Endogenous retinoids in the early avian embryo.

Vitamin A is required to rescue the vitamin A-deficient quail embryo from early death, but the endogenous presence of bioactive retinoids has not been documented in these embryos. The analysis of 2000 pooled stage 5-8 normal quail embryos described here provides for the first time direct evidence for the presence of endogenous all-trans-retinoic acid (5 nM) and 3,4-didehydroretinoic acid (4 nM), signaling molecules known to be potent ligands for nuclear retinoic acid receptors. The demonstration of all-trans-retinal (80 nM), all-trans-retinol (100 nM), 3,4-didehydroretinol (200 nM), and retiny

Retinoids: transport, metabolism, and mechanisms of action.

The retinoids comprise a family of polyisoprenoid lipids that includes vitamin A (retinol) and structurally related compounds. The biological activity of retinoids can be modified, for example, by changes in the molecules' state of oxidation and cis/trans isomerization. Their activity is also dependent on the levels of specific types of retinoid-binding proteins which exist in extracellular, cytosolic and nuclear compartments. The role of retinoids in gene expression represents an important biological function for this family of molecules. Retinoid-dependent modulation of gene expression is cr

Retinoids and the alcohol dehydrogenase gene family.

Alcohol dehydrogenase (ADH) is best known as the enzyme which catalyzes the reversible oxidation/reduction of ethanol/acetaldehyde. However, mammalian ADH has also been shown to function in vitro as a retinol dehydrogenase in the conversion of retinol (vitamin A alcohol) to retinoic acid, a hormone which regulates gene expression at the transcriptional level. It is clear that retinol must be converted to more active retinoid forms in order to fulfill its roles in growth, development, and cellular differentiation. An important unsolved issue in retinoid research is the control of retinoic acid

[Retinoids in therapy of granuloma anulare disseminatum].

We report on four patients with granuloma annulare disseminatum successfully treated with etretinate as basic medication. In two cases there was complete remission after treatment with initial doses of up to 0.9 mg/kg body weight etretinate. One patient did not respond to etretinate alone but the condition was cured when Re-PUVA therapy was administered in addition. In another patient, with widespread granuloma annulare, primary treatment with Re-PUVA was instigated, followed by low-dose monotherapy with etretinate. According to these findings, we propose a three-stage regimen of therapy. Afte

Retinoids and carotenoids in bovine pineal gland.

The retinoids and carotenoids in bovine pineal gland were analyzed by high-performance liquid chromatographic methods. The major retinoid observed was all-trans retinol, with an average level of 579 pmol/g wet tissue. In agreement with a previous report, retinal was not detected; however, we could not detect any retinyl ester in bovine pineal gland. Methods similar to those used in studying metabolism of vitamin A in the bovine eye were used to study the esterification and isomerization reactions of retinoids in the pineal gland. Neither esterification nor isomerization reactions occurred. The

Neutrophil function in psoriasis: effects of retinoids.

The present investigation focused on the oxidative response of polymorphonuclear neutrophil leukocytes in psoriasis, in particular pustular psoriasis and how this response was affected by different retinoid compounds. In the active phase of pustular psoriasis, the neutrophil chemiluminescence response to the chemotactic peptide f-met-leu-phe and to phorbol myristate acetate was enhanced and correlated to the development of pustules, whereas cells from psoriasis vulgaris patients showed normal chemiluminescence response. Retinoids, particularly tretinoin (= retinoic acid) and isotretinoin cause

Effects of topical retinoids on cytoskeletal proteins: implications for retinoid effects on epidermal differentiation.

In vivo effects of retinoids on epidermal differentiation were investigated by analyzing cytoskeletal proteins in rhino mice treated topically with all-trans-retinoic acid (RA) and other retinoids (13-cis-retinoic acid, etretinate, TTNPB). Non-disulfide-linked cytoskeletal proteins, including keratins from the epidermal "living layers," were first selectively extracted using 9.5 M urea; subsequently, keratins of the stratum corneum were isolated using 9.5 M urea plus a reducing agent. Gel electrophoresis and immunoblot analysis showed that urea extracts of epidermis from vehicle-treated skin w

Do topical retinoids cause acne to "flare"?

BACKGROUND: Guidelines support this use of topical retinoids as a fundamental part of acne treatment regimens. However, existing dogma holds that topical retinoids may initially worsen acne. PURPOSE: To review the available data from clinical trials for evidence of initial worsening of acne with topical retinoids. METHODS: A PubMed and Google Internet search was performed for sources indicating or refuting worsening of acne with topical retinoids. RESULTS: No primary data from clinical trials were identified to support the dogma of acne worsening secondary to topical retinoids. Available da

The mechanism of action of topical retinoids.

