Immunogenicity of a cell culture-derived inactivated vaccine against a common virulent isolate of grass carp reovirus.

Grass carp (Ctenopharyngodon idella) hemorrhagic disease, caused by grass carp reovirus (GCRV), is emerging as a serious problem in grass carp aquaculture. There is no available antiviral therapy and vaccination is the primary method of disease control. In the present study, the immunological effects and protective efficacy of an inactivated HuNan1307 vaccine in grass carp were evaluated. The GCRV isolate HuNan1307 was produced by replication onto the grass carp PSF cell line, and inactivated with 1% β-propiolactone for 60 h at 4 °C. Grass carp were injected with inactivated GCRV vaccine, foll

hnRNPM suppressed IRF7-mediated IFN signaling in the antiviral innate immunity in triploid hybrid fish.

Mammalian heterogeneous nuclear ribonucleoproteins M (hnRNPM) is a critical splicing regulatory protein that has been reported to negatively regulate the RLR signaling pathway by impairing the binding of RIG-I and MDA5 to viral RNA. To explore the role of hnRNPM in the antiviral innate immune response in teleost fish, the hnRNPM homologue of triploid fish (3nhnRNPM) has been cloned and identified in this paper. The CDS of 3nhnRNPM gene is composed of 2016 nucleotides and encodes 671 amino acids. 3nhnRNPM migrated around 71 kDa in immunoblotting assay and was mainly detected in the nucleus in n

Carbon nanotube-based nanocarrier loaded with ribavirin against grass carp reovirus.

Infectious diseases of viral origin cause major aquatic production losses in different parts of the world. Because of formidable barriers for gastrointestinal tract, skin and cell, large amounts of antiviral drugs have limited therapeutic effect. In the current study, functionalized single-walled carbon nanotubes (SWCNTs) were selected as a drug carrier to carry antiviral drug for the treatment of viral diseases on fish. The results show that increasing antiviral drug (ribavirin) intake was observed by SWCNTs carrier and therapeutic dosage to kill grass carp reovirus is significantly reduced.

Isolation of a virus serologically related to the bluetongue group from an outbreak of haemorrhagic disease among exotic deer in Saudi Arabia.

In February 1991 a severe haemorrhagic disease affected exotic deer aged over six months in the Al-Hofuf area of the eastern region of Saudi Arabia. The morbidity rate was 40 per cent and the case fatality rate was 60 per cent. The clinical signs were high temperature (up to 41.5 degrees C), nasal discharge, slight salivation and lacrimation, congestion of the conjunctivae, torticollis, tremors when trying to stand, recumbency and coma leading to death. Post mortem examination revealed a severe haemorrhagic disease. A virus, serologically related to the bluetongue serogroup, was isolated from

Nuclear scaffold attachment factor A functions as a potential viral recognition receptor involved in the antiviral immunity of grass carp (Ctenopharyngodon idella).

As one of the most primitive vertebrates, fish have evolved a distinct repertoire of viral recognition receptors. However, the existence of nuclear viral recognition receptors in fish remains uncertain. In this study, we identified a scaffold attachment factor A (SAFA), CiSAFA, as a potential nuclear recognition receptor for grass carp reovirus (GCRV) in grass carp (Ctenopharyngodon idella). CiSAFA shares high amino acid sequence similarity (70.3-99.6 %) and identity (56.1-98.6 %) with its counterparts from other vertebrates and contains three conserved domains, namely, SAP, SPRY, and AAA_33.

Preliminary study of BF/C2 on immune mechanism of grass carp against GCRV infection.

BF/C2 is a crucial molecule in the coagulation complement cascade pathway and plays a significant role in the immune response of grass carp through the classical, alternative, and lectin pathways during GCRV infection. In vivo experiments demonstrated that the mRNA expression levels of BF/C2 (A, B) in grass carp positively correlated with GCRV viral replication at various stages of infection. Excessive inflammation leading to death coincided with peak levels of BF/C2 (A, B) mRNA expression and GCRV viral replication. Correspondingly, BF/C2 (A, B) recombinant protein, CIK cells and GCRV co-incu

Rapid detection of infectious pancreatic necrosis virus (IPNV) by the enzyme-linked immunosorbent assay (ELISA).

The enzyme-linked immunosorbent assay (ELISA) was used to demonstrate the presence of infectious pancreatic necrosis virus (IPNV) antigen in cell cultures and in fish. Virus antigen could be detected in infected cell cultures before visible cytopathic effect (c.p.e.) was evident and cell cultures showing complete viral c.p.e. produced intense colour reactions. Virus antigen was detected in infected fry during and immediately after an epizootic, but although IPNV-carrier fish could be detected by the ELISA technique, the sensitivity of detection was not as great as that of isolation of the infe

The epizootiology of infectious bursal disease and prevention of it by immunization.

