Development Features of Ixodes ricinus × I. persulcatus Hybrids under Laboratory Conditions.

Widely distributed Ixodes ricinus and Ixodes persulcatus ticks transmit many pathogens of both medical and veterinary significance. The ranges of these tick species overlap and form large sympatric areas in the East European Plain and Baltic countries. It has previously been shown that crossing I. ricinus and I. persulcatus is possible, resulting in the appearance of sterile hybrids. In the present study, we analyzed the features of this hybrid's life cycle under laboratory conditions. For this purpose, virgin females of I. ricinus and I. persulcatus ticks were obtained in the laboratory, and

Ixodes ricinus tick lipocalins: identification, cloning, phylogenetic analysis and biochemical characterization.

BACKGROUND: During their blood meal, ticks secrete a wide variety of proteins that interfere with their host's defense mechanisms. Among these proteins, lipocalins play a major role in the modulation of the inflammatory response. METHODOLOGY/PRINCIPAL FINDINGS: Screening a cDNA library in association with RT-PCR and RACE methodologies allowed us to identify 14 new lipocalin genes in the salivary glands of the Ixodes ricinus hard tick. A computational in-depth structural analysis confirmed that LIRs belong to the lipocalin family. These proteins were called LIR for "Lipocalin from I. ricinus"

Replication of tick-borne encephalitis (TBE) virus in Ixodes ricinus ticks.

The influence of external factors on virus carriage of Ixodes ricinus ticks in laboratory and in nature was studied. In laboratory experiment, only one nymph was positive for the presence of virus on 120th day after metamorphosis. The virus titer was 10(2) mouse i.c. LD50/0.03 ml. Transmission experiments were negative. The nymphs were positive on 75th, 111th and 159th day after metamorphosis, always chilling in the field experiment. The titres of virus varied from the lowest detectable amount value to 10(3.6) mouse i.c. LD50/0.03 ml. The transmission of virus was positive in two cases.

[Serum antibodies against Ixodes ricinus Spirochaeta in acrodermatitis chronica atrophicans (Herxheimer)].

Using indirect immunofluorescence, IgG antibodies against the recently detected Ixodes-ricinus-spirochaeta, which causes erythema chronicum migrans could be demonstrated in all 21 persons with acrodermatitis chronica atrophicans. Titers were from 1 : 64 to 1 : 1024, specific IgM antibodies were demonstrable in only 5 patients in a titer of 1 : 64. Even after treatment with penicillin high IgG antibody titers of up to 1 : 1024 were found. Fourfold decreases could be found only once for each IgG and IgM antibodies.l Serology indicates that acrodermatitis chronica atrophicans as well as erythema

Qualitative profiling of the gut-specific chlamydial population in Ixodes ricinus ticks.

Members of the phylum Chlamydiota are obligate intracellular bacteria increasingly recognized across a wide range of arthropod hosts, including ticks. In this study, we investigated the diversity and distribution of chlamydiae in Ixodes ricinus ticks and their potential association with Lyme borreliosis spirochetes. A total of 250 questing nymphal and female I. ricinus ticks were collected from three recreational sites in Vienna, Austria. Individual tick guts were screened for chlamydiae using pan-Chlamydiota PCR assays targeting the 16S rRNA gene, followed by sequencing for taxonomic identifi

Detection of Borrelia burgdorferi (s.l.) in Ixodes ricinus ticks collected in Iceland.

BACKGROUND: Ixodes ricinus is the most common tick species throughout Europe; it can transmit various pathogens that can cause diseases in humans and animals. It is the principal vector of Borrelia burgdorferi sensu lato (s.l.) and tick-borne encephalitis virus (TBEV), and there is increasing concern about I. ricinus's potential to transmit pathogens to humans and animals in Iceland. The aim of this research is to determine whether I. ricinus ticks collected in Iceland carry B. burgdorferi (s.l) and, in a limited number of samples, other pathogens to better understand the potential health risk

Detection of Babesia species in questing Ixodes ricinus ticks in England and Wales.

Babesiosis, a disease in humans and animals is caused by piroplasms from the genus Babesia and is transmitted by ixodid ticks. Bovine babesiosis, commonly called redwater fever, is reported in cattle from many regions of the British Isles. The presence of Babesia in questing ticks in the United Kingdom (UK) and its potential impact on public and animal health has not been widely studied. Therefore, this study aimed to assess the presence of Babesia spp. in England and Wales using ticks collected over a six-year period. Questing Ixodes ricinus nymphs were collected at 20 recreational areas betw

Predicting habitat suitability for Ixodes ricinus and Ixodes persulcatus ticks in Finland.

BACKGROUND: Ticks are responsible for transmitting several notable pathogens worldwide. Finland lies in a zone where two human-biting tick species co-occur: Ixodes ricinus and Ixodes persulcatus. Tick densities have increased in boreal regions worldwide during past decades, and tick-borne pathogens have been identified as one of the major threats to public health in the face of climate change. METHODS: We used species distribution modelling techniques to predict the distributions of I. ricinus and I. persulcatus, using aggregated historical data from 2014 to 2020 and new tick occurrence data

Functional characterization of the insulin signaling pathway in the hard tick Ixodes ricinus.

