Applied Rheology as Tool for the Assessment of Chitosan Hydrogels for Regenerative Medicine.

The regeneration of soft tissues that connect, support or surround other tissues is of great interest. In this sense, hydrogels have great potential as scaffolds for their regeneration. Among the different raw materials, chitosan stands out for being highly biocompatible, which, together with its biodegradability and structure, makes it a great alternative for the manufacture of hydrogels. Therefore, the aim of this work was to develop and characterize chitosan hydrogels. To this end, the most important parameters of their processing, i.e., agitation time, pH, gelation temperature and concentr

Rheology and stability of acidified food emulsions treated with high pressure.

The stability and rheology of acidified model oil-in-water emulsions (pH 3.6 +/- 0.1) were evaluated before and after high-pressure treatments. Varying concentrations of canola oil (0-50% w/w), whey protein isolate, polysorbate 60, soy lecithin (0.1-1.5% w/w each), and xanthan (0.0-0.2% w/w) were chosen. Exposure to high pressures (up to 800 MPa for 5 min at 30 degrees C) did not significantly affect the equivalent surface mean diameter D[3,2], flow behavior, and viscoelasticity of the whey protein isolate and polysorbate 60-stabilized emulsions. Pressure treatments had negligible effects on e

Sickle cell rheology is determined by polymer fraction--not cell morphology.

Sickle cell disease pathophysiology is mediated by acute and chronic impairment of cell flexibility due to the formation of intracellular sickle hemoglobin (Hb S) polymer as cells are partially deoxygenated in the microcirculation. We have recently developed a method to measure the relationship between the formation of intracellular polymerized Hb S and cell filtration. In this study, we have used this method to examine whether sickle cell morphology, independent of Hb S polymer fraction, had an effect on cell rheology. We primarily use sickle trait (AS) and Hb S-beta(+)-thalassemia (S-beta(+)

Effect of perfusate rheology on the diastolic coronary pressure-flow relationship.

The hypothesis that rheological properties of the coronary perfusate account for the curvilinearity and high zero-flow pressure (Pf = 0) of the diastolic coronary-pressure flow relationship (DCPFR) was tested by measuring these relationships using coronary perfusates of varying rheological character. In 16 open-chest, heart-blocked dogs the left circumflex coronary artery was cannulated and perfused using an extra-corporeal circuit, and autoregulation was abolished with intracoronary adenosine. DCPFRs were constructed from data obtained at multiple steady-state levels of coronary pressure duri

Effect of alpha thalassaemia on the rheology of homozygous sickle cell disease.

A study of rheological determinants (plasma viscosity, whole-blood viscosity, and erythrocyte deformability) was made in 24 matched pairs of patients with homozygous sickle cell disease, with and without homozygous alpha-thalassaemia 2. Patients with coexisting alpha-thalassaemia showed a significant increase in erythrocyte deformability measured as filtration of washed erythrocytes through 5 micron diameter pores and also as viscosity of whole blood at high shear rate (230s-1) and standard haematocrit (0.45). This rheological advantage may explain the beneficial effect of alpha-thalassaemia 2

Rheology of liquid foods under shear flow conditions: Recently used models.

Proper modeling of flow or viscosity curves as a function of shear rate is a useful tool in any engineering activity. The rheology of foods depends on the composition, processing to which they have been subjected and the state of dispersion in which they are found. Liquid foods are complex biosystems, that show non-Newtonian behavior under flow conditions. This review presents models used in recent decades to describe the experimental rheological behavior of various liquid foods, ranging from Newtonian fluids to the most complex. Some non-Newtonian parameters such as those of the Ostwald-de Wa

Use of Dynamic Shear Rheology to Understand Soy Protein Dispersion Properties.

Soy flour dispersions are used as adhesives for bonding interior wood laminates, but the high viscosity of these dispersions requires low solids in the adhesive formulations; the greater water content causes excessive steam pressure during hot press manufacturing. This limits the utility of soy adhesives in replacing urea-formaldehyde adhesives; thus, understanding the cause of high soy viscosities is important. Lack of literature on aqueous soy flour dispersion rheology led to our dynamic rheology studies of these dispersions to understand high viscosity and the effect of various additives. E

Red blood cell rheology using single controlled laser-induced cavitation bubbles.

The deformability of red blood cells (RBCs) is an important property that allows the cells to squeeze through small capillary vessels and can be used as an indicator for disease. We present a microfluidic based technique to quantify the deformability of RBCs by stretching a collection of RBCs on a timescale of tens of microseconds in a microfluidic chamber. This confinement constrains the motion of the cell to the imaging plane of the microscope during a transient cavitation bubble event generated with a focused and pulsed laser. We record and analyze the shape recovery of the cells with a hig

Tunable yielding and emergent rheology in amorphous solids with active particle doping.

Mechanically stable glasses, notably well-annealed and ultrastable ones, typically exhibit brittle failure through shear-band formation. Whether this localization is an inevitable consequence of annealing or can be dynamically controlled remains unknown. We address this by doping amorphous solids with self-propelled particles that perform run-and-tumble or active Brownian dynamics. Through extensive simulations of a polydisperse model, we show that shear-band formation and propagation are governed by a competition among three timescales: the imposed shear time 1 / γ ° , the activity persiste

Rheology and Thermal Characterization of Lecithin-Based Kerosene Gels for In-Space Propulsion.