Transport of leftover feed from infectious disease (IBD) farms to susceptible flocks on clean premises is believed to have played an important role in the transmission of this disease to new farms and to new areas. Servicemen, caretakers, contaminated equipment and air were suspected in the spread of the disease, but these modes were not demonstrated. Litter, drinking water and samples of feed from feeders of IBD flocks were found to be infectious when added to rations of susceptible chickens but were not when added to drinking water. Infectious bursal agent (IBA) remained viable for at least

Impact of anthropogenic activities on the ecosystem and emergence of bat-borne zoonotic diseases.

About 70 % of zoonotic infectious diseases originate from wildlife reservoirs, particularly bats, primates, and rodents. Bats comprise about 20 % of all known mammal species worldwide and have been identified as reservoir and carrier hosts of various viral disease outbreaks. They also play a crucial role in viral adaptation and evolution. Given this inherent risk, this review focuses on how anthropogenic activities (habitat destruction, agricultural intensification, bushmeat hunting, and occupational exposure) and climate change are increasing the frequency and intensity of bat-human contact.

A modified IL-7 enhances the immune response to an inactivated GCRV-II vaccine in grass carp (Ctenopharyngodon idella).

Grass carp reovirus (GCRV) is a major pathogen causing hemorrhagic disease in grass carp (Ctenopharyngodon idella), posing a serious threat to aquaculture. However, the protective efficacy of current inactivated vaccines remains limited, underscoring the need for effective adjuvants. In this study, a modified grass carp interleukin-7 (mIL-7) was generated by mutating glutamine at position 130 to glutamic acid (Q130E), based on integrated structural and energetic analyses. Structural modeling suggested that Q130E mutation may improve local conformational stability and alter the predicted interf

Multi-tissue transcriptomics reveals age-dependent susceptibility to grass carp reovirus linked to metabolic alterations and extracellular matrix disruption in grass carp.

Grass carp (Ctenopharyngodon idella) is an important freshwater aquaculture species. Infection by grass carp reovirus (GCRV) can cause hemorrhagic disease and lead to major economic losses in aquaculture. Susceptibility to GCRV is strongly age dependent: juveniles (≤1 year old) are highly vulnerable, whereas fish aged three years or older show much greater tolerance. In this study, transcriptome sequencing was used to compare gene expression in the gill, intestine, head kidney, muscle, and spleen of three-year-old and six-month-old grass carp at pre-infection (Day 0) and at Days 4 and 7 post-i

Safety, immunogenicity, and broad-spectrum protective efficacy of a naturally attenuated GCRV-II strain against viral and bacterial challenges in grass carp.

Grass carp hemorrhagic disease (GCHD), caused by genotype II grass carp reovirus (GCRV-II), is a severe viral disease affecting Chinese aquaculture. In this study, a naturally attenuated GCRV-II strain, GCRV-GD2021, was isolated from healthy grass carp and evaluated as a live-attenuated vaccine candidate. The virus was confirmed as GCRV-II by immunofluorescence and S2 gene sequencing. Safety assessment demonstrated complete avirulence in grass carp, with no clinical signs or mortality observed. Serial in vivo blind passage confirmed that progressive viral load decline and no virulence reversio

Biochemical characterization and mucosal immune function of IgT in grass carp (Ctenopharyngodon idella).

Immunoglobulin T (IgT) is a teleost-specific immunoglobulin discovered to be specialized in mucosal immunity. For fish that live in water throughout their lives, eliciting a local mucosal immune response through immersion immunization is an important and highly suitable way for preventing fish diseases. However, due to the lack of corresponding antibodies against IgT, studies on its immune responses to pathogens and vaccines in commercial fish have largely been limited to rainbow trout. Thus, a valuable tool for assessing mucosal immune response to immersion vaccination is still lacking in mos

[Assessment of immunogenic activity of the cloned human rotavirus A WA strain.].

INTRODUCTION: Rotovirus infection (RVI) caused by the dsRNA-containing virus from genus Rotavirus, Reoviridae family, belonging to group A (RVA), is the cause of severe diarrhea in human and other mammalian species. Vaccination is the most effective way to reduce the incidence of RVI. At present, the effectiveness of using gnotobiotic piglets as a universal model for reproducing human rotavirus infection and assessing the quality of RVI vaccine preparations has been experimentally proven. OBJECTIVES: Evaluation of immunogenic activity of the cloned RVA Wa strain in the new-born Vietnamese pot

Orbivirus infection of the bovine fetus.

Direct inoculation of 85-125 day bovine fetuses with 4 serotypes of bluetongue virus (BTV) or 1 serotype of epizootic hemorrhagic disease virus (EHDV) resulted in either fetal death or teratogenesis. Three serotypes of BTV, and 1 of EHDV caused a similar cavitating encephalopathy, the severity of which likely reflected gestational age of the fetus at infection. Virus-specific antibody was readily detected in precolostral serum samples from all inoculated fetuses that survived until term, whereas, virus was not recovered from any of these calves. Calves with severe Orbivirus induced cerebral ma
