Acid-fast bodies in faecal smears stained by the modified Ziehl-Neelsen technique.

The modified Ziehl-Neelsen (ZN) stain has proved useful in the laboratory diagnosis of cryptosporidiosis and, more recently, for the laboratory diagnosis of cyclospora. Apart from cryptosporidia and cyclospora, many other organisms and artefacts are present in faeces, and may be seen in faecal smears stained by the modified ZN method. Described here is the presence of such organisms and artefacts in faecal samples submitted to the routine microbiology laboratory of a district general hospital. Over 6000 faecal smears were examined using the modified ZN method, with an incidence of cyclospora a

Progressive changes in coral reef communities with increasing ocean acidification.

Ocean acidification from increasing atmospheric CO2 is progressively affecting seawater chemistry, but predicting ongoing and near-future consequences for marine ecosystems is challenging without empirical field data. Here we quantify tropical coral reef benthic communities at 37 stations with varying exposure to submarine volcanic CO2 seeping, and determine the aragonite saturation state (ΩAr) where significant changes occur in situ. With declining ΩAr, reef communities displayed progressive retractions of most reef-building taxa and a proliferation in the biomass and cover of non-calcareous

Advances in Carrageenases: Molecular Engineering, Immobilization Techniques, and Biomedical Potential of Carrageenan Oligosaccharides.

Carrageenan is a sulfated polysaccharide extracted from red algae, widely utilized in the food and pharmaceutical industries due to its unique gelling properties and biological activities. However, its high molecular weight and low solubility limit certain high-value applications. Carrageenases specifically degrade carrageenan to produce low-molecular-weight oligosaccharides with excellent bioactivities, thereby overcoming this bottleneck. This review systematically summarizes the microbial sources of carrageenases and the classification characteristics of their three types (κ, ι, and λ) withi

Modeling with uncertainty quantification reveals the essentials of a non-canonical algal carbon-concentrating mechanism.

The thermoacidophilic red alga Cyanidioschyzon merolae survives its challenging environment likely in part by operating a carbon-concentrating mechanism (CCM). Here, we demonstrated that C. merolae's cellular affinity for CO2 is stronger than the affinity of its rubisco for CO2. This finding provided additional evidence that C. merolae operates a CCM while lacking the structures and functions characteristic of CCMs in other organisms. To test how such a CCM could function, we created a mathematical compartmental model of a simple CCM, distinct from those we have seen previously described in de

Extracts of North Sea macroalgae reveal specific activity patterns against attachment and proliferation of benthic diatoms: a laboratory study.

A variety of macroalgae (Ceramium rubrum, Corallina officinalis, Palmaria palmata, Mastocarpus stellatus, Fucus vesiculosus, Cladophora rupestris, Ulva sp.) were investigated by scanning electron microscopy to visualize epiphytic colonizers. The macroalgae differed in terms of their epiphytic coverage of bacteria, fungi and diatoms. Macroalgae, largely devoid of epiphytic diatoms, were hypothesized to employ effective antifouling means to reduce epiphytic coverage, whilst heavily fouled macroalgae were proposed to lack antifouling strategies. To test these hypotheses from an allelochemical per

The antifungal activity of extracts of Osmundea pinnatifida, an edible seaweed, indicates its usage as a safe environmental fungicide or as a food additive preventing post-harvest fungal food contamination.

In the present work, we explored the antifungal activity of the wild edible seaweed Osmundea pinnatifida (Rhodophyta) collected from the Portuguese coast, which is used as a food seasoning in Scotland, Ireland and Portugal. We performed a sequential extraction of the seaweed components with methanol, dichloromethane and n-hexane. These extracts showed an antifungal activity against Alternaria infectoria and Aspergillus fumigatus. The n-hexane fraction of the seaweed inhibited the sporulation of Alternaria infectoria at 30 μg mL-1 and induced a statistically significant (P < 0.001) decrease in

Aerobic transformation of cadmium through metal sulfide biosynthesis in photosynthetic microorganisms.

BACKGROUND: Cadmium is a non-essential metal that is toxic because of its interference with essential metals such as iron, calcium and zinc causing numerous detrimental metabolic and cellular effects. The amount of this metal in the environment has increased dramatically since the advent of the industrial age as a result of mining activities, the use of fertilizers and sewage sludge in farming, and discharges from manufacturing activities. The metal bioremediation utility of phototrophic microbes has been demonstrated through their ability to detoxify Hg(II) into HgS under aerobic conditions.

Bio-Guided Fractionation and Molecular Networking Reveal Fatty Acids to Be Principal Anti-Parasitic Compounds in Nordic Seaweeds.

Widespread use of antimicrobial drugs has led to high levels of drug-resistance in pathogen populations and a need for novel sources of anti-bacterial and anti-parasitic compounds. Macroalgae (seaweed) are potentially a rich source of bioactive compounds, and several species have traditionally been used as vermifuges. Here, we investigated the anti-parasitic properties of four common cold-water Nordic seaweeds; Palmaria palmata (Rhodophyta), Laminaria digitata, Saccharina latissima and Ascophyllum nodosum (Ochrophyta, Phaeophyceae). Screening of organic extracts against helminths of swine (Asc

Comparative efficiency of macroalgal extracts and booster biocides as antifouling agents to control growth of three diatom species.

The application of 'booster biocides' Diuron, Tolylfluanid and Copper thiocyanate inbantifouling paints, used to prevent development of biofouling, needs to be monitored before assessing their impacts on the environment. An alternative approach aims to propose eco-friendly and effective antifoulants isolated from marine organisms such as seaweeds. In this study, the effects of 'booster biocides' and the ethanol and dichloromethane extracts from a brown (Sargassum muticum) and a red alga (Ceramium botryocarpum) have been compared by algal growth inhibition tests of marine diatoms. Gut-protective efficacy of red algal galactans: The role of structure and molecular weight.

Linking structure, rheology, and bioactivity, this study demonstrates how red algal galactans can be tailored as functional food ingredients with gut-protective potential. Funoran and furcellaran were isolated from Gloiopeltis furcata and Furcellaria lumbricalis, respectively, and characterized using chromatographic and NMR spectroscopic techniques. Funoran was identified as a highly sulfated, methoxylated agaran (22.7% sulfate), whereas furcellaran exhibited a hybrid κ-/β-carrageenan structure (17.4% sulfate). High molecular weights (3955 and 2966 kDa) were observed and reduced via controlled

Effects of sunflower oil infusions of Asparagopsis taxiformis on in vitro ruminal methane production and biohydrogenation of polyunsaturated fatty acids.

Asparagopsis taxiformis inhibits ruminal methane (CH4) production due to its bromoform (CHBr3) content. The immersion of A. taxiformis in edible vegetable oils allows the extraction and stabilization of the highly volatile CHBr3 in the oil phase. The objectives of this study were to explore the effects of adding sunflower oils with increasing concentrations of CHBr3 on in vitro ruminal methanogenesis and biohydrogenation. Five batches of 48-h in vitro incubations were performed in 14 fermentation bottles, using rumen inocula collected shortly after the slaughter of young crossbred bulls and 1
