-averaged linear energy transfer (LD) of protons increased with increasing depth, i.e. LD = 3.5, 5.3 and 19.0 keV/microm. Dicentrics were scored in first division solid-stained metaphases. RESULTS: Dicentric yields measured in the

Chromosome aberration frequencies produced by a 70-MeV proton beam.

The effectiveness of a 70-MeV proton beam in the induction of chromosome aberrations was studied. We employed peripheral lymphocytes and analyzed the frequencies of dicentrics and rings after irradiation at doses ranging from 0.1 to 8.0 Gy at various depths within a Lucite phantom. The frequency of chromosome aberrations after irradiation with an unmodulated proton beam at 5 mm showed a dose-response relationship similar to that of 60Co gamma rays. However, irradiation at greater depths with the spread-out Bragg peak induced higher aberration frequencies at doses lower than those with gamma ra

The induction of chromosome aberrations in human lymphocytes by in vitro irradiation with beta particles from tritiated water.

The yields of unstable chromosome aberrations induced in human lymphocytes by beta particles from tritiated water ( HTO ) have been measured. HTO was mixed with heparinized blood in various amounts so that doses of 0.25 to 7.0 Gy were delivered in 20 min and 1.5 and 2.5 hr. After culturing for 48 hr, the dicentric yield was measured as a function of dose to the blood and compared with data previously reported for X rays. Using a linear-quadratic dose-effect relation to fit the experimental data, a significant linear contribution was found. The main difference between the coefficients for beta

Comparison of two biologically effective dose calculation models applied to single fraction stereotactic radiosurgery.

BACKGROUND: Recent studies suggest strong correlations between Biologically Effective Doses (BED) and single fraction stereotactic radiosurgery treatment outcomes, as demonstrated for vestibular schwannomas (VS), arterio-venous malformations and pituitary adenomas. The BEDs calculated in these studies consider an uniform dose delivery where the spatio-temporal aspects of dose delivery were neglected. PURPOSE: The aim of the study is to quantify the discrepancies between the BED values calculated with a simplified model of uniform dose delivery against the more complex model that incorporates

Enhanced RBE of Particle Radiation Depends on Beam Size in the Micrometer Range.

High-linear energy transfer (LET) radiation, such as heavy ions is associated with a higher relative biological effectiveness (RBE) than low-LET radiation, such as photons. Irradiation with low- and high-LET particles differ in the interaction with the cellular matter and therefore in the spatial dose distribution. When a single high-LET particle interacts with matter, it results in doses of up to thousands of gray (Gy) locally concentrated around the ion trajectory, whereas the mean dose averaged over the target, such as a cell nucleus is only in the range of a Gy. DNA damage therefore accumu

Detailed dosimetry calculation for in-vitro experiments and its impact on clinical BNCT.

PURPOSE: Boron Neutron Capture Therapy (BNCT) is a form of hadrontherapy based on the selective damage caused by the products of neutron capture in 10B to tumour cells. BNCT dosimetry strongly depends on the parameters of the dose calculation models derived from radiobiological experiments. This works aims at determining an adequate dosimetry for in-vitro experiments involving irradiation of monolayer-cultured cells with photons and BNCT and assessing its impact on clinical settings. Low LET protons focused to submicrometer shows enhanced radiobiological effectiveness.

This study shows that enhanced radiobiological effectiveness (RBE) values can be generated focusing low linear energy transfer (LET) radiation and thus changing the microdose distribution. 20 MeV protons (LET = 2.65 keV µm(-1)) are focused to submicrometer diameter at the ion microprobe superconducting nanoprobe for applied nuclear (Kern) physics experiments of the Munich tandem accelerator. The RBE values, as determined by measuring micronuclei (RBE(MN) = 1.48 ± 0.07) and dicentrics (RBE(D) = 1.92 ± 0.15), in human-hamster hybrid (A(L)) cells are significantly higher when 117 protons were foc

Chromosome aberrations induced by protons up to 31 MeV in cultured human cells.

Chromosome aberrations were induced in cultured human cells by proton beams of 31, 12, and 8 MeV. The frequencies of isochromatid breaks and dicentrics have been analysed as a function of proton energy and dose. Both effects are largely dependent on proton energy; isochromatid breaks increased linearly with the dose, whereas dicentrics show a definite parabolic behaviour. The experimental data were fitted to the analytic form Y = KDn and Y = alpha D + beta D2 and the best fitted values of the parameters are reported and discussed. The values of RBE for the isochromatid breaks are in the ratio

Revisiting fractionation in prostate carbon ion therapy: first comparative analysis of biologically effective dose using mMKM with diverse parameters.