Ticks are blood-feeding arachnids transmitting a variety of pathogens to humans and animals. A unique trait in tick physiology is their ability to engorge and digest large amounts of host blood, ensuring their high reproductive potential. Activation of the blood digestive machinery in the tick gut, as well as processes controlling maturation of ovaries, are triggered upon blood meal uptake by still largely unknown mechanisms. Sensing of the nutritional status in metazoan organisms is facilitated by the evolutionarily conserved Insulin Signaling Pathway (ISP) and the interlinked Target of Rapam

Evaluation of Disease Causality of Rare Ixodes ricinus-Borne Infections in Europe.

In Europe, Ixodes ricinus ticks transmit pathogens such as Borrelia burgdorferi sensu lato and tick-borne encephalitis virus (TBEV). In addition, there is evidence for transmission to humans from I. ricinus of Anaplasma phagocytophilum, Babesia divergens, Babesia microti, Babesia venatorum, Borrelia miyamotoi, Neoehrlichia mikurensis, Rickettsia helvetica and Rickettsia monacensis. However, whether infection with these potential tick-borne pathogens results in human disease has not been fully demonstrated for all of these tick-borne microorganisms. To evaluate the available evidence for a caus

First report of Borrelia miyamotoi in an Ixodes ricinus tick in Augsburg, Germany.

Borrelia miyamotoi, a spirochete found in the hard tick Ixodes ricinus, is thought to cause relapsing fever. The disease caused by this bacterium can manifest with high fever, fatigue and other symptoms. It may also lead to central nervous system involvement with symptoms similar to meningoencephalitis. DNA from ticks from the greater Augsburg region in Germany was subjected to qPCR for Borrelia spp., followed by nested PCR and subsequent sequencing for species identification of the qPCR positive samples. From 112 ticks, 20 were found to be positive for Borrelia. The DNA sequenced showed 50% B

Functional characterization of two defensin isoforms of the hard tick Ixodes ricinus.

BACKGROUND: The immune system of ticks is stimulated to produce many pharmacologically active molecules during feeding and especially during pathogen invasion. The family of cationic peptides - defensins - represents a specific group of antimicrobial compounds with six conserved cysteine residues in a molecule. RESULTS: Two isoforms of the defensin gene (def1 and def2) were identified in the European tick Ixodes ricinus. Expression of both genes was induced in different tick organs by a blood feeding or pathogen injection. We have tested the ability of synthetic peptides def1 and def2 to inhi

Efficacy of Ixodes ricinus as a vector of zoonotic babesiosis in Sinai Peninsula, Egypt.

Ticks and blood samples were collected every month from March 2009 through April 2010 from different sites in Sinai to detect babesial parasites using PCR assay based on nuclear small subunit rRNA gene. Ticks were found to contain babesial DNA. Sequence determination and analysis of amplified portions of nss-rDNA revealed their identity with B. bovis and a high degree of homology with B. bigemina and B. divergens. The results represent the first genetic evidence of different species of Babesia and identified the role of Ixodes ricinus as a vector of zoonotic B. microti infection. Rodent isolat

Influence of climatic factors on dynamics of questing Ixodes ricinus ticks in Slovenia.

Ixodes ricinus is a vector of pathogens that cause many diseases in Europe and Slovenia: tick-borne encephalitis (TBE), anaplasmosis, borreliosis, babesiosis and others. The risk for contracting these diseases depends strongly on the density of the infected questing ticks and many studies have investigated tick population dynamics and the parameters affecting them. They have shown a clear influence of climatic and landscape arrangements in the microhabitat on tick abundance and dynamics and therefore on transmission of pathogens important in human and veterinary medicine. In our study we asses

Presence of potentially pathogenic Babesia sp. for human in Ixodes ricinus in Switzerland.

We have designed and performed a new PCR method based on the 18S rRNA in order to individuate the presence and the identity of Babesia parasites. Out of 1159 Ixodes ricinus (Acari: Ixodidae) ticks collected in four areas of Switzerland, nine were found to contain Babesia DNA. Sequencing of the short amplicon obtained (411-452 bp) allowed the identification of three human pathogenic species: Babesia microti, B. divergens, for the first time in Switzerland, Babesia sp. EU1. We also report coinfections with B. sp. EU1-Borrelia burgdorferi sensu stricto and Babesia sp. EU1-B. afzelii.

Evidence for flavivirus(es) outside of the distribution area for Ixodes ricinus in Norway.

Small rodent (vole) sera were collected from three different locations in Norway. One of these was within the distribution area for Ixodes ricinus, and a tick-borne encephalitis (TBE) virus strain had been isolated from ticks collected there (Traavik & Mehl, 1977). The two other locations were outside the I. ricinus area, one in southern Norway, the other at nearly 70 degrees N. The sera were tested for TBE antibodies by hemagglutination inhibition (HI), hemolysis-in-gel (HIG) and complement fixation (CFT). All sera were also tested for HI antibodies to Uukuniemi (UUK) virus, and some positive

[Study of characteristics of distribution of the tick Ixodes ricinus over a large territory].

Attempts were undertaken to study the character of the distribution of I. ricinus over a large territory. Methodic principles of the land survey of arthropods distribution and laboratory processing of its results, that were used before for I. persulcatus, were found to be quite suitable for I. ricinus. It was established that hungry adults of I. ricinus have at least three types of the distribution throughout the forests of Lithuania. Types of the distribution can be ascertained by the results of the record of I. ricinus nymphs with a flag.