Gel fuels bridge the gap between liquid and solid propellants by improving handling and safety, while making them energy dense, an attractive fuel class for rocket and satellite propulsion. In this study, kerosene is gelled with lecithin, a naturally derived organic gellant and a low concentration of CaCl2, producing a fully combustible, optically transparent gel. CaCl2 promotes lecithin self-assembly into long worm-like micelles that form a mesh-like network and immobilize kerosene, yielding a viscoelastic material whose state shifts from near viscous-liquid to solid-like as gellant concentra

Diffusion NMR and Rheology of a Model Polymer in Bacterial Cell Lysate Crowders.

The intracellular milieu is crowded and heterogeneous, and this can have profound consequences for biomolecule motions and biochemical kinetics. Macromolecular crowding has been traditionally studied in artificial crowders like Ficoll and dextran or globular proteins such as bovine serum albumin. It is, however, not clear if the effects of artificial crowders on such phenomena are the same as the crowding that is experienced in a heterogeneous biological environment. Bacterial cells, for example, are composed of heterogeneous biomolecules with different sizes, shapes, and charges. Using crowde

Factors affecting the particle size distribution and rheology of brinzolamide ophthalmic suspensions.

Drug particle size distribution (PSD) and dispersion viscosity are two critical quality attributes that govern the performance of topical ophthalmic suspensions, such as suspension physical stability, ocular retention, and drug release characteristics.. An in-depth knowledge of the effects of formulation and manufacturing process on these critical quality attributes may facilitate the product and process development, quality control, as well as support regulatory policy and approval. The current study has investigated the effect of formulation and process parameters on the quality attributes o

Development and quantitative characterization of the precursor rheology of hyaluronic acid hydrogels for bioprinting.

Bioprinting technologies have tremendous potential for advancing regenerative medicine due to the precise spatial control over depositing a printable biomaterial, or bioink. Despite the growing interest in bioprinting, the field is challenged with developing biomaterials for extrusion-based bioprinting. The paradigm of contemporary bioink studies relies on trial-and-error methods for discovering printable biomaterials, which has little practical use for others who endeavor to develop bioinks. There is pressing need to follow the precedent set by a few pioneering studies that have attempted to

[Evaluation of blood rheology by patients with acute ischemic stroke with Mexidol administration].

UNLABELLED: To study rheological properties of blood in patients with acute ischemic stroke treated with mexidol. MATERIAL AND METHODS: Sixty patients with acute stroke, including 32 patients who received mexidol (500 mg/IV/20 days) and 28 people who received magnesium sulfate (2000 mg/IV/20 days) were examined. The control group included 20 people without a cardiovascular pathology. Blood rheology (viscosity of whole blood, plasma viscosity, hematocrit, aggregation and deformability of erythrocytes, fibrinogen level) was evaluated in patients three times: for the first 12 hours, 3-5 days and

Insoluble layer deposition and dilatational rheology at a microscale spherical cap interface.

The dilatational properties of insoluble monolayers are important for understanding the mechanics of biological systems and consumer products, but isolating the dilatational response of an interface is challenging due to the difficulties in separating dilatation from shear and other deformation modes. Oscillations of a microscale bubble radius are useful for generating purely dilatational flows, but the current deposition methods for insoluble layers onto fluid interfaces are not easily scaled down. In this paper, we describe a miscible solvent exchange procedure for generating insoluble layer

Effects of methacholine and uridine 5'-triphosphate on tracheal mucus rheology in mice.

We compared the action of methacholine (MCh) and uridine 5'-triphosphate (UTP) with and without pretreatment with the chloride channel blocker 4,4'-diisothiocyano-2,2'-stilbenedisulfonate (DIDS) on the transepithelial potential difference (PD), the mucus collection rate (MCR), and tracheal mucus rheology using anesthetized C57BL/6 mice. The cystic fibrosis transmembrane conductance regulator (CFTR) blocker 5-nitro-2-(3-phenylpropylamino)benzoate (NPPB) was also used as a pretreatment for MCh. After collecting baseline mucus for 1.5 h, mucus secretion was stimulated by instilling 5 microl of 10

The etiopathogenesis of premenstrual syndrome as a consequence of altered blood rheology: a new hypothesis.

The multisystem symptoms of premenstrual syndrome (PMS), the variable nature of their occurrence and their relatively rapid disappearance with the onset of bleeding are highly suggestive of a systemic factor as the causal agent of the disorder. Reports of increased blood viscosity and enhanced red blood cell (RBC) hydration provide a basis for proposing that PMS is the consequence of an aberration of the change in blood viscosity which occurs in the third week of the normal menstrual cycle. The seriousness of the viscosity-impaired capillary blood flow will be greatest in organs and capillary

Evaporation-controlled dripping-onto-substrate (DoS) extensional rheology of viscoelastic polymer solutions.

Extensional flow properties of polymer solutions in volatile solvents govern many industrially-relevant coating processes, but existing instrumentation lacks the environment necessary to control evaporation. To mitigate evaporation during dripping-onto-substrate (DoS) extensional rheology measurements, we developed a chamber to enclose the sample in an environment saturated with solvent vapor. We validated the evaporation-controlled DoS device by measuring a model high molecular weight polyethylene oxide (PEO) in various organic solvents both inside and outside of the chamber. Evaporation subs

A power-law rheology-based finite element model for single cell deformation.

Physical forces can elicit complex time- and space-dependent deformations in living cells. These deformations at the subcellular level are difficult to measure but can be estimated using computational approaches such as finite element (FE) simulation. Existing FE models predominantly treat cells as spring-dashpot viscoelastic materials, while broad experimental data are now lending support to the power-law rheology (PLR) model. Here, we developed a large deformation FE model that incorporated PLR and experimentally verified this model by performing micropipette aspiration on fibroblasts under