BACKGROUND: Carbon ion radiotherapy (CIRT) for prostate cancer has evolved from conventional regimens [66 Gy(RBE)/20 fractions (66G20), 63 Gy(RBE)/20 fractions (63G20)] toward hypofractionated schedules [57.6 Gy(RBE)/16 fractions (57.6G16), 51.6 Gy(RBE)/12 fractions (51.6G12), and 40 Gy(RBE)/4 fractions (40G4)]. Whether these are biologically equivalent remains unclear. To our knowledge, this is the first study to systematically evaluate the biological effects of these regimens using the modified microdosimetric kinetic model (mMKM) across diverse parameter settings. METHODS: Biologically effe

Radiobiological considerations in the design of fractionation strategies for intensity-modulated radiation therapy of head and neck cancers.

PURPOSE: The dose distributions of intensity-modulated radiotherapy (IMRT) treatment plans can be shown to be significantly superior in terms of higher conformality if designed to simultaneously deliver high dose to the primary disease and lower dose to the subclinical disease or electively treated regions. We use the term "simultaneous integrated boost" (SIB) to define such a treatment. The purpose of this paper is to develop suitable fractionation strategies based on radiobiological principles for clinical trials and routine use of IMRT of head and neck (HN) cancers. The fractionation strate

Assessment of contributors to radiation dose following intakes of rapidly excreted [14C]-compounds.

The International Commission on Radiological Protection default biokinetic model for the assessment of radiation dose received following intakes of unspecified [14C]-compounds (DCM) appears to overestimate the radiation doses delivered by many [14C]-compounds. The DCM does not specify the route of elimination and this review of the biokinetic data for four [14C]-compounds, [14C]sevelamer, [14C]colestipol, [14C]levetiracetam and [14C]ifetroban, that are rapidly lost from the human body via different excretory pathways, indicates that the excretory pathway is the critical determinant for both th

Creation, evolution, and future challenges of ion beam therapy from a medical physicist's viewpoint (Part 2). Chapter 2. Biophysical model, treatment planning system and image guided radiotherapy.

When an ion beam penetrates deeply into the body, its kinetic energy decreases, and its biological effect increases due to the change of the beam quality. To give a uniform biological effect to the target, it is necessary to reduce the absorbed dose with the depth. A bio-physical model estimating the relationship between ion beam quality and biological effect is necessary to determine the relative biological effectiveness (RBE) of the ion beam that changes with depth. For this reason, Lawrence Berkeley Laboratory, National Institute of Radiological Sciences (NIRS) and GSI have each developed t

RBE and weighting of absorbed dose in ion-beam therapy.

Absorbed dose is the fundamental quantity used to quantify the exposure of any biological system to ionizing radiation. However, the relationship between dose and biological effect is not unique but varies with fractionation and time factor(s), radiation quality and irradiation conditions. In radiation therapy, weighting factors are used to correlate absorbed dose and clinical effects when altering irradiation conditions, or for combining or comparing different technical modalities. For some well established therapy modalities (e.g. fractionated photon beam therapy), a general agreement on wei

Acute skin reactions observed in fractionated proton irradiation.

Between May 1985 and July 1987, 49 skin reactions of 43 patients treated by proton irradiation were observed at the Particle Radiation Medical Science Center (PARMS), The University of Tsukuba. Taking the peak skin score as an endpoint, the radiobiological effects [relative biological effectiveness (RBE) and time-dose relationships] of the proton beam in multi-fractionated treatments were estimated. Factors influencing the skin dose, such as the prescribed tumor dose, tumor site, and number of applied fields, were also analyzed. The following conclusions regarding acute skin reactions to the c

Dose distribution of neutron beam and chromosome analysis.

Chromosome analysis using peripheral lymphocytes is a sensitive and reliable indicator of the biological effect of radiation at low radiation doses. In the present study, using this chromosome aberration analysis, we tried to investigate the radiation dose distribution within and around the radiation field of a 6 MeV neutron beam. The efficient induction of dicentrics and rings at the irradiation of neutrons compared to those of gamma or X rays especially in a lower dose range, results in a dominant linear component in the linear quadratic model in the dose response relation of a chromosome ab

Chromosome aberrations in human lymphocytes induced by fission neutrons.

The dose-response relationships of dicentrics and excess acentrics were analysed after exposure of human lymphocytes to a mixed fission neutron-gamma-ray beam. From the analysis of exclusively first division cells a linear-quadratic relation was obtained for dicentrics with the ratio of linear and quadratic components, zeta, equal to 2.76 Gy. Over the range of doses studied (0.04-1.97 Gy) intratrack events therefore predominated. This also applied to acentrics which were linearly related to dose. At the lowest level of observed effect and dose, r.b.e. values with respect to 60Co gamma-rays of