UV irradiation of human skin sets in motion a complex sequence of events that causes damage to the dermal matrix. When topical tretinoin is applied to human skin, any collagen deficiency existing in photoaged skin is remedied at least partially, and the skin is primed to prevent further matrix degradation induced by solar UV. Retinoids, therefore, have become essential in the treatment and prevention of photoaging. This article describes the mechanism of action of retinoids, including how they are mediated through retinoic acid receptors (RARs) and retinoid X receptors (RXRs), how they block i

Oral retinoids--efficacy and toxicity in psoriasis.

Psoriasis is a disease of unknown aetiology, affecting approximately 1-3% of the population. In most cases involving relatively localized disease, patients are best managed with either topical therapy alone or topical therapy in combination with UV-phototherapy. However, approximately 35% of patients do not respond well to these conventional treatments or have moderate-to-severe disease requiring more aggressive forms of therapy. The second-generation retinoids, etretinate and its metabolite acitretin, are important additions to the armamentarium of agents used to treat these recalcitrant or s

Regulation and regulatory role of the retinoids.

Retinoids regulate differentiation and cellular growth, exerting their physiological action by interacting with two families of nuclear receptors, the retinoic acid receptors (RARs), and the retinoid X receptors (RXRs), which regulate gene expression by forming transcriptionally active heterodimeric RAR/RXR or homodimeric RXR/RXR complexes on DNA. Although RAR/RXR heterodimers form preferentially in vitro and in vivo, it does not exclude the possibility that RXR/RXR homodimers may regulate a distinct signaling pathway. Synthetic retinoids that selectively activate or antagonize retinoic acid r

Retinoids and the prevention of cervical dysplasias.

Women with abnormal cytology were matched with normal control subjects for age, parity, ethnicity, and socioeconomic class and participated in a blind case-control study focused on the role of nutrition in cervical dysplasia. Sucrose gradient ultracentrifugation studies for determination of the presence and concentration of the binding proteins for retinol and retinoic acid were performed on colposcopic biopsy tissue specimens. The nutritional survey revealed statistically significant differences for vitamins A and C and beta carotene. Retinol binding protein was absent or minimally detectable

Psoriatic erythroderma coexisting with erythema multiforme-like lesions induced by retinoids or retinoids combined with an antibiotic: case report.

Psoriasis is currently considered a multifactorial disease, which can coexist with many somatic and psychological disorders. We present the case of a 50-year-old woman referred to our department due to erythroderma with concomitant peculiar violaceous, polycyclic lesions most likely induced by medications. Past medical history revealed numerous systemic disorders, including metabolic syndrome, hypertension, cardiac insufficiency, obesity, and depression. Additional examinations and consultations demonstrated dyslipidemia, xanthelasma, incomplete block of the right branch of His bundle, thyreoc

Human skin retinoid-binding proteins and therapy with synthetic retinoids: a still unexplained link.

Human skin contains several proteins that could bind retinoids. One is the retinol-binding protein (RBP, the plasma carrier of natural vitamin A, retinol, which was found to also bind retinoic acid (RA) in vitro. A polyacrylamide gel electrophoresis technique followed by protein immunoblot analysis with an antiserum specific against RBP was developed. This technique allowed us to study the binding of several natural and synthetic retinoids to the plasma RBP; it was found that only natural retinoids bind to RBP in vitro; RA binding was found to induce major conformational changes in the protein

Systemic retinoids for treatment of recalcitrant IgA pemphigus.

IgA pemphigus is an exceedingly rare autoimmune blistering disorder, caused by IgA autoantibodies against desmosomal proteins. No treatment option has been found to be universally effective. The disease is often recalcitrant to oral steroids and immunosuppressants. Here, we describe the use of systemic retinoids for the treatment of recalcitrant IgA pemphigus in 3 cases. Although the use of acitretin has been reported before, we present for the first time the positive effects of alitretinoin in treatment of 2 patients with IgA pemphigus. Besides hyperlipoproteinaemia requiring use of hypolipid

In vitro inhibition of mumps virus by retinoids.

BACKGROUND: Mumps virus (MuV) is a highly infectious paramyxovirus closely related to measles virus (MeV). Despite the availability of a mumps vaccine, outbreaks continue to occur and no treatment options are available. Vitamin A and other naturally occurring retinoids inhibit the replication of MeV in vitro. METHODS: Anti-viral effects of retinoids were observed in cell culture using the myelomonocytic U937, NB4/R4, and Huh7/7.5 cells. Observations of anti-viral effect were quantified using TCID50 analysis. Molecular properties of the antiviral effect were analysed using quantitative RT-PCR
